Modeling the infection dynamics of bacteriophages in enteric Escherichia coli: estimating the contribution of transduction to antimicrobial gene spread.

Animal-associated bacterial communities are infected by bacteriophages, although the dynamics of these infections are poorly understood. Transduction by bacteriophages may contribute to transfer of antimicrobial resistance genes, but the relative importance of transduction among other gene transfer mechanisms is unknown. We therefore developed a candidate deterministic mathematical model of the infection dynamics of enteric coliphages in commensal Escherichia coli in the large intestine of cattle. We assumed the phages were associated with the intestine and were predominantly temperate. Model

Two routes of MexS-MexT-mediated regulation of MexEF-OprN and MexAB-OprM efflux pump expression in Pseudomonas aeruginosa.

The NfxC-type mutant of Pseudomonas aeruginosa produces the MexEF-OprN efflux pump and down-regulates expression of the quorum-sensing-dependent efflux pump MexAB-OprM and production of virulence factors in the presence of an active transcriptional regulator, MexT. Consequently, these cells are resistant to chloramphenicol and hypersusceptible to β-lactam antibiotics. An upper negative regulator, MexS, has been assumed to inactivate MexT in wild-type strains, hence shutting down production of the MexEF-OprN pump. This observation was, however, reported in only one clinical strain and not confi

Molecular characteristics of extended-spectrum β-lactamases in clinical isolates of Enterobacteriaceae from the Philippines.

β-Lactamases, including extended-spectrum β-lactamases (ESBLs) and AmpC β-lactamases, are major resistance mechanisms of Enterobacteriaceae. Emergence of plasmid-mediated quinolone resistance (PMQR) determinants in ESBL-producing isolates poses a global threat. The molecular characterisitcs of ESBL and PMQR determinants in the Philippines are not well characterized. In this study, we investigated ESBLs and AmpC β-lactamases in clinical isolates of Enterobacteriaceae from the Philippines, and analyzed the association between ESBL and PMQR genes. A total of 91 amoxicilin non-susceptible Enteroba

Antimicrobial resistances of extraintestinal pathogenic Escherichia coli isolates from swine in China.

Antibiograms and relevant genotypes of porcine extraintestinal pathogenic Escherichia coli (ExPEC) isolates (n = 315) recovered between 2004 and 2007 in China were assessed. Among the 14 antimicrobials tested, the most prevalent resistance was to ampicillin, trimethoprim, sulfadimidine, tetracycline, neomycin, streptomycin, kanamycin, ciprofloxacin and ofloxacin (ranging from 81.9 to 100%). Forty-six multiresistant patterns were found. For each antimicrobial agent, ampicillin resistance was primarily mediated by bla(TEM,) streptomycin resistance by strA and strB, kanamycin/neomycin resistance

Genome-wide single nucleotide polymorphism typing method for identification of Bacillus anthracis species and strains among B. cereus group species.

As an issue of biosecurity, species-specific genetic markers have been well characterized. However, Bacillus anthracis strain-specific information is currently not sufficient for traceability to identify the origin of the strain. By using genome-wide screening using short read mapping, we identified strain-specific single nucleotide polymorphisms (SNPs) among B. anthracis strains including Japanese isolates, and we further developed a simplified 80-tag SNP typing method for the primary investigation of traceability. These 80-tag SNPs were selected from 2,965 SNPs on the chromosome and the pXO1

Characterisation of MRSA from Malta and the description of a Maltese epidemic MRSA strain.

Malta has one of the highest prevalence rates of methicillin-resistant Staphylococcus aureus (MRSA) in Europe. However, only limited typing data are currently available. In order to address this situation, 45 MRSA isolates from the Mater Dei Hospital in Msida, Malta, were characterised using DNA microarrays. The most common strain was ST22-MRSA-IV (UK-EMRSA-15, 30 isolates). Sporadic strains included ST36-MRSA-II (UK-EMRSA-16, two isolates), PVL-positive ST80-MRSA-IV (European Clone, one isolate), ST228-MRSA-I (Italian Clone/South German Epidemic Strain, one isolate) and ST239-MRSA-III (Vienna

cagA vacA alelles and babA2 genotypes of Helicobacter pylori associated with gastric disease in Brazilian adult patients.

Helicobacter pylori is an important human pathogen that causes chronic gastritis and is associated with development of peptic ulcer disease and gastric malignancies. The vacuolating cytotoxin (vacA), cagA gene, and babA2 gene are important virulence factor involving gastric diseases. Eighty-nine Helicobacter pylori-positive gastric biopsies were analyzed by polymerase chain reaction and Southern blotting for H. pylori detection and genotyping with primer pairs from each virulence gene. Fifty-three strains (59%) were common vacA genotype s1/m1, and only 14 (16%) were s2/m2, 12% of strains was f

Protection against scrub typhus by a plasmid vaccine encoding the 56-KD outer membrane protein antigen gene.

The 56-kD outer membrane protein of Orientia tsutsugamushi has previously been shown to be the immunodominant antigen in scrub typhus infections. Its gene was cloned into the DNA vaccine vector pVR1012 as a vaccine candidate (pKarp56). The in vitro expression of this 56-kD antigen by pKarp56 was confirmed in tissue culture by an indirect fluorescence assay and Western blot analysis. The initial antibody responses of mice immunized with varied doses of the pKarp56 were barely detected, but increases were observed after each of three subsequent booster immunizations. Although no protection was o

Type III secretion systems and the evolution of mutualistic endosymbiosis.

The view that parasites can develop cooperative symbiotic relationships with their hosts is both appealing and widely held; however, there is no molecular genetic evidence of such a transition. Here we demonstrate that a mutualistic bacterial endosymbiont of grain weevils maintains and expresses inv/spa genes encoding a type III secretion system homologous to that used for invasion by bacterial pathogens. Phylogenetic analyses indicate that inv/spa genes were present in presymbiotic ancestors of the weevil endosymbionts, occurring at least 50 million years ago. The function of inv/spa genes in

Carbadox, a veterinary growth promoter exhibits anticlostridial activity through induction of oxidative stress in Clostridioides difficile.

BACKGROUND: Clostridioides difficile is the leading cause of antibiotic-associated diarrhea in hospitalized patients and is classified as an urgent public health threat. Current therapies, while effective, are limited by antibiotic-induced dysbiosis, treatment failure, and high recurrence rates, underscoring the need for novel therapeutics. Carbadox (CRX), an antibacterial growth promoter, has recently been identified as a potential anti-C. difficile inhibitor. This study aimed to comprehensively evaluate the anti-C. difficile activity, mechanism of action, and in vivo efficacy of CRX. METHOD

Genomic and proteomic characterization of SuMu, a Mu-like bacteriophage infecting Haemophilus parasuis.

BACKGROUND: Haemophilus parasuis, the causative agent of Glässer's disease, is prevalent in swine herds and clinical signs associated with this disease are meningitis, polyserositis, polyarthritis, and bacterial pneumonia. Six to eight week old pigs in segregated early weaning herds are particularly susceptible to the disease. Insufficient colostral antibody at weaning or the mixing of pigs with heterologous virulent H. parasuis strains from other farm sources in the nursery or grower-finisher stage are considered to be factors for the outbreak of Glässer's disease. Previously, a Mu-like bacte

Lineage-dependent Microbial Shifts in the Reproductive Tract of Black Soldier Flies (Hermetia illucens) Reveal Novel Mating-linked Associations.

Reproductive microbiota is known to modulate host immunity, reproductive physiology, and longevity, yet their interactions with host genetics, sex, and mating status remain relatively underexplored in most insects. Some studies suggest that species (and even populations) can vary in the microbial communities associated with their reproductive tissues. Sexually transmitted microbes can also directly influence population growth and reproductive fitness, making them relevant even for applied insect management strategies. The economically relevant, Black Soldier Fly (BSF; Hermetia illucens), is kn

Gut Microbiome Diversity, Functional Potential, and Ecological Relevance of Hottentotta tamulus.

Scorpions are ancient arachnids of medical and ecological importance; they prey on insects and other arthropods while serving as prey to birds and reptiles. In this study, we reported for the first time on the characterization of the gut intestinal microbiome of Hottentotta tamulus native to northeastern Pakistan. The scorpions were identified on a morphological basis and the Cytochrome c oxidase subunit 1 gene sequence, while the gut microbiome was characterized through full-length 16S rRNA (V1-V9) Nanopore sequencing. The gut microbiota exhibited low to moderate alpha diversity with Chao1 an

Improved Detection and Sequencing of Rickettsia spp. DNA in South African Wildlife.

BACKGROUND: Rickettsia spp. are obligate intracellular bacteria of the spotted fever group (SFG), typhus group (TG) and transitional group, with diverse vectors and significant zoonotic potential. While Rickettsia africae is well documented in South African ticks, the role of wildlife hosts in pathogen maintenance remains unclear. METHODS: We re-screened 73 spleen samples opportunistically collected from 11 wild mammal species that previously tested negative for Rickettsia by reverse line blot hybridization (RLB). The samples were collected from Kruger National Park, Timbavati Private Nature

Strain-specific qPCR assay to track the vaginal colonization of the probiotic Lacticaseibacillus rhamnosus TOM 22.8 strain orally administered.

BACKGROUND: Over the past decade, research on the female reproductive tract microbiota has grown significantly and the use of probiotics has gained growing interest in restoring vaginal microbial balance. In this context, probiotic lactobacilli, topically or orally administered, emerged as a valuable alternative or a complementary approach to conventional antimicrobial therapies. Although topical application allows for a fast and direct action on the altered vaginal ecosystem, the oral intake provides long-term benefits due to the passage through the gastrointestinal tract. METHODS: In the pre

Coxiella burnetii detected in Rhipicephalus sanguineus ticks collected in Portuguese autochthonous livestock.

Coxiella burnetii is a bacterium that causes coxiellosis in animals and Q fever in humans. While ruminants are the main reservoirs, the role of ticks in their transmission is still uncertain. This study looked for C. burnetii in ticks collected from two autochthonous breeds in Portugal: Churra Galega Mirandesa sheep and Garrano horses. A total of 555 ticks were tested, including 100 Rhipicephalus sanguineus sensu (s.) stricto (s.) from sheep and 455 Rhipicephalus bursa from horses. Coxiella burnetii was found in 7 % of R. sanguineus s.s., but not in R. bursa. Phylogenetic analysis confirmed th

Using Digital PCR to Unravel the Occurrence of Piroplasmids, Bartonella spp., and Borrelia spp. in Wild Animals from Brazil.

Piroplasmids (Babesia spp., Rangelia spp., Theileria spp., Cytauxzoon spp.) are tick-borne apicomplexan protozoa that infect, depending on the species, erythrocytes and leucocytes in a wide variety of mammals and birds. The genera Bartonella and Borrelia include vector-borne bacteria that can infect and cause disease in both animals and humans. Detection of hemotropic bacteria and piroplasmids in wild animals is often challenging due to low bacteremia or parasitemia. Digital (d)PCR has proven to be an effective modality for the detection and quantification of DNA of hemotropic pathogens with l

Bacterial recolonization of hospital sink biofilms.

BACKGROUND: Hospital sink drains are known reservoirs for many pathogens that cause healthcare-associated infections (HAIs). Drain bacteria can migrate up to the drain cover and then spread to surrounding surfaces and patients through droplet dispersal during sink use. Therefore, cleaning sink surfaces represents a key intervention strategy to limit transmission between drains and patients. AIMS: In this study, we aimed to: (1) characterize microbial community taxonomy and abundance in sink components and (2) evaluate the kinetics and sources of bacterial recolonization onto sink surfaces aft
