Central nervous system effects of the neurohypophyseal hormones and related peptides.

This review of the CNS effects of the neurohypophyseal hormones and related neuropeptides discusses recent data illustrating the significance of these principles in brain function, synthesis, distribution, in particular in extrahypothalamic brain structures, binding sites, and signal transduction. Binding sites for vasopressin of the vascular V1a type have been found in the CNS and there is evidence for the existence of a subtype of the antidiuretic V2 receptor in the brain. Also two types of oxytocin binding sites have been detected. One widely distributed throughout the CNS is comparable to

[Recent development in research and management of cancer anorexia-cachexia syndrome].

Cachexia is among the most debilitating and life-threatening aspects of cancer, and is more common in children and elderly patients. Associated with anorexia, fat and muscle tissue wasting, psychological distress, and a lower quality of life, cachexia arises from a complex interaction between the cancer and the host. This process results from a failure of the adaptive feeding response seen in simple starvation and includes cytokine production, release of lipid-mobilizing and proteolysis-inducing factors, and alterations in intermediary metabolism. Cytokines play a pivotal role in long-term inh

Sleep restriction induces histoarchitectural and cellular remodeling in the rat distal colon: a peripheral consequence of circadian disruption.

Sleep restriction disrupts circadian organization and represents a behavioral condition with systemic consequences that extend beyond the central nervous system. In this study, we evaluated structural and cellular changes in the distal colon of rats exposed to sustained sleep restriction. Sixteen male Rattus norvegicus were assigned to a control group (n = 8) or a sleep-restricted group (n = 8) and subjected to a modified multiple-platform protocol for 21 days. Blood samples were collected for inflammatory assessment, and the distal colon was analyzed using histochemical and immunohistochemica

Non-invasive Assessment of Social Isolation Induced Stress in Tharparkar Calves (Bos indicus).

Accurate and welfare-friendly methods for stress detection in livestock are essential for advancing animal husbandry practices. This study investigated the efficacy of fecal cortisol metabolites (FCM) as a non-invasive biomarker for assessing stress induced by social isolation in calves. Sixteen Tharparkar calves were subjected to an 8-day experimental period, which was divided into three distinct phases: pre-isolation (3 days), isolation (2 days) and reunion (3 days). The experimental design consisted of four replications, each with four calves. Behavioral and physiological observations were

Changes in the orexin system, hypothalamic-pituitary-adrenal axis and circadian rhythm in juvenile-stressed rats.

The orexin system has been demonstrated to play various physiological roles, such as in feeding, sleep, and autonomic neural function. Clinically, orexin receptor antagonists are used as hypnotics, and several clinical trials have suggested their efficacy in major depressive disorder. We investigated the orexin system using juvenile-stressed (3-week foot shock) rats, an animal model of depression. 3-week foot shock rats had significantly reduced prepro-orexin mRNA in the hypothalamus, but not orexin A protein or orexin A -like immunoreactive cells. In contrast, orexin receptor type 1 mRNA and

Silencing of the T-type voltage-gated calcium channel α1 subunit by fungus-mediated RNAi altered the structure of F-actin and caused defective behaviors in Ditylenchus destructor.

BACKGROUND: T-type calcium channels, characterized as low-voltage activated (LVA) calcium channels, play crucial physiological roles across a wide range of tissues, including both the neuronal and nonneuronal systems. Using in situ hybridization and RNA interference (RNAi) techniques in vitro, we previously identified the tissue distribution and physiological function of the T-type calcium channel α1 subunit (DdCα1G) in the plant-parasitic nematode Ditylenchus destructor. METHODS AND RESULTS: To further characterize the functional role of DdCα1G, we employed a combination of immunohistochemis

Household prallethrin-based insecticide toxicity on different C. elegans life stage: A possible sign of Huntington Disease.

Household insecticide is largely used for insect and ectoparasite control, in city centers as well as in the countryside. The pyrethroids are the most used class of insecticide, these compounds in low doses have low toxicity for mammalians, in comparison to other compounds, with insecticide effects. The contact of these compounds in sublethal doses begins in early life and many cases, in intrauterine life. Considerable diseases still with undefined etiology, such as neurodegenerative conditions, and Huntington's Disease (HD) is one of them. HD is related to overexpression of Polyglutamine (Pol

Peripheral Nerve Resident Macrophages and Schwann Cells Mediate Cancer-Induced Pain.

Although macrophages (MΦ) are known to play a central role in neuropathic pain, their contribution to cancer pain has not been established. Here we report that depletion of sciatic nerve resident MΦs (rMΦ) in mice attenuates mechanical/cold hypersensitivity and spontaneous pain evoked by intraplantar injection of melanoma or lung carcinoma cells. MΦ-colony stimulating factor (M-CSF) was upregulated in the sciatic nerve trunk and mediated cancer-evoked pain via rMΦ expansion, transient receptor potential ankyrin 1 (TRPA1) activation, and oxidative stress. Targeted deletion of Trpa1 revealed a k

Clinical and molecular characterization of individuals with recurrent genomic disorder at 10q22.3q23.2.

The identification of genomic imbalances in young patients can affect medical management by allowing early intervention for developmental delay and by identifying patients at risk for unexpected medical complications. Using a 105K-feature oligonucleotide array, we identified a 7.25 Mb deletion at 10q22.3q23.2 in six unrelated patients. Deletions of this region have been described in individuals with cognitive and behavioral abnormalities, including autistic features, and may represent a recurring genetic syndrome. All four patients in this study for whom clinical information was available had

Mind versus metabolism in the control of food intake and energy balance.

In a restrictive food environment, the homeostatic control system regulates body weight and adiposity with remarkable precision. However, this regulation appears to break down in many genetically predisposed individuals under conditions prevailing in the modern era characterized by a sedentary lifestyle and easy availability of large portions of palatable and calorically dense food. The nervous system is the main interface by which food-related environmental factors influence the regulatory process. Thus, focusing on the neural systems located in the telencephalon dealing with environmental fa

Muricidal suppression by histidine and its antagonism by alpha-fluoromethylhistidine in thiamine deficient rats.

Male Wistar rats maintained on a thiamine deficient diet showed mouse-killing behavior (muricide). On the 30th day of experimental feeding, the incidence of muricide was 70%. Intraperitoneal (ip) injection of L-histidine, a precursor of brain histamine, suppressed the muricide induced by thiamine deficiency in a dose dependent manner. The ED50 for muricidal suppression by histidine alone was 155 mg/kg (95% confidence limits; 107.6-223.2 mg/kg). Rotarod performance was concurrently examined as an index of behavioral toxicity. High doses of histidine produced insignificant changes in rotarod per

Reframing Mother-Child Closeness and Cooperative Caregiving: A Comparative Study of Two Hunter-Gatherer Populations.

OBJECTIVES: Caregiving during early childhood emerges from a dynamic interplay between maternal and allomaternal contributions. This study aimed to challenge polarized portrayals of hunter-gatherer childcare as either "mother-centered" or "cooperative" and provide empirical evidence for an integrative cross-cultural approach. MATERIALS AND METHODS: We compared caregiving structures in two hunter-gatherer populations positioned at contrasting ends of a continuum of child-rearing strategies: the !Xun in southern Africa, portrayed as emphasizing mother-child closeness, and the Baka in central Af

Nutritional and Lifestyle Behaviors and Their Influence on Sleep Quality Among Spanish Adult Women.

Background: Sleep is a fundamental component of health, and deprivation has been linked to numerous adverse outcomes, including reduced academic and occupational performance, greater risk of accidents, and increased susceptibility to chronic diseases and premature mortality. Dietary and lifestyle behaviors are increasingly recognized as key determinants of sleep quality. Women are particularly susceptible to sleep disturbances due to hormonal fluctuations and psychosocial factors. However, women remain underrepresented in sleep research. This study aims to examine the associations between slee

Association of body mass index with progression from binge-eating behavior into binge-eating disorder among adolescents in the United States: A prospective analysis of pooled data.

The association between body mass index (BMI) and binge-eating disorder (BED) is well-established. However, data on the extent to which BMI is associated with progression from binge-eating behavior into BED among adolescents are limited, which was the aim of this investigation. Participants were 9964 U.S. adolescents from the Adolescent Brain Cognitive Development (ABCD) Study, aged 9-13 at the time of study enrollment. A computerized parent-reported assessment was used to establish adolescents' binge-eating behaviors and BED. Cox proportional hazards models adjusting for sociodemographic cova

Loss of Snord116 impacts lateral hypothalamus, sleep, and food-related behaviors.

Imprinted genes are highly expressed in the hypothalamus; however, whether specific imprinted genes affect hypothalamic neuromodulators and their functions is unknown. It has been suggested that Prader-Willi syndrome (PWS), a neurodevelopmental disorder caused by lack of paternal expression at chromosome 15q11-q13, is characterized by hypothalamic insufficiency. Here, we investigate the role of the paternally expressed Snord116 gene within the context of sleep and metabolic abnormalities of PWS, and we report a significant role of this imprinted gene in the function and organization of the 2 m

Cardiometabolic Risk in First Episode Psychosis Patients.

Previous research in patients with schizophrenia in European and USA population groups has demonstrated a high prevalence of metabolic syndrome and disease progression (~35%-40%) and increased risk for cardiovascular disease and long-term mortality. Limited research has determined the prevalence of existing cardiometabolic risk factors at onset of a first episode psychosis. This cross-sectional study presents a clinical overview of the cardiometabolic risk profile in young people with first episode psychosis in the UK. Forty-six participants (72% male) clinically diagnosed with first episode p
