Visceral Obesity and Its Complications: The Role of Bioelectrical Impedance Analysis in Longevity Medicine.

Background: Visceral obesity is increasingly recognised not as a simple excess of adipose tissue, but as a systemic pathological condition characterised by profound metabolic, endocrine, and immune dysregulation. Visceral adipose tissue (VAT) operates as an autonomous neuro-immune-endocrine organ whose dysfunctional expansion drives insulin resistance, atherogenesis, and accelerated cellular ageing through mechanisms converging on chronic low-grade sterile inflammation, referred to as inflammaging. Objectives: This narrative review integrates evidence across four domains: (1) the multi-system

Targeting the Jun-Irf8-CD36 axis attenuates fibrotic scar formation and promotes functional recovery after spinal cord injury.

BACKGROUND: Fibrotic scar formation constitutes a significant pathological obstacle that impedes neural regeneration and long-term functional recovery following spinal cord injury (SCI). However, the spatial distribution of key pro-fibrotic mediators within lesion scars and the upstream regulatory mechanisms driving fibroblast activation remain inadequately defined. This study aims to characterize CD36-associated fibrotic remodeling after SCI and to determine whether targeting the c-Jun-Irf8-CD36 axis could attenuate scar formation, improve the regenerative microenvironment, and promote functi

Perimenopause as an obesogenic sensitive period: Contributions to elevated cardiovascular risk.

BACKGROUND: Perimenopause, encompassing the period of progressive menstrual irregularity preceding a woman's final menstrual cycle and extending 12 months thereafter, is associated with an acceleration in atherosclerotic cardiovascular disease (CVD) risk, attributable in part to redistribution of body fat. During perimenopause, even in the context of minimal-to-modest weight gain, women experience an expansion of visceral adipose tissue. Concomitantly, women often experience a reduction in gluteofemoral subcutaneous adipose tissue. This pattern of fat redistribution is associated with a greate

Bone Marrow Aspirate Concentrate With Two-Staged-Revision ACL Reconstruction.

BACKGROUND: The incidence of anterior cruciate ligament (ACL) injuries, reconstructions, and re-ruptures has rapidly increased. Patients with failed ACL reconstructions have been reported to suffer from far worse outcomes as compared with those with primary reconstructions, prompting the advancement of surgical and biologic techniques. Effective treatment of re-tears has been shown to be achieved either utilizing a 1-stage or 2-staged approach, with the latter preferred if the patient presents with significant bone loss, previously malpositioned tunnels, or unacceptable tunnel expansion. A telomere-to-telomere genome reveals arachidonic acid as a key regulator of hepatic lipid metabolism during hibernation.

Elucidating the molecular mechanisms governing mammalian hibernation is crucial for understanding energy homeostasis and physiological adaptation to extreme environments. In this study, we present the first high-quality telomere-to-telomere (T2T) reference genome (2.69 Gb) of the Daurian ground squirrel (Spermophilus dauricus), a typical hibernating mammal. The assembled genome comprises 19 chromosomes, nine of which are completely gapless chromosomes, and exhibits a scaffold N50 of 160 Mb. Comparative genomic analysis with 17 hibernating and non-hibernating mammals revealed lineage-specific e

Feeder-free generation of functional dendritic cells from human pluripotent stem cells.

BACKGROUND: The scarcity of primary conventional dendritic cells (cDCs) and the limited functionality of monocyte-derived dendritic cells (moDCs) have long hindered mechanistic and translational studies in human dendritic cell (DC) biology and immunotherapy. METHODS: We developed a feeder-free differentiation platform to generate CD1c+CD141+ human pluripotent stem cell-derived conventional dendritic cells (hPSC-cDCs) to provide a scalable source of DCs with defined properties. A Design-of-Experiments (DoE) optimization strategy was applied to refine cytokine and serum conditions, with the goa

Single-cell profiling reveals MIF-mediated immune evasion and CD8 + T cell exhaustion in relapsed multiple myeloma.

Multiple myeloma (MM) is a heterogeneous hematologic cancer marked by clonal plasma cell expansion in the bone marrow. Although treatments have improved, relapse remains common due to clonal evolution, tumor microenvironment changes, ultimately leading to drug resistance. This study utilized single-cell RNA sequencing data analysis to explore immune microenvironment dynamics during MM progression and relapse, aiming to uncover key pathways and cellular interactions correlated with disease relapse. ScRNA-seq data from 20 MM patients (10 primary, 2 remission, 8 relapsed) from the GEO dataset GSE

HEATR1 deficiency enhanced gastric cancer cell PANoptosis to boost anti-tumor immunity.

BACKGROUND: Despite the promise of immune checkpoint inhibitors (ICIs) for gastric cancer (GC) treatment, limited cytotoxic T cell infiltration remains a major barrier to therapeutic efficacy. This study investigates whether targeting HEAT repeat containing 1 (HEATR1) can induce immunogenic PANoptosis to enhance anti-tumor immunity. METHODS: Bioinformatics analysis of TCGA-STAD cohort, syngeneic mouse models, and patient-derived organoids (PDOs) were employed to assess HEATR1 function. Mechanistic studies examined STAT1/IRF1 signaling, PANoptosis markers, and dendritic cell (DC) activation. T

The global outbreak of Oropouche virus: surveillance of global trends.

Oropouche virus (OROV) is a re-emerging arthropod-borne virus of the genus Orthobunyavirus (family Peribunyaviridae) that poses an increasing public health concern in South and Central America and the Caribbean. First identified in Trinidad in 1955, OROV has caused recurrent outbreaks, with recent resurgence associated with geographic expansion and increased urban transmission. In 2024 alone, more than 11,600 confirmed cases were reported in the Region of the Americas, with continued transmission documented in early 2025. The virus is primarily transmitted in urban settings by the biting midge

Lymphoid Organs Remodeling in Non-Obese Diabetic Goto-Kakizaki Rats Immunized With SARS-CoV-2 Antigens.

The COVID-19 pandemic revealed reduced vaccine efficacy and worse outcomes in people with type 2 diabetes mellitus (T2DM). However, how T2DM, independent of obesity, alters immune responses and lymphoid organ architecture remains unclear. We evaluated antibody production and lymphoid organ morphology following SARS-CoV-2 antigen immunization in Goto-Kakizaki (GK) rats, a non-obese T2DM model, to evaluate the isolated effect of diabetes. Ten-week-old male Wistar and GK rats received three intramuscular injections of SARS-CoV-2 antigens at 15-day intervals. Two antigen formulations were used: ch

Establishment and functional analysis of an inducible Apoc2-knockout mouse model to investigate the role of Apoc2 in normal hematopoiesis.

Apolipoprotein C2 (APOC2) has been identified as a potential therapeutic target for acute myeloid leukemia, but its impact on normal hematopoiesis remains unclear. Here we generated a global inducible Apoc2 knockout (KO) mouse model by breeding R26-CreERT2 mice with mice homozygous for Apoc2 lox sites (Apoc2fl/fl) over three generations. Global Apoc2 KO was induced in 4-8-week-old mice by 5 consecutive days of intraperitoneal administration of tamoxifen. Successful Apoc2 KO was confirmed by measuring Apoc2 mRNA levels, showing 80-99% reduction in the liver, bone marrow, spleen and blood tissue

Animal Age-Dependent Susceptibility of Mouse Oocytes to Zearalenone-Induced Developmental Impairment: Roles of Pharmacokinetic Exposure and Intrinsic Oocyte Sensitivity.

Background: Zearalenone (ZEN) exposure poses health risks to both humans and animals. Although evidence indicates that ZEN significantly impairs oocyte developmental competence, the underlying mechanisms remain largely unclear. While it is recognized that prepubertal animals are particularly vulnerable to ZEN and that juvenile in vitro embryo transfer represents a promising strategy for accelerating genetic progress, the potential impact of ZEN on developmental competence of prepubertal oocytes remains uninvestigated. Methods: In vivo, we administered graded oral doses of ZEN to female mice at

Semaglutide alleviates age-related dry eye disease by restoring lacrimal gland structure and function.

The prevalence of age-related dry eye disease (DED) is rapidly increasing with global aging, and progressive lacrimal gland (LG) dysfunction represents a central pathogenic mechanism for which targeted therapies remain limited. Semaglutide (Sema), a glucagon-like peptide-1 receptor agonist widely used in metabolic diseases, has recently shown anti-aging effects in multiple tissues, yet its effects on LG aging and age-related DED have not been defined. Here, using a naturally aged mouse model, we show that long-term Sema treatment significantly alleviates DED-related phenotypes and preserves LG

Construction of biomimetic hydroxyapatite bilayer scaffolds via DLP 3D printing for bio-root regeneration.

Digital light processing (DLP) 3D printing of hydroxyapatite (HA) bioceramics offers a promising strategy for fabricating complex, patient-specific bio-root scaffolds with high resolution and bioactivity. However, achieving adequate mechanical integrity while guiding the formation of a periodontal ligament (PDL)-like enthesis through structural optimisation remains a major challenge. Inspired by interfacial micromorphological cues that regulate both intrinsic mechanical properties and cellular responses, this study proposed a novel bone-root integrated design for a DLP 3D-printed bio-root scaf

Genome-wide characterization and cold-responsive expression profile of Rab GTPases and their regulatory proteins in O. rufipogen.

The DXWR genome was found to contain 44 OrRabs. Most of the cold-activated OrRab genes belong to the RabA/Rab11 group, with OrRab11B3 the most strongly induced gene in DXWR. Dongxiang wild rice (Oryza rufipogon Griff., DXWR), the wild rice species distributed at the highest latitude, exhibits significantly greater cold tolerance than cultivated rice, making it a valuable genetic resource for enhancing cold resistance in domesticated varieties. While previous studies in Arabidopsis thaliana have shown that members of the Rab gene family-encoding small GTP-binding proteins-are regulated by cold

Host-viral interaction of HBV infection revealed by single-cell transcriptome jointly profiling the viral replication state.

BACKGROUND AND AIMS: Hepatitis B virus (HBV) infection exhibits marked cellular heterogeneity, which conventional poly(A)-based single-cell RNA sequencing fails to resolve this heterogeneity owing to the overwhelming host transcript background. To overcome this, we developed B-BEST (HBV can BE Seen on host Transcriptome), a targeted sc/snRNA-seq approach using custom beads conjugated with HBV-specific probes to simultaneously quantify 5 viral genomic regions (S, X, pgRNA, rcDNA, cccDNA). APPROACH AND RESULTS: We validated B-BEST in HepAD38 cells and integrated it with long-read sequencing, sp

Chronic Intermittent Hypoxia Induces Hippocampal Cellular Remodeling through microglial polarization imbalance and neural functional disconnection.

BACKGROUND: Cognitive impairment in Obstructive Sleep Apnea-Hypopnea Syndrome (OSAHS) is primarily driven by chronic intermittent hypoxia (CIH), yet the systemic, dynamic responses of hippocampal cell types to CIH are unclear. METHODS: Using single-cell RNA sequencing (scRNA-seq), we analyzed hippocampal tissues from mice under normoxia or after 2 or 6 weeks of CIH. Integrated analyses assessed cellular composition, pseudotemporal trajectories, intercellular communication, and cell-type-specific responses. RESULTS: CIH induced time-dependent hippocampal reorganization, characterized by a bip

Wnt Stimulation and Inhibition in the Development and Phenotype of Patient-Derived Gallbladder Organoids.

BACKGROUND: Gallbladder cancer (GBC) is a highly lethal malignancy with limited experimental models to study disease biology or evaluate therapeutic responses. Although canonical Wnt activation is commonly used for patient-derived organoid (PDO) development and expansion, gallbladder PDOs have also been generated under Wnt-inhibitory conditions. No comparative assessments have determined how Wnt pathway modulation influences gallbladder PDO development, phenotype, or drug response. METHODS: This study systematically compared the impact of canonical Wnt activation (WNTAct medium containing CHI

Brain-derived neurotrophic factor stimulates hypothalamic and gonadal reproductive hormones and oocyte maturation in zebrafish.

IN BRIEF: The brain-derived neurotrophic factor (BDNF) has been shown to regulate metabolism in zebrafish. This research shows that Bdnf is a regulator of female zebrafish reproduction and reproductive hormones. ABSTRACT: BDNF is a peptide widely known for its role in neurogenesis and synaptic plasticity. Its expression in non-neuronal tissues has been reported. In mammals, it is involved in ovarian development, follicle growth, oocyte maturation and early embryonic development. In zebrafish, it was demonstrated that Bdnf increases food intake and regulates metabolism. Reproduction and metabo

Human Pancreatic Cancer Single-Cell Atlas Reveals Association of CXCL10+ Fibroblasts and Basal Subtype Tumor Cells.

PURPOSE: Pancreatic ductal adenocarcinoma (PDAC) patients with tumors enriched for the basal-like molecular subtype exhibit enhanced resistance to standard-of-care treatments and have significantly worse overall survival compared with patients with classic subtype-enriched tumors. It is important to develop genomic resources, enabling identification of novel putative targets in a statistically rigorous manner. EXPERIMENTAL DESIGN: We compiled a single-cell RNA sequencing (scRNA-seq) atlas of the human pancreas with 229 patient samples aggregated from publicly available raw data. We mapped cel
