Separation of triterpenoid saponins from the root of Bupleurum falcatum by counter current chromatography: the relationship between the partition coefficients and solvent system composition.

A separation method using counter current chromatography coupled with an evaporative light-scattering detection system was developed to purify five triterpenoid saponins from the roots of Bupleurum falcatum. The methanol extract was loaded onto a Diaion® HP20 column and fractionated by a methanol and water gradient elution. The saikosaponin-enriched fraction was obtained by elution with 100% methanol. The two-phase solvent systems used for separation were composed of chloroform/methanol/isopropanol/water at a volume ratio of 60:60:1:60 and 6:6:1:6. The relationship between the isopropanol rati

Mast cell degranulation after a single dose of gliadin in the jejunum of patients with coeliac disease.

The exact pathomechanism of jejunal damage caused by gliadin in coeliac disease has not been clarified. 0.3 g of gluten per kg bodyweight was administered to 14 children affected by coeliac disease, being on gluten-free diet, five hours before jejunal biopsy. There was no change in the number of intraepithelial lymphocytes and eosinophils while cellular infiltration of the lamina propria exhibited a marked increase and the number of mast-cells per tissue unit significantly decreased as compared with controls. It is concluded that mast-cells may play an important role in the mechanism of gluten

Rice straw sample preparation protocol for cell wall major constituent analysis via Raman spectroscopy.

Raman spectroscopy (RS) is a rapid, label-free, and non-destructive technique for analyzing molecular and spectral characteristics of biological samples. Here, we present a reproducible protocol for glycerol-based sample preparation and RS analysis of long-term air-dried rice straw. We describe steps for preparing samples, acquiring spectral data, and preprocessing data. Our workflow enables reproducible spectral acquisition and semi-quantitative assessment of relative Raman spectral markers associated with major plant cell wall components using standard laboratory procedures. Spectral preproc

Tetrahydrofuran- and dihydrofuran-containing nor-clerodane diterpenoids from Teucrium viscidum: Isolation, stereochemical assignment, and anti-hepatic fibrosis activity.

Ten previously undescribed nor-clerodane diterpenoids, teuvinoids A-J (1-10), were isolated from the whole plants of Teucrium viscidum. Comprehensive structural elucidation, including unambiguous determination of absolute configurations, was accomplished using spectroscopic analysis, electronic circular dichroism, and single-crystal X-ray diffraction. Teuvinoids A (1) and B (2) are particularly noteworthy for featuring an unusual 2,3,4,5-tetrahydroxylated tetrahydrofuran moiety. Compounds 1 and 2 exhibited anti-hepatic fibrosis effects in TGF-β1-induced LX-2 hepatic stellate cells, as demonstr

Anthraquinones of the roots of Pentas micrantha.

Pentas micrantha is used in the East African indigenous medicine to treat malaria. In the first investigation of this plant, the crude methanol root extract showed moderate antiplasmodial activity against the W2- (3.37 μg/mL) and D6-strains (4.00 μg/mL) of Plasmodium falciparum and low cytotoxicity (>450 μg/mL, MCF-7 cell line). Chromatographic separation of the extract yielded nine anthraquinones, of which 5,6-dihydroxylucidin-11-O-methyl ether is new. Isolation of a munjistin derivative from the genus Pentas is reported here for the first time. The isolated constituents were identified by NM

The hepatoprotective activity of kinsenoside from Anoectochilus formosanus.

Carbon tetrachloride (CCl(4)) causes chronic hepatitis, featuring an increase in hepatic hydroxyproline, spleen weight and serum GPT levels and a decrease in plasma albumin levels. Crude extracts of fresh whole plants of Anoectochilus formosanus showed inhibition of chronic hepatitis induced by CCl(4) in mice. Bioactivity-guided fractionation and spectroscopic analysis revealed that kinsenoside was the most active compound. In an in vitro study, the LD(50) values for H(2)O(2)-induced cytotoxicity in BALB/c normal liver cells were significantly higher after kinsenoside pretreatment than after v

[Variation in evolutionary unstable regions of the chloroplast genome in plants obtained in anther culture of dihaploid wheat lines].

In dihaploid wheats, two evolutionarily unstable regions of the chloroplast genome were examined. These regions include the following genes, changes in which could be associated with albinism in anther culture: rbcL, encoding the large Rubisco subunit; psaA, encoding p700 apoprotein Ia; petA, encoding cytochrome f; atpB and atpE, encoding respectively beta and epsilon subunits of the CF1 ATPase complex; trnE, encoding glutamine tRNA; and cemA, encoding a cell membrane protein. Using PCR, we have shown that atpB was the gene most often not detected in the lines examined. These results suggest t

Ferulic acid stabilizes a solution of vitamins C and E and doubles its photoprotection of skin.

Ferulic acid is a potent ubiquitous plant antioxidant. Its incorporation into a topical solution of 15%l-ascorbic acid and 1%alpha-tocopherol improved chemical stability of the vitamins (C+E) and doubled photoprotection to solar-simulated irradiation of skin from 4-fold to approximately 8-fold as measured by both erythema and sunburn cell formation. Inhibition of apoptosis was associated with reduced induction of caspase-3 and caspase-7. This antioxidant formulation efficiently reduced thymine dimer formation. This combination of pure natural low molecular weight antioxidants provides meaningf

Exploring the therapeutic potential of momordica balsamina: A focus on anticancer and nephroprotective effects.

BACKGROUND: Despite being considered a gold standard chemotherapeutic drug, cisplatin is associated with dose-limiting toxicity, especially nephrotoxicity. This highlights the need for safe and effective adjuvant agents that protect against cisplatin-induced toxicity without affecting its anticancer activity. This study investigated the potential of Momordica balsamina aqueous extract (MBAE) as an adjuvant to reduce cisplatin-induced kidney cell damage and death. METHODS: The phytochemical analysis of the extract was done using Gas Chromatography-Mass Spectrometry. The effects of the extract

Analyses of the Cell Wall Modification of Wheat Cultivars Infested with Diuraphis noxia.

Wheat infested with Diuraphis noxia (Russian wheat aphid: RWA) respond by inducing a range of defense processes at the morphological, physiological, molecular, and biochemical levels. The physiological response includes the cell wall modification, which is generally weakened by the RWA infestation in the susceptible cultivar, while in the resistant cultivar, the cell wall modification leads to a more robust and strengthened wall. Direct measurements of the cell wall have been a challenge, but in the last decade, extraction methods for cell wall components, i.e., cellulose, hemicellulose and li

TipQuant: a robust algorithm for quantitative analysis of spatiotemporally dynamic activities in tip-growing cells.

Cell polarity and tip growth rely on the dynamic spatial organization of signaling and structural components. Quantitative characterization of these spatiotemporal dynamics is critical for understanding polarized cell growth, yet manual quantification is labor-intensive and existing computational tools often lack the flexibility and robustness needed to analyze molecular and structural dynamics in tip-growing cells. Tip Quantification (TipQuant) identifies the cell apex by detecting the site of maximum expansion and automatically quantifies fluorescence distribution along the plasma membrane a

Oleanolic acid ameliorates dextran sodium sulfate-induced colitis in mice by restoring the balance of Th17/Treg cells and inhibiting NF-κB signaling pathway.

In a preliminary experiment, it was found that oleanolic acid (OA), which is widely distributed in food and medicinal plants, inhibited interleukin (IL)-6/tumor growth factor beta-induced differentiation of splenic T cells into Th17 cells. Moreover, OA induced the differentiation of splenic T cells into Treg cells. Therefore, we examined the anti-inflammatory effect of OA in mice with dextran sodium sulfate (DSS)-induced colitis. Oral administration of OA significantly inhibited DSS-induced colon shortening, macroscopic score, and myeloperoxidase activity. Treatment with OA inhibited DSS-induc

Anticoagulants prevent monocrotaline-induced hepatic parenchymal cell injury but not endothelial cell injury in the rat.

Monocrotaline (MCT) is a pyrrolizidine alkaloid plant toxin that produces hepatotoxicity in humans and animals. Human exposure to MCT occurs through consumption of contaminated grains and herbal medicines. Administration of MCT to rats stimulates activation of the coagulation system and fibrin deposition in the liver. Fibrin deposition occurs simultaneously with endothelial cell damage and prior to hepatic parenchymal cell injury. Accordingly, the hypothesis that activation of the coagulation system is required for MCT-induced liver injury was tested. Treatment of rats with either heparin or w
