Carprofen, a carbazole derivative, has been found to be highly potent as anti-inflammatory agent. After a single dose of 100 mg, plasma caprofen peaked in 1 to 2 hours, and t 1/2 was estimated to be 4.4 +/- 1.4 hours, based on a monoexponential fit of the data. Cumulative urinary excretion of total carprofen in 24 hours was almost 30% of the dose, chiefly as glucuronide conjugate. With simultaneous administration of carprofen and probenecid, peak plasma carprofen doubled. The t 1/2 was somewhat prolonged, and urinary excretion was significantly reduced. Phencyclidine (PCP) impaired spatial alternation performance in rats. This effects was mimicked by antimuscarinic anticholinergics (scopolamine, atropine) and PCP derivatives (ketamine, cyclohexamine) but not by a variety of other agents. Muscarinic cholinergic agonists antagonized PCP. Impairment of spatial alteration performance by PCP appears to be mediated, at least in part, by and anticholinergic action; this is the first instance of a behavioral effect of PCP that can be largely attributed to a specific mechanism.

The influence of treatment with allopurinol (5 mg/kg/die for 15 days) on T/2 of tolbutamide and rifomycin-SV intravenously administered, has been studied in 10 healthy volunteers. Using oxygen uptake as an index of the physiological response of isolated parietal cells, the interactions between histamine and gastrin and between histamine and carbamylcholine and the effects of atropine and metiamide on these interactions have been studied. Our review of published material, manufacturer's files, and submissions to the United States Food and Drug Administration indicates that cimetidine is safe for short term use (up to 8 weeks). This judgment is based on a total experience with over 3000 patients. Clinically insignificant elevations of serum creatinine occur in a high percentage of patients, persisting until the end of therapy, then disappearing. Gynecomastia occurred in some patients on long term therapy.

A GLC assay to quantitate the methaqualone metabolite 2-methyl-3-(2-hydroxymethylphenyl)-4(3H)-quinazolinone was developed. Standard curves were linear, and recovery of the metabolite from tissue homogenates averaged 89%. The joint toxic action of mixtures of perchloroethylene with benzene or toluene was tested at five dose levels for each of six binary combinations with 20 rats per dose. LD50 data with 95% fiducial limits are reported for each mixture and tests for additive joint toxicity were made. The results indicate that the mixtures of perchloroethylene and benzene gave values for the LD50 which were slightly less than additive while perchloroethylene and toluene mixtures had LD50 values which could not be predicted by an additive model.
