Sex-specific prediction models for aortic aneurysm integrating traditional clinical risk factors and proteomic profiles: a large-scale prospective study from the UK Biobank.

BACKGROUND: This study aimed to systematically map the sex-specific clinical and proteomic risk profiles of aortic aneurysm (AA), elucidate its molecular mechanisms, and develop a sex-specific protein risk prediction score. METHODS: Based on the UK Biobank, we adopted a sex-stratified strategy to assess associations between traditional clinical factors and AA in 471,660 participants, and performed proteomics analysis on 49,887 participants with plasma protein data. Mediation analysis was used to explore the molecular mechanisms by which clinical risks drive AA. Finally, sex-specific protein r

Integrative Genetic and Bioinformatic Analysis of Androgen Pathway Gene Polymorphisms, Testosterone, and PSA Levels in Prostate Cancer.

BACKGROUND: Prostate cancer (PCa) is genetically a complicated disease with mediation by variations in genes coding for androgen biosynthesis and signaling genes. In the current study, we investigate the associations of key polymorphisms in the androgen pathway with serum testosterone and prostate-specific antigen (PSA) levels, while also exploring their potential functional impact using bioinformatics analyses. METHODS: A total of 314 patients with prostate adenocarcinoma and 287 age-matched healthy controls were genotyped for ten SNPs across CYP17A1, HSD3B1, HSD3B2, SRD5A2, and AR genes usi

Violence exposure and quality of life among partners and parents of individuals with substance use problems.

BACKGROUND: Substance use problems (SUP) have significant adverse consequences not only for individuals with such problems, but also for their loved ones. One important concern is exposure to violence among affected family members. This study investigates whether close family members, specifically partners or parents, of individuals with SUP, including alcohol, face increased risks of physical and sexual violence and emotional abuse within the last 5 years. We also examined a hypothesized mediation model linking SUP exposure, violence, and quality of life (QoL). METHODS: We conducted a cross-

Nonlinear Relationship Between Lactate/Albumin Ratio and 28-Day ICU Mortality in Patients With Congestive Heart Failure: Insights Into Inflammatory and Metabolic Interplay.

BACKGROUND: Congestive heart failure (CHF) among critically ill patients is a significant cardiovascular disorder that is linked to elevated mortality rates. The lactate/albumin ratio (LAR) is a readily available clinical metric that may provide valuable information regarding the metabolic and nutritional conditions. However, its association with mortality in patients with CHF remains unexplored. This study aimed to investigate the association between LAR and 28-day intensive care unit (ICU) mortality among critically ill patients with CHF. METHODS: Using the eICU Collaborative Research Datab

Associations of gut bacterial classes Clostridia and Deltaproteobacteria with type 2 diabetes and Alzheimer's disease: A two-sample Mendelian randomization study.

Using a 2-sample Mendelian randomization (MR) approach, we evaluated MR evidence for genetically proxied effects of 10 pre-specified gut bacterial classes on type 2 diabetes mellitus (T2DM) and Alzheimer disease (AD), and assessed MR evidence for a direct genetically proxied effect of T2DM on AD. Mediation by T2DM was explored where feasible. We performed 2-sample MR using MiBioGen gut microbiota GWAS summary statistics as exposure, and DIAMANTE (T2DM) and European AD GWAS data as outcomes. Inverse-variance weighted (IVW) MR was the primary analysis, with MR-Egger, weighted median, and weighte

Propensity score matched cohort study on magnesium supplementation and mortality in critically ill patients with HFpEF.

Heart failure with preserved ejection fraction (HFpEF) emerges as a singular subclass of heart failure, bereft of specific therapeutic options. Magnesium, an indispensable trace element, is essential to the preservation of cardiac integrity. However, the association between magnesium supplementation and mortality in HFpEF patients remains unclear. This study extracted HFpEF patient data from the MIMIC-IV database between 2008 and 2019. Propensity score matching was conducted to ensure that patients receiving magnesium supplementation (including magnesium sulfate and magnesium oxide) were balan

Immune, blood-brain barrier, and metabolic biomarkers mediate gut-brain axis crosstalk in alzheimer's disease.

BACKGROUND: Gut microbiota may influence Alzheimer's disease (AD) pathogenesis by modulating host homeostasis. However, population-based causal evidence linking gut dysbiosis to Alzheimer's disease pathogenesis, especially via immune, vascular, and metabolic pathways, remains insufficient. METHODS: We performed Mendelian randomization (MR) and colocalization analysis on 629 gut microbiota features and 2,103 immune, blood-brain barrier (BBB), and metabolic biomarkers regarding the risk of AD and cerebrospinal fluid (CSF) pathological biomarkers. RESULTS: We identified that mucin-degraders, sh
