Moderate Elevation of Ambient Temperature Alleviates Growth Reduction Caused by Phosphorus Deficiency in Lotus japonicus Under Hydroponic Conditions.

High-temperature stress driven by global warming and phosphorus (P) deficiency resulting from the depletion of phosphate rock reserves pose serious threats to modern agriculture. However, our understanding of the combined effects of these stresses on plants remains limited. In this study, we employed a hydroponic culture system to investigate the physiological and transcriptomic responses of the model legume Lotus japonicus to simultaneous moderate high-temperature and low-P conditions. Both elevated temperature and low-P individually reduced shoot biomass. Under low-P conditions, shoot biomas

A case of renal hypouricemia type 1 associated with compound heterozygous URAT1/SLC22A12 variants A209V and F379L: clinical evidence for their pathogenic significance.

Renal hypouricemia (RHUC) is an inherited renal tubular disorder characterized by low serum uric acid levels and excessive urinary uric acid excretion due to impaired urate reabsorption in proximal tubules. Type 1 RHUC (RHUC1) is caused by pathogenic variants in URAT1/SLC22A12. In Japanese patients, recurrent variants such as W258X and R90H account for a substantial proportion of RHUC1 cases; however, the clinical significance of rare URAT1 variant combinations remains incompletely understood. A 69-year-old woman was referred to our hospital for evaluation of renal dysfunction and hypouricemia

ER-Nε-lysine acetylation machinery in neurological disorders: A context-dependent regulator of proteostasis.

Despite extensive characterization of cytosolic and nuclear acetylation, the functional relevance of luminal ER-Nε-lysine acetylation in the nervous system remains poorly understood. This modification is mediated by a coordinated metabolic-secretory axis involving acetyl-CoA availability and the ER-resident acetyltransferases ATase-1 (NAT8B) and ATase-2 (NAT8), which depend on acetyl-CoA import into the ER via the transporter SLC33A1 (AT-1). Upstream, the citrate transporters SLC25A1 and SLC13A5 contribute to shaping the cytosolic acetyl-CoA pool. Emerging evidence identifies this pathway as a

Pt/Ce-Sm-Ox catalysts for stable steam-assisted biogas reforming: comparative performance and carbon accumulation.

Steam-assisted biogas reforming can tune syngas composition, but long-term operation is limited by catalyst deactivation and carbon accumulation. To compare the effects of Ce-, Sm-, and combined Ce/Sm-containing oxide environments under identical conditions, Pt/CeO2, Pt/Sm2O3, and Pt/Ce-Sm-Ox catalysts containing a nominal Pt loading of 1 wt% were prepared by a citrate-assisted sol-gel method. Under a CH4/CO2/H2O/N2 feed ratio of approximately 3:2:2.7:1, Pt/Ce-Sm-Ox achieved approximately 91 % CH4 conversion and 55 % CO2 conversion at 800 °C. During the 100 h test, the CH4 and CO2 conversions

Targeting Dyslipidemia in Ischemic Stroke: Pathophysiology, Clinical Evidence, and Future Perspectives.

Dyslipidemia is a major risk factor for atherosclerosis, and high low-density lipoprotein (LDL) cholesterol is closely associated with the onset of atherosclerotic cardiovascular disease (ASCVD), including coronary artery disease. Numerous large-scale clinical trials have demonstrated that LDL cholesterol-lowering therapy, primarily involving statins, reduces not only coronary events, but also stroke, and has been established as a cornerstone of cardiovascular prevention. However, ischemic stroke is a group of diseases comprising different pathologies, such as atherothrombotic stroke, cardioem

Integrative single‑cell and bulk transcriptomic profiling reveals MMP1 as a core effector and CTSB as a putative collaborator in ATBC‑aggravated pulmonary fibrosis.

BACKGROUND: Acetyl tributyl citrate (ATBC) is a widely used environmentally friendly plasticizer, and exposure via the respiratory tract is common in the general population. However, its pulmonary toxicity and underlying molecular mechanisms remain poorly understood. METHODS: An integrative strategy combining compound target prediction, IPF transcriptome screening, Bootstrap LASSO stability selection, and protein‑protein interaction network topological analysis was used to identify core targets. Single-cell transcriptomics was applied to dissect the cellular localization and functional roles

Phase transfer studies of iron oxide nanoparticles: characterization based insights for applications.

This study systematically explores the synthesis and functionalization of iron oxide nanoparticles (IONPs) synthesized via thermal decomposition methods using three common precursors: iron(iii) oleate (FeOl), iron pentacarbonyl [Fe(CO)5], and iron acetylacetonate [Fe(acac)3]. Comprehensive characterization was employed to evaluate the morphology, crystal structure, and magnetic properties of the synthesized IONPs, alongside four phase transfer strategies. Among the investigated methods, Fe(acac)3-based synthesis predominantly yielded Fe3O4 nanoparticles with superior crystallinity and magnetic

Therapeutic targeting of lipid metabolism in cancer: Mechanisms, pathways, and translational prospects.

Cancer cells undergo intense metabolic reprogramming to provide fast proliferation, persistence, immune evasion, and resistance to therapy. Malignant cells often rely on exogenous lipids and maintaining strict metabolic regulation. Cancer cells stimulate de novo lipogenesis, lipid uptake, and storage pathways even in nutrient-limited microenvironments. This review combines current knowledge of cancer types, highlights vital enzymatic regulators and translational prospects. A comprehensive literature review was conducted, focusing on the complex relationship between lipid metabolic pathways and

Suppressive Genetic Interactions Between Haploinsufficient Mitochondrial Genes Encoded in the 22q11.2 Microdeletion Locus Define Brain and Cardiac Phenotypes.

Genomic copy number variations, such as the 22q11.2 microdeletion syndrome, cause pleiotropic disorders that affect diverse organ systems and disrupt neurodevelopment. Deletions of the 22q11.2 locus reduce the dosage of up to 46 protein coding genes, raising questions about the identity of haploinsufficient genes and their genetic interactions contributing to 22q11.2 phenotypes. Here, we dissect functional and molecular relationships between two genes encoded within the 22q11.2 locus: the mitochondrial ribosomal protein gene MRPL40 and the mitochondrial citrate transporter SLC25A1. We show tha

Single-phase silver nanoparticles from white tea: colloidal quality, mass-specific potency and bacterial envelope permeabilisation.

Silver nanoparticles (AgNPs) are broad-spectrum antibacterial materials that remain active against clinically relevant pathogens, but tea-mediated syntheses have focused predominantly on green tea and have generally emphasised potency without linking particle properties to downstream damage of the bacterial envelope. Here, we synthesised AgNPs using white tea (Camellia sinensis) and benchmarked them against a citrate-reduced preparation produced and assayed in parallel. X-ray diffraction identified face-centred-cubic Ag in both preparations; the white-tea product showed no crystalline silver-s

Simultaneous Inhibition of ACLY and OGDH Has a Synergistic Effect on Hepatocellular Carcinoma Cell Lines.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related mortality worldwide. Current treatments offer limited efficacy and no definitive cure, underscoring the urgent need for more selective and effective therapeutic strategies. This study investigated the synthetic lethality caused by co-targeting two metabolic genes, ATP citrate lyase (ACLY) and oxoglutarate dehydrogenase (OGDH), in HCC cells. Using valproic acid (VPA) and bempedoic acid (BA) as pharmacological inhibitors of OGDH and ACLY, respectively, we observed a strong synergistic effect in inhibiting the proliferation

Metabolic profiling identifies fertility markers in bull sperm†.

Early and accurate assessment of bull fertility is critical for the success of artificial insemination (AI) programs in cattle production. However, current selection tools, including genomic predictions and standard semen evaluations, offer limited reliability in forecasting field fertility outcomes. To address this limitation, we explored the sperm metabolome as a potential source of novel fertility-associated biomarkers. Using high-throughput untargeted metabolomics, we profiled frozen-thawed sperm from Holstein-Friesian bulls with high (n = 12) and low (n = 12) adjusted fertility scores, ea

Molecular interactions and viscosity regulation mechanism of IgG4 antibody in high-concentration solutions.

A major bottleneck in developing high-concentration (≥ 100 mg/mL) monoclonal antibody (mAb) formulations for subcutaneous injection is the exponential increase in viscosity, often exceeding the 20 mPa·s clinical threshold value. Using an IgG4 antibody (pI ≈ 6.9) as the model in a pH 5.5 sodium citrate buffer, we innovatively used amino acids and salts (AAs/salts) as "molecular probes" to investigate their viscosity-reducing efficacy, mechanisms, and effects on the formulation stability. Compared with the control group without viscosity-reducing agents, L-Arginine (Arg) achieved a maximum visco

Integrated transcriptomic and metabolomic analysis of copper sulfate-induced hepatotoxicity in Oreochromis niloticus via glutathione depletion and energy metabolic reprogramming.

Copper is a widespread heavy metal pollutant in aquatic ecosystems, but the systemic molecular mechanisms of copper-induced hepatotoxicity in fish remain incompletely understood. Here, we integrated histopathological examination, biochemical assays, transcriptomics, and metabolomics to investigate the hepatotoxic effects of copper sulfate (CuSO₄) exposure in Oreochromis niloticus. CuSO₄ exposure caused typical pathological damage, including hepatocyte swelling, vacuolar degeneration, and pyknosis. Biochemical assays revealed significantly reduced activities of superoxide dismutase (SOD) and ca

Distinct and indispensable roles of cell viability and extracellular polymeric substances in Pb mineralization by Sinorhizobium sp. C10.

Microbially induced phosphate mineralization (MIPP) is a promising strategy for immobilizing lead (Pb) in contaminated environments. This study aimed to determine whether cell viability and EPS are indispensable for Pb mineralization by Sinorhizobium sp. C10, a strain previously shown to form lead phosphate minerals. We compared live versus heat-inactivated cells, employed a mild sodium citrate method to strip EPS while preserving cell viability, and determined Pb removal efficiency, alkaline phosphatase (ALP) activity, inorganic phosphate (PO43-) release, and mineral phase composition.
