Endothelialized human umbilical cord mesenchymal stem cell-derived exosomes enhances angiogenesis and stabilizes the blood-spinal cord barrier after spinal cord injury.

Restoration of vascular structure and function is pivotal for neurological recovery following spinal cord injury (SCI), yet repairing the bl … ) via lentiviral modification. In vivo, these modified Exos preferentially localized to neovascular endothelial cells, promoted angiogenesis, reinforced BSCB integrity, and improved neurological outcomes in SCI mouse models. Proteomic profiling identified key angiogenic and barrier‑stabilizing factors carried by RGD‑E‑UCMSC‑Exos, including MMP2, FLT1, TIMP1, GAS6, …

Knockdown of Long Non-coding RNA-MALAT1 Ameliorates Diabetic Lower Limb Atherosclerotic Disease Through MiR-17-5p-Mediated Endothelial Cell Pyroptosis.

We observed the expression of long non-coding RNA (lncRNA) MALAT1 and microRNA (miR)-17-5p in patients with diabetic lower extremity atherosclerosis (LEAD) and EA. hy926 human endothelial cells (EA. Hy926 cells). We further investigated whether knockdown of MALAT1 (sh-MALAT1) could protect endothelial cells and improve the occurrence of atherosclerosis through miR-17-5p, aiming to dissect the underlying mechanism. Patients with type 2 diabetes were stratified into two groups: those with lower extremity atherosclerotic lesions (LEAD group) and those without (T2DM group). For in vitro studies, E…

A Case of Corneal Endothelial Cell Loss After PreserFlo MicroShunt Implantation Requiring Device Removal and Ahmed Glaucoma Valve Implantation With Tube Insertion Into the Vitreous Cavity.

We report a case of progressive endothelial cell loss (ECL) following PreserFlo MicroShunt (PMS) (Santen Pharmaceutical, Osaka, Japan) implantation, which was successfully managed by PMS removal and the pars plana placement of Ahmed glaucoma valve (AGV) implantation. An 82-year-old man with primary open-angle glaucoma underwent PMS implantation combined with cataract surgery. Preoperative endothelial cell density (ECD) was 2,148 cells/mm², and intraocular pressure (IOP) was 21 mmHg despite five topical IOP-lowering medications. One week postoperatively, the IOP normalized to 11 mmHg without me

The effect of concomitant beta-blocker use on survival in patients with metastatic renal cell carcinoma treated with a vascular endothelial growth factor receptor inhibitors in the first line.

PURPOSE: Vascular endothelial growth factor (VEGF) inhibition is one of the cornerstones of treatment in the treatment of metastatic renal cell carcinoma (mRCC). Since RCC is a disease of advanced age and hypertension as a side effect of VEGF receptor inhibitors, beta-blocker use is common in these patients. We aimed to compare the treatment efficacy and survival results in case of concomitant use of these two drugs due to the inhibition of VEGF in beta-blockers. METHODS: A total of 121 patients with a diagnosis of mRCC who used sunitinib or pazopanib in first-line therapy were included in th

Serum mRNA levels of cytokeratin-19 and vascular endothelial growth factor in oral squamous cell carcinoma and oral potentially malignant disorders using RT-PCR.

BACKGROUND: Oral cancers, which include tumors of the oral cavity, salivary glands, and pharynx, are becoming increasingly prevalent worldwide. Squamous cell carcinoma accounts for over 90% of malignant oral lesions, with oral squamous cell carcinoma (OSCC) being notably common in the Indian subcontinent and other regions of Asia. This is especially true in South-Central Asia, including Sri Lanka, where it is particularly prevalent among men. This study aims to evaluate the levels of Vascular Endothelial Growth Factor-A (VEGF-A) and Cytokeratin-19 (CK-19) mRNAs in whole blood as a potential me

Complete remission of reactive cutaneous capillary endothelial proliferation caused by the programmed cell death-1 inhibitor camrelizumab achieved through thalidomide monotherapy: A case report.

Reactive cutaneous capillary endothelial proliferation (RCCEP) is a common adverse effect of the anti-programmed cell death-1 (PD-1) monoclonal antibody camrelizumab and usually occurs on the skin. This condition causes bleeding nodules of varying severity depending on disease grade; these affect a person's appearance and quality of life. The exact mechanism remains elusive and its occurrence in visceral organs has not been previously reported, to the best of our knowledge. Furthermore, there is currently a lack of standard, uniform treatments. The present study reported on a patient who exper

Endothelial microparticles delivering microRNA-155 into T lymphocytes are involved in the initiation of acute graft-versus-host disease following allogeneic hematopoietic stem cell transplantation.

Endothelial microparticles (EMPs) upregulation has been observed in acute graft-versus-host disease (aGVHD) after allogeneic hematopoietic stem cell transplantation (allo-HSCT). However, the role of EMPs remains unclear. We found that EMPs derived from TNF-α-stimulated human umbilical vein endothelial cells (EA.hy926) concentrated more microRNA-155 (miR-155) compared with maternal cells. The miR-155 levels in MPs from peripheral blood of aGVHD patients and mice were remarkably elevated and significantly higher than the levels in plasma. Moreover, the rising peak of miR-155 in MPs occurred sign…

Distinct Endothelial Cell Responses in the Heart and Kidney Microvasculature Characterize the Progression of Heart Failure With Preserved Ejection Fraction in the Obese ZSF1 Rat With Cardiorenal Metabolic Syndrome.

BACKGROUND: The combination of cardiac and renal disease driven by metabolic risk factors, referred to as cardiorenal metabolic syndrome (CRMS), is increasingly recognized as a critical pathological entity. The contribution of (micro)vascular injury to CRMS is considered to be substantial. However, mechanistic studies are hampered by lack of in vivo models that mimic the natural onset of the disease. Here, we evaluated the coronary and renal microvasculature during CRMS development in obese diabetic Zucker fatty/Spontaneously hypertensive heart failure F1 hybrid (ZSF1) rats. METHODS AND RESUL

Mesenchymal stem cell-derived microparticles ameliorate peritubular capillary rarefaction via inhibition of endothelial-mesenchymal transition and decrease tubulointerstitial fibrosis in unilateral ureteral obstruction.

INTRODUCTION: Microparticles (MPs) derived from kidney-derived mesenchymal stem cells (KMSCs) have recently been reported to ameliorate rarefaction of peritubular capillaries (PTC) in ischemic kidneys via delivery of proangiogenic effectors. This study aimed to investigate whether KMSC-derived MPs show anti-fibrotic effects by ameliorating endothelial-to-mesenchymal transition (EndoMT) in human umbilical vein endothelial cells (HUVEC) in vitro and by preserving PTC in kidneys with unilateral ureteral obstruction (UUO) in vivo. METHODS: MPs isolated from the supernatants of KMSC were co-cultur…
