Blood-brain barrier permeability varies by brain region and APOE4 status and correlates with brain microstructure among high-AD risk groups.

BACKGROUND: Although strong evidence exists for blood-brain barrier (BBB) disruption in Alzheimer's disease (AD), substantial uncertainty remains regarding its role. We address gaps and inconsistencies in the literature by examining regional variation in BBB permeability among cognitively normal older adults enriched for AD risk, the influence of genetic risk and its interactions with amyloid-β and sex, and the relationships between BBB breakdown and brain microstructure. Additionally, we compare two methods of quantifying BBB permeability. METHODS: Dynamic contrast-enhanced magnetic resonanc

Intersecting stigma of substance use and child welfare system involvement: Understanding stigma drivers, experiences, and outcomes from a socioecological perspective.

BACKGROUND: Substance use in parents is a common reason for child welfare system involvement (CWS). Stigma associated with substance use and disorders (SU/D) continues to negatively impact individuals; however, less is understood about the intersecting stigma of CWS involvement and SU/D. The current study seeks to understand parents' experiences of stigma related to SU/D and CWS involvement as well as the drivers and consequences of stigma at multiple socioecological levels using the Health Stigma and Discrimination Framework. METHODS: Qualitative data analysis was conducted on semi-structure

Multi-region spatial transcriptomics reveals region specific differences in response to amyloid beta (Aβ) plaque induced changes in Alzheimer's Disease (AD).

Alzheimer's disease (AD) is the leading cause of dementia affecting 55 million people worldwide. The pathological hallmarks of AD, beta-amyloid (Aβ) plaques and neurofibrillary tangles (NFT), follow distinct stereotypical patterns of progression across brain regions and trigger a multicellular response that ultimately leads to neuronal loss and cognitive decline. Despite the uniform spread of Aβ plaque across the cortex during AD progression, different regions demonstrate varying levels of vulnerability and resilience to temporal Aβ plaque induced changes, such as NFT accumulation. There is a

Effect of Dietary Neutral Detergent Fiber Intake on Improving Fecal Consistency in Gestating Gilts.

Constipation is a common issue in sows, characterized by dry and hard feces or prolonged absence of defecation, which exerts adverse effects on the overall health and reproductive performance of sows. While dietary neutral detergent fiber (NDF), due to its physicochemical properties, can enhance satiety and promote gastrointestinal health. The present study aims to determine the effect of different dietary NDF intake levels on reducing constipation without impairing the reproductive capacity of gestating sows. A total of 110 primiparous crossbred gilts (Landrace × Yorkshire) were randomly assi

Temperament correlates of stigma resistance among patients with mood disorders: a cross-sectional study.

BACKGROUND: In the realm of mental health, stigma presents a barrier to well-being and social acceptance. However, amidst societal prejudices, stigma resistance emerges as a vital concept, reflecting individuals' capacity to challenge negative stereotypes and maintain a positive self-concept. This paper explores the dynamics of stigma resistance, its determinants, and its implications for mental health outcomes, focusing specifically on mood disorder patients. METHODS: Adult patients with mood disorders who provided written informed consent were consecutively recruited. Data were collected be

Fighting Autism with Fatty Acids: Maternal Omega-3 Shields the Developing Brain from VPA-Induced Behavioral and Neurochemical Damage.

Background/Objectives: Autism spectrum disorder is a psychological condition characterized by symptoms such as repetitive stereotypic behaviors and social interaction/communication difficulties. It is known that omega-3 deficiency during brain maturation may cause learning disabilities and motor impairment. Therefore, we examined the effects of omega-3 treatment during gestation and/or lactation on autism-related behavioral and molecular deficits in a valproic acid (VPA)-rat model. Methods: Female Wistar rats were divided into five groups: control, VPA (500 mg/kg at G12.5), VPA+omega-3 (gestat

Development of an Opioid Stigma Scale for People Living With Chronic Pain.

PURPOSE: High levels of stigma among patients with chronic pain contribute to negative psychological well-being. Existing measures have not focused on opioid stigma among individuals with chronic pain who may have legitimate need for opioid therapy. DESIGN: We developed a measure of opioid stigma that captures two constructs: perceived stigma (awareness of others' negative attitudes) and internalized stigma (application of stereotypes to oneself) among individuals with chronic pain. METHODS: We recruited 180 participants with chronic pain via the online platform CloudResearch to modify an ex

The Field of Gender Through Metaphors: The Dilemma of Female and Male Referees in the Minds of Football Fans.

The perception of football as a male-dominated sport by society, coupled with the socio-cultural and economic barriers faced by women, has constrained their presence in the domain of football and revealed the manifestation of gender norms within the sport. This exclusion further masculinizes sport, negatively affecting social unity and cohesion, and deepening inequality within sport. Within this context, the study seeks to reveal how football fans perceive female and male referees through metaphorical representations. Participants, selected using purposive sampling, are individuals who regular

Elites' perceptions of women's representation in the Omani media.

INTRODUCTION: This study investigates how Omani elites perceive women's representation in national media and how structural, cultural, and political forces shape gender narratives in the Sultanate. It explores how women's visibility is instrumentalized to project modernization while maintaining patriarchal authority, situating the Omani case within broader Gulf Cooperation Council (GCC) media-gender dynamics. METHODS: The research draws on 38 semi-structured elite interviews conducted between 2021 and 2025 with parliamentarians, journalists, academics, activists, and government officials. Int

Postnatal propionic acid exposure disrupts hippocampal agmatine homeostasis leading to social deficits and cognitive impairment in autism spectrum disorder-like phenotype in rats.

Autism Spectrum Disorder (ASD) is a complex neurodevelopmental condition characterized by a range of symptoms including impaired social interaction and cognitive deficits. Although the exact pathogenesis of ASD is not well established, recent clinical findings suggest a decline in levels of biogenic amine agmatine in autistic patients. The present study was designed to investigate the impact of postnatal propionic acid (PPA) exposure on hippocampal agmatine homeostasis in male rat pups and to explore a new therapeutic intervention for ASD using agmatine as a biological target. PPA is commonly

Sleep loss suppresses the motivation to prepare for sleep.

In the period preceding sleep, humans and other animals display a stereotyped repertoire of behaviors-including hygiene-related activities and preparing a place to sleep. Evidence suggests that this pre-sleep phase actively contributes to sleep initiation and quality. Nonetheless, individuals can sometimes fall asleep without preparation, even under undesirable circumstances. These abrupt transitions into sleep can have severe consequences, particularly in high-risk environments. Although progress has been made in identifying neuronal populations controlling sleep-wake states and mechanisms re

Therapeutic effects of striatal dopaminergic modulation on idiopathic dystonia and OCD in humans: insights from the striosome hypothesis.

Emerging evidence suggests that striatal striosomes play a key role in the dopaminergic regulation of motor and mental action selection processes, with impairments leading to repetitive stereotyped movements (dystonias), thoughts (obsessions), and behaviors (compulsions). To explore this hypothesis therapeutically, we investigated how idiopathic dystonia and obsessive-compulsive disorder (OCD) respond to a novel dopaminergic treatment using low-dose L-DOPA combined with chlorpromazine (CPZ), which can primarily enhance striosomal D1 dopamine receptor (D1R) signaling in humans. The therapeutic

Multivariate Resting-State Functional Connectivity Features Linked to Transdiagnostic Psychopathology in Early Psychosis.

BACKGROUND: Early psychosis (EP) is characterized by neurobiological changes, including alterations in resting-state functional connectivity (RSFC). We now understand that symptoms and neural changes may overlap across EP diagnostic categories. However, the relationship between RSFC patterns and transdiagnostic symptom dimensions remains poorly understood. METHODS: We used partial least squares correlation to examine multivariate relationships between whole-brain RSFC and clinical symptoms in 124 patients with EP (ages 16-35 years) diagnosed with schizophrenia, schizoaffective disorder, or a

Mediating Role of Self-Efficacy in the Association Between Fatigue and Depressive Symptoms in Females with Rheumatoid Arthritis.

Background and Objectives: Extant research on the relationship between fatigue and depression in people with rheumatic diseases portrays a divergent picture. While caring for persons with this medical condition, one issue that represents individual confidence in carrying out specific tasks, namely self-efficacy level, has attracted significant attention. Yet, the information regarding whether self-efficacy may pose a clue linking these two major symptoms is still unknown. The aim of this study, therefore, is to examine whether self-efficacy mediates the association between fatigue and depressi

Bud, branch, breathe! Building a mammalian lung over space and time.

Many mammalian organs, such as the mammary and lachrymal glands, kidney and lungs develop by the process known as branching morphogenesis. An essential feature of this process is the reciprocal interaction between the inner branched tubular epithelium and the surrounding mesenchyme to optimize the final amount of epithelial tissue that is generated for specific functions. To achieve this expansion the initial epithelial population undergoes repeated rounds of bud formation, branch outgrowth and tip bifurcations, with each repertoire requiring dynamic changes in cell behavior. The process of br

Myopathology and Immune Profile of Granulomatous Myositis in Sarcoid Myopathy.

AIMS: Sarcoid myopathy (SaM) is characterised by granulomatous myositis (GM) and can overlap with inclusion body myositis (IBM), a late-onset chronic idiopathic inflammatory myopathy with a still enigmatic pathogenesis. As GM can occur in different clinical contexts, we aimed to examine the histomorphologic features and gene expression profiles in cases of definite SaM that may inform diagnostic and therapeutic considerations. METHODS: We performed a multidimensional characterisation of muscle biopsy specimens from patients with 'pure SaM' (n=17), SaM with concomitant IBM (SaM-IBM) (n=2), inc

Hippocampus diffusivity abnormalities in classical trigeminal neuralgia.

INTRODUCTION: Patients with chronic pain frequently report cognitive symptoms that affect memory and attention, which are functions attributed to the hippocampus. Trigeminal neuralgia (TN) is a chronic neuropathic pain disorder characterized by paroxysmal attacks of unilateral orofacial pain. Given the stereotypical nature of TN pain and lack of negative symptoms including sensory loss, TN provides a unique model to investigate the hippocampal implications of chronic pain. Recent evidence demonstrated that TN is associated with macrostructural hippocampal abnormalities indicated by reduced sub
