Preliminary studies on the inflammatory actions of the venoms of some Australian spiders.

Experiments were performed using an anaesthetised rat model to investigate the local inflammatory responses produced by intradermal injections of crude venom gland extracts from a number of Australian spiders, namely, Phonognatha graeffei, Delena cancerides, Isopeda montana, Badumna insignis, Lampona cylindrata, Steatoda grossa, S. capensis Hann. All of the venom gland extracts tested, with the exception of that from S. capensis, produced increases in vascular permeability consistent with acute inflammatory responses. The responses primarily involved the activation of 5-HT receptors, since the

Salicylates for ulcerative colitis--their mode of action.

Delivery of 5-aminosalicylic acid to the colon by sulphasalazine, other azo-bonded compounds and controlled-release preparations is introduced in the context of metabolism by epithelial cells and therapeutic efficacy in ulcerative colitis. Potential modes of action are then reviewed, including actions on luminal bacteria, epithelial cell surface receptors, cellular events (such as nitric oxide release or butyrate oxidation), electrolyte transport and epithelial permeability. Evidence for an influence of salicylates on circulating and lamina propria inflammatory cells is presented, as well as a

Cytokines as therapeutic and diagnostic agents.

Cytokines are key mediators of immunity and inflammation. These proteins or glycoproteins act as communication signals between different populations of leukocytes but neither their effects nor their production are restricted to immune cells. In the last few years many new cytokines and their effects have been discovered and it was agreed that when the amino acid sequence of a new cytokine was established it would be assigned the name interleukin (IL), with an added number. The detection of cytokines in disease states promises to provide useful information for diagnostic purposes. The therapeut

[Growth factors in the pathogenesis of progressive systemic sclerosis].

Scleroderma is a connective tissue disorder characterized by vascular lesions, fibrosis and inflammation. The pathogenesis of this disease is not clear. A vascular lesion, possibly caused by deposition of immune complexes or by release of cytotoxic factors, seems to be at the origin of the disease. As a consequence, platelet adhesion and activation might occur in sclerodermic patients. The observation that platelet might release, upon aggregation, a potent mitogenic factor, named Platelet Derived Growth Factor (PDGF) has focused interest on platelets as the potential mediators of the fibrotic

The Inflammatory Basis of Acne Pathogenesis.

BACKGROUND: Acne is a common inflammatory and genetically influenced skin disease. Previous genome-wide association studies and epigenome-wide association studies have demonstrated that genetic factors constitute an important basis for acne susceptibility, while recurrent and persistent local inflammatory responses contribute to its chronic and relapsing nature. SUMMARY: Infection with Cutibacterium acnes, activation of Toll-like receptors and the NLRP3 inflammasome, and dysregulation of the IL-6/STAT3 and Notch signaling pathways induce the release of inflammatory mediators and immune activa

TLR7 and TLR9 have overlapping but distinct roles in systemic lupus erythematosus and Sjögren's disease.

Endosomal toll-like receptors (TLRs), such as TLR7 and TLR9, are key mediators of autoimmunity. Systemic lupus erythematosus (SLE or lupus) and SjD are distinct diseases that are both characterized by heightened activation of TLR7 and TLR9 signaling networks that serve as potent modulators of chronic inflammation. This review will provide an overview of the role of these receptors in lupus and SjD, with a focus on recent mechanistic insights in the field. We will compare and contrast the roles of TLR7 and TLR9 in lupus and SjD, with a focus on the importance of B cell activation in disease. Mo

African swine fever virus B66L drives extracellular mitochondrial release to promote systemic inflammation in mice.

Systemic inflammation is a hallmark of viral infection, but the upstream signals that initiate it remain poorly defined. Here we show that extracellular mitochondria act as inflammatory mediators during infection by vesicular stomatitis virus, influenza A virus, rabies virus, herpes simplex virus 1 and African swine fever virus (ASFV). In ASFV-infected primary porcine alveolar macrophages, the viral protein B66L promotes the capture of damaged mitochondria by autophagosomes while blocking their fusion with lysosomes, causing mitochondria to accumulate outside cells. Extracellular mitochondria

Mechanistic investigation of DDT/DDE in MASLD/MASH pathogenesis: An integrated network toxicology and transcriptomics approach.

Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) and Metabolic Dysfunction-Associated Steatohepatitis (MASH) pose significant public health challenges, yet the role of environmental pollutants remains inadequately characterized. Here, we systematically dissect the hepatotoxic mechanisms of DDT and its metabolite DDE through a multi-platform strategy. Network toxicology and PPI analysis pinpointed IL-6, ALB, AKT1, TNF, and TP53 as central mediators, with molecular docking validating DDT's high-affinity binding. Transcriptomic and KEGG pathway analyses uncovered broad dysregulati

Fecal Microbiota Transplantation in Reducing Uremic Toxins Accumulation in Kidney Disease: Current Understanding and Future Perspectives.

During the past decades, the gut microbiome emerged as a key player in kidney disease. Dysbiosis-related uremic toxins together with pro-inflammatory mediators are the main factors in a deteriorating kidney function. The toxicity of uremic compounds has been well-documented in a plethora of pathophysiological mechanisms in kidney disease, such as cardiovascular injury (CVI), metabolic dysfunction, and inflammation. Accumulating data on the detrimental effect of uremic solutes in kidney disease supported the development of many strategies to restore eubiosis. Fecal microbiota transplantation (F

Adipose tissue macrophages: implications for obesity-associated cancer.

Obesity is one of the most serious global health problems, with an incidence that increases yearly and coincides with the development of cancer. Adipose tissue macrophages (ATMs) are particularly important in this context and contribute to linking obesity-related inflammation and tumor progression. However, the functions of ATMs on the progression of obesity-associated cancer remain unclear. In this review, we describe the origins, phenotypes, and functions of ATMs. Subsequently, we summarize the potential mechanisms on the reprogramming of ATMs in the obesity-associated microenvironment, incl

Involvement of Cytokines in the Pathogenesis of Diabetic Macular Edema.

Diabetic macular edema (DME) is a critical complication of diabetic retinopathy, a condition that arises from the breakdown of the blood-retinal barrier and the consequent increase in vascular permeability. Over the years, attempts have been made to treat DME by various approaches, including laser photocoagulation, steroid triamcinolone acetonide, and vitrectomy. However, treatment was unsatisfactory until research identified vascular endothelial growth factor (VEGF) as a factor in the pathogenesis of DME. Intraocular anti-VEGF agents show good efficacy in DME. Nevertheless, in some patients t

T Cell-Mediated Nasal Hyperresponsiveness in Allergic Rhinitis.

Allergic rhinitis patients suffer various symptoms such as sneezing, runny nose, and nasal congestion. As disease severity and chronicity progress, nasal hyperresponsiveness (NHR) develops in those patients. During the generation of a mouse allergic rhinitis model, we discovered that immunized mice developed NHR upon repeated nasal antigen challenge. Using genetically modified mice and an originally developed T cell-transferred mouse model, we confirmed the critical role of CD4+ T cells after differentiation into several helper subsets in NHR. On the other hand, immunoglobulin E/mast cell-depe
