Simultaneous UHPLC-MS/MS profiling of eight phosphate metabolites reveals divergent metabolic reprogramming under hypoxic stress.

Phosphate-containing metabolites serve as critical regulators of energy homeostasis and signal transduction under hypoxic stress. However, their simultaneous quantification is technically challenging due to high polarity and non-specific adsorption. In this study, a sensitive UHPLC-MS/MS method was established for the simultaneous quantification of eight key phosphate metabolites, including adenosine triphosphate (ATP), phosphoenolpyruvate (PEP) and glucose-1-phosphate (P1G), in biological matrices. By compressing the analytical run time to <4 min per sample, this high-throughput platform demo

Simple and sensitive cellulose paper-based microextraction of 25 steroid esters from human serum/plasma for doping control purposes.

Blood is increasingly used as a biological matrix in the doping control field; however, the limited volume typically available for analysis poses a significant challenge, particularly when multiple tests must be carried out on the same sample. This study proposes a simple and efficient microextraction protocol based on the use of unmodified cellulose for the extraction of 25 steroid esters from just 20 µL of serum or plasma. Sample preparation consisted of spotting 20 µL of serum or plasma on a cellulose card, followed by a double extraction of the target analytes with 500 µL of methanol and s

Simultaneous Quantification of Mycophenolic Acid and Its Glucuronide Metabolite in PBMCs from Kidney Transplant Recipients by LC-MS/MS.

BACKGROUND: Mycophenolic acid (MPA), administered as mycophenolate mofetil (MMF) or enteric-coated mycophenolate sodium (EC-MPS), is the first-line immunosuppressant for kidney transplant patients. Traditional plasma-based therapeutic drug monitoring (TDM) for MPA fails to accurately reflect intracellular drug exposure at the pharmacological action site. PURPOSE: This study aimed to establish and validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for simultaneous quantification of MPA and its glucuronide metabolite, MPAG, in peripheral blood mononuclear cells (PBMCs)

Development and validation of a UPLC-MS/MS method for real-time neuropharmacokinetic monitoring of iboga alkaloids in rat brain.

Iboga alkaloids, including ibogaine, its primary metabolite noribogaine, and the naturally occurring congener ibogamine, have shown promise for the treatment of substance use and other neuropsychiatric disorders but are limited by cardiotoxic effects. Safer synthetic analogs, such as oxa-noribogaine, are under active development; however, validated analytical methods for quantifying these compounds in brain extracellular fluid are lacking. Existing approaches that rely on tissue homogenate analysis reflect total tissue drug content, encompassing intracellular, protein-bound, and vascular compa

The Value of LC-MS/MS in Apparent Bilateral Aldosterone Suppression in Adrenal Venous Sampling for Primary Aldosteronism.

CONTEXT: Adrenal venous sampling (AVS) is recommended for subtyping primary aldosteronism (PA); however, it is unreliable when apparent bilateral aldosterone suppression (ABAS) occurs, defined as aldosterone/cortisol ratio in the adrenal vein being bilaterally lower than that in the inferior vena cava. The value of liquid chromatography-tandem mass spectrometry (LC-MS/MS) in PA subtyping when ABAS occurs is unclear. OBJECTIVE: This work aimed to determine whether LC-MS/MS can reduce the occurrence of ABAS and misdiagnosis of PA subtyping. METHODS: Aldosterone and cortisol in AVS samples from

SPE-LC-MS/MS optimisation using response surface methodology for quantification of pharmaceuticals, pesticides and organic UV filters included in water protection guidelines.

The environmental concern about micropollutants (MPs) in aquatic environments has grown significantly in recent decades due to their widespread occurrence and potential risks to ecosystems and human health. Monitoring of MPs is crucial for understanding the status of aquatics systems, supporting risk assessments, developing elimination strategies, and informing regulatory decision-makers. However, due to the very low concentrations of MPs (usually ranging between ng L-1 and μg L-1 levels), their quantification requires highly sensitive and precise analytical methods. Solid phase extraction (SP

HPLC-MS/MS-based detection of Pseudomonas aeruginosa secondary metabolites and their application in metabolomics.

An HPLC-MS/MS method was developed for determining Pseudomonas aeruginosa metabolites in culture media and cosmetics. The samples were dispersed in saturated sodium chloride, purified by C18-SPE after acetonitrile extraction, filtered through a PTFE membrane, and then analyzed. The mobile phase system used was 0.1 % formic acid-methanol. Positive and negative ions are scanned simultaneously, and multiple reaction monitoring (MRM) mode was used for detection. The results revealed that the linear correlation coefficients (r) of the 17 metabolic substances were all greater than 0.99 within the li

Identification of potential biomarkers for diabetic nephropathy via UPLC-MS/MS-based metabolomics.

OBJECTIVE: Diabetes mellitus (DM) is a prevalent chronic disease, with diabetic nephropathy (DN) being a significant complication. Early detection of DN is critical for effective management. Current diagnostic methods, such as urinary albumin-to-creatinine ratio (uACR) and estimated glomerular filtration rate (eGFR), have limitations. Metabolomics offers a promising alternative by identifying specific metabolic signatures associated with DM and DN. This study aimed to identify potential metabolic biomarkers of DN using metabolomics. METHODS: A total of 100 participants were recruited, includi

Development of an Optimized LC-MS Workflow for Host Cell Protein Characterization to Support Upstream Process Development.

Host cell proteins (HCPs) coexpressed during the production of biotherapeutics can affect the safety, efficacy, and stability of the final product. As such, monitoring HCP populations and amounts throughout the production and purification process is an essential part of the overall quality control framework. Mass spectrometry (MS) is used as an orthogonal method to enzyme-linked immunosorbent assays (ELISA) for the simultaneous identification and quantification of HCPs, particularly for the analysis of downstream processes. In this study, we present an MS-based analytical protocol with improve
