Imunomodulative effect of liposomized muramyltripeptide phosphatidylethanolamine (L-MTP-PE) on mice with alveolar echinococcosis and treated with albendazole.

The effect of liposomized muramyltripeptide phosphatidylethanolamine (L-MTP-PE) administered separately or with anthelmintic albendazole (ABZ) on cellular immunity of mice with alveolar echinococcosis was studied. The proliferative activity of splenic T and B lymphocytes was the most stimulated after combined L-MTP-PE + ABZ therapy [from weeks 8 to 14 post-infection (p.i.)] that also induced a long-term development of protective Th1 response (the highest serum concentration of IFN-gamma from weeks 8 to 18 p.i.). On the contrary, Th2 response (cytokine IL-5) in infected mice treated with L-MTP-

Annexin B1 from Taenia solium metacestodes is a newly characterized member of the annexin family.

We previously reported cloning of the Taenia solium annexin B1 gene from a metacestode cDNA expression library and demonstrated that it acts as a protective antigen for effective vaccine development against cysticercosis. In the present study we produced recombinant annexin B1 and antiserum against the protein to investigate its structural and functional properties. Western blotting of metacestode fractions indicated that T. solium annexin B1, similar to vertebrate annexins, associates with acid phospholipids in the presence of Ca(2+). This property was confirmed by the recognition of apoptoti

Blood coagulation in falciparum malaria--a review.

Falciparum malaria infection influences blood coagulation by various interacting pathobiological mechanisms, the most important being the overwhelming response of the host to sepsis resulting in a cytokine storm. In addition, the parasite infects the red cells leading to changes in the red cell phospholipid composition which supports blood coagulation. Red cells infected with Plasmodium falciparum also adhere to deeper tissue capillary endothelium leading to profound damage to endothelial cells leading to further activation. This results in widespread consumption of platelets and activation of

Effects of environmental stress on the condition of Littorina littorea along the Scheldt estuary (The Netherlands).

The condition of the periwinkle Littorina littorea, expressed in terms of its shell morphology, reproductive impairment (i.e. female sterility/intersex, male penis shedding), trematode infestation load, lipid reserves and dry/wet weight ratio, was determined in function of environmental stress along the polluted Western and relatively clean Eastern Scheldt estuary (The Netherlands). The upstream increasing pollution and decreasing salinity levels along the Western Scheldt estuary (Fig. 1) are reflected in the dry/wet weight ratio and lipid content of the periwinkles. Compared to the Eastern Sc

Detection of excretory/secretory coproantigens in experimental hookworm infection.

This report describes the detection of hookworm excretory/secretory (ES) antigens in soluble hamster fecal extracts by an enzyme-linked immunosorbent assay (ELISA). A rabbit polyclonal IgG antibody against Ancylostoma ceylanicum ES was used to capture hookworm coproantigens that were then detected using pooled, high-titer, infected hamster serum. The ELISA was capable of detecting ES proteins over a range of 10 ng/mL to 10 mug/mL when the antigens were diluted in buffer or uninfected fecal extract, and ES could be detected in infected hamster feces at dilutions up to 1:256. Examination of the

Control of tsetse flies and trypanosomes using molecular genetics.

TSETSE FLIES (DIPTERA: Glossinidae) are important agricultural and medical vectors transmitting the African trypanosomes, the agents of sleeping sickness disease in humans and various diseases in animals (nagana). While the prevalence of disease has increased to epidemic proportions, lack of a mammalian vaccine and affordable and effective drugs have hindered disease control. Trypanosomiasis management relies heavily on the control of its single insect vector, the tsetse fly. Despite the effectiveness of some of these tools, their impact on disease control has not been sustainable due to their

Schistosoma mansoni: larval damage and role of effector cell(s) in the synergy between vaccine immunity and praziquantel treatment.

A number of authors have demonstrated that the schistosomicidal compound, Praziquantel (Pzq), depends for its action upon the immune status of the host (Sabah et al. 1985; Brindley & Sher, 1987; Doenhoff et al. 1987). We have attempted to define the synergistic interaction between immuno- and chemotherapy further, using the murine irradiated vaccine model of schistosomiasis mansoni. In vaccinated mice, resistance operates in the skin but not the lungs; drug targeted towards lung-stage worms exacerbates lung-phase immunity, however, as depicted by the increased number and size of inflammatory r

Glycoconjugates of the intestinal epithelium of the domestic fowl (Gallus domesticus): a lectin histochemistry study.

A lectin histochemical study was performed on formalin-fixed paraffin-embedded tissues of duodenum, jejunum, ileum, caecum and colon from six fasted and six non-fasted 8-week-old chickens (Gallus domesticus). The purpose of this study was to identify in situ the pattern of carbohydrate residues present on the luminal surface of the intestinal epithelium. Ten biotinylated lectins with different sugar specificities were used as probes, and avidin-biotin-peroxidase complex (ABC) was used as a 'visualant'. The most significant finding was the binding pattern of Lens culinaris agglutinin to various

The role of let-7b in the inhibition of hepatic stellate cell activation by rSjP40.

BACKGROUND: Hepatic stellate cells (HSCs) are one of the main cell types involved in liver fibrosis induced by many factors, including schistosomes. Previous studies in our lab have shown that recombinant P40 protein from Schistosoma japonicum (rSjP40) can inhibit HSC activation in vitro. Let-7b is a member of the let-7 microRNA family and plays an inhibitory role in a variety of diseases and inflammatory conditions. In this study, we investigated the role of let-7b in the inhibition of HSC activation by rSjP40. METHODS: Expression of let-7b was detected by quantitative real-time PCR. A dual

Intestinal helminth infection promotes IL-5- and CD4+ T cell-dependent immunity in the lung against migrating parasites.

The ability of helminths to manipulate the immune system of their hosts to ensure their own survival is often credited with affecting responses to other pathogens. We undertook co-infection experiments in mice to determine how infection with the intestinal helminth Heligmosomoides polygyrus affected the parasitological, immunological and physiological outcomes of a primary infection with a distinct species of helminth; the lung migratory parasite Nippostrongylus brasiliensis. We found that migrating N. brasiliensis larvae were killed in the lungs of H. polygyrus-infected mice by a process invo

Yip1A, a novel host factor for the activation of the IRE1 pathway of the unfolded protein response during Brucella infection.

Brucella species replicate within host cells in the form of endoplasmic reticulum (ER)-derived vacuoles. The mechanisms by which the bacteria are sequestered into such vacuoles and obtain a continuous membrane supply for their replication remain to be elucidated. In the present study, we provided several lines of evidence that demonstrate the mechanism by which B. abortus acquires the ER-derived membrane. First, during Brucella infection, the IRE1 pathway, but not the PERK and ATF6 pathways, of the unfolded protein response (UPR) was activated in a time-dependent manner, and the COPII vesicle

Fc gamma receptor IIa-H131R polymorphism and malaria susceptibility in sympatric ethnic groups, Fulani and Dogon of Mali.

It has been previously shown that there are some interethnic differences in susceptibility to malaria between two sympatric ethnic groups of Mali, the Fulani and the Dogon. The lower susceptibility to Plasmodium falciparum malaria seen in the Fulani has not been fully explained by genetic polymorphisms previously known to be associated with malaria resistance, including haemoglobin S (HbS), haemoglobin C (HbC), alpha-thalassaemia and glucose-6-phosphate dehydrogenase (G6PD) deficiency. Given the observed differences in the distribution of FcγRIIa allotypes among different ethnic groups and wit

Comparative haematological parameters of HbAA and HbAS genotype children infected with Plasmodium falciparum malaria in Yemen.

BACKGROUND: Sickle haemoglobin (HbS) is known to offer considerable protection against falciparum malaria. However, the mechanism of protection is not yet completely understood. In this study, we investigate how the presence of the sickle cell trait affects the haematological profile of AS persons with malaria, in comparison with similarly infected persons with HbAA. This study is based on the hypothesis that the sickle cell trait plays a protective role against malaria. METHODS: Children from an endemic malaria transmission area in Yemen were enrolled in this study. Hematological parameters

Pathogenic role of B cells and antibodies in murine Leishmania amazonensis infection.

Leishmania amazonensis infection, occurring predominantly in Central and South America, can manifest itself in several forms, including those of cutaneous and diffuse cutaneous leishmaniasis. The outcome of L. amazonensis infection depends largely on host immune responses to the parasites. While CD4+ T cell activation is a prerequisite for pathogenesis in L. amazonensis-infected mice, the roles of B cells and their antibody production are unclear. In this study, we provide evidence suggesting that B cells and antibodies are involved in disease pathogenesis. We documented a correlation between

Gene-for-gene defense of wheat against the Hessian fly lacks a classical oxidative burst.

Genetic similarities between plant interactions with microbial pathogens and wheat interactions with Hessian fly larvae prompted us to investigate defense and counterdefense mechanisms. Plant oxidative burst, a rapid increase in the levels of active oxygen species (AOS) within the initial 24 h of an interaction with pathogens, commonly is associated with defenses that are triggered by gene-for-gene recognition events similar to those involving wheat and Hessian fly larvae. RNAs encoded by Hessian fly superoxide dismutase (SOD) and catalase (CAT) genes, involved in detoxification of AOS, increa

Structure of the human S100A12-copper complex: implications for host-parasite defence.

S100A12 is a member of the S100 family of EF-hand calcium-modulated proteins. Together with S100A8 and S100A9, it belongs to the calgranulin subfamily, i.e. it is mainly expressed in granulocytes, although there is an increasing body of evidence of expression in keratinocytes and psoriatic lesions. As well as being linked to inflammation, allergy and neuritogenesis, S100A12 is involved in host-parasite response, as are the other two calgranulins. Recent data suggest that the function of the S100-family proteins is modulated not only by calcium, but also by other metals such as zinc and copper.

Effect of oral zinc supplementation upon Taenia crassiceps murine cysticercosis.

The effect of zinc supplementation on Taenia crassiceps murine cysticercosis was studied in susceptible BALB/cAnN mice. Female offspring of mice supplemented with high zinc throughout gestation and lactation were intraperitoneally infected with T. crassiceps cysticerci. Offspring from nonsupplemented mothers were used as controls. Significantly fewer parasites were recovered from zinc-supplemented mice (Zsm) 30 days after infection. Increased resistance was not related to the IgG antibody response. At early stages of infection, T cells from Zsm proliferated to T. crassiceps antigens, whereas c

Determinants for resistance and susceptibility to microfilaraemia in Litomosoides sigmodontis filariasis.

Filarial infections of humans are chronic diseases. Despite an ongoing immune response, adult filariae continuously produce their offspring, the microfilariae (Mf), which are able to persist in sufficient numbers to ensure transmission. In this study, host- and parasite-derived factors, which contribute to persistence of Mf, were investigated using the filariasis model of Litomosoides sigmodontis in mice. Different strains of mice were found to differ widely in their capability to eliminate circulating Mf. Studies of congenic mouse strains showed that early and rapid clearance of Mf was mediat

Pathogen-tick-host interactions: Borrelia burgdorferi and TBE virus.

Both tick-borne encephalitis (TBE) virus and Borrelia burgdorferi depend for their survival on a tick-borne transmission cycle involving diverse vertebrate hosts and ixodid (primarily Ixodes) tick species. Increasing evidence strongly suggests that a key factor in successful transmission is the ability of these pathogens to exploit the pharmacological properties of their vector tick's saliva. Proteins and other chemicals, secreted in tick saliva, control the vertebrate hosts' haemostatic, inflammatory and immune responses in order to facilitate blood-feeding. Such bioactive saliva molecules in
