An Observational Case-Control Study of the Anti-Inflammatory Effects of Bromelain on Peripheral Blood Mononuclear Cells from Patients with Systemic Lupus Erythematosus.

BACKGROUND: Bromelain is a proteolytic enzyme that has been investigated as an adjuvant for treating autoimmune disorders. However, data regarding the role of bromelain in systemic lupus erythematosus is limited. In the present study, we examined the effect of bromelain on the mononuclear cells of SLE patients, precisely its anti-inflammatory properties. MATERIALS AND METHODS: Peripheral blood mononuclear cells (PBMCs) from SLE patients and healthy controls were stimulated with bromelain (10μg/ml), lipopolysaccharide (10μg/ml), or without any stimulant and cultured for 12 hours. The supernata

Safety and efficacy of low-dose, cord blood-derived CD19 CAR-NK cells with 4-1BB co-stimulation in refractory systemic lupus erythematosus.

BACKGROUND: B cell-targeting chimeric antigen receptor (CAR) T cell therapy has shown efficacy in autoimmune diseases but is limited by toxicity, complex manufacturing, and high cost. Umbilical cord blood (UCB)-derived CAR-natural killer (CAR-NK) cells offer an alternative "off-the-shelf" platform with a potentially superior safety profile. We developed a UCB-derived CD19-targeting CAR-NK product incorporating the 4-1BB costimulatory domain (CD19-BBz) and evaluated its safety and efficacy in refractory systemic lupus erythematosus (SLE). This study was registered at ClinicalTrials.gov (Clinica

Case Report: Equine allogeneic umbilical cord blood mesenchymal stromal cells (CB-MSC) as adjunctive therapy in a foal with septic arthritis and osteomyelitis.

Mesenchymal stromal cells (MSCs) are recognized for their potent anti-inflammatory, antibacterial, and immunomodulatory properties, making them a promising therapeutic option for combating antibiotic resistance and biofilm-associated infections. This report describes the successful treatment of septic arthritis and osteomyelitis in a foal using equine allogeneic cord blood-derived MSCs (CB-MSCs) in combination with antibiotic therapy. An 8-day-old Thoroughbred filly initially presented with septic arthritis of the right tibiotarsal joint, pneumonia, and omphalophlebitis/arteritis. Subsequently

Peripheral Immune Reprogramming Characterizes a Low C-Peptide Subgroup of Type 2 Diabetes: Transcriptomic Profiling of Peripheral Blood Mononuclear Cells and Systemic Inflammation.

BACKGROUND: Type 2 diabetes (T2DM) is clinically heterogeneous. A subgroup with markedly reduced C-peptide has poorer glycemic stability and different therapeutic needs, but the associated peripheral immune features and practical non-invasive markers are not well defined. We investigated whether T2DM with low C-peptide (T2DM-LowC) is associated with a distinct peripheral immune profile. METHODS: In this single-center, cross-sectional study, patients with T2DM were propensity score-matched 1:1 by disease duration into low C-peptide (T2DM-LowC, n=109) and preserved C-peptide (T2DM-PresC, n=109)

Setmelanotide alleviates diabetes-associated cognitive impairment by interrupting the binding of ICAM-1 and JIP1 in endothelial cells of the blood-brain barrier.

Diabetes-associated cognitive impairment (DCI) is highly prevalent worldwide, but the mechanisms underlying this disorder remain unclear. Here we show that disruption of the blood-brain barrier (BBB) under hyperglycemic conditions is critical to the initiation and perpetuation of cognitive dysfunction. Proteomic analyses of brains from diabetic patients and non-diabetic patients revealed a substantial increase in the expression of intercellular adhesion molecule 1 (ICAM-1), which was confirmed with immunohistochemistry and immunoblotting. This upregulation was linked to cognitive deficits in p

Detectability of hepatitis B virus DNA in peripheral blood mononuclear cells following long-term nucleos(t)ide analogue therapy-induced sustained viral suppression.

OBJECTIVE: Hepatitis B virus (HBV) can persist in peripheral blood mononuclear cells (PBMCs) as an extrahepatic reservoir in treatment-naive patients; however, its persistence during long-term nucleos(t)ide analogue therapy with sustained serum viral suppression remains unclear. This study aimed to evaluate the frequency and predictors of HBV DNA detectability in PBMCs of chronic hepatitis B (CHB) patients following long-term nucleos(t)ide analogue therapy-induced sustained serum viral suppression. METHODS: Eighty-eight CHB patients, on long-term tenofovir disoproxil fumarate ( n = 44) or ent

Real-world comparison of COBE2991 and LOVO Med devices for the post-thaw washing of peripheral blood stem cells and donor lymphocyte infusions.

Post-thaw washing of cryopreserved hematopoietic stem cell products is essential to reduce dimethyl sulfoxide concentration and toxicity. This procedure is routinely performed in French cell therapy units. Following the discontinuation of the COBE2991 device, alternative technologies such as the LOVO Med automated closed system have been evaluated. We retrospectively compared the COBE2991 and LOVO Med for washing peripheral blood stem cells (PBSC) products and donor lymphocyte infusions (DLIs). A total of 946 products processed between September 2021 and October 2023 were included. The paramet

Long-Term Transcriptomic Reprogramming in Peripheral Blood Mononuclear Cells of Severe COVID-19 Survivors Reveals Pro-Oncogenic Signatures and Cancer-Associated Hub Genes.

This study examined the long-term transcriptomic reprogramming in peripheral blood mononuclear cells (PBMCs) of severe COVID-19 patients and its effects for cancer development. RNA sequencing was conducted on PBMCs obtained from healthy controls, COVID-19 patients without pneumonia, and COVID-19 patients exhibiting severe pneumonia one year post-infection. Differential gene expression analysis identified a sustained pro-oncogenic molecular signature, especially among severe COVID-19 patients. Functional enrichment analysis revealed a substantial enrichment of cancer-associated pathways, encomp

The Effect of Intrauterine Autologous Peripheral Blood Mononuclear Cells (PBMCs) Administration in Modulating the Immunologic Profile Aberrations in Repeated Implantation Failure (RIF) Women.

PURPOSE: Considering the immune-related etiology of RIF, administration of an immunomodulatory approach such as autologous PBMCs seems to be helpful in modifying the dysregulated immune responses. METHODS: 100 RIF women were divided into the PBMC receiving and the control group. Blood sampling was conducted 48 h before and 2 weeks after PBMC administration. The frequency of Th17, Treg, and NK cells, the expression level of related transcription factors and miRNAs, and the concentration of inflammatory and anti-inflammatory cytokines were evaluated pre- and post-treatment. Hospital Coordination and Protocols Using Serum and Peripheral Blood Cells from Patients and Healthy Donors in a Longitudinal Study of Guillain-Barré Syndrome.

Background/Objectives: Guillain-Barré syndrome (GBS) is a rare autoimmune peripheral neuropathy that affects both the myelin sheaths and axons of the peripheral nervous system. It is the leading cause of acute neuromuscular paralysis worldwide, with an annual incidence of less than two cases per 100,000 people. Although most patients recover, a small proportion do not regain mobility and even remain dependent on mechanical ventilation. In this study, we refer to the analysis of samples collected from GBS patients at different defined time points during hospital recovery and performed by a medi

NINJ1-mediated plasma membrane rupture of pyroptotic endothelial cells exacerbates blood-brain barrier destruction caused by neutrophil extracellular traps in traumatic brain injury.

Brain endothelial cell (bEC) dysfunction is the main factor of blood-brain barrier (BBB) breakdown, which triggers a vicious cycle of aggravating traumatic brain injury (TBI) pathogenesis. Previous studies have revealed that neutrophil extracellular traps (NETs) released by neutrophils can lead to BBB disruption, but there is a lack of research on the underlying mechanisms after TBI. Here, excessive NETs were found in both contused brain tissue and circulation following TBI. We found that NETs could activate the TLR4/NF-κB pathway to induce bEC pyroptosis, which led to BBB disruption after TBI

Oxidative stress, senescence and mRNA decay in peripheral blood mononuclear cells of women with gestational diabetes mellitus-link to increased cardiovascular disease risk.

AIMS: Women who develop gestational diabetes mellitus (GDM) have a pancreatic β-cell defect that cannot compensate for the insulin resistance of pregnancy. The glucose intolerance may, in addition to direct effects, negatively impact lipid metabolism, promote vascular dysfunction and contribute to cardiovascular disease (CVD). We assessed associations between oxidative stress and senescence markers with indices of glucose and lipid metabolism in women developing GDM and with CVD risk after 5 years. MATERIALS AND METHODS: The oxidative stress and senescence markers NRF2, GPX4, TERC, XRN1 and D

Pulsed electric field (PEF)-processed 6-shogaol-rich ginger extract protects β-Thalassemic red blood cells from iron-induced oxidative stress and hemolysis.

β- thalassemia is the genetic blood disorder characterized by ineffective beta-globin chain synthesis. The increased iron absorption and frequently blood transfusion result in iron overload and the production of reactive oxygen species (ROS). ROS causes lipid peroxidation and membrane damage in red blood cells (RBCs), culminating in hemolysis and anemia. This study investigated the protective effects of ginger extract against iron induced oxidative damage in thalassemic RBCs. The pulsed electric field (PEF) technique with high-voltage electric pulses, was used for ginger extraction. The PEF si…

Sustained antiviral response against in vitro HIV-1 infection in peripheral blood mononuclear cells from people with chronic myeloid leukemia treated with ponatinib.

HIV-1 infection cannot be cured due to long-lived viral reservoirs formed by latently infected CD4+ T cells. "Shock and Kill" strategy has been considered to eliminate the viral reservoir and achieve a functional cure but the stimulation of cytotoxic immunity is necessary. Ponatinib is a tyrosine kinase inhibitor (TKI) clinically used against chronic myeloid leukemia (CML) that has demonstrated to be effective against HIV-1 infection in vitro. Several TKIs may induce a potent cytotoxic response against cancer cells that makes possible to discontinue treatment in people with CML who present lon
