Localization of puff sites adjacent to the plasma membrane: functional and spatial characterization of Ca2+ signaling in SH-SY5Y cells utilizing membrane-permeant caged IP3.

The Xenopus oocyte has been a favored model system in which to study spatio-temporal mechanisms of intracellular Ca2+ dynamics, in large part because this giant cell facilitates intracellular injections of Ca2+ indicator dyes, buffers and caged compounds. However, the recent commercial availability of membrane-permeant ester forms of caged IP3 (ci-IP3) and EGTA, now allows for facile loading of these compounds into smaller mammalian cells, permitting control of [IP3]i and cytosolic Ca2+ buffering. Here, we establish the human neuroblastoma SH-SY5Y cell line as an advantageous experimental syst

Pharmacological activation of plasma-membrane KATP channels reduces reoxygenation-induced Ca(2+) overload in cardiac myocytes via modulation of the diastolic membrane potential.

1. The opening of cardiac plasma-membrane ATP-sensitive K(+) channels (pmK(ATP)) can protect the heart against ischaemia/reperfusion injury. We recently demonstrated that the resting membrane potential (E(m)) of ventricular myocytes strongly modulates reoxygenation-induced Ca(2+) overload. This led to the hypothesis that activation of pmK(ATP) can influence the extent of chemically induced hypoxia (CIH)/reoxygenation Ca(2+) overload via hyperpolarization of the diastolic membrane potential of ventricular myocytes. 2. The membrane potential (E(m)) of isolated rat myocytes was determined using t

Modulated drug release using iontophoresis through heterogeneous cation-exchange membranes. 2. Influence of cation-exchanger content on membrane resistance and characteristic times.

An implantable drug delivery method using iontophoresis through cation-selective membranes was further developed. Heterogeneous cation-exchange membranes (HCMs) were prepared by mixing conductive sulfonated polystyrene beads into a nonconductive silicone rubber matrix. The membrane resistivity and lag time to steady-state transport of two salts, (+/-)-phenylpropanolamine hydrochloride (PPA) and NaCl, were evaluated during constant current iontophoresis at 37 degrees C as a function of the resin content in the HCMs. A continuous decline in membrane resistivity was observed as fractional resin c…

Comet Assay for Genotoxicity Evaluation of Salmonella Outer Membrane Vesicles.

Outer membrane vesicles (OMVs) from different Gram-negative bacteria, including Salmonella spp., have appeared as potential acellular vaccine candidates for their role in host-pathogen interaction by delivering virulence factors and modulating immune response. Given this host-cell contact, it is crucial to assess their genotoxic effects to ensure safety as vaccine candidates. The Comet assay, a sensitive and effective method for detecting single-cell DNA strand breaks, offers an effective approach for evaluating such effects. This study explores the application of the Comet assay as a highly s

The Membrane Continuum Governing Cadmium Uptake, Partitioning, and Detoxification in Plants.

Cadmium (Cd) contamination threatens agro-ecological safety and food security; thus, elucidating mechanisms of Cd uptake and accumulation in plants is key to breeding low-Cd crops. Within the defensive network plants deploy against Cd stress, the plasma membrane and endomembrane system act as physical barriers to metal influx and as the core regulatory hub governing signal perception, transmembrane transport, and vacuolar compartmentalization for detoxification. This review examines the physiological and molecular mechanisms by which the cell membrane system influences Cd transport and accumul

Membrane Formulation Impacts Vesicle-Based Artificial Cell Functions.

Engineering vesicle-based artificial cells capable of mimicking cellular processes is a central objective in synthetic biology. However, while giant unilamellar vesicles (GUVs) have become a popular artificial cell platform, the impact of fundamental design features remains obscure. Here, we assembled GUVs composed primarily of two phospholipids: POPC, a phosphatidylcholine widely employed in GUV preparation, and DOPC, a phosphatidylcholine with a lower melting temperature and higher fluidity. We found that the phospholipid mixture leads to an increase in average GUV size (compared to POPC onl

Metabolic profiles of IgA nephropathy, membranous nephropathy, and diabetic nephropathy.

INTRODUCTION: Immunoglobulin A nephropathy (IgAN), membranous nephropathy (MN), and diabetic nephropathy (DN) are prominent contributors to chronic kidney disease burden. Our main objective was to contribute to the understanding of the metabolic profiles of these three major types of nephropathies and identify potential metabolic biomarkers. MATERIAL AND METHODS: Kidney samples of 20 sex- and age-matched patients with biopsy-proven IgAN, MN, DN, and controls without any kidney diseases were included. Ultra-high-performance liquid chromatography-mass spectrometry analysis was conducted. The t-

A generalizable method for the purification and biophysical characterization of bacterial membrane receptors.

Membrane-embedded bacterial receptors are challenging to express and purify in soluble form, yet their isolated domains are essential tools for structural and ligand-discovery studies. Pseudomonas aeruginosa relies on the TonB-dependent heme receptor HasR for iron acquisition, a process central to its pathogenicity. Here, we report a robust strategy for the recombinant expression, purification, and biophysical characterization of the two soluble HasR domains directly involved in heme uptake: the N-terminal plug and the Secretin/TonB short N-terminal domain. Each domain was expressed individual

Deep Dehydrated Layered Vermiculite Membrane for Selective Lithium Separation.

Two-dimensional (2D) layered membranes hold great promise in water treatment due to their simple film formation, tunable interlayer spacing, and designable surface chemistry. However, their practical application of aqueous ion sieving is largely constrained by swelling problems. Here, we report a deep dehydration strategy to suppress reswelling after ion-intercalation by removing free and bound water within the interlayer channels. The deep dehydration drives the interlayer free spacing below a critical threshold, enhancing van der Waals and electrostatic interactions, thereby energetically in…

Clustering hinders APP α-secretase processing in the plasma membrane.

Alzheimer's disease is associated with the extracellular accumulation of neurotoxic Aβ-peptides in the brain. Aβ-peptides are produced via an amyloid precursor protein (APP) cleavage pathway that is initiated by the β-secretase. The majority of APP circumvents the amyloidogenic pathway due to α-secretase cleavage at the plasma membrane. In this study, we set out to identify potential mechanisms limiting α-cleavage. We employed isolated cell membranes to study α-secretase cleavage in the absence of APP delivery to the plasma membrane and internalization. We distinguish a readily cleavable and a…

Behind the membranous curtain-lipid dynamics and functions in coronaviral replication.

Lipids are naturally occurring hydrophobic biomolecules characterized by remarkable structural diversity. This includes various types of head groups, varying fatty acid chain lengths, degrees of unsaturation, and stereochemical configurations. Such variability enables lipids to serve multiple biological functions, such as forming membranes, storing energy, and facilitating signaling. Given their diverse roles, it is not surprising that approximately 5% of genes in eukaryotic cells are involved in lipid biosynthesis pathways. The multifunctional nature of lipids also makes them attractive targe…

Light-Controlled Conductive Molecular Wire in an Artificial Lipid Membrane.

In this work, we investigate a novel photochromic azobenzene derivative, TC827, which exhibits a unique photoelectric response when embedded in artificial lipid membranes. By combining spectroscopic, electrochemical, and molecular simulation analyses, we demonstrate that TC827 forms light-controlled "molecular wires" in its trans conformation, resulting in a significant increase in membrane conductivity under dark conditions, which is fully reversible, as light-induced cis isomerization suppresses the conductivity enhancement. This behavior is fundamentally different from previously studied am…

Lipid ciliology: specialized ciliary membrane lipids in physiology and disease.

Primary cilia are the microtubule-based sensory organelles. The unique lipid makeup of the ciliary membrane strictly controls their signaling ability, to orchestrate tissue formation and homeostasis. Emerging evidence has demonstrated that lipids play important roles in cilia formation and cilia-related signaling, solidifying the previously proposed concept of "lipid ciliology." The ciliary membrane exhibits a highly specialized lipid composition, including enrichment in cholesterol, sphingolipids, and specific phosphoinositides, compared with the surrounding plasma membrane. Recent studies ha
