Cortistatin attenuates inflammatory pain via spinal and peripheral actions.

Clinical pain, as a consequence of inflammation or injury of peripheral organs (inflammatory pain) or nerve injury (neuropathic pain), represents a serious public health issue. Treatment of pain-related suffering requires knowledge of how pain signals are initially interpreted and subsequently transmitted and perpetuated. To limit duration and intensity of pain, inhibitory signals participate in pain perception. Cortistatin is a cyclic-neuropeptide that exerts potent inhibitory actions on cortical neurons and immune cells. Here, we found that cortistatin is a natural analgesic component of the

Acidosis dilates brain parenchymal arterioles by conversion of calcium waves to sparks to activate BK channels.

RATIONALE: Acidosis is a powerful vasodilator signal in the brain circulation. However, the mechanisms by which this response occurs are not well understood, particularly in the cerebral microcirculation. One important mechanism to dilate cerebral (pial) arteries is by activation of large-conductance, calcium-sensitive potassium (BK(Ca)) channels by local Ca(2+) signals (Ca(2+) sparks) through ryanodine receptors (RyRs). However, the role of this pathway in the brain microcirculation is not known. OBJECTIVE: The objectives of this study were to determine the mechanism by which acidosis dilate

Extracellular heat shock protein 70 induces cardiomyocyte inflammation and contractile dysfunction via TLR2.

BACKGROUND: Toll-like receptors (TLRs) are expressed on cardiomyocytes and recognize pathogen-associated molecular patterns. Whether endogenous molecules produced by tissue injury (damage associated molecular patterns, DAMPs) can induce cardiomyocyte inflammation via TLR signalling pathways and/or reduce cardiomyocyte contractility is unknown. METHODS AND RESULTS: Primary cardiomyocytes isolated from nuclear factor κ B (NFκB)-luciferase knock-in mice were used to assess NFκB signalling. DAMPs, HSP60, HSP70 and HMGB1, increased NFκB transcriptional activity compared to controls. HSP70 stood ou

Rosuvastatin inhibits norepinephrine-induced cardiac hypertrophy via suppression of Gh.

Statins have recently been shown to produce anti-cardiac hypertrophic effects via the regulation of small GTPases. However, the effects of statins on G protein-mediated cardiac hypertrophy, which is the main pathway of cardiac hypertrophy, have not yet been studied. We sought to evaluate whether statin treatment directly suppresses cardiac hypertrophy through a large G protein-coupled pathway regardless of the regulation of small GTPases. Using neonatal rat cardiomyocytes, we evaluated norepinephrine-induced cardiac hypertrophy for suppressibility of rosuvastatin and the pathways involved by a

Effects of short-term food deprivation on orexin-A-induced intestinal bicarbonate secretion in comparison with related secretagogues.

Studies of gastrointestinal physiology in humans and intact animals are usually conducted after overnight fast. We compared the effects of orexin-A, vasoactive intestinal polypeptide (VIP), melatonin, serotonin, uroguanylin, ghrelin and prostaglandin E(2) (PGE(2)) on duodenal bicarbonate secretion in fed and overnight fasted animals. This review is a summary of our findings. Secretagogues were administered by intra-arterial infusion or luminally (PGE(2)). Enterocyte intracellular calcium ([Ca(2+)](i)) signalling was studied by fluorescence imaging. Total RNA was extracted, reverse transcripted

The involvement of phosphatidylinositol 3-kinase in crystal induced human neutrophil activation.

OBJECTIVE: We investigated whether phosphatidylinositol (PI) 3-kinase is involved in the signal transduction pathway leading to neutrophil activation by inflammatory microcrystals. METHODS: Neutrophil chemiluminescence and degranulation responses to opsonized crystals were measured in the presence of selective inhibitors known to inhibit PI 3-kinase activity in neutrophils. RESULTS: Wortmannin and LY 294002, 2 selective inhibitors of PI 3-kinase, were shown to inhibit neutrophil activation induced by plasma opsonized crystals of calcium pyrophosphate dihydrate (CPPD) [both monoclinic (M) and

Nimodipine represses AMPK phosphorylation and excessive autophagy after chronic cerebral hypoperfusion in rats.

Chronic cerebral hypofusion (CCH) after bilateral carotid artery occlusion (2VO) causes cognitive damage and neuronal degeneration in the cortex and hippocampal CA1 area, and influences the oxygen and glucose supply in the brain which often results in metabolic alterations and oxidative stress. AMP-activated protein kinase (AMPK) phosphorylation, a sensor of cellular energy status, directs metabolic adaptation to support cellular growth and survival after CCH. Autophagy is also likely to be involved in metabolic adaptation and plays an important role in neuronal deterioration and cognitive dec

MR imaging of tophaceous gout.

OBJECTIVE: MR imaging is not routinely used for evaluation of tophaceous gout. However, gout may present clinically in an atypical, unusual, or confusing manner. A gouty tophus occasionally mimics an infectious or neoplastic process, and MR imaging may be obtained under these circumstances. The purpose of this study was to determine the MR imaging characteristics of intraosseous and soft-tissue tophi. MATERIALS AND METHODS: We identified 13 MR imaging examinations performed during a 27-month period on nine patients with gouty arthritis. All were men 42-70 years old. T1-, proton density-, and

Post-Translational Protein Deimination Signatures in Serum and Serum-Extracellular Vesicles of Bos taurus Reveal Immune, Anti-Pathogenic, Anti-Viral, Metabolic and Cancer-Related Pathways for Deimination.

The bovine immune system is known for its unusual traits relating to immunoglobulin and antiviral responses. Peptidylarginine deiminases (PADs) are phylogenetically conserved enzymes that cause post-translational deimination, contributing to protein moonlighting in health and disease. PADs also regulate extracellular vesicle (EV) release, forming a critical part of cellular communication. As PAD-mediated mechanisms in bovine immunology and physiology remain to be investigated, this study profiled deimination signatures in serum and serum-EVs in Bos taurus. Bos EVs were poly-dispersed in a 70-5

Diversity of potassium channels in neuronal dendrites.

Complex computations in the nervous system begin with electrical signals generated in single neurons. Such signals include action potentials mediated by the opening of voltage-dependent ion channels, and synaptic potentials arising from neurotransmitter receptor activation. The amplitude, waveform, and propagation of action potentials and synaptic potentials influence cellular signaling in profound ways, and are largely determined by activities of ion channels in the cell membrane. The location and properties of ion channels therefore play critical roles in shaping electrical signaling in the

PRKAR2A/RIIα as a candidate spatial cAMP-PKA signal router in triple-negative breast cancer metastasis: evidence boundaries, competing models and decisive experiments.

Metastatic relapse remains the principal lethal event in triple-negative breast cancer (TNBC), yet the contribution of PRKAR2A, which encodes the type IIα regulatory subunit (RIIα) of protein kinase A (PKA), remains unresolved. RIIα creates a mechanistic duality: it restrains catalytic PKA within the holoenzyme while recruiting type II PKA to A-kinase anchoring protein (AKAP) microdomains. A targeted critical narrative search identified no eligible published study in the searched sources that causally linked PRKAR2A to a TNBC metastasis endpoint. Direct evidence is limited to four association

An integrated workflow for long-term fiber photometry analysis.

Long-term fiber photometry enables measurement of neural dynamics across hours to days, but these recordings create analytical and reproducibility challenges that are not well addressed by tools developed for short, stimulus-locked experiments. Here we present a software environment for long-term photometry analysis organized around a structured guided workflow for configuring, reviewing, executing, and inspecting analyses. The workflow makes consequential analysis choices visible by allowing correction strategies and event-detection settings to be previewed and configured for individual regio

Heavy metal toxicity as a driver of endoplasmic reticulum stress and dysfunction.

Heavy metals like lead, cadmium, arsenic, mercury and chromium are persistent environmental contaminants recognised for their harmful effects on human health. Among their various toxicological effects, recent studies have identified endoplasmic reticulum (ER) as a critical target of HM-induced cellular damage. This review explores the contribution of HMs in disrupting ER homeostasis, with a particular focus on ER dysfunctions, ER stress mechanisms, calcium regulation, and ER-mitochondria crosstalk. ER stress occurs when its protein-folding capacity is exceeded, activating the unfolded protein

Piezo1 in digestive homeostasis and disease: Barrier function, immune signalling, cellular injury and metabolism.

BACKGROUND: Piezo1 is a mechanosensitive cation channel that converts stretch, compression, and shear stress into calcium-dependent signals. This narrative review evaluates how Piezo1 contributes to digestive physiology and disease and why divergent outcomes arise across experimental contexts. MAIN BODY: We searched PubMed/MEDLINE, Embase, Web of Science, and Scopus from database inception to 15 July 2026, prioritising primary studies that used Piezo1-specific genetic or pharmacological interventions. Piezo1-specific evidence supports roles in epithelial junctional adaptation, mucus productio

A PIEZO-Gated Mechanotransduction Timing Window Governs Scar Commitment in Wound Healing.

Cutaneous scarring remains a major clinical challenge, often resulting in impaired function, altered tissue mechanics, and poor aesthetic outcomes. Most current anti-fibrotic therapies target late-stage fibrosis, when extracellular matrix deposition and myofibroblast persistence are already established, limiting regenerative benefit. We propose that scar formation is governed not by gradual accumulation of fibrotic signals but by an early mechanically regulated commitment event, that precedes overt fibrosis. We introduce the concept of a PIEZO-gated mechanotransduction timing window, a finite

MicroRNA mediated regulation in early-onset cardiac hypertrophy: Insights from the hypertrophic heart rat model.

Cardiac hypertrophy is a pathological response to increased myocardial stress and a key contributor to heart failure. The Hypertrophic Heart Rat (HHR) is a well-established model for investigating primary cardiac hypertrophy in the absence of hypertension. This study aimed to characterise the role of microRNAs and their downstream regulatory pathways in neonatal HHRs to identify mechanisms contributing to early-onset hypertrophy. Heart tissue from 2-day-old HHRs and Normal Heart Rats (NHRs) was analysed using microarray profiling to identify differentially expressed miRNAs and mRNAs. Gene Set

Insight into the Neuroprotective Effect of Genistein-3'-Sodium Sulfonate Against Neonatal Hypoxic-Ischaemic Brain Injury in Rats by Bioinformatics.

Therapeutic hypothermia (TH) is the only intervention approved for the treatment of neonatal hypoxic-ischaemic encephalopathy (HIE), but its treatment window is narrow (within 6 h after birth), and its efficacy is not ideal. Thus, alternative treatments are urgently needed. Our previous studies showed that genistein-3'-sodium sulfonate (GSS), a derivative of genistein (Gen), has a strong neuroprotective effect in rats with ischaemic stroke, but its role in HIE is unclear. A hypoxia-ischaemia (HI) brain injury model was established in neonatal male Sprague‒Dawley (SD) rats. Twenty-four hours af

The Myrosinases TGG1 and TGG2 Function Redundantly in Reactive Carbonyl Species Signaling in Arabidopsis Guard Cells.

Myrosinase (β-thioglucoside glucohydrolase, enzyme nomenclature, EC 3.2.1.147, TGG) is a highly abundant protein in Arabidopsis guard cells, of which TGG1 and TGG2 function redundantly in abscisic acid (ABA)- and methyl jasmonate-induced stomatal closure. Reactive carbonyl species (RCS) are α,β-unsaturated aldehydes and ketones, which function downstream of reactive oxygen species (ROS) production in the ABA signalling pathway in guard cells. Among the RCS, acrolein is the most highly reactive, which is significantly produced in ABA-treated guard cells. To clarify the ABA signal pathway downst

TaCPK2-A, a calcium-dependent protein kinase gene that is required for wheat powdery mildew resistance enhances bacterial blight resistance in transgenic rice.

Calcium-dependent protein kinases (CPKs) are important Ca2+ signalling components involved in complex immune and stress signalling networks; but the knowledge of CPK gene functions in the hexaploid wheat is limited. Previously, TaCPK2 was shown to be inducible by powdery mildew (Blumeria graminis tritici, Bgt) infection in wheat. Here, its functions in disease resistance are characterized further. This study shows the presence of defence-response and cold-response cis-elements on the promoters of the A subgenome homoeologue (TaCPK2-A) and D subgenome homoeologue (TaCPK2-D), respectively. PROKR2 missense mutations associated with Kallmann syndrome impair receptor signalling activity.

Kallmann syndrome (KS) combines hypogonadism due to gonadotropin-releasing hormone deficiency, and anosmia or hyposmia, related to defective olfactory bulb morphogenesis. In a large series of KS patients, ten different missense mutations (p.R85C, p.R85H, p.R164Q, p.L173R, p.W178S, p.Q210R, p.R268C, p.P290S, p.M323I, p.V331M) have been identified in the gene encoding the G protein-coupled receptor prokineticin receptor-2 (PROKR2), most often in the heterozygous state. Many of these mutations were, however, also found in clinically unaffected individuals, thus raising the question of their actua
