SiNiSan Ameliorates the Depression-Like Behavior of Rats That Experienced Maternal Separation Through 5-HT1A Receptor/CREB/BDNF Pathway.

Background: Early adverse life stress is an important dangerous factor in the development of psychiatric disorders, particularly depression. … response element-binding protein (CREB), p-CREB and brain-derived neurotrophic factor (BDNF) in the hippocampus were examined by Western blot, and in situ 5-HT1A receptor expression was measured by IHC. Results: Young and adult MS rats exhibited depression-like behavior. However, the depression-like behavior was ameliorated by SNS in both age groups. The leve …

Protective influences of N-acetylcysteine against alcohol abstinence-induced depression by regulating biochemical and GRIN2A, GRIN2B gene expression of NMDA receptor signaling pathway in rats.

Evidences have indicated a high degree of comorbidity of alcoholism and depression. N-acetylcysteine (NAC) has shown its clinical efficiency … timated by using commercially available kits. Serotonin concentrations were measured in the plasma, hippocampus and pre-frontal cortex using the rat ELISA kit. The expression of GRIN1, GRIN2A, GRIN2B genes for the N-methyl d-aspartate receptors (NMDAR) subunits in the hippocampus and the prefrontal cortex were also examined by reverse-transcription quantitati …

Aquaporins and Roles in Brain Health and Brain Injury.

In the literature screening, aquaporins were found in the cerebral structures including the pia mater, choroid plexus, ependyma, piriform cortex, hippocampus, dorsal thalamus, supraoptic and suprachiasmatic nuclei, white matter and subcortical organ. Among these, the most common are AQP1, AQP4, and AQP9. The roles of aquaporins have been demonstrated in several diseases such as cerebral edema, various central nervous system tumors, Alzheimer's Disease and epilepsy. In this review, the relationship between brain/brain-injury and aquaporin, has been reviewed.

Vascular type I aldosterone binding sites are physiological mineralocorticoid receptors.

In vitro, Type I receptors have high and equivalent affinity for aldosterone, corticosterone and cortisol: in vivo, physiological mineralocorticoid target tissues (kidney, colon, parotid) are highly aldosterone-selective, in contrast with hippocampus and heart. In the present study we show that the mesenteric vascular arcade is similarly highly aldosterone-selective in vivo, and in vitro shows considerable levels of 11 beta OH steroid dehydrogenase activity, previously postulated as the mechanism whereby glucocorticoids are excluded from physiological mineralocorticoid receptors.

Neuropathologic Characterisation of Mitochondrial Membrane Protein-Associated Neurodegeneration (MPAN) With Coexisting α-Synuclein and Tau Pathology in a Young Adult.

Mitochondrial membrane protein associated neurodegeneration (MPAN) is a rare and genetically defined subtype of neurodegeneration with brain iron accumulation (NBIA) caused by pathogenic variants in C19orf12. We report an autopsy case in a young adult with a homozygous C19orf12 missense variant. Neuropathological examination demonstrated iron deposition within the globus pallidus, widespread neuroaxonal spheroids, and extensive α-synuclein pathology of the brainstem, limbic and neocortical regions. In addition, we identified focal tau pathology in hippocampus, and entorhinal cortex.

Impaired semantic memory during acute transient global amnesia.

As a clinical model of hippocampal dysfunction, transient global amnesia (TGA) causes reversible memory disturbance. While episodic memory deficits in TGA patients have been extensively described, data regarding semantic memory involvement are sparse and contradictory. We report impaired semantic fluency performance in 16 patients with hippocampal lesions on MRI during acute TGA compared to their performance one day later and to that of 20 healthy subjects. Our findings support the involvement of the hippocampus in semantic retrieval.

Spider toxin (JSTX) blocks glutamate synapse in hippocampal pyramidal neurons.

Effects of spider toxin (JSTX)--a specific blocker of glutamate receptors--on single pyramidal neurons of the hippocampus were studied using tissue slices in vitro. JSTX blocked the synaptic response in CA1 pyramidal cells evoked by Schaffer collateral stimulation without affecting the antidromic spike potential. The toxin suppressed glutamate-induced cell firings whereas it had little effect on aspartate-induced responses. The results suggest that glutamate is a neurotransmitter of the Schaffer collateral input to CA1 pyramidal neurons.

Behavioral and Fluorescent-Based Immunohistochemistry Protocols for Examining Antidepressant-Like Effects of Melatonin in Mice.

Antidepressants are characterized by their ability to decrease despair behavior assessed in mice as a decrease in immobility time in the forced swimming test (FST) (antidepressant-like behavior). This behavioral parameter is associated with increased neurogenesis in the dentate gyrus of the hippocampus of the rodents summitted to this test. Herein, we describe an optimized protocol used to characterize the melatonin antidepressant-like effect associated with its pro-neurogenic activity after an acute and a triple administration to mice measured by the FST and fluorescence-based immunohistochem…

Aerosol Delivery of Curcumin Reduced Amyloid-β Deposition and Improved Cognitive Performance in a Transgenic Model of Alzheimer's Disease.

We report a novel approach for the delivery of curcumin to the brain via inhalation of the aerosol for the potential treatment of Alzheimer's disease. The percentage of plaque fraction in the subiculum and hippocampus reduced significantly when young 5XFAD mice were treated with inhalable curcumin over an extended period of time compared to age-matched nontreated counterparts. Further, treated animals demonstrated remarkably improved overall cognitive function, no registered systemic or pulmonary toxicity associated with inhalable curcumin observed during the course of this work.
