Small Heterodimer Partner Negatively Regulates TLR4 Signaling Pathway of Titanium Particles-Induced Osteolysis in Mice.

Arthroplasty has been widely performed worldwide. However, peri-prosthetic osteolysis and aseptic loosening induced by macrophages activated by wear particles still remain a predominant cause of long term prosthetic failure. Our study aimed to identify the role of small heterodimer partner (SHP) in secretion of proinflammatory cytokines by macrophages through Toll-like Recepters (TLR)s signaling pathway activated by wear particles both in vivo and in vitro. The effect of SHP on activation of TLR4 pathway and secretion of cytokines was observed in RAW264.7 cells and SHP gene over-expressed mice

Demineralized dentin and enamel matrices as suitable substrates for bone regeneration.

BACKGROUND: In recent decades, tooth derivatives such as dentin (D) and enamel (E) have been considered as potential graft biomaterials to treat bone defects. This study aimed to investigate the effects of demineralization on the physical-chemical and biological behavior of D and E. METHODS: Human D and E were minced into particles (Ø<1 mm), demineralized and sterilized. Thorough physical-chemical and biochemical characterizations of native and demineralized materials were performed by SEM and EDS analysis and ELISA kits to determine mineral, collagen type I and BMP-2 contents. In addition, M

Effects of hypothalamic leptin gene therapy on osteopetrosis in leptin-deficient mice.

Impaired resorption of cartilage matrix deposited during endochondral ossification is a defining feature of juvenile osteopetrosis. Growing, leptin-deficient ob/ob mice exhibit a mild form of osteopetrosis. However, the extent to which the disease is (1) self-limiting and (2) reversible by leptin treatment is unknown. We addressed the first question by performing histomorphometric analysis of femurs in rapidly growing (2-month-old), slowly growing (4-month-old) and skeletally mature (6-month-old) wild-type (WT) and ob/ob male mice. Absent by 6 months of age in WT mice, cartilage matrix persist

The Pathophysiological Sequence of Glucocorticoid-Induced Osteonecrosis of the Femoral Head in Male Mice.

In search of the sequence of pathogenic events leading to glucocorticoid-induced osteonecrosis, we determined the molecular, biomechanical, cellular, and vascular changes in the femur of C57BL/6 mice receiving prednisolone for 14, 28, or 42 days. The femoral head, but not the distal femur, of mice treated for 14 days showed a decrease in the expression of the hypoxia-inducible factor (Hif)-1α and vascular endothelial growth factor (VEGF), the number of osteoblasts, and bone formation rate and strength and showed an increase in osteoclasts. These changes were accompanied by conversion of the no…

Apical External Root Resorption and Repair in Orthodontic Tooth Movement: Biological Events.

Some degree of external root resorption is a frequent, unpredictable, and unavoidable consequence of orthodontic tooth movement mediated by odontoclasts/cementoclasts originating from circulating precursor cells in the periodontal ligament. Its pathogenesis involves mechanical forces initiating complex interactions between signalling pathways activated by various biological agents. Resorption of cementum is regulated by mechanisms similar to those controlling osteoclastogenesis and bone resorption. Following root resorption there is repair by cellular cementum, but factors mediating the transi

Root Resorption: Simplifying Diagnosis and Improving Outcomes.

Root resorption is a condition resulting in the progressive loss of dental hard tissue. It may occur both within the root and upon the external aspect of the root. Diagnosis can be difficult and management challenging. Understanding the pathology is critical to understanding why and when this disease occurs and what the best management techniques involve. With such knowledge practitioners can confidently diagnose resorption, discuss prognoses and management strategies with the patient and either refer or begin treatment. Early intervention is paramount in improving outcomes. As such, if practi

Osteonecrosis of the Jaw-a Bone Site-Specific Effect of Bisphosphonates.

A known complication that can occur in patients using bisphosphonates (BPs) is osteonecrosis of the jaw (ONJ). ONJ features bone exposure that may be associated with severe pain, swelling, local infection, and pathological fracture of the jaw. Current literature indicates that a complex combination of factors is necessary to induce ONJ. Several hypotheses about the pathophysiology of ONJ were previously reported. Here, we review these hypotheses and introduce new ideas and suggestions on this topic, focusing on bone site-specific cells, and the effect that BPs and other anti-resorptive drugs h

Follicular dendritic cell-secreted protein may enhance osteoclastogenesis in periodontal disease.

PURPOSE OF THE STUDY: Follicular dendritic cell-secreted protein (FDC-SP) has been found to be expressed in periodontal ligament (PDL), a layer of soft connective tissue between tooth root and alveolar bone, and involved in immunoreaction. This study was performed to explore the potential role of FDC-SP in periodontal disease. MATERIALS AND METHODS: The human periodontal ligament cells (hPDLCs) were stimulated with Porphyromonas gingivalis (P. gingivalis) lipopolysaccharide (LPS) and FDC-SP expression was examined by real-time PCR and western blot. Then this molecule was overexpressed or sile

The manifestation of cocaine-induced midline destructive lesion in bone tissue and its identification in human skeletal remains.

Cocaine-induced midline destructive lesion (CIMDL) is a condition that may arise in response to chronic insufflation ("snorting") of cocaine. It is clinically diagnosed when the nasal septum, lateral nasal walls, and/or hard palate show signs of destruction in association with cocaine use. Although its true incidence is unknown, CIMDL is not an uncommon clinical finding amongst intranasal cocaine abusers and is likely to be encountered by forensic anthropologists and medical examiners working worldwide. Given the preponderance of drug abusers amongst the subjects of forensic casework, the abil

Root resorption of primary molars without successor teeth. An experimental study in the beagle dog.

Tooth agenesis is a common craniofacial congenital malformation in humans, but little is known about the mechanisms of root resorption in this condition. The purpose of this study was to investigate the mechanisms of root resorption in primary molars without successors. An animal model without permanent tooth germs was established by surgery in beagles. The times of onset of primary molar root resorption, with and without successors, were compared. The distribution of immune cells, odontoclasts, and their activating factors were determined by histochemistry and immunohistochemistry. Root resor

[Encounter of cancer cells with bone. Development of bone metastasis-specific targeting therapy].

Bone, as well as liver and lungs, is one of the most preferential metastatic targets for solid cancers including breast, prostate, and lung cancers. Control of the bone metastasis is clinically important ; because they are frequently associated with bone pain and pathological fracture which greatly diminish the quality of life for patients. Interaction between cancer cells and microenvironment of metastatic site play an important role for establishment of cancer metastasis. The understanding of their mechanisms has been improved in recent years, and they will be new therapeutic strategies to c

Bisphosphonate-related osteonecrosis of the jaw: position paper from the Allied Task Force Committee of Japanese Society for Bone and Mineral Research, Japan Osteoporosis Society, Japanese Society of Periodontology, Japanese Society for Oral and Maxillofacial Radiology, and Japanese Society of Oral and Maxillofacial Surgeons.

Bisphosphonates (BPs) have been widely, efficiently, and safely used for the treatment of osteoporosis, malignant hypercalcemia, bone metastasis of solid cancers, and multiple myeloma bone diseases. Accumulating recent reports describe that surgical dental treatments in patients with cancer or osteoporosis who have been receiving intravenous or oral BPs are associated with osteonecrosis of the jaw (bisphosphonate-related osteonecrosis of the jaw, BRONJ). The accurate incidence, clinical backgrounds, and pathogenesis of BRONJ have been unclear and appropriate approaches for prevention and treat

Quantifiable excess of bone resorption in monoclonal gammopathy is an early symptom of malignancy: a prospective study of 87 bone biopsies.

To determine if excessive osteoclastic-mediated bone resorption (BR) is an early tumor-induced event in multiple myeloma (MM), BR was assessed at first presentation on quantitative bone biopsy in 87 individuals evaluated for monoclonal gammopathy of undetermined significance (MGUS) and reinterpreted according to the presenting features and subsequent follow-up evaluation. As a reference population, 48 patients with previously untreated overt MM were evaluated under similar conditions. The median level of BR was significantly higher in 48 overt MM versus 87 MGUS patients (12.2% v 5.1% [normal l

Childhood renal osteodystrophy.

The processes of growth and remodeling that characterize growing bone are vulnerable to the biochemical effects of uremia. These processes are affected by the alterations in vitamin D metabolism and hyperparathyroidism that commonly occur with chronic renal insufficiency. Uremia interferes with cartilaginous developments as well. These changes, which are similar radiographically to vitamin-D-deficiency rickets, differ in their basic histological and biochemical evolution. They account for growth failure and deformity of long bones, which contribute to the short stature of children with renal i

Molecular recognition of glycosaminoglycans and their sulfated deep-sea bacteria-derived mimetics, diabolican and infernan, by RANK, RANKL and OPG.

Maintenance of bone function and remodelling is a strictly controlled physiological process that is modulated by bone forming and resorbing, mediated by osteoblasts and osteoclasts respectively. A signalling axis involved in controlling this process involves receptor activator of nuclear factor kappa B ligand, receptor activator of nuclear factor kappa B and osteoprotegerin. The two former proteins are vital in osteoclastogenesis whilst osteoprotegerin serves a modulatory role, interfering with the interaction of the other two proteins. The binding of these proteins by glycosaminoglycans has b…
