Protein-fermentation biomarkers in plasma and urine differ between bovine plasma protein and whey protein isolate in a randomized fully controlled dietary intervention.

BACKGROUND: Microbial fermentation of undigested proteins in the large intestine can produce potentially harmful metabolites. Although protein intake increases large intestinal protein inflow, the effects of dietary protein composition and digestibility are largely unknown. OBJECTIVES: This study aimed to investigate the effects of protein sources differing in amino acid (AA) composition and digestibility on protein-fermentation biomarkers (primary outcomes), digesta transit, and gut microbiome composition (secondary outcomes), by contrasting protein and precursor AA delivery into the large i

Evaluation of exogenous phytase in high-phytate diets for broiler chickens and pigs.

There is a gap in the understanding of the relationship between dietary phytate levels and the relative efficacy of phytase to improve amino acid (AA) digestibility in pigs and chickens. Two experiments were conducted to investigate the effect of exogenous phytase on standardized ileal digestibility (SID) of AA and the apparent ileal digestibility (AID) of P in both standard- (SP) and high-phytate (HP) diets for broilers and swine. There were either 40 cages of Cobb 500 male broilers or 10 crossbred barrows (35 kg) fitted with ileal T-cannulas. Both studies were allotted to five dietary treatm

Phosphorus equivalency of exogenous phytase relative to phosphorus in monosodium phosphate in broiler chickens.

The objective of this study was to estimate phosphorus (P) equivalency of exogenous phytase relative to P from monosodium phosphate (MSP) in broiler chickens. To attain this objective, the impact of dietary MSP or exogenous phytase on growth performance, ileal digestibility of P, and bone characteristics in broiler chickens was assessed. Six experimental diets consisting of a P-deficient basal diet based on corn and soybean meal, basal diet plus 0.9 or 1.8 g/kg of inorganic P from MSP, and basal diet plus 500, 1,000, or 2,000 FYT/kg of exogenous phytase. Calcium to P ratio in all diets was mai

Low-calcium diets increase duodenal mRNA expression of calcium and phosphorus transporters and claudins but compromise growth performance irrespective of microbial phytase inclusion in broilers.

The hypothesis that dietary inclusion of microbial phytase improves apparent calcium (Ca) digestibility thereby allowing a lower dietary Ca inclusion without compromising growth performance was tested. One-day-old male Ross 308 broilers (25 birds/pen, 9 pens/treatment) were assigned to 8 experimental diets containing one of 4 dietary Ca to retainable P (rP) ratios (1.3, 1.8, 2.3, and 2.8) with (1,000 FTU/kg) or without microbial phytase. On d 21 to 23, digesta from different intestinal segments of 8 birds per pen were collected to determine apparent Ca and P digestibility. Mid duodenal mucosa

Incidental findings of thickening luminal gastrointestinal organs on computed tomography: an absolute indication for endoscopy.

OBJECTIVES: Incidental findings of thickened luminal GI organs (LGIO) on CT are not uncommon. However, the significance of these findings is unclear. Because of the lack of scientific data, there are no clinical guidelines for the evaluation of these radiologic abnormalities. Our objective was to determine whether endoscopic evaluation of these findings revealed significant abnormalities. METHODS: This study evaluated all incidental findings of thickened LGIO in a large medical center from October, 1997 to March, 1999 that were followed by endoscopic examinations. RESULTS: Ninety-six percent

Gummivory and gut morphology in two sympatric callitrichids (Callithrix emiliae and Saguinus fuscicollis weddelli) from western Brazilian Amazonia.

A comparative analysis of the gastrointestinal tracts of wild-caught marmosets, Callithrix emiliae, and tamarins, Saguinus fuscicollis weddelli, was undertaken in order to evaluate the degree of specialisation for digestion of plant exudates. Compared to S.f. weddelli, C. emiliae exhibits a reduced small intestine and a relatively large, compartmentalised caecum in which gum is probably fermented. The apparent specialisation of the digestive tract in C. emiliae correlates with that of its dentition, which is adapted for gouging the bark of gum-producing plants. A similar degree of specialisati…

The bacterial flora of neonates with congenital abnormalities of the gastro-intestinal tract.

The development of the bacterial flora of neonates with congenital abnormalities of the gastro-intestinal tract was studied in 31 infants during the first 10 days of life. Specimens were collected from the umbilicus, mouth and gastro-intestinal tract on the pre-operative day, at operation and on post-operative days 1, 2, 3, 5, 7 and 10. Bacteria were isolated semi-quantitatively on a variety of plain and selective media and identified by conventional methods.Staphylococcus albus was the predominant species isolated from the umbilicus; it was recovered from 24 of the 31 babies. The viridans gro

Automated strain-to-peptide conversion: a high-throughput proteome analysis platform empowering rational design of microbial cell factories.

Mass spectrometry-based proteomics offers a powerful tool for characterizing enzyme expression in engineered strains, yet rapid generation of large strain libraries creates proteomic analysis bottlenecks. The critical limitation lies in manual sample preparation-protein extraction, denaturation, reduction, desalting, and digestion-which is time-consuming and risks compromising reproducibility. To overcome this bottleneck, we developed a novel "strain-to-peptide conversion" (SPC) strategy for high-throughput proteome profiling in microbial cell factories. This automated workflow integrates bact…

Isolation and Dissection Procedures for EGC Isolation by Human Mucosal Biopsies.

This chapter provides a detailed guide for isolating and dissecting enteric glial cells (EGCs) from human mucosal biopsies. EGCs play crucial roles in gut homeostasis and neuroimmune communication, making their isolation a valuable endeavor in neurogastroenterology research. The method outlined here aims to enable competent scientists, even those unfamiliar with the technique, to successfully isolate EGCs at their first attempt. It outlines a systematic approach for the collection, digestion, mechanical dissection, and purification of cells from human mucosal biopsies, primarily focused on gas…

GC-rich regions affect amplification efficiency of neighboring amplicons in qPCR and digital PCR.

DNA polymerase chain reaction (PCR) has been widely used for the quantitation of DNA and RNA samples. Multiple factors can affect amplificat … -rich region, with the nearest amplicon yielding only 9% of the reference value. Restriction enzyme digestion that physically separated amplicons from the GC-rich region restored efficiency to approximately 100%. In qPCR, the effect on quantitation depended on whether the standard curve material and the sample shared similar neighboring GC-rich sequence con …
