Cyclobenzaprine-related adverse events: a comprehensive pharmacovigilance analysis using the FDA Adverse Event Reporting System.

BACKGROUND: Cyclobenzaprine, a centrally-acting muscle relaxant, is widely used for the treatment of musculoskeletal pain. Despite its efficacy, concerns regarding its safety profile, particularly adverse events (AEs), have been increasingly reported. This study aims to comprehensively analyze cyclobenzaprine-related AEs using the FDA Adverse Event Reporting System (FAERS) database to identify potential safety signals and inform clinical practice. METHODS: A retrospective pharmacovigilance study was conducted using FAERS data from Q1 2004 to Q3 2024. Reports involving cyclobenzaprine as the p

Assessment of ultra-processed food consumption and pro-inflammatory dietary load in adults diagnosed with fibromyalgia syndrome: A case-control study.

Background: Fibromyalgia syndrome (FMS) is a chronic condition marked by widespread musculoskeletal pain, persistent fatigue, and cognitive impairment. Its pathophysiology involves central sensitization, neuroinflammation, oxidative stress, and gut microbiota dysbiosis. Emerging evidence suggests that dietary factors-particularly the consumption of ultra-processed foods (UPFs)-may exacerbate these mechanisms and contribute to increased symptom burden. Aims/Objectives: This study aimed to examine the dietary patterns, inflammatory potential of the diet, and body composition profiles in adults d

Deep Learning in Scaphoid Fracture Detection and Healing Prediction: A Systematic Review of Artificial Intelligence Applications in Hand Surgery.

Scaphoid fractures are the most common carpal bone injuries and continue to pose diagnostic and therapeutic challenges due to their tendency for delayed union or non-union. Interpretation variability and delayed detection remain key contributors to missed injuries and adverse outcomes. Deep learning (DL) models, particularly convolutional neural networks (CNNs), have shown strong performance in musculoskeletal imaging and offer potential to improve diagnostic accuracy and prognostication in hand surgery. A systematic review of PubMed, Embase, and PMC was conducted covering January 2015 to Marc

Sonographic Signatures of Immune Checkpoint Inhibitor-Associated Musculoskeletal Adverse Events.

Background: Immune checkpoint inhibitors (ICIs) transformed cancer treatment, producing significant survival benefits. However, ICIs can trigger toxicities called immune-related adverse events (irAEs), including inflammatory arthritis (IA) and polymyalgia rheumatica (PMR)-like syndromes. Our study aimed to systematically further characterize musculoskeletal ultrasound (MSKUS) findings in patients with ICI-IA and ICI-PMR, collectively referred to as "MSK-irAEs", and explore the role of US in guiding treatment. Methods: The authors conducted a comprehensive chart review for patients receiving IC

Cognitive-motor interference during walking with modified leg mechanics: a dual-task walking study.

BACKGROUND: The use of mobile exoskeletons as assistive walking devices has the potential to affect the biomechanics of the musculoskeletal system due to their weight and restricted range of motion. This may result in physical and cognitive load for the user. Understanding how lower extremity loading affects cognitive-motor interference is crucial for the design of wearable devices, including powered exoskeletons, and the development of effective training interventions. OBJECTIVE: This study aims to examine the effects of modified leg mechanics on cognitive-motor interference in dual-task wal

Evaluating the representativeness of a cohort study of low back pain: Using electronic health record data to make direct comparisons of study participants with non-participants from the study population.

Representativeness is an important component of generalizability. Few studies have rigorously examined the representativeness of randomized trials or observational studies of pain or musculoskeletal conditions with regards to a wide range of factors beyond age, sex, race, and ethnicity. We conducted the first study of a pain condition that uses individual-level data to directly compare the enrolled study sample to the population from which it was drawn. We used electronic health record data from the Veterans Affairs health system to compare participants in an observational study of low back pa

Transcranial direct current stimulation is more effective than pregabalin in controlling nociceptive and anxiety-like behaviors in a rat fibromyalgia-like model.

OBJECTIVES: Despite the fact that fibromyalgia, a widespread disease of the musculoskeletal system, has no specific treatment, patients have shown improvement after pharmacological intervention. Pregabalin has demonstrated efficacy; however, its adverse effects may reduce treatment adherence. In this context, neuromodulatory techniques such as transcranial direct current stimulation (tDCS) may be employed as a complementary pain-relieving method. Consequently, the purpose of this study was to evaluate the effect of pregabalin and tDCS treatments on the behavioral and biomarker parameters of ra

Developing and testing a framework for coding general practitioners' free-text diagnoses in electronic medical records - a reliability study for generating training data in natural language processing.

BACKGROUND: Diagnoses entered by general practitioners into electronic medical records have great potential for research and practice, but unfortunately, diagnoses are often in uncoded format, making them of little use. Natural language processing (NLP) could assist in coding free-text diagnoses, but NLP models require local training data to unlock their potential. The aim of this study was to develop a framework of research-relevant diagnostic codes, to test the framework using free-text diagnoses from a Swiss primary care database and to generate training data for NLP modelling. METHODS: Th
