Genomic and metabolomics analysis: unlocking the biocontrol mechanism of Trichoderma virilente Z53 against anthracnose in Pleione yunnanensis.

Anthracnose caused by Colletotrichum spp. is harmful to orchidaceous plants including Pleione yunnanensis. The use of microorganisms for biocontrol is considered safe and effective. This study demonstrated that Trichoderma virilente Z53 not only exhibits strong antagonistic activity against 12 phytopathogenic fungal species but also promotes maize seed germination and seedling growth under in vitro conditions and effectively suppresses anthracnose disease in Pleione yunnanensis. Pot studies demonstrated that Z53, when used as a soil inoculant, significantly promoted plant height, root length,

Microbiota-driven epigenetic programming of local immunity.

The maintenance of local immunity requires stable tissue adaptation, implying that persistent environmental cues must continuously reinforce niche-specific immune programs. Microbiota-derived metabolites are increasingly recognized as such cues, acting not merely as metabolic byproducts but as epigenetic inputs that regulate histone modifications, DNA and histone methylation, and non-coding RNA networks. Through these epigenetic mechanisms, microbial metabolites reprogram key immune populations by promoting tolerogenic or immunostimulatory states, tuning effector and memory differentiation, an

The Role of Genomics in the Development and Treatment of Multiple Myeloma: Understanding the Challenges and Opportunities.

This review aims to provide a comprehensive overview of recent advances in the genomics of multiple myeloma and their clinical implications. Multiple myeloma is a hematologic malignancy originating from bone marrow plasma cells, characterized by the infiltration of pathological plasma cells, osteolytic bone lesions, and the presence of monoclonal immunoglobulins in serum and/or urine. Multiple myeloma exhibits significant genetic heterogeneity, which is the core reason for the considerable variability in patient prognosis, differential treatment responses, and the eventual development of drug

Ventricular arrhythmia and Noonan syndrome with leucine zipperlike transcription regulator 1 mutations: expanding the phenotype with a case report and review of the literature.

BACKGROUND: Noonan syndrome is characterised by typical facial features, short stature, CHDs, and other comorbidities, which are caused by germline mutations in genes coding for components of the Ras-mitogen-activated protein kinase pathway. Noonan syndrome is an inherited disease involving multiple systems, but ventricular arrhythmia in Noonan syndrome is rarely reported. CASE SUMMARY: Here we report a 15-year-old patient with leucine zipperlike transcription regulator 1-associated Noonan syndrome, who has CHD (left ventricular hypertrophy with left ventricular outflow tract obstruction), ve

Extracellular vesicle-mediated drug resistance in cancer.

Drug resistance remains a major challenge to successful cancer treatment. Although cell-autonomous genetic, molecular, and cellular modifications have historically been linked to resistance, there is growing evidence that extracellular vesicles (EVs) can promote the intercellular transfer of resistance-associated components. With a focus on the ways in which EV cargo modifies treatment response, this review critically investigates the role of EVs in therapeutic resistance in breast cancer, lung cancer, leukemia, glioblastoma, pancreatic cancer, and melanoma. The effects of major EV-associated

FNIP1 variants are associated with favourable metabolism in 1 million humans.

Altered energy metabolism is a shared driver across cardiometabolic diseases-the leading cause of death globally1. Energy metabolism varies between individuals and is partly heritable2-9. Here, to investigate the genetic basis of energy metabolism, we perform an exome-sequencing analysis of 1,032,116 people from America, Europe and Asia, and estimate associations between rare protein-coding variants and the ratio of triglyceride to high-density-lipoprotein cholesterol (TG:HDL)-an energy-state biomarker that we associate with diverse cardiometabolic risk factors and diseases. We identify 59 ind

Rare AFG3L2 and POLG Variants Suggest a Role for Mitochondrial Dysfunction in Enteric Neuronal Vulnerability in Idiopathic Achalasia.

Idiopathic achalasia is a rare esophageal motility disorder characterized by the selective degeneration of inhibitory myenteric neurons. Its genetic basis remains poorly defined. We investigated whether rare coding variants may contribute to disease susceptibility. Exome sequencing was performed in 31 individuals with idiopathic achalasia and seven unaffected relatives. Candidate variants were prioritized using phenotype-driven filtering and assessed through in silico and structural analysis of publicly available gene expression and single-cell transcriptomic datasets. No pathogenic or likely

Incident psoriasis in atopic dermatitis: a large-scale cohort study of disease- and treatment-associated risks.

BACKGROUND: Clinical and genetic evidence on the association between atopic dermatitis (AD) and subsequent psoriasis remains conflicting, and it is unclear whether this risk is modified by systemic treatments. Recent reports suggest that type 2-targeted biologics may unmask psoriasis in patients with AD, but data are limited. We thus aimed to assess whether AD is associated with incident psoriasis and whether this risk differs by systemic treatment, particularly biologics versus conventional systemic immunosuppressants (cvIS). METHODS: Scoping analyses informed a locked analytic design, prere

Exploratory Association of 5' and 3' UTRs Variants in TYMS and CCND1 with Severe Hematologic Toxicity in Pediatric Acute Lymphoblastic Leukemia.

Mexico has one of the highest incidences of childhood acute lymphoblastic leukemia worldwide and also reports one of the lowest 5-year overall survival rates; one cause is elevated treatment-related mortality from severe hematologic toxicity, a critical side effect during chemotherapy. Although association studies have identified common genetic variants linked to toxicity, the Mexican population remains incompletely explored. We performed targeted next-generation sequencing of genes involved in methotrexate and mercaptopurine metabolism in 96 Mexican children with ALL, stratified by hematologi

Disruption of local CTCF binding impairs renin expression and blood pressure response in Dahl salt-sensitive rats.

Most genetic variants associated with any complex trait are found in non-coding regions. Multiple studies have reported enrichment in single nucleotide polymorphisms (SNPs) found in specific sequences bound by CCCTCF-binding factor (CTCF). CTCF binds DNA to establish boundaries, mediate chromatin interactions and topological domains, and recruit factors to regulate local gene expression. Hypertension is one such disease with enrichment of SNPs in CTCF-binding sites; however, it is not clear whether disruption of CTCF binding to any specific site in the genome could be sufficient to alter blood

MicroRNA profiles in multiple sclerosis: A review of the literature.

Multiple sclerosis (MS) is an inflammatory demyelinating disorder that affects the central nervous system. Although its exact cause remains unknown, it is known that an autoimmune response, triggered by interactions between genetic predisposition and environmental factors, leads to an attack against the myelin sheath. MicroRNAs (miRNAs), small non-coding regulatory RNAs that modulate post-transcriptional gene expression, may play an important role in MS pathophysiology, affecting immunological pathways such as T-cell differentiation, macrophage activation, and apoptosis regulation. Studies hav

Comparative Genomic Analysis of 66 Bacteriophages Infecting Morganella morganii Strains.

Bacteriophages are viruses that specifically target bacteria and play a crucial role in influencing bacterial evolution and the transmission of antibiotic resistance. In this study, we explored the genomic profiles of 66 bacteriophages that infect Morganella morganii, an opportunistic pathogen associated with difficult-to-treat nosocomial and urinary tract infections. Our findings highlight the extraordinary diversity within this phage population, reflected in their genomic features, evolutionary relationships, and potential contributions to bacterial pathogenicity. The 66 phage genomes exhibi

Synaptic and Non-Synaptic Functions of PTPRD: A Receptor Tyrosine Phosphatase at the Crossroads of Neural Circuitry and Metabolism.

Protein-tyrosine phosphatase receptor-type D (PTPRD) is an adhesion-coupled phosphatase that translates extracellular binding codes into intracellular phosphotyrosine signaling from embryogenesis through adulthood. Alternative inclusion of the Ig-domain mini-exons meA and meB tailors the ectodomain surface, thereby dictating high-affinity engagement with IL1RAPL1, IL1RAP, Slitrks, LRFN4/5 (SALM3/5), neuroligin-3, and other postsynaptic partners. Intracellularly, the catalytically active D1 domain and scaffold-like D2 module, anchored to liprin-α, coordinate presynaptic vesicle release, postsyn

LRRK2 as a Potential Disease-Modifying Target in Sporadic Parkinson's Disease.

A growing understanding of the role that leucine-rich repeat kinase 2 (LRRK2) plays in Parkinson's disease (PD) supports continued focus on this enzyme as a therapeutic target for PD. Accumulating evidence suggests that there are phenotypic, neuropathologic, and biological similarities between sporadic PD (sPD) and familial forms in which LRRK2 variants are inherited in an autosomal-dominant pattern with variable penetrance (LRRK2-PD). Further, genome-wide association studies have found specific non-coding variants that are risk factors for sPD. In this review, we describe the current state of

Global emergence of Acinetobacter baumannii International Clone 12 predominantly found in the Middle East.

Infections caused by carbapenem-resistant Acinetobacter baumannii (CRAB) are of great concern, as mortality is high, and treatment options are very limited. Despite having among the highest rates reported worldwide, scarce genomic data are available on CRAB strains from the Middle East. Here, we report the global emergence of a novel International Clone (IC), designated IC12, based on the epidemiological, phenotypic and genome sequencing data (short reads and long reads) of a set of 60 A. baumannii isolates belonging to multilocus sequence type 158 (Pasteur scheme). IC12, prevailing in the Mid

Evolutionary hotspots of structural variation drive inter-individual differences in the expression of fusion transcripts in the human brain.

Alternative splicing is a well-known contributor to transcriptomic diversity between individuals. Less explored are inter-gene splicing events, which generate fusion transcripts-RNA molecules combining exons from two or more distinct genes. In cancer, fusion transcripts frequently result from somatic structural rearrangements, yet it remains unclear to what extent germline structural variants in non-cancer tissues contribute to fusion transcript formation and inter-individual variability. We conducted a genome-wide fusion transcript analysis in post-mortem human brain tissues from 312 individu
