Pronounced cholinergic but only moderate purinergic effects in isolated atrial and ventricular heart muscle from cats.

1. The effects of cholinergic and purinergic stimulation on action potential, force of contraction and 86Rb efflux were investigated in cat atrial and/or ventricular heart muscle. 2. Acetylcholine and carbachol exerted a concentration-dependent negative inotropic effect in cat atrial heart muscle. Carbachol 10 mumol l-1 completely abolished the force of contraction and increased the rate constant of 86Rb efflux 2-3 fold, whereas the action potential duration was shortened to about 1/10 of its length under control conditions. 3. The effects of acetylcholine and carbachol in cat atrial heart mus

[Recovery of impaired respiratory activity in cats by thyroliberin analog (PR-546) devoid of hormonal properties].

Synthetic analogue thyroliberin (PR-546) devoid of hormonal activity, recovered respiration in conditions of hyperventilation, two-sided vagotomy and hypoxia, caused by the 40-70% bleeding. PR-546 (in doses 100-500 micrograms/kg) intravenously injected was shown to abolish diagram activity decreased effect after hyperventilation. Injection of PR-546 to vagotomized cats in 1.5-2 h, increased breathing rate. The breathing activity resumed after long breaks in respiration as a result of peptide injection. The responses to analogue introductions is similar to the thyroliberin effect.

Clonal evolution following chemotherapy-induced stem cell depletion in cats heterozygous for glucose-6-phosphate dehydrogenase.

The number of hematopoietic stem cells necessary to support normal hematopoiesis is not known but may be small. If so, the depletion or damage of such cells could result in apparent clonal dominance. To test this hypothesis, dimethylbusulfan [2 to 4 mg/kg intravenously (IV) x 3] was given to cats heterozygous for the X-linked enzyme glucose-6-phosphate dehydrogenase (G-6-PD). These cats were the daughters of domestic X Geoffroy parents. After the initial drug-induced cytopenias (2 to 4 weeks), peripheral blood counts and the numbers of marrow progenitors detected in culture remained normal, al…

Effect of removal of lacrimal and third eyelid glands on Schirmer tear test results in cats.

Schirmer tear tests I and II values for 10 healthy cats were 16.2 +/- 3.8 mm/min and 13.2 +/- 3.4 mm/min, respectively. Surgical removal of the lacrimal gland or the third eyelid gland from the left eye resulted in decreased Schirmer tear test-I values. Clinical signs of keratoconjunctivitis sicca were not seen until the lacrimal and third eyelid glands were removed, at which time the Schirmer tear test-I value was close to 0 mm/min. Conjunctival glands (accessory lacrimal glands of Krause and Wolfring) appeared to contribute little to tear production in these cats.

Effects of stimulating the sympathetic innervation in bursts on submandibular vascular and secretory function in cats.

1. The effects of continuous stimulation of the peripheral end of the ascending cervical sympathetic nerve were compared with those of intermittent stimulation, so arranged as to deliver the same total number of impulses, in cats under chloralose anaesthesia. 2. Continuous stimulation caused a flow of saliva at 5-10 Hz, but not at 2 Hz. In contrast, the same total number of impulses delivered intermittently in bursts elicited a prompt secretion at a frequency as low as 20 Hz for 1 s at 10 s intervals (corresponding to 2 Hz continuously) and a significantly higher rate of secretion at 50 Hz in …

Role of medullary inspiratory neurones in the control of the diaphragm during oesophageal stimulation in cats.

1. The effect of oesophageal distension and swallowing on the activity of medullary respiratory neurones was recorded in decerebrate, spontaneously breathing cats. The distension, produced by inflating a balloon in the thoracic portion of the oesophagus, was of sufficient magnitude to induce inhibition of the peri-oesophageal part of the crural diaphragm, with little effect on the respiratory function of the diaphragm as measured by the activity in the C5 branch of the phrenic nerve. 2. 424 neurones were tested. They were located bilaterally, in the region of the nucleus tractus solitarius (do…

Virion polypeptide specificity of immune complexes and antibodies in cats inoculated with feline infectious peritonitis virus.

Immune complexes purified from sera and ascitic fluids of cats after inoculation with feline infectious peritonitis (FIP) virus contained proteins and proteolytic fragments of the peplomer, nucleocapsid, and envelope polypeptides; in addition, host proteins were demonstrated in the immune complexes. Free (uncomplexed) antibodies against the 3 classes of virion polypeptides were detected and quantitated; the weakest and latest response was directed against the peplomer protein. Immunofluorescence titers showed the best correlation with the antibody response directed against the envelope polypep…

Antiepileptic effects of globulin-N, an intact human immunoglobulin and its tissue-distribution in kindled cats.

The antiepileptic effects of globulin-N, an intact human immunoglobulin was examined, using the amygdaloid kindled animals prepared by the method of Goddard et al. [1969] and Wada et al. [1974 b]. In the non-treated kindled cats, kindled convulsion and after-discharge (AD) were simultaneously created at 24-hour intervals by electrically stimulating generalized seizure-triggering threshold (GST) in amygdala, hippocampus and neocortex. The kindled cats, treated intravenously with 200 mg/kg of globulin-N, the generalized convulsion and AD produced by GST-stimulation disappeared entirely from 30 m…

Comparison of the cardiovascular effects of meptazinol and naloxone following haemorrhagic shock in rats and cats.

The cardiovascular effects of the opioid mixed agonist-antagonist, meptazinol, and the opioid antagonist, naloxone, have been evaluated in conscious rats, anaesthetized rats and anaesthetized cats following the induction of haemorrhagic shock. The mean arterial pressure of conscious rats decreased by 17-29 mmHg following a haemorrhage of 20% of blood volume. Meptazinol (17 mg kg-1, i.m.) administered after haemorrhage evoked a rapid and sustained increase in mean arterial pressure to pre-haemorrhage levels. Naloxone (10 mg kg-1, i.v.) also increased mean arterial pressure to a level significan…

Trigeminal projections to supratentorial pial and dural blood vessels in cats demonstrated by horseradish peroxidase histochemistry.

Anatomical and clinical observations suggest that supratentorial vascular structures contain afferent projections from the trigeminal ganglia. To characterize this innervation, horseradish peroxidase (HRP) and HRP conjugated to wheat germ agglutinin were applied to the pial and dural arteries and sinuses of 33 cats. HRP was restricted to the site of interest by applying it dissolved in a viscous polymer, polyvinyl alcohol (PVA), to achieve slow release and minimize diffusion. The ganglia of cranial nerves V, VII, IX, and X and the superior cervical ganglia (SCGs) were examined bilaterally for …

Visual evoked potentials to light flash in cats: is a frontal sinus reference electrode truly indifferent?

In flaxedilized cats, visual cortex evoked potentials (VEPs) to an intensity series of brief light flashes were recorded differentially using either a screw over frontal sinus or an insect pin in neck muscle as the reference electrode. At the same time EPs from the cornea (the ERG) and frontal sinus were recorded differentially against the neck muscle electrode. The EP recorded from the frontal sinus screw was a miniature of the ERG recorded from the cornea, with the same latencies and shape as the ERG but only 10-30% of its amplitude. At a fast presentation rate (1/sec) the amplitude of the E…

The release of prostanoids during the acute pulmonary response to E. coli endotoxin in anaesthetized cats.

1 The administration of E. coli endotoxin (2 mg/kg i.v.) to anaesthetized cats results in a characteristic acute pulmonary response. This consists of increases in pulmonary artery pressure and airways resistance and a reduction in lung compliance. 2 Plasma concentrations of prostaglandin E2 (PGE2), PGF2 alpha, thromboxane B2 and 6-keto PGF1 alpha were measured by radioimmunoassay in aortic and pulmonary arterial blood samples before, during and after the acute pulmonary response to endotoxin. 3 Endotoxin administration resulted in the rapid release of PGF2 alpha and thromboxane B2 from the lun…

Respiratory and cardiovascular depressant effects of nabilone, N-methyllevonantradol and delta 9-tetrahydrocannabinol in anesthetized cats.

Drug effects were examined in cats anesthetized with a mixture of pentobarbital and barbital injected i.p. Respiratory observations were analyzed according to effects produced on 1) chemoregulatory responsiveness determined by changes in tidal volume resulting from CO2 inhalation or measured during isocapnic stabilization, and on 2) mechanoreflex control of respiratory frequency through the vagus nerves. Blood pressure and heart rate were recorded concomitantly. Cardiovascular effects were manifested as dose-related hypotension and bradycardia that were generally response-limited by contrast w…

The effects of parasympathetic blocking agents on bladder electromyograms and function in conscious and anaesthetized cats.

Voiding induced in conscious cats by infusion of sterile saline into the bladder via a chronically implanted bladder catheter was stable over many months. Artefact-free recordings of electrical activity obtained from the bladder neck and dome of these preparations during bladder filling and voiding showed characteristic voiding electromyograms but did not permit a functional differentiation. Both voiding and the associated electromyogram were abolished by the ganglion blocking agent, pentolinium. Hyoscine or methyl atropine did not affect the electromyogram but impaired the ability of cats to …

Effect of centrophenoxine on acetylcholine release in perfused cerebral ventricles of cats under dynamic electrophysiological control.

The effects of centrophenoxine on the release of acetylcholine and on the changes in the bioelectrical activity are determined in experiments on non-anaesthesized cats subjected to perfusion of the anterior horn of the lateral cerebral ventricle and simultaneous recording of the bioelectrical activity of cortical and subcortical structures. Centrophenoxine is tested in doses of 25, 50 and 100 mg/kg intravenously. Most characteristic changes are found to occur after the dose of 50 mg/kg, when centrophenoxine markedly increases the amount of the released acetylcholine and changes the bioelectric…
