Effect of 3-[p-(trans-4-aminomethylcyclohexylcarbonyl)-phenyl] propionic acid hydrochloride on gastric mucosal blood flow.

The effects of 3-[p-(trans-4-aminomethylcyclohexylcarbonyl)phenyl]propionic acid hydrochloride (TEI-5103, TG-51) on gastric mucosal blood flow (GMBF) in rats, rabbits, and dogs were studied by using hydrogen gas clearance, cross-thermocouple, and 51Cr-labelled erythrocyte methods. TEI-5103 (5 mg/kg i.v.) increased GMBF, measured by hydrogen gas clearance method, in anesthetized rats. At 1-10 mg/kg i.v. it also increased GMBF, measured by cross-thermocouple method, in anesthetized rabbits and dogs. The increasing effect of TEI-5103 was more potent than that of cetraxate. By the intragastric rou

Pathogenesis of the mucosal hyperplasia in self-filling blind loops of rat jejunum: a morphometric study in germ free animals.

Bacterial overgrowth and high intraluminal concentrations of deconjugated bile acids are thought to be responsible for mucosal hyperplasia in self-filling blind loops of rat jejunum. To investigate this hypothesis further we have assessed the three dimensional architecture of these loops created in germ free animals without or with di- or mono-association of different bacterial species. It was found that mucosal hyperplasia develops in the absence of any bacterial contamination and that bacterial association does not lead to a more pronounced mucosal proliferation. This implies that other mech

Effects of sodium 5-methoxysalicylate on macromolecule absorption and mucosal morphology in a vascularly perfused rat gut preparation in vivo.

The effect of sodium 5-methoxysalicylate on absorption was assessed by measurement of the appearance of test compounds in the portal blood output of a perfused rat gut model. The test compounds were a hexapeptide analogue of somatostatin, insulin, and horseradish peroxidase. Considerable amounts of sodium 5-methoxysalicylate were present in the portal blood 10 min after intraduodenal administration. When co-administered with sodium 5-methoxysalicylate (60 mg), a marked increase in the concentrations of the test substances occurred at t = 15 min which lasted for a further 15 min. Quantities of

Serum antibody responses to indigenous oral mucosal antigens and selected laboratory-maintained bacteria in recurrent aphthous ulceration.

Sera from subjects with recurrent aphthous ulceration (RAU) and control subjects were tested for relative levels of IgM, IgG, and IgA antibodies against eight selected laboratory-maintained bacteria, including Streptococcus sanguis which has been implicated in the etiopathogenesis of RAU. There were no differences in relative serum antibody levels for any isotype against any bacteria between control and RAU groups. RAU subjects with active lesions were then paired with control subjects, and each serum was tested against sedimentable material derived from the oral mucosa of each pair member. Th

Cytoprotecting-effect of cimetidine: experimental evidence in the rat gastric mucosal lesions induced by intragastric administration of necrotizing agents.

Contradictory data have been found on cimetidine-induced gastric cytoprotection. In our paper the effect of cimetidine administered intraperitoneally in doses of 2.5, 10 and 50 mg X kg-1, on: 1. The gastric acid secretory responses in 1 and 4 hr pylorus-ligated rats, and 2. The rat gastric mucosal lesions induced by intragastric administration of 0.6 M HCl, 0.2 M NaOH, 96% ethanol and 25% NaCl is discussed. It was found that: 1. Cimetidine dose-dependently inhibited the gastric acid secretion in 1 and 4 hr pylorus-ligated rats, but a dose of 2.5 mg X kg-1 did not exert any significant inhibiti

Na+-dependent co-transport of alpha-methyl D-glucoside across the mucosal border of rabbit descending colon.

(1) The uptake and bidirectional fluxes of 1-alpha-methyl D-glucoside were studied in isolated rabbit colonic mucosa. (2) The uptake of alpha-methyl D-glucoside was linear over the first 30 min and reached maximum after 1 h; was a saturable function of sugar concentration and was Na+-dependent. (3) An increase in sugar uptake across the mucosal border and net transepithelial sugar flux across sheets of colon was observed in the presence of 10(-4) M amiloride. (4) Phlorizin (10(-4) M) inhibited sugar uptake into the tissue water and abolished net sugar flux. Amiloride-stimulated sugar uptake wa

Inhalable epitope vaccine 1022-SpaA V3 establishes nasal mucosal fortress against zoonotic Streptococcus suis.

Streptococcus suis serotype 2 (SS2), a zoonotic pathogen, can breach the respiratory barriers and cause systemic infections in both humans and swine. Current vaccines against SS2 are mainly formulated as an injectable product and primarily induce serum IgG antibodies. However, there is an urgent need for inhalable vaccines that induce the mucosal immune response. Leveraging immunoinformatics, we designed a multi-epitope vaccine targeting conserved epitopes from two virulence factors of SS2, SSU05-1022 (IgA1 protease) and SpaA (pilus-associated protein). Four cytotoxic T-lymphocyte (CTL), seven

Interferon-λ inhibits immunopathology and boosts T cell-mediated immunity in a mouse model of mucosal viral infection.

Interferon lambda (IFN-λ) has emerged as a critical inducer of immune regulation in addition to its mucosal antiviral properties. However, the cellular and molecular mechanisms that govern its dual role during viral infections remain unclear. Here, we discovered that mucosal delivery of IFN-λ abrogates the herpes simplex virus type 2-induced (HSV-2-induced) vaginal tissue pathology observed in the absence of type I IFN signaling, through drastically inhibiting IL-6 production. At the same time, we demonstrated that IFN-λ rescued local production of CD4+ T cell-dependent IFN-γ, which is essenti

Comparison of Primary Endoscopic Submucosal Dissection versus Primary Endoscopic Mucosal Resection for Barrett's Esophageal Neoplasia Treatment.

Background Endoscopic resection of Barrett's esophageal neoplasia is commonly performed using either endoscopic submucosal dissection (ESD) or endoscopic mucosal resection (EMR). While submucosal dissection enables en bloc resection, institutional approaches vary. We compared clinical, histopathologic, and oncologic outcomes between a primary submucosal dissection strategy and a mucosal resection-based strategy with selective use of submucosal dissection. Methods This retrospective cohort study included all patients with Barrett's esophageal neoplasia treated at two European tertiary referral

Mitochondria-associated endoplasmic reticulum membranes facilitate gastric epithelial injury in hypoxia-induced gastric mucosal lesions.

INTRODUCTION: Hypoxia following mucosal congestion in clinical practice causes a wide range of gastrointestinal symptoms and induces profound structural and functional perturbations in mitochondria-associated ER membranes (MAMs), which are subcellular compartments whose integrity and stability are regulated by FUN14 domain-containing protein 1 (FUNDC1). OBJECTIVES: To investigate the effect of hypoxia-induced MAMs formation on gastric mucosal lesions in patients with severe hepatic disorders. METHODS: Liver cirrhosis (LC) and hepatic failure (HF) were selected as representative severe hepati

Biopsy-derived colonoid air-liquid interface monolayers reveal persistent mucosal defects in ulcerative colitis patients.

BACKGROUND: The inflammatory bowel disease (IBD) ulcerative colitis (UC) is characterized by colonic mucosal inflammation and barrier dysfunction. We hypothesized that UC causes persistent defects in mucosal homeostasis, evident even in the absence of active inflammation, contributing to disease chronicity. METHODS: To test our hypothesis, we grew patient biopsy-derived sigmoid colonoids into air-liquid interface (ALI) monolayers, characterizing them through microscopy, proteomics, bulk RNA Sequencing (RNAseq), and their susceptibility to UC patient-isolated Escherichia coli pathobiont p19A.

Microbiota-derived IPA protects against colitis by regulating intestinal HMGCS2-mediated ketogenesis to facilitate mucosal healing.

The gut microbiota sustains intestinal homeostasis, yet how microbial metabolites direct epithelial repair remains unclear. Here we identify indole-3-propionic acid (IPA), a tryptophan-derived bacterial metabolite, as a key regulator of mucosal healing. IPA activates PPARα in intestinal epithelial cells, enhancing transcription of the ketogenic enzyme HMGCS2 and boosting β‑hydroxybutyrate (BHB) production. BHB in turn stimulates LGR5⁺ intestinal stem cells, accelerating epithelial regeneration. Using germ-free models and the IPA‑producer Peptostreptococcus russellii, we show that dietary trypt

Regimen-Related Oral Mucosal Injury: Pathobiology, Clinical Consequences and Rationale for Intervention Across Cancer Treatment Modalities.

PURPOSE OF REVIEW: Mucosal injury of the alimentary tract (mucositis) has been a consistent, markedly symptomatic, and clinically disruptive side effect of conventional cytotoxic chemotherapy and radiotherapy regimens since their inception. Despite its impact, mucositis remains a significant unmet clinical need as available treatment options are sparse. This review uses oral mucositis as a predicate regimen-related toxicity to provide a comparative discussion of the pathobiology, clinical features, and risk factors of regimen-related mucosal injury by therapy class. RECENT FINDINGS: The alime…

ADP-ribosylation factor 1 sustains regulatory T cell function and mucosal immune homeostasis in ulcerative colitis.

Dysregulation of regulatory T cells (Tregs) is a hallmark of immune imbalance in ulcerative colitis (UC), in which their suppressive function relies heavily on post-translational modifications and the stabilization of the transcription factor Foxp3. However, the upstream regulatory mechanisms remain poorly understood. Here, we identify ADP-ribosylation factor 1 (Arf1) as a key regulator of Treg function. Clinical samples from UC patients revealed an inverse correlation between ARF1 expression in Tregs and disease severity. Using a Treg-specific Arf1 knockout mouse model, we demonstrate that Ar

Enrichment of Candida associated with dysbiosis contributes to mucosal CD4+FOXP3+ regulatory T cell accrual and their dysfunction in aging.

Age-associated T cell dysfunction is a defining feature of inflammaging and immunosenescence, the progressive decline in immune competence observed with advancing age. Here we identified the association between aging (defined as age >60) and fungal dysbiosis, notably characterized by increased colonization of Candida species in the oral mucosa. There is also a notable enrichment of other taxa related to the order Saccharomycetales in older individuals. In contrast, younger individuals exhibit a greater abundance of Cryptococcus, Yarrowia, Kluyveromyces, and various Incertae sedis lineages. Fur
