[HBV pol/S gene mutations in chronic hepatitis B patients receiving nucleoside/nucleotide analogues treatment].

Chronic hepatitis B (CHB) is an important public health problem affecting over 240 million people all around the world. The aim of the treatment in chronic hepatitis B is to prevent progression to cirrhosis and liver cancer. Interferons (standard and peginterferon) (Peg-IFN) and nucleoside/nucleotide analogues (NAs) are widely used in the treatment of CHB. The use of long-term therapy can however result in drug resistant mutations, which can lead to treatment failure. In patients with chronic hepatitis B, in addition to primary drug resistance mutations in the pol gene, compensatory mutations

Altered topoisomerase I activity and recombination activating gene expression in a human leukemia cell line resistant to doxorubicin.

We examined the expression of the genes encoding topoisomerases I and II and those associated with V(D)J [variable(diversity)joining] recomb … ecombination although RAG1 gene is overexpressed. The mechanism of the RAG1 gene activation was not gene amplification, and no rearrangement was detected in the RAG1 gene locus. RAG1 presents homology with the yeast gene HPR1, itself homologous to yeast topoisomerase I and responsible for the control of recombination in somatic cells. Since DNA topoisomerases are th …

RGS Family Remodeling in Breast Cancer: A Meta-Analysis and Multi-Omics Profiling of Prognostic Biomarkers.

BACKGROUND: Breast cancer (BC) is a heterogeneous malignancy with diverse molecular subtypes and variable clinical outcomes. Despite diagnostic and therapeutic advances, recurrence and metastasis contribute to poor prognosis in subsets of patients. Regulators of G protein signaling (RGS) proteins, negative modulators of G protein-coupled receptor (GPCR) pathways, influence tumor progression, but their expression profiles, genomic alterations, immune associations, and prognostic roles in BC remain incompletely understood. This study systematically investigated the RGS family to identify potenti

Transcriptome-based selection and validation of optimal house-keeping genes for skin research in goats (Capra hircus).

BACKGROUND: In quantitative real-time polymerase chain reaction (qRT-PCR) experiments, accurate and reliable target gene expression results are dependent on optimal amplification of house-keeping genes (HKGs). RNA-seq technology offers a novel approach to detect new HKGs with improved stability. Goat (Capra hircus) is an economically important livestock species and plays an indispensable role in the world animal fiber and meat industry. Unfortunately, uniform and reliable HKGs for skin research have not been identified in goat. Therefore, this study seeks to identify a set of stable HKGs for t

Hodgkin's lymphoma: the role of cell surface receptors in regulation of tumor cell fate.

UNLABELLED: The hallmark of Hodgkin's lymphoma (HL) are mononucleated Hodgkin's cells and multinucleated Reed-Sternberg (HRS) cells, which usually account for only about 1% of cells in the tumor tissue. The majority of HRS cells in classical HL are derived from germinal centre B cells that have acquired disadvantageous Ig variable chain gene mutations and escaped from apoptosis. Due to reprogramming of gene expression, these lymphoma cells have lost the expression of most B-cell specific genes and acquired expression of multiple genes that are typical for other hematopoietic cells. HRS cells a

Analysis of cytokine gene polymorphisms in recipient's matched with living donors on acute rejection after renal transplantation.

Despite advances in immunosuppressive therapy in last few years, allograft rejection still remains the concern for kidney graft failure. Cytokines are key mediators in the induction and effector phases of all immune and inflammatory responses. They are not allospecific so both recipient as well as donor cells may be subjected to cytokine changes. We sought to ascertain whether IL-1B -511, IL-1B +3954, TNF-A -308, TGF-B Codon 10 and 25, IL-2 -330, IL-6 -174, IL-10 -1082, IL-10 -819 (SNPs), IL-1RN, IL-4 (VNTR) and TGF-B C-del (deletion) genes in two hundred subjects including recipients and thei

Isolation of a novel iris-specific and leucine-rich repeat protein (oculoglycan) using differential selection.

PURPOSE: To identify and characterize genes expressed in the iris. METHODS: A human adult iris cDNA library was constructed and subjected to a differential selection screen to identify genes preferentially expressed in iris or trabecular tissue versus those expressed in lymphoblasts. Selected cDNAs were partially sequenced. Novel cDNAs were chosen for further analysis. The cDNAs were localized within chromosomes using a radiation hybrid (RH) mapping panel. The tissue expression profile of each cDNA was found through computer-based searches. One novel cDNA was subjected to 5' rapid amplificati

Molecular characterization of the A52 murine hepatocellular carcinoma cell line.

BACKGROUND: To combat hepatocellular carcinoma (HCC) disease heterogeneity and growing mortality, there is an urgent need for targeted and personalized therapeutics. While syngeneic mouse models are commonly used for preclinical validation of these therapeutics, the lack of genetically characterized murine cell lines adds uncertainty to the study of drug-gene interactions in these models. We previously generated a novel murine cell line, A52, from a diethylnitrosamine (DEN)-induced adiponectin-knockout mouse model. Here, we present a comprehensive genomic and transcriptomic characterization of

In situ perfusion with MCC950 modulates the pyroptosis-related axis and DAMP-TLR4 signaling in kidneys from brain-dead donor.

Brain death (BD), followed by static cold storage (SCS), induces sterile inflammation in donor kidneys, leading to molecular injury that compromises graft quality. This response is associated with activation of the NLRP3 inflammasome and pyroptosis related pathways. We investigated whether pharmacological inhibition of NLRP3 with MCC950, administered during in situ renal perfusion, during SCS, or through a combined strategy, could attenuate BD-induced inflammation exacerbated by SCS. Male Wistar rats (n = 36) were subjected to BD followed by 6 h of donor maintenance and subsequent 16 h of SCS.

Highly efficient intranasal H5N1 vaccine based on a safe vesicular stomatitis virus (VSV) vector.

Recombinant vesicular stomatitis virus (rVSV) is an increasingly popular vaccine vector, but it still faces challenges such as difficulties in construction, low in vitro amplification titers, safety concerns, and uncertain mucosal immune effects. Therefore, there is an urgent need to construct new VSV vectors for next-generation vaccines with high immunogenicity and excellent safety. In this study, we independently constructed a high-expression 6-9G recombinant replication-competent VSV vector. Based on the 6-9G vector, site-directed mutations were introduced into the M gene, including deletio

Distal-less homeobox 5 transactivated by MYCN contributes to progression of neuroblastoma through AKT signaling pathway.

Neuroblastoma is one of the highest incidence solid tumors of childhood. Therapy for neuroblastoma with MYCN proto-oncogene (MYCN) amplification is of great challenge, due to its aggressive invasion and chemoresistance. In our study, differentially expressed genes (DEGs) between MYCN-amplified and non-amplified neuroblastoma tissues were analyzed using "limma" R package. Then, DEGs were screened by silencing or enhancing expression of MYCN in neuroblastoma cells, and distal-less homeobox 5 (DLX5) was identified to be the downstream target gene. The positive correlation between DLX5 and MYCN ex

Extramedullary myeloma is genomically complex and characterized by near-universal MAPK pathway alterations.

Extramedullary disease (EMD) is associated with an inferior prognosis and lower response rates to conventional multiple myeloma (MM)-directed therapies compared to MM without EMD. A deeper understanding of the molecular landscape and underlying drivers of EMD is essential to identify potential targets for novel therapeutic strategies. To address this, we performed whole-exome sequencing on EMD tumor tissue from 18 unique patients and bone marrow aspirates (BMAs) from 20 patients at the time of EMD development. Notably, paired EMD and BMA samples were collected from 6 patients at the point of E

Development and validation of a carnitine cycle and transport disorders (CCD) panel: an ONT-compatible multi-gene diagnostic kit for newborn and selective screening.

Carnitine transport and cycle disorders (CCD) are a group of metabolic disorders characterized by either carnitine depletion or dysfunction in the carnitine cycle, a critical process for the transport of fatty acids into the mitochondria and their subsequent β-oxidation. Clinically, CCD can manifest with a wide range of symptoms, including hypoketotic hypoglycemia, which may be accompanied by signs of liver dysfunction, hepatic steatosis, myopathy and cardiomyopathy. Biochemical diagnosis typically involves measuring carnitine and acylcarnitine levels in blood, alongside organic acid profiling
