Theoretical insights into the cooperative effects of rigid π-bridges and auxiliary acceptors on the photovoltaic properties of D-π-A'-π-A sensitizers.

CONTEXT: The photovoltaic performance of dye-sensitized solar cells (DSSCs) is critically governed by the energy-level alignment, excited-state charge-transfer dynamics, and dye-TiO2 interfacial interaction of the sensitizer. In this work, nine D-π-A'-π-A organic dyes based on an aminofluorene donor, 2-(2,4-difluorophenyl)pyridine as the auxiliary acceptor (A'), and cyanoacrylic acid as the terminal acceptor, with thiophene, selenophene, or acetylene as the π-bridge, were theoretically investigated to elucidate the cooperative effects of π-bridge engineering and substituent modification on pho

A compact five-gene immuno-fibrotic profile distinguishing systemic sclerosis subtypes with PXDN validated as a fibrosis-associated target.

INTRODUCTION: Systemic sclerosis (SSc) is a heterogeneous autoimmune disease characterized by a paucity of reliable biomarkers for accurate diagnosis and subtype stratification. The considerable clinical variability between diffuse and limited cutaneous subtypes underscores an urgent need for molecular tools that can dissect this heterogeneity and guide therapeutic strategies. METHODS: To address this, we employed a multi-step computational approach. Initially, Weighted Gene Co-expression Network Analysis (WGCNA) was applied to the GSE130955 dataset to identify disease-associated modules. Thi

Anti-Kasha Fluorescence in Molecular Entities: Central Role of Electron-Vibrational Coupling.

According to Kasha's rule, the emission of a photon in a molecular system always comes from the lowest excited state. A corollary of this rule (i.e., the Kasha-Vavilov rule) states that the emission spectra are independent of the excitation wavelength. Although these rules apply for most of the molecular systems, violations of these rules are often reported. The prototypical case of a Kasha's rule violation is the fluorescence observed from S2 in azulene. Thanks to the advances in both theoretical and experimental research, other types of anomalous fluorescence (e.g., excitation energy transfe

Neuromolecular imaging, a nanobiotechnology for Parkinson's disease: advancing pharmacotherapy for personalized medicine.

Evaluating each patient and animal as its own control achieves personalized medicine, which honors the hippocratic philosophy, explaining that "it is far more important to know what person has the disease than what disease the person has." Similarly, individualizing molecular signaling directly from the patient's brain in real time is essential for providing prompt, patient-based treatment as dictated by the point of care. Fortunately, nanotechnology effectively treats many neurodegenerative diseases. In particular, the new medicinal frontier for the discovery of therapy for Parkinson's diseas

Computational analysis of pathogenic nsSNPs in the human TNF gene and their role in neuroinflammation in Adrenoleukodystrophy.

Adrenoleukodystrophy (ALD) is an X-linked neurodegenerative disorder caused by pathogenic variants in the ABCD1 gene and characterized by progressive demyelination and neuroinflammatory responses. Tumor necrosis factor-alpha (TNF-α) is an important mediator of inflammatory signaling and has been implicated in the neuroinflammatory environment associated with ALD. However, the structural and functional consequences of genetic variations within TNF remain insufficiently understood. The study focuses to find high-risk non-synonymous single nucleotide polymorphisms (nsSNPs) in the TNF gene and the

Computational Drug Design of Natural Product-Based Azole Hybrids for Multifactorial Diseases: Success Stories.

Complex diseases such as cancer, neurodegenerative disorders, and metabolic syndromes involve multiple interconnected biological pathways, often rendering single-target therapies ineffective. Conventional drugs frequently lead to undesirable side effects and contribute to Multidrug Resistance (MDR). Natural products, known for their structural diversity and relatively lower toxicity, have emerged as promising candidates for the development of multi-target therapeutic agents. Hybrid molecules, which combine pharmacophores from natural and synthetic origins, offer a novel strategy to simultaneou

Spatial multi-omics-guided drug development in inflammatory bowel disease: from mucosal immune niches to precision therapy.

Crohn's disease and ulcerative colitis are now treated with an expanding set of biologics and small molecules, yet treatment selection remains largely empirical. Primary non-response, secondary loss of response, incomplete mucosal healing, fibrostenosis, and fistulizing disease show that conventional clinical, endoscopic, and bulk molecular markers do not capture how disease is organized within tissue. This review argues that the inflamed intestine is better viewed as a set of spatially structured mucosal immune niches rather than as uniform inflammation, with epithelial, myeloid, lymphoid, st
