Antibacterial agents with two-photon absorption are expected to play a significant role in biomedical science. Herein, two novel organotin complexes, HLSn1 and HLSn2, based on coumarin were designed, synthesized, and systematically investigated. It was found that these complexes possessed suitable two-photon-active cross sections in the near-infrared region. Moreover, complex HLSn1 could efficiently inhibit the growth of Gram-negative Escherichia coli and Gram-positive Bacillus subtilis, especially the latter with a minimum inhibitory concentration (MIC; 90%) of 2 ± 0.14 μg mL-1, which is lowe

This study describes the application of at-line nanofractionation to the screening of snake venoms for antibacterial activity against Gram-positive and Gram-negative bacteria, the detection of proteins of interest, and their partial or full identification. A method was developed to identify bioactive peptides in crude snake venoms based on reversed-phase liquid chromatography (LC), with parallel nanofractionation onto 384-well plates and mass spectrometry (MS). Bioactivity assays were based on a resazurin-reduction assay. Accurate masses of the bioactive peptides were determined, and peptides …

BACKGROUND/AIMS: The increased resistance of Helicobacter pylori to antibiotics has increased the need to develop new treatments for this bacterium. The aim of our study was to identify new drugs with anti-H. pylori activity.
METHODS: We screened a small molecule library-the library of pharmacologically active compounds (LOPAC), which includes 1,280 pharmacologically active compounds-to identify inhibitors of H. pylori growth. The minimal inhibitory concentrations (MICs) of antibiotics against multidrug-resistant H. pylori strains were determined using the agar dilution method.
RESULTS: We i

Curcumin is well known for its antimicrobial and anti-inflammatory properties. However, since systemic absorption and bioavailability of curcumin from gastrointestinal tract is considerably poor, synthetic curcuminoids are being developed as better alternatives. Two curcumin derivatives: 3,5-bis(benzylidene)-4-piperidone (EF24) and EF24-dimer linked via diethylenetriaminepentacetic acid (EF2DTPA), were included in this study. We investigated the antibacterial activity of EF24 and EF2DTPA against Gram-negative (Escherichia coli) and Gram-positive (Enterococcus faecalis, Staphylococcus aureus) b

Overwhelming Post-Splenectomy Infection (OPSI or PSS), most frequently caused by encapsulated Gram-positive pathogens, is a complication after splenectomy. Reasons for splenectomy include trauma, or malignant and non-malignant hematologic diseases. OPSI-inducing bacteria are mainly Streptococcus pneumoniae and less frequently Haemophilus influenzae, Neisseria meningitides and Gram-negative bacilli. There exist very efficient--albeit often neglected--strategies, how to prevent infections in patients after splenectomy. These include vaccination, prophylactic antibiotics (always for 3 years durin

Cefditoren pivoxil, a new-third generation cephalosporin antibiotic that has recently been granted approval in Spain, shows important activity over a large part of the pathogens causing skin, soft tissue and respiratory tract infections, including Gram-negative and Gram-positive bacteria. Cefditoren has also been shown to be stable against hydrolysis by many common beta-lactamases. Data from in vitro studies and clinical trials show this antibiotic as an oral formulation with an intrinsic activity against Haemophilus influenzae, Moraxella catarrhalis and Streptococcus pneumoniae equivalent to

Respiratory infections are a common source of morbidity and mortality, with pneumonia being the number one cause of death from infectious disease in Western industrialized countries. Initial antibiotic therapy of upper and lower respiratory infections is often empiric, being directed at the pathogens that are most likely to be present. Leading pathogens in respiratory infections are S. pneumoniae, H. influenzae and M. catarrhalis, which have developed considerable resistance problems against previous standard antibiotics like beta-lactams, macrolides and tetracyclines in the last decade. Newly

During a 7-month period beginning February 1989 a total of 3387 bacteria were isolated aerobically from clinical materials at Al-Adan Hospital laboratory. Their susceptibilities to 14 commonly used antibiotics were analysed. Escherichia coli was the most frequently isolated organism among all Gram-negative bacilli. In general, members belonging to Enterobacteriaceae presented higher resistance to ampicillin, trimethoprim-sulphamethoxazole, and cephalothin, the exception being Salmonella spp. Of all the aerobic Gram-positive cocci, Staphylococcus aureus was the most common isolate and showed ex

Fifty consecutive episodes of septicemia were studied in 41 children who had acute lymphoblastic leukemia. Seventy-six percent of these episodes occurred when the absolute granulocyte count was 200/mm3 or less and were caused by gram-negative enteric and gram-positive mucocutaneous bacteria. In eight patients, Streptococcus pyogenes was isolated at the time when ALL was diagnosed. Multiple anaerobic and aerobic isolates from a single blood culture were associated with abdominal distress, whereas Streptococcus pneumoniae and Hemophilus influenzae septicemia occurred in associated with respirato

ATP-binding cassette (ABC) transporters constitute a large family of proteins present in all domains of life. They are powered by dynamic ATPases that harness energy from binding and hydrolyzing ATP through a cycle that involves the closing and reopening of their two ATP-binding domains. The LptB2FGC exporter is an essential ABC transporter that assembles lipopolysaccharides (LPS) on the surface of Gram-negative bacteria to form a permeability barrier against many antibiotics. LptB2FGC extracts newly synthesized LPS molecules from the inner membrane and powers their transport across the peripl…

Periprosthetic joint infection (PJI) is a serious complication following total hip arthroplasty and is most commonly caused by Gram-positive organisms. Infections involving anaerobic bacteria or multidrug-resistant Gram-negative pathogens are rare and particularly challenging to manage. We report a case of periprosthetic hip infection in a female patient in whom Fusobacterium nucleatum (F. nucleatum) was initially isolated, followed by Acinetobacter baumannii (A. baumannii) later in the treatment course. Initial management with debridement, antibiotics, and implant retention (DAIR) was unsucce

Urinary tract infections (UTIs) are among the most common bacterial infections worldwide. Although Gram-negative bacteria predominate, Gram-positive cocci (GPC) are increasingly recognized as significant uropathogens. This retrospective study analyzed 655 patients with culture-confirmed GPC UTIs in a hospital in Central Portugal (2018-2022) to identify predictors of multidrug resistance (MDR). MDR was defined as resistance to three or more antimicrobial classes. The overall MDR prevalence was 83.7%, with Enterococcus spp. showing the highest MDR prevalence (98.2%). Multivariate analysis identi…

Implant-associated infections (IAIs) caused by multidrug-resistant (MDR) pathogens are incredibly challenging to treat due to their strong resistance to conventional antibiotics. Here, we developed efficient biofilm-preventive multilayer coatings against the MDR strain Acinetobacter baumannii (A. baumannii) by leveraging the synergistic antimicrobial activity of the antimicrobial peptide protamine (PR) and graphene oxide (GO) for medical implants. First, the anti-biofilm activity of the PR/GO multilayer coatings was successfully demonstrated against both Gram-negative and Gram-positive bacteri

Racemosol, a natural phenolic compound, is known for its antimicrobial potential, yet experimental studies remain limited. In this study, two new racemosol derivatives (4 and 5) and four known compounds (1-3, 6) were isolated from the bark of Bauhinia malabarica and structurally elucidated using spectroscopic analyses. Most of isolated compounds exhibited notable activity against Gram-positive bacteria, including Staphylococcus aureus, Bacillus subtilis, and Listeria monocytogenes, while showing limited effects on Gram-negative strains. Racemosol (1) and its derivatives (2, 4, and 6) displayed
