Music-STAR trial: study protocol for a randomised controlled trial to assess the impact of live and recorded music-based interventions on preoperative stress, mood, pain and biomarkers in neurosurgical patients.

INTRODUCTION: Neurosurgical patients commonly experience significant preoperative anxiety, which can negatively impact surgical outcomes and recovery trajectories. Music, as a powerful neurophysiological modulator of the human stress response, offers a non-pharmacological, non-invasive and cost-effective approach to anxiolysis by stabilising the hypothalamic-pituitary-adrenal axis and autonomic nervous system. The goal of this study is to evaluate the effects of live and recorded music on preoperative stress, serum-level and plasma-level biomarkers, perceived pain, anxiety and mood states in n

Premenstrual syndrome (PMS) symptom emergence across the cycle in relation to hormonal and psychological components in a community sample.

For many individuals, the days leading up to menstruation are associated with aversive mood, behavioral, and physical symptoms, a condition known as premenstrual syndrome (PMS). Severe levels of these symptoms can lead to clinical diagnosis of premenstrual dysphoria (PMDD). Although prior research has uncovered important psychological and hormonal predictors of PMS/PMDD symptoms in clinical populations, more research is needed to understand how these factors relate in a subclinical population. A test of the menstrual timing and hormonal links to symptom experience alongside factors related to

Milk and infant plasma hormone concentrations predict sweet and fat taste acceptance and later food intake.

Early-life exposure to metabolic hormones may influence the development of taste acceptance and subsequent intake of hyperpalatable foods (HPF) in infants, but evidence integrating hormonal, behavioral, and dietary data in humans is limited. Therefore, our objective was to determine whether concentrations of insulin, leptin, cortisol, and liver-expressed antimicrobial peptide 2 (LEAP2) in human milk and infant plasma at 6 months predict infant sweet and fat taste acceptance and HPF intake later in infancy. In this prospective cohort study, human milk and infant plasma were collected at 6 month

Associations between hair endocannabinoid concentrations and parental depressive symptoms: A longitudinal study of mothers, fathers, and their offspring up to two years postpartum.

BACKGROUND: About one in five mothers and one in ten fathers experience postpartum depression, with adverse consequences for family health and child development. The aetiology of postpartum depressive symptoms (PPDS) is characterised within a biopsychosocial framework, yet the underlying biological mechanisms remain incompletely understood. The endocannabinoid system (ECS), which interacts with the glucocorticoid system and is implicated in the pathophysiology of depressive disorders, has not yet been investigated in relation to PPDS. Moreover, longitudinal investigations including both mother

Effects of static and dynamic canine-assisted intervention in young adults: sex differences in hormonal, EEG, HRV, and emotional responses.

As mental health disorders like depression and anxiety rise, animal-assisted intervention (AAI) has emerged as a promising complementary approach, recognized for its psychological benefits. However, the underlying physiological mechanisms remain largely unexplored. The present study aims to investigate the impact of animal-assisted intervention on physiological, psychological changes and self-reported measurements. Thirteen young adults participated in three activities: meditation, static interaction with a dog, and dynamic interaction with a dog. Salivary hormone levels were measured by ELISA

Registered Report: How does art impact pain and stress? Exposure to multimodal art (Music + Visual) and music alone enhances pain tolerance more than visual art, but neither art form impacts autonomic or endocrine markers.

While the pain- and stress-reducing effects of music are well investigated, effects of visual art and the combination of both modalities (music and visual art) are much less explored. We tested the (1) pain- and (2) stress-reducing effects of a multimodal (music + visual art) aesthetic experience-expecting stronger effects than single modal aesthetic experiences (music or visual art)-and, in an exploratory manner, (3) investigated underlying mechanisms of aesthetic experience and (4) individual differences. In a repeated-measures design (music, visual art, multimodal aesthetic experience, cont

Facile and scalable fabrication of molecularly imprinted polymer (MIP) sensors on poriferous laser-engraved graphene electrodes for stress monitoring.

Monitoring cortisol levels is essential for understanding the body's response to stress. Traditional cortisol testing is confined to centralized labs, and current portable platforms are based on slow and complex assays. Here, we introduce a portable, disposable, non-invasive, and sensitive electrochemical sensor strip created using electropolymerized molecularly imprinted polymers (eMIPs) on laser engraved graphene (LEG) electrodes for rapid, simple, and reliable salivary cortisol detection. Herein, the characteristics of LEGs generated on a polyimide (PI) film with different laser processing

Evaluation of visfatin levels in patients with hyperemesis gravidarum.

The severe end of the clinical spectrum of nausea and vomiting during pregnancy is referred to as hyperemesis gravidarum (HEG). Although its exact pathophysiology has not yet been fully elucidated, multifactorial mechanisms are known to play a role. Visfatin is a proinflammatory adipocytokine primarily secreted by visceral adipose tissue and is also produced by the placenta and amniotic membranes during pregnancy. This study aimed to investigate the association between HEG and serum visfatin levels in a prospective observational case-control study. A total of 90 pregnant women were included in

Inflammatory Markers Link Steroid Profiles to Bone Status in Patients with Autonomous Cortisol Secretion.

CONTEXT: Autonomous cortisol secretion (ACS) is a common cause of endogenous glucocorticoid-induced osteoporosis. Cortisol excess alone, however, does not fully explain altered bone status, and other steroid hormone changes, including androgen deficiency, may contribute. ACS is also associated with an inflammatory state, which may relate to changes in bone status and steroid profiles. PURPOSE: To investigate whether inflammatory markers are associated with bone status and steroid profiles in ACS and explore their potential mediating role. METHODS: Inflammatory markers and bone status were as

Urinary free cortisol analyses: Enhancing their clinical performance in Cushing's syndrome management by means of LC-MS/MS.

24 h urinary free cortisol (UFC) analysis constitutes one of the three first level recommended tests in Cushing's syndrome (CS) diagnostic confirmation work up. However, it occasionally leads to inaccurate results due to the use of immunoassays (IAs) or the concomitant administration of exogenous glucocorticoids, among others. This study aimed to develop a rapid and accurate LC-MS/MS method which may ultimately replace the use of IAs, and also provide relevant clinical information through the simultaneous monitoring of UFC, cortisone, and dexamethasone. An LC-MS/MS method based on direct injec

Clinical and Metabolic Outcomes of Adrenalectomy Versus Conservative Management in Mild Autonomous Cortisol Secretion.

Adrenalectomy and conservative management are therapeutic approaches for mild autonomous cortisol secretion; however, their comparative clinical impact in routine practice remains uncertain. We aimed to evaluate real-world hormonal, clinical, and metabolic outcomes according to the treatment strategy in patients with mild autonomous cortisol secretion. This single-center retrospective observational study included consecutive patients with adrenal incidentaloma fulfilling guideline-based diagnostic criteria for mild autonomous cortisol secretion between January 2015 and December 2024. Substance use involvement moderates the effect of cortisol response on white-matter microstructure longitudinally.

INTRODUCTION: Stress contributes to substance use (SU) involvement (SUI) and differences in brain white matter (WM). However, longitudinal relationships involving SUI, stress hormones, and WM are not well understood. This study examined the moderating role of SUI on the prospective association between cortisol response (CR) and WM microstructure. METHODS: Data were drawn from a larger longitudinal study on SU disorder (SUD) aetiology. Participants were enrolled at ages 10-12 years when salivary cortisol was assayed. The Drug Use Chart assessed cumulative involvement with 10 substance categori

Hair cortisol concentrations to picture the dysregulation of the hypothalamic-pituitary-adrenocortical axis in panic disorder.

Panic attacks are characterized by intense feelings of uncontrollable threat and induce experiences of intense stress. As one of the major physiological stress systems of the body, the hypothalamic-pituitary-adrenocortical axis (HPA) may be a key to our understanding of a biological basis of this anxiety disorder. While recent studies have found evidence for short-term HPA changes in panic disorder with flattened cortisol responses to stimulation, no data have been available on tonic, i.e., long-term HPA activity in these patients. The current study therefore investigated the cumulative cortis

Risk factors for adrenal insufficiency after adrenalectomy for mild autonomous cortisol secretion.

BACKGROUND: Although unilateral adrenalectomy can improve metabolic outcomes associated with mild autonomous cortisol secretion, it can also be associated with postoperative adrenal insufficiency. We aimed to look at the incidence and risk factors of suspected adrenal insufficiency in these patients. METHODS: Adults undergoing unilateral adrenalectomy for mild autonomous cortisol secretion, defined by cortisol >1.8 μg/dL following 1-mg post-dexamethasone suppression test (post-DST), were included. Patients with adrenocortical carcinoma or Cushing syndrome were excluded. Suspected adrenal insu

Ectopic ACTH-dependent Cushing syndrome due to mature ovarian teratoma.

Ectopic adrenocorticotropic hormone (ACTH) secretion accounts for 10%-20% of ACTH-dependent Cushing syndrome, with ovarian teratomas being an exceptionally rare source. We present a female in her early 40s with severe Cushing syndrome due to ectopic ACTH secretion from a mature ovarian teratoma. She presented with progressive weight gain, moon facies, dorsocervical fat pad, proximal muscle weakness, hypertension and new-onset diabetes. Biochemical evaluation confirmed ACTH-dependent hypercortisolism with elevated cortisol, plasma ACTH, late-night salivary cortisol, urinary free cortisol and fa

Gamma-aminobutyric acid attenuates cortisol-induced damage in human colorectal adenocarcinoma cells via Nrf2 signaling.

Long-term psychological stress is associated with increased intestinal epithelial permeability. In the human central nervous system, gamma-aminobutyric acid (GABA), a non-protein amino acid found in bacteria, plants, and animals, acts as an inhibitory neurotransmitter that controls the cardiovascular system, reduces blood pressure, enhances mood, and encourages sleep. It is still unclear how GABA controls the function of the colon epithelial barrier under long-term stress. This study explored the potential of GABA to ameliorate cortisol-induced damage in human colorectal adenocarcinoma cells (

Association of prenatal alcohol exposure and prenatal depressive symptoms with offspring hair cortisol in childhood and adolescence.

BACKGROUND: Prenatal alcohol exposure (PAE) and maternal depressive symptoms are associated with hypothalamic-pituitary-adrenal (HPA) axis alterations in the offspring. The present study investigated long-term associations of both risk factors on the offspring’s hair cortisol concentration (HCC) in childhood and adolescence. METHODS: The HCC of n = 94 children was assessed at primary school age (T1, M = 7.7 years, SD = 0.81) and in early adolescence (T2, M = 13.3 years, SD = 0.30). PAE was operationalized by maternal self-report and the meconium alcohol metabolite ethyl glucuronide (EtG), appl
