Nanoparticles as antibiotic-delivery vehicles (ADVs) overcome resistance by MRSA and other MDR bacterial pathogens: The grenade hypothesis.

OBJECTIVES: The aim of this study was to examine how the concentrated delivery of less effective antibiotics, such as the β-lactam penicillin G, by linkage to nanoparticles (NPs), could influence the killing efficiency against various pathogenic bacteria, including methicillin-resistant Staphylococcus aureus (MRSA) and other multidrug resistant (MDR) strains. METHODS: The β-lactam antibiotic penicillin G (PenG) was passively sorbed to fluorescent polystyrene NPs (20nm) that were surface-functionalized with carboxylic acid (COO--NPs) or sulfate groups (SO4--NPs) to form a PenG-NP complex. Anti

Therapeutics and delivery vehicles for local treatment of osteomyelitis.

Osteomyelitis, or the infection of the bone, presents a major complication in orthopedics and may lead to prolonged hospital visits, implant failure, and in more extreme cases, amputation of affected limbs. Typical treatment for this disease involves surgical debridement followed by long-term, systemic antibiotic administration, which contributes to the development of antibiotic-resistant bacteria and has limited ability to eradicate challenging biofilm-forming pathogens including Staphylococcus aureus-the most common cause of osteomyelitis. Local delivery of high doses of antibiotics via trad

Nanoantibiotics: A Novel Rational Approach to Antibiotic Resistant Infections.

BACKGROUND: The main drawbacks for using conventional antimicrobial agents are the development of multiple drug resistance due to the use of high concentrations of antibiotics for extended periods. This vicious cycle often generates complications of persistent infections, and intolerable antibiotic toxicity. The problem is that while all new discovered antimicrobials are effective and promising, they remain as only short-term solutions to the overall challenge of drug-resistant bacteria. OBJECTIVE: Recently, nanoantibiotics (nAbts) have been of tremendous interest in overcoming the drug resis

Red blood cell-hitchhiking boosts delivery of nanocarriers to chosen organs by orders of magnitude.

Drug delivery by nanocarriers (NCs) has long been stymied by dominant liver uptake and limited target organ deposition, even when NCs are targeted using affinity moieties. Here we report a universal solution: red blood cell (RBC)-hitchhiking (RH), in which NCs adsorbed onto the RBCs transfer from RBCs to the first organ downstream of the intravascular injection. RH improves delivery for a wide range of NCs and even viral vectors. For example, RH injected intravenously increases liposome uptake in the first downstream organ, lungs, by ~40-fold compared with free NCs. Intra-carotid artery inject

Drug Target Validation Methods in Malaria - Protein Interference Assay (PIA) as a Tool for Highly Specific Drug Target Validation.

BACKGROUND: The validation of drug targets in malaria and other human diseases remains a highly difficult and laborious process. In the vast majority of cases, highly specific small molecule tools to inhibit a proteins function in vivo are simply not available. Additionally, the use of genetic tools in the analysis of malarial pathways is challenging. These issues result in difficulties in specifically modulating a hypothetical drug target's function in vivo. OBJECTIVE: The current "toolbox" of various methods and techniques to identify a protein's function in vivo remains very limited and th

Formulations of deoxycholic for therapy: a patent review (2011 - 2014).

INTRODUCTION: Deoxycholic acid (DOCA) is involved in many physiological functions and has been used in various fields of pharmaceutical formulations as a natural active solubilizing and permeation-enhancing agent. Although DOCA has been suggested to be a promoter of colon cancer, it has also been used extensively as a starting material to obtain new derivatives for potential therapeutic applications. AREA COVERED: In this review, we focus on patents and research reports from 2011 to 2014 related to pharmaceutical formulations and therapeutic applications using DOCA and its derivatives as surf

Herbal medicines for the prevention and treatment of Alzheimer's disease.

Alzheimer's disease (AD) is a chronic neurodegenerative disorder and is the most common cause of progressive dementia in aging. Research on AD therapy has been partly successful in terms of developing symptomatic treatments, but there have been a number of failures with regard to developing disease-modifying therapies. The pathogenesis of AD remains unclear and the present one-drug, one-target paradigm for anti-AD treatment appears to be clinically unsuccessful. In many countries, traditional herbal medicines are used to prevent or treat neurodegenerative disorders, and some have been develope

Sodium channel blockers for neuropathic pain.

IMPORTANCE OF THE FIELD: The voltage-gated sodium channels (VGSCs) play a fundamental role in controlling cellular excitability and their abnormal activity is related to several pathological processes, including cardiac arrhythmias, epilepsy, neurodegenerative diseases, spasticity, chronic and neuropathic pain. In particular, neuropathic pain (e.g., postherpetic and trigeminal neuralgia, diabetic neuropathy and spinal cord injury) is a serious clinical problem that affects a high percentage of the world population. Because an altered sodium channel isoform expression profile has been considere

Chemokines as novel therapeutic targets in autoimmune thyroiditis.

Chemokines, a group of proinflammatory chemotactic cytokines, function to recruit leukocytes to inflammation sites, but they also play important roles in tumor growth, angiogenesis, organ sclerosis, and autoimmunity. In recent years, increasing evidence has accumulated to support the concept that thyroid epithelial cells (TEC) as well as the lymphocytes infiltrating into the thyroid are capable of producing CC and CXC chemokines. They, in turn, promote the initiation and maintenance of an inflammatory response, resulting in the development of autoimmune thyroiditis. This review focused on the

Age-related macular degeneration: a target for nanotechnology derived medicines.

Despite the fact that the retina is a fairly accessible portion of the central nervous system, there are virtually no treatments for early age-related macular degeneration (AMD). AMD is a degenerative retinal disease that causes progressive loss of central vision and is the leading cause of irreversible vision loss and legal blindness in individuals over the age of 50. Both environmental and genetic components play a role in its development. AMD is a multifactorial disease with characteristics that include drusen, hyperpigmentation and/or hypopigmentation of the retinal pigment epithelium (RPE

Mass transport properties of progesterone and estradiol in model microemulsion formulations.

PURPOSE: The purpose of these studies was to determine the extent to which drug loading influences the mass transport characteristics of poorly soluble steroids from model microemulsion formulations in vitro. METHODS: Two conditions of drug loading in the microemulsions were tested, "near saturation" and "constant loaded." Mass flux of (3)H-labelled progesterone or estradiol was measured in a side-by-side diffusion chamber from microemulsions consisting of Brij 97, Miglyol 812 and water. Pulsed gradient NMR was used to measure the diffusivities of all components. The thermodynamic activity an

Effective antitussives for the cough patient: an unmet need.

Chronic cough is a prevalent symptom in the community but it is likely that only a small fraction of chronic coughers seek medical attention. Chronic cough can be controlled by addressing the 'cause' of the cough, but not all cough is controlled using this approach; an 'idiopathic' cough or cough of unknown aetiology is becoming more well-recognized. In these patients and in those whose cough has not responded to treatment of the cause(s), there is a lack of efficacious antitussive therapies ('non-specific' antitussives). Even in those whose cough is controlled by treatment of the cause, an ef
