Cellular long noncoding RNAs (lncRNAs) have been shown to participate in diverse biological pathways, including the regulation of virus infections. However, their functions in the hepatitis B virus (HBV) life cycle still need to be explored. Here, we conducted a loss-of-function screen targeting 1779 human lncRNAs and identified 19 lncRNAs associated with HBV infection. One lncRNA, named MAPKAPK5-AS1 (lnc-MAAS), was identified as a host factor required for HBV infection. Our data suggest that lnc-MAAS is involved in the conversion of HBV relaxed circular DNA into covalently closed circular DNA

Posttranslational modifications (PTMs) are crucial regulators of protein functions, localization, and interactions, contributing to nearly all aspects of cellular physiology. Conventional analytical approaches, such as mass spectrometry (MS), have achieved high sensitivity and coverage, yet they often require complex sample preparation, enrichment, and high-end instrumentation. Nanopore has emerged as a promising single-molecule approach for PTM detection, offering label-free, high-throughput, and potentially portable analysis capabilities. Despite remarkable progress in nucleic acid sequencin

Phase separation is a fundamental principle of cellular organization that typically leads to two coexisting phases: a dense one, where intermolecular interactions are stronger and molecules are more tightly packed, and a dilute, less packed phase, with weaker interactions and lower molecular concentration. This process drives the formation of both lateral membrane domains (rafts) and liquid-like protein and nucleic acid condensates. This Review explores the dynamic interplay between biomolecular condensates and membrane lipid domains and how phase separation occurs in the three-dimensional (bu

CRISPR-Cas systems use RNA-guided proteins for adaptive immunity through a mechanism whose origin is unknown. Here we report the discovery of Viral Interference Programmable Repeat (VIPR) systems consisting of a Vipr protein more ancient than CRISPR-Cas and vrRNAs comprising alternating GGY/NN motifs. Unlike canonical guide RNAs that base pair with nucleic acid targets using an uninterrupted sequence, vrRNAs recognize double-stranded DNA through a noncontiguous code in which the variable NNs of each repeat collectively specify a target that itself contains a gapped recognition sequence. Analys

Human norovirus is a major acute gastroenteritis pathogen with high genotypic diversity, mutation, and recombination rates. Existing sequencing methods for monitoring its genetic variations are limited by low sensitivity and contamination risk. This study optimized our previously established Micro Target Hybrid Capture System (MT-Capture) by increasing nucleic acid input, streamlining library preparation, and introducing a deployable USER enzyme-based contamination prevention system. The optimized protocol completes library preparation and capture within 7 hours. Compared with multiplex PCR se

Sample preparation is essential in chemical analysis as a critical determinant of analytical sensitivity, selectivity, and throughput, and is often the most resource-intensive stage of an analytical workflow. Microscale solid-phase extraction (µSPE) helps to address the sample preparation limitations associated with conventional SPE by enabling analyte enrichment and matrix cleanup at reduced volumes, with integration potential that traditional formats cannot match. This Critical Review assesses recent advances across six µSPE modalities: reverse-phase, ion exchange, immunoaffinity, aptamer-ba

Endogenous, or 'self', vs. microbial, or 'non-self', RNA sensing can tip the scales between immune pathology and effective immunity. Cells are equipped to sense RNA, fundamental to trigger an innate immune response to clear viral infection that should not generate a harmful immune response against endogenous RNA. Multiple chemical modifications in RNA fine-tune its cellular sensing and are exploited by pathogens to evade immunity. Likewise, perturbations triggering self RNA sensing cause immune pathologies. This underscores the clinical need for a better understanding of self RNA recognition.

Cell surface glycoRNAs have recently emerged as an unexpected class of RNA-glycan conjugates. Their discovery challenges the long-standing view that RNA is confined to intracellular compartments. These small noncoding RNAs, modified with N- or O-linked glycans and displayed on the outer leaflet of the plasma membrane, create a unique molecular interface recognizable by both glycan-binding and nucleic acid-sensing immune receptors. Recent studies suggest that glycoRNAs engage multiple immune receptor families, including P-selectin, Siglecs, Toll-like receptors (TLRs), and lectins, to influence

Influenza A virus (IAV) remains a major focus of global public health concern, as it can cause influenza-like illness and severe respiratory infections. Therefore, achieving accurate and sensitive detection of IAV would greatly facilitate disease diagnosis and control. The two main conventional technical methods for IAV detection that are reviewed in this article are viral nucleic acid amplification analysis and immunological assays. At the same time, this review pays special attention to the cross-integration of these two types of techniques with modern engineering technologies, and systemati

Catalytic DNA molecules (DNAzymes) offer a programmable means to manipulate RNA without genomic modification. Although DNAzymes have been thoroughly characterized in culture and in various animals, their utility in the prevalent model system, Caenorhabditis elegans, has not been explored. Here, we examine the feasibility of catalytic DNA-mediated RNA targeting in C. elegans. Cholesterol-conjugated 8-17 DNAzymes were efficiently delivered to intestinal cells following ingestion, with no visibile toxic effects on growth or health, reducing targeted reporter fluorescence and endogenous gene funct

Nanomedicine has transformed therapeutic strategies by enabling precise delivery of nucleic acid-based drugs, including small interfering RNA (siRNA), messenger RNA (mRNA), and antisense oligonucleotides. A landmark achievement is Onpattro (patisiran), the first FDA-approved RNAi therapy, which employs lipid nanoparticles (LNPs) to silence transthyretin in hereditary amyloidosis. Its approval validates RNAi as a viable therapeutic modality and underscores the central role of nanocarriers in clinical translation. Despite this success, barriers such as nanoparticle stability, targeted delivery,

Escalating antibiotic resistance and poor options for many parasitic infections, including neglected tropical diseases, demand new antimicrobial strategies. G-quadruplexes (G4s), which are noncanonical nucleic acid structures, are emerging targets in microbial genomes. We summarize evidence for G4 formation in bacteria and parasites, which is provided by phenotypic and chemical-biology datasets, whereas evidence in support of fungal G4 targeting remains nascent, with fewer studies and limited mechanistic validation. After integrating genomics and bioinformatics data, we argue that microbial G4

Schistosomiasis, caused by Schistosoma species, is notoriously difficult to accurately diagnose with conventional methods. In this study, we present an innovative biosensor that integrates CRISPR-Cas12a technology with nucleic acid aptamers for the highly sensitive detection of Schistosoma japonicum eggs. The biosensor leverages a Y-shaped DNA structure (Y-DNA) that incorporates an aptamer specific to S. japonicum eggs, along with an activator DNA and a segment for immobilization on magnetic nanomaterials. Upon target recognition, the Y-DNA releases the activator, which triggers the collateral

Lipid nanoparticles (LNPs) have become an important platform for nucleic acid delivery. However, LNP-mediated delivery to non-hepatic organs and specific cell types remains a non-negligible challenge. As a key component of LNPs, ionizable lipids were rationally designed to adjust the LNPs properties to achieve organ-targeted delivery. The use of the Ugi four-component reaction (Ugi-4CR) as a one-pot multicomponent synthesis strategy to construct ionizable lipid molecules offers advantages, as it enables multidimensional structural diversity of ionizable lipids. We use isocyanides with cyclic t

Inorganic phosphate (Pi) is an essential nutrient for all organisms. It has critical functions in lipid and nucleic acid synthesis, protein signaling and bone growth. Loss-of-function mutations in Pi transporters lead to embryonic and neonatal lethality. Here, we show that the only known Pi exporter, XPR1, is critical for the development of fetal macrophages in the liver and the spleen. Single-cell RNA-seq and flow cytometry analyses in conditional mice lacking Xpr1 in hematopoietic and/or CD206+ cells revealed loss of the Kupffer cell transcriptional program and a shift in the development of

The nucleocapsid (N) protein of SARS-CoV-2 performs multiple functions essential for viral replication and host adaptation. Here, we identify ionic strength as a critical determinant of its dual nucleic acid-manipulating activities. Under low-salt conditions (≤100 mM NaCl), SARS-CoV-2 N primarily promotes strand annealing, whereas at high ionic strength (≥300 mM NaCl), its helicase activity is additively enhanced. SARS-CoV-2 N also preferentially acts on DNA rather than RNA substrates and modulates the helicase activity of Nsp13 in an ion-dependent manner-inhibiting it under low-salt condition

Emerging swine enteric coronaviruses (SeCoV) pose significant threats to global pork security, with zoonotic porcine deltacoronavirus (PDCoV) demonstrating cross-species transmission potential. Here, we identified a new PDCoV strain, CHN/GD/2023, which was isolated from porcine intestinal epithelium, showing phylogenetic clustering with China-endemic variants through viral spike (S) protein evolutionary tracing. A vaccine candidate for PDCoV was produced by genetically fusing the receptor-binding domain (RBD) of CHN/GD/2023 to the C-terminus of the AP205 gene. The purified AP205-RBD successful

Influenza A virus (IAV) infections can cause life-threatening illness in humans. The severity of disease is directly linked to virus replication in the alveoli of the lower respiratory tract. In particular, the lytic death of infected alveolar epithelial cells (AECs) is a major driver of influenza severity. Recent studies have begun to define the molecular mechanisms by which IAV triggers lytic cell death. Z-form nucleic-acid-binding protein 1 (ZBP1) senses replicating IAV and drives programmed cell death (PCD) in infected cells, including apoptosis and necroptosis in AECs and pyroptosis in my

Osteoporotic bone regeneration is challenging due to impaired bone formation. Tetrahedral DNA nanostructures (TDN), promising nucleic acid nanomaterials, have garnered attention for their potential in osteoporotic mandibular regeneration owing to their ability to enhance cellular activity and promote osteogenic differentiation. Osteoblasts play a critical role in bone regeneration; however, intracellular delivery of TDN into osteoblasts remains difficult. In this study, a novel osteoblast-targeted CH6 aptamer-functionalized TDN (TDN-CH6) is aimed to develop for osteoporotic mandibular regenera
