[The mode of action of bisacodyl on the smooth muscle of the small and the large intestine of the guinea pig].

4,4'-Diacetoxy-diphenyl-(pyridyl-2)-methan (Bisacodyl, Dulcolax, 12--240 mug/ml) was shown to initiate dose-dependently contracile responses in the guinea pig isolated terminal ileum and taenia coli (a preparation of pure longitudinal muscle fibers). In contrast to muscle contractions induced by 1 mug/ml acetylcholine (ACh) and 1 mug/ml histamine, respectively bis-acodyl-induced contractile responses were antagonized neither by the anticholinergic agent atropine (6 mug/ml) nor by the antihistaminic compound pheniramine (6 mug/ml). On the other hand, bisacodyl inhibited contractile responses in

2026 update on the COVADIS diagnostic criteria of international standardization for microvascular angina.

Microvascular angina, in the context of ischemia with non-obstructed coronary arteries (INOCA), is a highly prevalent clinical condition in cardiovascular medicine. Although it remains frequently underdiagnosed, clinical research and practice have made substantial progress in the last 10 years. This includes better characterisation of affected patient cohorts, new data on the prevalence of coronary vasomotor disorders and improved diagnostic assessments (non-invasive and invasive). The aim of this paper is to update our previous 2018 recommendations for the standardization of diagnostic criter

Structural and mechanistic insights into OCTN1-mediated transport and cancer cell proliferation.

The Novel Organic Cation Transporter 1 (OCTN1) facilitates the uptake of endogenous metabolites and therapeutic drugs across diverse tissues. Recognized as a key transporter for the antioxidant ergothioneine (ET), OCTN1 also mediates the cellular entry and release of various cationic compounds. Dysregulation of OCTN1 has been implicated in cancer progression and drug interaction. In non-neuronal systems, OCTN1-mediated acetylcholine (ACh) transport has been implicated in promoting cancer progression by enhancing tumor cell proliferation and invasion. Despite its physiological and clinical rele

Integrating synthesis, pharmacological evaluation, and molecular dynamics simulation of novel 8-substituted theophylline hybrids as potential PDE-4B inhibitors, bronchodilators and antibacterial.

Methylxanthines, particularly theophylline, have long served as effective bronchodilators for severe asthma. Emerging evidence links bacterial infection to asthma pathogenesis, motivating the search for multifunctional therapeutic agents. In this study, a series of 8-substituted 1,3-dimethylxanthines bearing aryl and heteroaryl groups were synthesized and structurally characterized by NMR, elemental analysis, and HR-ESI-MS. Most compounds exhibited significant bronchodilator activity in an acetylcholine-induced guinea pig model, surpassing theophylline. The compounds demonstrated potent in vit

Neurotransmitter regulatory networks in persistent postural-perceptual dizziness: potential mechanisms and therapeutic implications.

Persistent postural-perceptual dizziness (PPPD) is a chronic functional vestibular disorder characterized by persistent non-spinning dizziness, subjective unsteadiness, and symptom exacerbation induced by upright posture, movement, and complex visual motion. Abnormal vestibular sensory reweighting, increased visual dependence, postural hypervigilance, and comorbid anxiety are considered to contribute to the persistence of symptoms. Neurotransmitter systems, including serotonin, norepinephrine, dopamine, gamma-aminobutyric acid, glutamate, histamine, acetylcholine and neuropeptides, may partici

Perivascular adipose tissue from female rats fed a high-fat diet impaired mesenteric artery vasodilation.

High-fat diet (HFD) is a cardiovascular risk factor that may disproportionately affect women compared to men. HFD alters perivascular adipose tissue (PVAT), which surrounds arteries and regulates vascular function. This study investigated the sex-specific effects of HFD on PVAT-mediated vascular dysfunction. Male and female Sprague-Dawley rats were fed a normal chow diet (NCD) or HFD for 16 weeks. Thoracic aortic PVAT was isolated, and PVAT adipokine expression and inflammatory markers were measured. We then assessed PVAT-conditioned media effects on vasodilation, endothelial oxidative stress,

Aberrant myocardial calcium cycling: pathogenic mechanisms and targeted therapeutics for heart failure.

Cardiovascular diseases rank as the global primary cause of mortality. Heart failure (HF) represents a progressive cardiac syndrome triggered by structural or functional myocardial damage that compromises ventricular systolic output. Intracellular calcium homeostasis underpins cardiomyocyte excitation-contraction coupling. Calcium overload and dysregulated calcium handling act as central pathogenic forces driving HF progression. Sympathetic β1-adrenergic receptor (β1-AR) and parasympathetic M2 muscarinic acetylcholine receptor (M2-AChR) coordinate dual regulatory signaling to control cardiac c

Multicentric prognostic observational study on biomarker profile in immunotherapy-naïve patients with highly active generalized myasthenia gravis (PROGNO-MG): a study protocol.

BACKGROUND: Myasthenia gravis has a variable disease course. Early identification of patients with a highly active disease is crucial to prevent myasthenic exacerbations and crisis, necessitating more aggressive immunotherapy. However, no biomarkers currently exist to identify patients at risk for highly active disease course. OBJECTIVE: The PROGNO-MG study aims to identify blood-based biomarker signatures associated with a highly active disease course in immunotherapy-naïve patients with acetylcholine-receptor-antibody positive (AChR+) generalized Myasthenia gravis (gMG). METHODS: This is a

Anti-VGCC negative and Anti-SOX1 positive Lambert-Eaton myasthenic syndrome as the initial presentation of small cell lung cancer: a case report.

Lambert-Eaton myasthenic syndrome (LEMS) is a rare autoimmune disorder characterized by impaired acetylcholine release at the neuromuscular junction, leading to proximal muscle weakness, autonomic dysfunction, and diminished or absent tendon reflexes. Here, we present a rare case of a 73-year-old male smoker with small cell lung cancer (SCLC), whose initial manifestation was LEMS. Notably, the patient tested seronegative for anti-voltage-gated calcium channel (VGCC) antibodies but was seropositive for anti-SOX1 antibodies. The diagnosis of LEMS was supported by characteristic clinical symptoms

Botulinum neurotoxin: from molecular pathogenesis to emerging countermeasures.

Botulism is a severe neuroparalytic syndrome caused by botulinum neurotoxins (BoNTs)-among the most potent biological agents-with an estimated human lethal dose (LD50) of approximately 1 ng/kg. By cleaving soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) proteins at cholinergic nerve terminals, BoNTs block acetylcholine release, producing potentially fatal flaccid paralysis. Despite advances in supportive care, current therapeutic options remain limited to antitoxin administration, which is effective only against circulating toxin and cannot neutralize intracellula

Behavioral Manipulation of Ixodes scapularis by Ehrlichia muris eauclairensis: Implications for Tick-Borne Disease Transmission.

Tick-borne diseases pose significant risks to both animals and humans, with emerging pathogens like Ehrlichia muris eauclairensis (EME) underscoring the need for a deeper understanding of pathogen-vector interactions and tick fitness. This study investigates the impact of EME on Ixodes scapularis nymphs, revealing significant behavioral changes in EME-positive ticks. These ticks exhibited increased movement speed, faster bite site-seeking for attachment, and prolonged feeding durations compared to control ticks. Proteomic analyses of the tick synganglion during resting and feeding phases ident

Modeling amyotrophic lateral sclerosis with amniotic membrane-derived mesenchymal stem cells: A novel approach for disease modeling.

Advancement of therapeutics for neurodegenerative diseases like amyotrophic lateral sclerosis (ALS) has been predominantly hampered by the dearth of relevant disease models. Despite numerous animal models, significant challenges remain in correlating these with human disease complexities. In this study, the ALS model was created using amniotic membrane-derived mesenchymal stem cells (AM-MSCs) which were differentiated into motor neurons (MN) with specific MN induction media and transiently transfected with mutated human SOD1 G93A plasmid to induce ALS-like condition. Characterization included

Astrocyte-to-neuron H2O2 signalling supports long-term memory formation in Drosophila and is impaired in an Alzheimer's disease model.

Astrocytes help protect neurons from potential damage caused by reactive oxygen species (ROS). While ROS can also exert beneficial effects, it remains unknown how neuronal ROS signalling is activated during memory formation, and whether astrocytes play a role in this process. Here we discover an astrocyte-to-neuron H2O2 signalling cascade in Drosophila that is essential for long-term memory formation. Stimulation of astrocytes by acetylcholine induces an increase in intracellular calcium ions, which triggers the generation of extracellular superoxide (O2•-) by astrocytic NADPH oxidase. Astrocy

The neuro-immune axis in cancer: from mechanisms to therapeutic opportunities.

In recent years, neural mechanisms involved in tumor initiation, progression, and immune regulation have garnered growing attention, fueling the rapid advancement of the burgeoning interdisciplinary domain of neuro-oncology. As a central component of physiological homeostasis, the neurophysiological system also exerts significant influence on the development and dynamics of the tumor microenvironment via complex neuro-immune signaling. Studies have shown that neurotransmitters, neuropeptides, and their mediated signaling pathways can directly or indirectly affect the functional state of tumor-

Seasonal variation in myasthenia gravis incidence.

INTRODUCTION: Environmental factors may contribute to myasthenia gravis (MG) development, sometimes with seasonal patterns of exposure. However, whether seasonality has an impact on MG incidence remains unclear. We aimed to investigate the association between seasonality and MG onset. METHODS: We reviewed data of MG patients with acetylcholine receptor (AChR)-IgG and disease onset between January 2010-December 2019, from two Italian cohorts: 1) an hospital-based cohort and 2) a population-based cohort. MG cases were assigned to four season-trimesters based on month of onset to determine seaso

Association between peripheral and coronary microvascular function and the impact of myocardial bridging.

The reactive hyperemia index (RHI) is a marker of peripheral microvascular function influenced by both endothelium-dependent and independent mechanisms. Myocardial bridging (MB) may impact coronary microvascular function, but its effect on the relationship between RHI and coronary microvascular function remains unclear. In this cross-sectional study, 38 patients underwent noninvasive RHI assessment alongside invasive coronary microvascular function testing. MB was identified via coronary angiography, while endothelium-dependent and independent coronary microvascular function were evaluated usi
