A number of studies have suggested B7-H1, a B7 family member, inhibits T cell responses. Therefore, its expression on nonlymphoid tissues has been proposed to prevent T cell-mediated tissue destruction. To test this hypothesis, we generated transgenic mice that expressed B7-H1 on pancreatic islet beta cells. Surprisingly, we observed accelerated rejection of transplanted allogeneic B7-H1-expressing islet beta cells. Furthermore, transgenic B7-H1 expression broke immune tolerance, as some of the mice spontaneously developed T cell-dependent autoimmune diabetes. In addition, B7-H1 expression inc

Although interleukin-2 (IL-2) and -15 (IL-15) share two receptor subunits and many functions, at times they provide contrasting contributions to T-cell-mediated immune responses. IL-2, through its pivitol role in activation-induced cell death (AICD), is involved in peripheral tolerance through the elimination of self-reactive T cells. In contrast, in general IL-15 manifests antiapoptotic actions and inhibits IL-2-mediated AICD. IL-15 stimulates persistence of memory phenotype CD8+ T cells, whereas IL-2 inhibits their expression. Humanized monoclonal antibodies that recognize IL-2Ra, the privat

Transplantation of cultured postnatal human thymus was performed in a patient with complete DiGeorge syndrome. Biopsy of the graft 3 mo after implantation revealed normal CD1+ thymocytes in thymic cortical epithelial regions and CD1- thymocytes in thymic medullary epithelial regions, respectively. HLA analysis of graft thymocyte and thymic microenvironment components demonstrated that developing thymocytes and thymic macrophages were recipient derived, while thymic epithelial components were of donor origin. The patient, who initially had no T cells and had profoundly defective T cell function

The MHC class I molecules play a pivotal role in triggering cellular immune responses, binding and presenting intracellularly derived peptide antigens. Studies of MHC class I expression revealed a complex regulatory mechanism that integrates tissue-specific and hormonal modulation. Dynamic regulation occurs in the thyroid, in response to hormonal repression by TSH and stimulation by thyroid hormone. This dynamic cycle provides the basis for proposing the model that such regulation is important to maintain tolerance to self-antigens in tissues synthesizing large amounts of secretory proteins. F

The central factor in the pathogenesis of preeclampsia appears to be placental ischaemia which is supposed to be caused by an initial defective placentation. The impaired placental function leads to generalized endothelial cell dysfunction, which in its turn gives rise to hypertension, proteinuria, oedema, thrombocytopenia and hypoperfusion, especially of liver and kidneys. The link between placental ischaemia and endothelial dysfunction could consist of a toxic circulating 'factor X' and the multifactorial origin of the defective placentation. Impaired development of immunological tolerance m

Overt malnutrition in children with cancer is seen with surprising frequency: up to 37.5% in a group of patients with disease metastatic to or from bone, and 17.5% in a group of newly diagnosed patients with abdominal or pelvic tumors. It appears more frequent in some cancers. e.g., Ewing's sarcoma, than in others, e.g., osteosarcoma. Criteria for diagnosis of overt malnutrition are applicable to the child with cancer. Such overt malnutrition can be successfully and safely treated with intravenous hyperalimentation (IVH). Marginal malnutrition is a state that can be inferred from clinical beha

Melanoma remains a lethal malignancy with frequent resistance to immune checkpoint blockade therapy. Clinical evidence associates elevated PAK4 expression with poor response to anti-PD-1 treatment in melanoma patients. While tumor-derived exosomes are recognized as key immunosuppressive mediators, the role of PAK4 in regulating exosome secretion to induce immunosuppression remains incompletely understood. Here, we demonstrate that PAK4-mediated phosphorylation of Annexin-A2 (ANXA2) at Ser26 promotes its nuclear-to-cytoplasmic translocation, facilitating the formation of S100A10-ANXA2 heterotet

Cancer-associated fibroblasts (CAFs) shape the immune microenvironment in triple-negative breast cancer (TNBC), yet widely used CAF markers such as fibroblast activation protein (FAP) are not CAF-specific and are predominantly upregulated in activated stroma, limiting early-window stratification. Reasoning that tumors hijack tissue-repair programs, we investigated CD201 (PROCR), a marker of multipotent fibroblast progenitors that can diverge toward inflammatory and myofibroblastic lineages, as an earlier stromal state. Single-cell RNA sequencing of 10 TNBC patients, supported by cross-species

BACKGROUND: Lymphopenia and lymphocytic metabolism have been individually associated with poor prognosis in sepsis; however, the relationship between these factors remains poorly understood. We evaluated whether lymphocyte count and lymphocytic mitochondrial metabolism are associated with the development of persistent inflammation, immunosuppression, and catabolic syndrome (PICS).
METHODS: A prospective cohort study was conducted in four clinical-surgical intensive care units (ICUs). We included critically ill patients with sepsis requiring vasopressor support and assessed mitochondrial metab

Disease tolerance is a defence strategy essential for survival of infections, limiting physiological damage without killing the pathogen1,2. The disease course and pathology an infection may cause can change over the lifespan of a host due to the structural and functional physiological changes that accumulate with age. Because successful disease tolerance responses require the host to engage mechanisms that are compatible with the disease course and pathology caused by an infection, we predicted that this defence strategy would change with age. Animals infected with a 50% lethal dose (LD50) o

INTRODUCTION: Sepsis is associated with immunosuppression, predisposing patients to secondary infections. Many treatments routinely used for infections have immunomodulatory effects, including antibiotics. We therefore assessed the immunomodulatory effects of beta-lactam antibiotics on monocyte and lymphocyte immunophenotype.
METHODS: Peripheral blood mononuclear cells isolated from Emergency Department patients with bacterial infection were incubated with narrow-spectrum (amoxicillin and cefuroxime) or broad-spectrum (piperacillin-Tazobactam and meropenem) beta-lactam antibiotics at low and

BACKGROUND: Mycobacterium tuberculosis (MTB) evades host immunity and maintains chronic infection, in part by reprogramming macrophage function. The chemokine MCP-1 and its receptor CCR2 play a key role in attracting monocytes and immunological modulation, but their exact involvement in MTB pathogenesis is unknown.
METHODS: Using the H37Ra-infected mouse model, the expression of MTB virulence marker ESAT-6 and autophagy marker Beclin-1 was assessed. Transcriptome analysis was performed to identify CCR2-related gene expression changes and enriched pathways. In addition, the effects of CCR2 ant

Reactive oxygen species (ROS) promote genomic instability and fuel oncogenic signaling in cancer, but antioxidant therapies have so far failed to improve, or worsen, cancer outcomes. Emerging data suggest that T cells depend on ROS for signal transduction. In this study, we show that tumors exploit this dependency, releasing antioxidant enzymes into the tumor environment to suppress T cell-mediated antitumor immunity. The interstitial fluid of tumors possesses potent antioxidant activity, associated with enrichment of the antioxidant enzyme peroxiredoxin 1 (PRDX1). Extracellular PRDX1 deprives

Tolerogenic dendritic cells (tolDCs) that dampen T cell responses can be induced from blood monocytes in vitro using factors such as Vitamin D3 (VitD), dexamethasone, IL-10, or rapamycin. However, challenges remain in obtaining robust and efficient generation of cell therapy-based tolDCs without compromising their viability. We recently reported that CCR2-dependent recruitment of monocytic cells, with the capacity to dampen T-helper responses, occurs in mice treated with a single-stranded oligonucleotide (ssON). Here, we investigated the effects of this immunomodulatory noncoding ssON on diffe

OBJECTIVE: Rheumatoid arthritis (RA) is a common inflammatory autoimmune disease. Previous studies have emphasized tolerogenic dendritic cells(tolDCs) could attenuate inflammatory lesions by inducing specific immune tolerance in RA animal models, but the mechanism still needs further investigation. This study focused on revealing the effects of tolDCs on the TLR4/MyD88/NF-κB signaling pathway that mediates inflammation.
METHODS: Bone marrow-derived tolDCs were induced by IL-4, GM-CSF and NF-κB Oligonucleotide Decoys. The DC-specific molecule OX-62 and co-stimulatory molecules CD80 and CD86 on

Toleration of a semi-allogeneic fetus in the mother's uterus as well as tolerance after allogeneic hematopoietic stem cell transplantation (HSCT) appear to share some immunologic concepts. The existence of microchimeric cells, and the original idea of a bidirectional cell trafficking between mother and child during pregnancy have been known for decades. Today, origins and mechanisms of persistence of microchimeric cells are intensively being elucidated. Both, the translation of the phenomenon of feto-maternal immune tolerance to donor choice or prevention of graft-versus-host disease (GvHD) in

BACKGROUND: Guillain-Barré syndrome (GBS) is an auto-inflammatory peripheral nerve disease. Dendritic cell-mediated T cell polarization is of pivotal importance in demyelinating lesions of peripheral nerves and nerve roots. However, the regulatory function of VX-509 (Decernotinib)-modified tolerogenic dendritic cells (VX-509-tolDCs) during immune remodeling following GBS remains unclear. Here, we used experimental autoimmune neuritis (EAN) as a model to investigate these aspects of GBS.
METHODS: DCs were treated with varying concentrations of VX-509 (0.25, 1, and 4 μM) or served as a control

Aire controls immunological tolerance by driving promiscuous expression of a large swath of the genome in medullary thymic epithelial cells (mTECs). Its molecular mechanism remains enigmatic. High-resolution chromosome-conformation capture (Hi-C) experiments on ex vivo mTECs revealed Aire to have a widespread impact on higher-order chromatin structure, disfavoring architectural loops while favoring transcriptional loops. In the presence of Aire, cohesin complexes concentrated on superenhancers together with mediator complexes, while the CCCTC-binding factor (CTCF) was relatively depleted from

In this study, we studied the long-term proliferation trajectory of myeloid-derived suppressor cells (MDSCs) in murine sepsis model and investigated whether swertianolin could modulate the immunosuppressive function of MDSCs. A murine sepsis model was established by cecal ligation and perforation (CLP), according to the Minimum Quality Threshold in Pre-Clinical Sepsis Studies (MQTiPSS) guidelines. The bone marrow and spleen of the mice were collected at 24 h, 72 h, 7 and 15 d after sepsis induction. The proportions of monocytic-MDSCs (M-MDSCs; CD11b+LY6G-LY6Chi) and granulocytic-MDSCs (G-MDSC,

The autoimmune disease type 1 diabetes is predominantly mediated by CD8+ cytotoxic T-cell destruction of islet beta cells, of which islet-specific glucose-6-phosphatase catalytic subunit-related protein (IGRP)206-214 is a dominant target antigen specificity. Previously, we found that a liposome-based antigen-specific immunotherapy encapsulating the CD4+ T-cell islet epitope 2.5mim together with the nuclear factor-κB inhibitor calcitriol induced regulatory T cells and protected from diabetes in NOD mice. Here we investigated whether the same system delivering IGRP206-214 could induce antigen-sp
