Fetal, infant, and child mortality in a context of alcohol use.

Alcohol is an important element in the causal chain of risk for fetal, infant, and childhood mortality. Mortality risk is influenced by interactions of alcohol with other environmental and genetic factors and temporal periods of susceptibility. In this paper we discuss four time periods (preconceptual, gestational, infancy, and childhood) where alcohol use may create a context of risk. Alcohol use in one period increases the risk of alcohol use in subsequent periods. Gestational alcohol use can influence risk of mortality from abuse of mother/fetus, can cause other adverse outcomes, and can re

Effects of acamprosate on ethanol-seeking and self-administration in the rat.

BACKGROUND: Acamprosate (calcium acetyl homotaurinate) has been used clinically to treat relapse in alcoholics. In rats, it has been shown to decrease ethanol, but not water, self-administration after ethanol deprivation. METHODS: To further investigate the effect of acamprosate on reinforced behaviors in rats, the present experiment used: (1) both ethanol and sucrose reinforcer solutions to better assess the distinct effects of acamprosate on ethanol-directed behaviors, and (2) an operant model that procedurally separates the "cost" to begin drinking from consuming the reinforcer solutions t

Alcohol and cancer.

A great number of epidemiological data have identified chronic alcohol consumption as a significant risk factor for upper alimentary tract cancer, including cancer of the oropharynx, larynx, and the esophagus, and for the liver. In contrast to those organs, the risk by which alcohol consumption increases cancer in the large intestine and in the breast is much smaller. However, although the risk is lower, carcinogenesis can be enhanced with relatively low daily doses of ethanol. Considering the high prevalence of these tumors, even a small increase in cancer risk is of great importance, especia

Myoclonic alcoholic epilepsy.

We describe 2 patients with chronic alcoholism who over a period of several years developed epilepsy associated with a cerebellar syndrome and action myoclonus. The clinical picture suggested progressive myoclonic epilepsy, but subsequent investigations did not confirm this diagnosis. Myoclonus worsened during abstinence and was relieved during indulgence. We believe that this special clinical syndrome should be recognized as a complication of chronic alcoholism.

Effects of liver disease on red blood cell acetaldehyde in alcoholics and non-alcoholics.

High endogenous levels of acetaldehyde were found in the blood (mainly in red blood cells) of patients with liver disease of various etiologies. The presence of severe liver injury increased the blood acetaldehyde response to ethanol consumption and resulted in a more prolonged persistence of high levels after ethanol withdrawal.

[Personality characteristics in students at risk of alcoholism].

The aim of the study was to establish personality traits differentiating between persons endangered with alcohol habituation and those that did not abuse alcohol. With Engs rating scale 225 men and 73 women in danger of alcohol habituation were found among a group of 1255 students. Significant differences in the adaptation level of the affected group--as compared to the unaffected group--were found using MMPI. The differences concerned mainly the lack of emotional stability, poor self-control, high level of hostility and low tolerance to frustration.

Selective suppression of schedule-induced ethanol drinking by antialcoholic drugs in rats.

Effects of disulfiram and calcium cyanamide, antialcoholic drugs, on schedule-induced ethanol drinking as well as on schedule-controlled response (lever-pressing) under a fixed interval 1 min schedule of food reinforcement were investigated in Wistar strain rats. When ethanol solution was available, the schedule-induced ethanol drinking decreased depending on the ethanol concentration (2-8%). However, the dose of ethanol intake during the 1 hr experimental session was at maximum (2.8 g/kg) when 4% ethanol solution was available. Thereafter, 4% ethanol solution was used in the experiment for st

A single dose of ethanol suppresses rat embryo development in vivo.

In humans and animal models, maternal ethanol consumption during pregnancy results in a variety of fetal defects collectively termed the fetal alcohol syndrome (FAS). Limited follow-up studies suggest that FAS children fail to achieve normal physical or mental development despite significant postnatal intervention. Although the complete FAS appears to result only when chronic, excessive alcohol consumption occurs throughout pregnancy, several investigators have suggested that ethanol consumption at intermediate levels may induce components of FAS. The defect most consistently observed in neona

Production of physical dependence on ethanol by a short drinking episode each day.

A 3-hr schedule-induced ethanol polydipsia regimen was used in rats to elevate blood alcohol concentration to a single intoxicating peak each day. After 3 weeks, and again after 3.5 months, animals were tested for the presence of physical dependence by exposure to a brief auditory stimulus (key shaking) at 7 and 11 hr after ethanol polydipsia. Withdrawal signs were observed only at 11 hr when blood ethanol levels had returned to zero. No such signs were observed when animals were made water polydipsic. While sufficient, continuous elevation of blood ethanol concentration is not necessary for t

Alcohol intake and ethanol intoxication in the rate: effect of a 6-OHDA-induced lesion of the ascending noradrenaline pathways.

The ascending noradrenaline (NA) pathways were lesioned by injecting 6-hydroxydopamine (6-OHDA) 16 micrograms/4 microliters bilaterally into the posterior mesencephalon in male Long Evans rats. Another group of rats was pretreated with protriptyline (25 mg/kg), a NA uptake blocking agent, 15 min before they received the intracerebral injections of 6-OHDA. The controls received the vehicle only. Spectrofluorimetric determination of the catecholamine concentrations in various parts of the brain revealed a marked degeneration of the ascending NA systems in the group receiving 6-OHDA. Unexpectedly

Alcohol dependence and phobias: clinical description and relevance.

Of 102 alcoholics admitted to an alcoholism treatment unit one third (33) were clinically rated as having a disabling agoraphobia and/or social phobia, and a further third (37) as having less disabling phobic symptoms of either or both kinds. Questionnaires and self-report symptom scales were used to validate the clinical ratings. Alcohol problem screening tests and consumtpion levels confirmed the severity of alcohol dependence. In a group of 44 phobic alcoholics who reports and physical dependence significantly more frequently than the converse. The prevalence of agoraphobia and social phobi

[Effect of central effective substances on alcohol preference].

The alcohol preference has been used as a model of drug habituation in animal experiments in order to examine the influence of altered synaptic efficiency of central transmission systems following acute receptor reactions with agonists or antagonists as well as by alterations of receptor sensitivity in mice. Ethanol in 10 per cent solution as the only beverage over 4 weeks produces an ethanol preference quotient 1 in the free choice of ethanol solution and water. Control animals without ethanol exposure before the preference test are only drinking water and refuse ethanol. A single injection o

Survival Outcomes in Young Nonsmoking-Nondrinking Individuals With Oral Cavity Squamous Cell Carcinoma.

OBJECTIVE: To determine the impact of age and presence of traditional risk factors (smoking and drinking) on survival outcomes in a large cohort of patients with oral cavity squamous cell carcinoma (OCSCC). METHODS: This was a single-center, retrospective review from 2012 to 2019. Patients were divided by age (≤ 40 vs. > 40 years old) and whether they had significant tobacco and/or alcohol use history. Univariate and multivariate analyses were conducted to compare overall and disease-specific survival between the four groups. RESULTS: There were 630 patients, and median follow-up was 52.5 mo
