Noninvasive prenatal test for FGFR3-related skeletal dysplasia based on next-generation sequencing and plasma cell-free DNA: Test performance analysis and feasibility exploration.

OBJECTIVE: To explore the feasibility and accuracy of a noninvasive prenatal test for fibroblast growth factor receptor 3 (FGFR3)-related skeletal dysplasia based on next-generation sequencing (NGS) of plasma cell-free DNA. METHOD: Fragmented genome DNA (gDNA) of fetuses with achondroplasia (ACH) and thanatophoric dysplasia type I (TD I) was mixed with postdelivery maternal plasma cell-free DNA to generate spiked samples of different modeled fetal fractions. Multiplex polymerase chain reaction was used to amplify the 19 FGFR3 loci, and the amplification products were then sequenced by NGS to

Combined cardiotocographic and ST event analysis: A review.

ST-analysis of the fetal electrocardiogram (ECG) (STAN(®)) combined with cardiotocography (CTG) for intrapartum fetal monitoring has been developed following many years of animal research. Changes in the ST-segment of the fetal ECG correlated with fetal hypoxia occurring during labor. In 1993 the first randomized controlled trial (RCT), comparing CTG with CTG + ST-analysis was published. STAN(®) was introduced for daily practice in 2000. To date, six RCTs have been performed, out of which five have been published. Furthermore, there are six published meta-analyses. The meta-analyses showed tha

Urinary outflow obstruction increases apoptosis and deregulates Bcl-2 and Bax expression in the fetal ovine bladder.

During organogenesis, net growth of tissues is determined by a balance between proliferation, hypertrophy, and apoptotic death. Human fetal bladder outflow obstruction is a major cause of end-stage renal failure in children and is associated with complex pathology in the kidney and lower urinary tract. Experimental manipulation of the fetal sheep urinary tract has proved informative in understanding the pathobiology of congenital obstructive uropathy. In this study we used an ovine model of fetal bladder outflow obstruction to examine effects on apoptotic cell death in the developing urinary b

Developmental and behavioral effects of fetal and neonatal exposure to the phenylpyrazole pesticide fipronil in mice.

Fipronil (FPN), a phenylpyrazole pesticide widely used in agriculture and households, acts as a noncompetitive antagonist of gamma-aminobutyric acid (GABA)-gated chloride channels. Although originally considered relatively safe for mammals at low doses, FPN is metabolized into fipronil sulfone (FPNS), a more toxic and persistent metabolite. Both FPN and FPNS have been detected in human biological samples, raising concerns about chronic exposure during critical developmental windows. In this study, pregnant mice were administered FPN orally at 0.43 mg/kg/day from gestational day 1.5 through wea

Emerging ultrasound-based imaging of the placenta during complications of pregnancy.

The placenta is an essential organ that develops throughout pregnancy, facilitating the exchange of nutrients, waste products, hormones, and antibodies between the fetal and maternal circulations. Insufficient development of placental structure and function is associated with various pregnancy disorders, including preeclampsia (PE), fetal growth restriction (FGR), gestational diabetes mellitus, preterm birth, and potential fetal cardiovascular diseases. Analyzing the spectrum of placental pathology in both normal and abnormal placentas helps to better understand disease pathologies and reduce

Birth Defects Related to Prenatal Exposure to Essential and Toxic Elements.

Background All naturally occurring metals and other elements are present in at least trace concentrations in soil, water, air, and organisms. Accumulation of these elements can lead to developmental delays, neurological damage, and even death. Although the toxicities associated with metals such as lead or mercury are well-demonstrated, little data exists regarding birth defects and maternal environmental exposures to other trace essential or toxic elements. Methodology A prospective case-control study of 23 patients delivering at the University of Mississippi Medical Center with prenatally dia

Associations of per- and polyfluoroalkyl substances (PFAS) exposure and thyroid hormone concentration during pregnancy in the LIFECODES cohort.

BACKGROUND: Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals widely used in consumer products such as firefighting foams and non-stick cookware. Increasing evidence links PFAS exposure to endocrine disruption and metabolic disease. Pregnancy is a particularly vulnerable period, and disruption of maternal thyroid hormones is of concern given their critical role in metabolism and fetal brain development. METHODS: We examined associations between prenatal PFAS exposure and maternal thyroid hormone concentrations among 434 participants in the LIFECODES birth cohort in Boston. Pl

Prenatal exposure to wildfire fine particulate matter and fetal growth outcomes in the United States: Findings from the ECHO Cohort.

BACKGROUND: Increasing exposure to wildfire fine particulate matter (PM2.5) may pose unique risks to fetal development. We assessed associations between prenatal wildfire PM2.5 exposure and fetal growth outcomes, including birth weight at term, birth weight for gestational age z-scores, small for gestational age, and large for gestational age in the U.S. Environmental Influences on Child Health Outcomes Cohort. METHODS: The study included 20,034 births (2006-2020) from 30 cohort sites nationwide. We assigned daily wildfire PM2.5 at census-tract resolution and summarized average concentrations…

Tim1 Deficiency Mediates Gestational Hyperglycemia-Related Syncytiotrophoblast Dysfunction and Fetal Growth Restriction.

Gestational hyperglycemia (GHG) causes fetal growth restriction (FGR), while its mechanism remains incompletely understood. Defects in the syncytiotrophoblast, which is pivotal for maternal-fetal substance exchange, adversely affect fetal development. Whether dysfunction in syncytiotrophoblast is involved in GHG-related FGR remains unclear. In this study, we used an STZ-induced GHG mouse model and found that GHG-induced FGR (44.5% reduction in fetal weight) was associated with a 28.3% decrease in placental efficiency. Immunofluorescence and transmission electron microscopy examinations reveale…

Fetal exposure to persistent organic pollutants and childhood autism risk.

Exposure to persistent organic pollutants (POPs) during fetal development may increase the risk of autism spectrum disorder, but previous studies have relied on maternal pregnancy samples as proxies instead of directly measuring fetal exposures. Here, we used naturally shed deciduous teeth from autism cases and typically developing controls born between 1998 and 2014 from the California, USA, population-based case-control Childhood Autism Risk from Genetics and the Environment study. Using gas chromatography-tandem mass spectrometry, we quantified fetal second and third trimester and postnatal…

Longitudinal Ultrasound Imaging of an Individual Fetus In Utero Using an Echogenic Marker.

OBJECTIVE: High frequency ultrasound imaging has become an important tool for the study of pregnancy disorders in mouse models as it enables detailed, noninvasive characterization of in utero development. However, the need to reliably identify individual fetuses over time presents a challenge when using this approach for longitudinal assessments. Large litter sizes and frequently changing fetal positions within the abdomen across gestation have the potential to confound such measurements. METHOD: The current study presents a minimally invasive method to longitudinally track a specific mouse f

Elevated levels of exogenous prolactin promote inflammation at the maternal-fetal interface via the JAK2/STAT5B signaling axis.

The placenta is a unique organ with various immunological and endocrinological roles that modulate maternal and fetal physiology to promote maternal-fetal tolerance, pregnancy maintenance, and parturition at term. During pregnancy, the hormone prolactin (PRL) is constitutively secreted by the placenta and is necessary for implantation, progesterone support, fetal development, and overall immune modulation. While PRL is essential for pregnancy, studies suggest that elevated levels of serum PRL (hyperprolactinemia) are associated with adverse pregnancy outcomes, including miscarriage, preterm bi…

Meiosis and retinoic acid in the mouse fetal gonads: An unforeseen twist.

In mammals, differentiation of germ cells is crucial for sexual reproduction, involving complex signaling pathways and environmental cues defined by the somatic cells of the gonads. This review examines the long-standing model positing that all-trans retinoic acid (ATRA) acts as a meiosis-inducing substance (MIS) in the fetal ovary by inducing expression of STRA8 in female germ cells, while CYP26B1 serves as a meiosis-preventing substance (MPS) in the fetal testis by degrading ATRA and preventing STRA8 expression in the male germ cells until postnatal development. Recent genetic studies in the

Analysis of characteristic features in ultrasound diagnosis of fetal limb body wall complex during 11-13+6 weeks.

BACKGROUND: Limb body wall complex (LBWC) is a fatal malformation characterized by major defects in the fetal abdominal or thoracic wall, visceral herniation, significant scoliosis or spina bifida, limb deformities, craniofacial deformities, and umbilical cord abnormalities (short or absent umbilical cord). Early diagnosis of this condition is of great clinical significance for clinical intervention and pregnancy decision-making. With the rapid development of fetal ultrasound medicine, early pregnancy (11-13+6 wk) standardized prenatal ultrasound examinations have been widely promoted and appl

Maternal iron supplementation during pregnancy affects placental function and iron status in offspring.

BACKGROUND: Iron deficiency and overload during pregnancy damage to maternal and fetal health. Placenta as an organ for the transport of nutrients between mother and fetus protects fetus from the harmful effects of iron deficiency and iron overload through regulation of placental iron homeostasis. METHODS: To determine the effect of dietary iron supplementation during pregnancy on reproduction and the mechanism of placental iron regulation, we designed dietary high iron (HI: 344 mg/kg), medium iron (MI: 40 mg/kg), low iron (LI: 2 mg/kg) groups of pregnant female mice fed ferrous citrate 2 wee

Interstitial Cells of Cajal and Neural Structures in the Human Fetal Appendix.

BACKGROUND/AIMS: The interstitial cells of Cajal (ICC) are located within and around the digestive tract's muscle layers. They function as intestinal muscle pacemakers and aid in the modification of enteric neurotransmission. The appendix's unique position requires an appropriate contraction pattern of its muscular wall to adequately evacuate its contents. We investigated the development and distribution of nervous structures and ICC in the human fetal appendix. METHODS: Specimens were exposed to anti-c-kit (CD117) antibodies to investigate ICC differentiation. Enteric plexuses were examined

Distal villous lesions are clinically more relevant than proximal large muscular vessel lesions of placental fetal vascular malperfusion.

BACKGROUND: Fetal vascular malperfusion (FVM) can be diagnosed on placental examination based on histology of distal placental villi and large muscular placental vessels. While histology of both those placental compartments can be low grade or high grade, it is not known if these are clinically equivalent. This retrospective study aimed to compare the impact of placental distal villous and large vessel FVM lesions on clinical and placental phenotypes. METHODS: Clinical and placental phenotypes of 479 consecutive ≥20 weeks of gestation at delivery cases of placental FVM were analyzed among 3 g

Cytomegalovirus infection in pregnancy - An update.

Cytomegalovirus (CMV) is a ubiquitous DNA virus with a global seroprevalence of 83 %. It is the most common pathogen causing teratogenic congenital infection. It is therefore a major public health concern. Maternal infection is associated with congenital CMV (cCMV), the leading cause of non-genetic sensorineural hearing loss. cCMV also causes impairment of cognitive development and cerebral palsy. Transmission of CMV occurs through direct contact with bodily fluids such as saliva, urine or semen from someone who is actively shedding the virus. Transmission rates are higher after primary infect

The fatal fetal tumor: a geneticist's perspective.

Epignathus is an extremely rare oral teratoma which leads to high mortality in the early neonatal period. Various theories have been put forward for the genesis of such a tumor, though none is completely convincing. A genetic basis is not well established for the tumor. Microdeletions/duplications, as well as single gene disorders, have been known to cause epignathus, all with additional malformations. Evidence of single gene involvement in an isolated epignathus is lacking. We present a case of a 19-week-fetus with oro-pharyngeal teratoma detected on the level II ultrasound. The couple was co
