Effects of oxidative stress and GDNF on patients with bipolar disorder: a prospective study.

BACKGROUND: Bipolar disorder (BD) is a common mental disorder characterized by significant cognitive dysfunction, the mechanisms of which remain unclear. Oxidative stress and glial cell line-derived neurotrophic factor (GDNF) influence the pathophysiology of BD. Their specific roles, particularly concerning cognitive function during manic episodes, are unclear. The serum levels of superoxide dismutase (SOD) and malondialdehyde (MDA), and GDNF were biochemically assayed in patients with bipolar mania before and after treatment to explore their associations with cognitive function. METHODS: A t

Effects of Berberine on Diabetes and Cognitive Impairment in an Animal Model: The Mechanisms of Action.

Diabetes is a group of metabolic disorders with an increased risk of developing cognitive impairment and dementia. The hippocampus in the forebrain contains an abundance of insulin receptors related to cognitive function and plays an important role in the pathophysiology of neurodegenerative disorders. Berberine from traditional Chinese medicine has been used to treat diabetes and diabetic cognitive impairment, although its related mechanisms are largely unknown. In this study, a STZ diabetes rat model feeding with a high-fat diet was used to test the effects of berberine compared with metform

Relationship between chemotherapy use and cognitive impairments in older women with breast cancer: findings from a large population-based cohort.

BACKGROUND: Several small scale clinical trials indicated a possible relationship between chemotherapy administration and the increased risk of cognitive impairments in patients with breast cancer, but little information was available from large population-based cohort studies. METHODS: We studied 62,565 women who were diagnosed with stages I-IV breast cancer at age ≥65 years from 1991 through 2002 from 16 regions in the Surveillance, Epidemiology and End Results program who were free of cognitive impairments at diagnosis with up to 16 years of follow-up, and also studied 9752 matched cohort

Chemogenetic Manipulation of mPFC Neurons Counteracts Cognitive Deficits But Leaves Sleep Disturbances Unaffected in a Mouse Model of Chronic Intermittent Hypoxia.

Chronic intermittent hypoxia (CIH), a hallmark of obstructive sleep apnea, gives rise to cognitive deficits and sleep disruption. To investigate the role of the medial prefrontal cortex (mPFC), male mice were subjected to CIH for 4 weeks (8 h/day). Brain-wide Fos B screening revealed specific hyperactivation in the infralimbic (IL) subregion of the mPFC. Following chemogenetic virus injection into the mPFC, cognitive function was assessed using novel object/location recognition and the Morris water maze, sleep architecture was recorded via EEG/EMG, and synaptic markers were examined by Western

The causal involvement of the BDNF-TrkB pathway in dentate gyrus in early-life stress-induced cognitive deficits in male mice.

Cognitive dysfunction is a significant, untreated clinical need in patients with psychiatric disorders, for which preclinical studies are needed to understand the underlying mechanisms and to identify potential therapeutic targets. Early-life stress (ELS) leads to long-lasting deficits of hippocampus-dependent learning and memory in adult mice, which may be associated with the hypofunction of the brain-derived neurotrophic factor (BDNF) and its high-affinity receptor, tropomyosin receptor kinase B (TrkB). In this study, we carried out eight experiments using male mice to examine the causal inv

Functional outcome assessment in bipolar disorder: A systematic literature review.

OBJECTIVES: Functional impairment is an important driver of disability in patients with bipolar disorder (BD) and can persist even when symptomatic remission has been achieved. The objectives of this systematic literature review were to identify studies that assessed functioning in patients with BD and describe the functional scales used and their implementation. METHODS: A systematic literature review of English-language articles published between 2000 and 2017 reporting peer-reviewed, original research related to functional assessment in patients with BD was conducted. RESULTS: A total of

Cognitive function in aging cocaine smokers.

BACKGROUND: Little is known about the functional status of older drug users, who may pose challenges to public health systems in coming years. Here, we assessed cognitive function in aging cocaine smokers compared to demographically matched controls. METHODS: A total of 22 non-treatment-seeking aging (50-60 years old) cocaine smokers (⩾twice/week; ⩾15 years of weekly use) and 19 controls completed a comprehensive cognitive battery. Controls with cannabis, tobacco, and alcohol use were included to better match the cocaine users. All cocaine users, and current cannabis- or alcohol-using control

A cognitive and affective pattern in posterior fossa strokes in children: a case series.

AIM: Posterior fossa strokes account for about 10% of ischaemic strokes in children. Although motor and dysautonomic symptoms are common, to our knowledge cognitive and affective deficits have not been described in the paediatric literature. Our aim, therefore, was to describe these symptoms and deficits. METHOD: In a retrospective study, we included all cases of posterior fossa strokes in children occurring at a single centre between 2005 and 2007, and investigated cognitive and affective deficits. RESULTS: Five males aged 3 to 14 years met the inclusion criteria. They all presented very ea

Can the pattern of neuropsychological improvement obtained with cholinergic drugs be used to infer a cholinergic mechanism in other nootropic drugs?

1. Enhancement of episodic memory and reduction of intrusion errors are considered as the most characteristic outcome of cholinergic drugs administration in AD patients. 2. Since the nootropic drugs Piracetam and Oxiracetam are deemed to act through a cholinergic mechanism, we checked whether AD patients treated with these drugs would show the same pattern of neuropsychological improvement. 3. Results were negative, since (a) episodic memory showed a similar degree of improvement both in patients treated with these drugs and in patients treated with placebo; (b) the number of intrusions tended

[A Case with Multiple Comorbidities of Obsessive-Compulsive and Related Disorders].

Obsessive-compulsive and related disorders (OCRDs) have been introduced in a revision to DSM-5 as a novel category that is distinct from other anxiety disorders in DSM-IV. OCRDs consist of 5 primary disorders: obsessive-compulsive disorder (OCD), body dysmorphic disorder (BDD), hoarding disorder (HD), skin picking disorder (SPD), and hair pulling disorder (HPD), which share core clinical features such as preoccupation or recurrent thoughts and/or repetitive behaviors. Repetitive behaviors in BDD and HD can be differentially characterized by the presence of cognitive components associated with

A comparison of the nature and correlates of panic attacks in the context of Panic Disorder and Social Anxiety Disorder.

UNLABELLED: Panic attacks occurring outside of Panic Disorder are not well-understood despite their inclusion as a diagnostic specifier in the Diagnostic and Statistical Manual for Mental Disorders (DSM-5). This study compares panic attacks in the context of Panic Disorder compared to social anxiety in terms of their symptom frequency, severity, and clinical correlates. METHOD: Participants (n=404) were interviewed using the Anxiety Disorders Interview Schedule (ADIS-IV-L; Brown et al., 1994), from which we analyzed interviewer ratings of panic attacks and panic attack symptoms, as well as ot

[Sleep disorders and oxidative stress].

Sleep disorders represent a significant medical and social issue, closely linked to the development of oxidative stress and cognitive impairment. This review summarizes current evidence for a bidirectional association between sleep disorders and oxidative stress. Sleep deprivation and chronic insomnia are associated with the accumulation of reactive oxygen species, mitochondrial dysfunction, neuroinflammation, and lipid damage, which together create a cycle that exacerbates sleep disturbances and contributes to cognitive impairment. The role of intermittent hypoxia in obstructive sleep apnea s
