Repeated restraint stress-induced increase in post-surgical somatosensory hypersensitivity and affective responding is mediated by β-adrenergic receptor activation and spinal NLRP3-IL1β signalling in male rats.

Pre-surgical stress is a well-recognised risk factor for persistent post-surgical pain, and while the precise underlying neurobiological mechanisms remain unknown, neuro-immune interactions are believed to play a pivotal role. Here, we investigated the effect of repeated restraint stress (RRS) on post-surgical somatosensory hypersensitivity and affective responding in male rats and examined underlying mechanisms. We showed that RRS induced behavioural despair, reduced body weight gain and elevated faecal corticosterone levels in male Sprague-Dawley rats. Following paw incision surgery, animals

Resveratrol protects against triclosan-induced impairment of colonic barrier function in chickens.

Triclosan (TCS) is a widely used antimicrobial agent, and chronic exposure has been reported to impair intestinal barrier function and induce oxidative stress and inflammation. Resveratrol (RSV), a natural polyphenolic compound with antioxidant and anti-inflammatory properties, may protect against TCS-induced intestinal injury. This study investigated the protective effects of RSV on TCS-induced colonic damage in chickens and the underlying mechanisms. In a 4-week trial, 100 10-week-old chickens were randomly assigned to five groups (n = 20 per group): control (CON), solvent control (VCON), TC

IL-Mediated Macrophage Polarization Axis: From Inflammatory Positive Feedback Loop to Precision Biomarkers and Therapeutic Targets for Allergic Rhinitis-A Narrative Review.

This narrative review comprehensively elaborates the complicated interleukin (IL)-macrophage polarization axis as the core pathogenesis of allergic rhinitis (AR), focusing on layered molecular regulatory mechanisms covering inflammatory signaling, metabolic reprogramming and epigenetic modulation. Pro-inflammatory IL-1β, IL-6, IL-17 and TNF-α bind to TLR receptors to activate NF-κB, NLRP3 inflammasome and PI3K/Akt cascades, triggering M1 macrophage overactivation and acute nasal congestion, rhinorrhea via robust inflammatory mediator release. By contrast, anti-inflammatory IL-4 and IL-10 predo

A Decoy-Receptor-Armed Biomimetic Nanotherapeutic With Inherent Tropism for Conserved Pathogenic Macrophages for Treating Osteoarthritis and Intervertebral Disc Degeneration.

Osteoarthritis (OA) and intervertebral disc degeneration (IVDD) are debilitating musculoskeletal disorders driven by shared inflammatory pathologies, yet lack effective disease-modifying therapies. Pro-inflammatory macrophages are central to their pathogenesis, but their selective therapeutic modulation remains a major challenge. Here, we first used single-cell transcriptomics to identify a conserved, pathogenic interleukin-1β (IL-1β)+ macrophage signature fueled by a shared nuclear factor-κB (NF-κB)-inflammation axis in human OA and IVDD. We engineered a macrophage-biomimetic nanosystem (GM1-

Ligand-specific duality of aryl hydrocarbon receptor signaling in cognitive health: from environmental neurotoxicity to microbiome-mediated neuroprotection.

The aromatic hydrocarbon receptor (AhR) is a key molecular interface integrating environmental chemical signals with host-microbiome metabolism, with profound effects on brain function. This review systematically addresses the ligand-specific duality of AhR signaling in cognitive health, comparing the predominantly neurotoxic signaling driven by environmental polycyclic aromatic hydrocarbons (PAHs) with the predominantly neuroprotective signaling mediated by gut microbiota-derived tryptophan metabolites. However, this dichotomy is context-dependent rather than absolute. PAHs activate AhR in a

Body mass index and gastrointestinal inflammation: Bio-molecular pathophysiology.

Overweight is recognized as a worldwide healthcare problem. Obesity has increased in recent decades and has been considered a risk factor for many gastrointestinal (GI) disorders. Recent scientific evidence has documented the association between being overweight and GI manifestations. Body mass index (BMI) is a simple, globally used anthropometric measure, but its role in GI inflammation remains incompletely elucidated and can be challenging to study. Current knowledge suggests that higher BMI is linked to a chronic low-grade pro-inflammatory state ("metainflammation") and several GI-relevant

β-Caryophyllene protects against ischemic stroke by inhibiting H3K9 and H3K18 lactylation-mediated cellular pyroptosis.

BACKGROUND: Ischemic stroke is a common and severe cerebrovascular disease with high mortality and disability. Accumulating evidence indicates that β-caryophyllene (BCP) exerts neuroprotective effects against cerebral ischemic injury; however, the precise underlying mechanisms remain largely unexplored. METHODS: Focal cerebral ischemia/reperfusion (I/R) mouse models were established in vivo and oxygen-glucose deprivation/reoxygenation (OGD/R) was conducted in BV2 microglial cells and primary microglia in vitro. RESULTS: We demonstrated that BCP administration significantly reduced cerebral i

Cannabidiol selectively attenuates lipotoxic immunometabolic inflammation in human macrophages.

The role of saturated fatty acid-induced immunometabolic stress in macrophage dysfunction during metabolic disease remains incompletely understood, particularly the interplay between inflammatory signaling and intracellular lipid handling. We employed a tightly controlled palmitic acid (PA)-based lipotoxicity model in PMA-differentiated U937-derived human macrophage-like cells to investigate how lipid excess reshapes inflammatory responses and to evaluate the modulatory effects of cannabidiol (CBD). PA exposure induced a metabolically stressed yet viable macrophage phenotype, characterized by

Mitochondrial DNA Instability and Neuroinflammation: Connecting the Dots Between Base Excision Repair and Neurodegenerative Disease.

Neurons have exceptionally high energy demands, sustained by thousands to millions of mitochondria per cell. Each mitochondrion depends on the integrity of its mitochondrial DNA (mtDNA), which encodes essential electron transport chain (ETC) subunits required for oxidative phosphorylation (OXPHOS). The continuous, high-level ATP production by OXPHOS generates reactive oxygen species (ROS) that pose a significant threat to the nearby mtDNA. To counter these insults, neurons rely on base excision repair (BER), the principal mechanism for removing oxidative and other small, non-bulky base lesions

Effects of New P2X7R Antagonists on Retinal Inflammatory Degenerative Conditions.

Purinergic receptor (P2X7R), an adenosine triphosphate (ATP) gated nonselective cation channel, is an emerging target for several neurodegenerative conditions, including retinal degeneration. In particular, P2X7R has an established role in multiple inflammatory and immune responses, being implicated in the formation and activation of NLRP3 inflammasome leading to inflammatory priming and release of immune cytokines. Despite several efforts in P2X7R antagonist design, no drugs have reached the market yet. In this perspective, through an integrated in silico and in vitro approach, we aimed to re

Functional role of deubiquitinating enzyme USP39 in inhibiting pyroptosis in human acute myeloid leukemia.

Acute myeloid leukemia (AML) exhibits a five-year overall survival rate below 30%, with its substantial genetic heterogeneity/clonal evolution complicating therapeutic strategies. Pyroptosis, inflammatory programmed cell death via Gasdermins holds therapeutic potential for AML. Deubiquitinase USP39 highly expressed in leukemia and linked to poor prognosis regulates pyroptosis in AML cells via an unclear mechanism. This study aims to explore this mechanism. The TCGA, GTEx, and GSE138702 datasets were applied for analysis of USP family genes differentially expressed. The biological behavior chan

The Redox-Inflammation Axis in Atherosclerosis and Ischemic Stroke: Mechanisms, Biomarkers, and Translational Challenges.

Cardiovascular diseases (CVDs) remain the leading cause of mortality worldwide and are driven by the interplay between oxidative stress and chronic inflammation. Although these processes have been extensively investigated, their integration into a unified mechanistic framework and their translational relevance for biomarker-guided precision medicine remain insufficiently addressed. This narrative review integrates mechanistic, biomarker, and translational perspectives on the redox-inflammation axis in atherosclerosis and acute ischemic stroke, critically evaluating oxidative and inflammatory b

Beyond dysbiosis: microbial metabolites as key remodelers of nasal mucosal immune tolerance in chronic rhinosinusitis.

Chronic rhinosinusitis (CRS) is a heterogeneous inflammatory disorder of the nasal and paranasal sinus mucosa affecting approximately 11% of adults worldwide. Although compositional dysbiosis of the sinonasal microbiome has historically dominated etiological discourse, this framework inadequately accounts for the mechanistic complexity of mucosal immune dysregulation in CRS. Emerging evidence positions microbial metabolites, rather than microbial identity per se, as the proximate immunological mediators of commensal microbiota-host crosstalk. This review presents a comprehensive analysis of th

The myocardial ischemic cascade network and multi-target synergistic interventions: From molecular mechanisms to therapeutic innovations.

Myocardial ischemic injury involves a multi-layered pathological cascade driven by interconnected energy metabolism disorders, calcium overload, oxidative stress, mitochondrial dysfunction, and inflammatory responses. Ischemia-hypoxia impairs mitochondrial oxidative phosphorylation, causing ATP depletion, acidosis, and calcium overload. Reperfusion exacerbates injury through ROS burst, mPTP opening, and NLRP3 inflammasome activation, leading to pro-inflammatory cytokine release. Sustained endoplasmic reticulum stress promotes apoptosis via the PERK/CHOP pathway, forming a vicious cycle with ox

Cytokine associated neuroinflammation in Parkinson's disease: Molecular pathways, therapeutic targets, and translational insights.

Parkinson's disease (PD) is a progressive neurodegenerative disorder in which neuroinflammation plays a key role. An imbalance between pro- and anti-inflammatory cytokines has been observed in both experimental models and PD patients. The inflammatory mediators activate signaling pathways that lead to oxidative stress, excitotoxicity, blood-brain barrier (BBB) disruption, gut dysbiosis, and hypothalamic-pituitary-adrenal axis (HPA-axis) dysregulation. Increased levels of pro-inflammatory cytokines such as tumor necrosis factor-α (TNF-α), Interleukin-1β (IL-1β), Interleukin-6 (IL-6), and others

Pharmacological basis of Edaravone-Dexborneol neuroprotection: Integrated modulation of oxidative stress, neuroinflammation, and ferroptosis.

Edaravone-Dexborneol (EDB) is a rationally engineered neuroprotective formulation that integrates the free-radical scavenger edaravone with the monoterpenoid (+)-borneol in a multimodal 4:1 M ratio. This combination was developed to overcome the translational limitations of single-target neuroprotectants by combining potent antioxidative activity with enhanced blood-brain barrier permeability, anti-inflammatory signaling, and improved pharmacokinetic behavior. This review provides a mechanistic synthesis of the multimodal pharmacology of EDB, emphasizing its associated modulation of oxidative

The Role of Galectin-3 in Liver Inflammation and Fibrosis.

Galectin-3, a β-galactoside-binding lectin encoded by LGALS3, is increasingly recognized as a central mediator in liver disease and inflammation. Galectin-3 has the unique ability to cross-link glycosylated molecules, resulting in a broad range of functions, from receptor clustering to influence downstream signaling, to extracellular matrix stabilization and the regulation of cell adhesion. In the liver, it regulates fibrogenesis through hepatic stellate cell activation, oxidative stress regulation, apoptosis, and extracellular matrix deposition. Immunologically, galectin-3 contributes to leuk

Targeting coronaviral inflammation: aptamer-based strategies for emerging threats.

Coronaviruses have repeatedly emerged in recent years, causing significant and ongoing threats to global public health. The development of therapeutic agents and strategies capable of responding to future outbreaks caused by emerging coronavirus variants remain an ongoing priority. Here, we engineered a single-stranded DNA aptamer (NApt8-3) that selectively binds to the conserved nucleocapsid (N) protein shared among multiple coronaviruses, including SARS-CoV-2 (wild-type, beta, omicron variant), SARS-CoV, MERS-CoV, HCoV-OC43 and HCoV-229E, and strongly inhibits N protein-induced inflammatory

Immune-metabolic positive feedback model in COPD: cross-mechanisms and potential intervention strategies.

Chronic obstructive pulmonary disease (COPD) is a common chronic condition characterized by chronic bronchitis and/or emphysema with airflow obstruction, which can progress to cor pulmonale and respiratory failure. Associated with abnormal inflammatory responses to harmful gases and particulate matter, it carries high rates of disability and mortality, with a global prevalence among individuals aged 40 and older reaching 9%-10%. It is often regarded as a clinical and molecular model of accelerated lung aging. Age-related drift in immune function and metabolism plays a central part in this proc
