The growing crisis of multidrug-resistant (MDR) Gram-negative bacteria requires that current technologies permit the rapid detection of extended-spectrum β-lactamase (bla(ESBL)) and Klebsiella pneumoniae carbapenemase (bla(KPC)) genes. In the present study, we assessed the performance characteristics of a commercially available nucleic acid microarray system for the detection of bla(ESBL) and bla(KPC) genes directly from positive blood cultures. Using blood cultures (BCs) that contained Gram-negative bacilli identified by Gram staining, we isolated bacterial DNA using spin columns (BC-C) and r

The best screening method for detecting heteroresistant vancomycin-intermediate Staphylococcus aureus (hVISA) remains unclear. Using population analysis profiling utilizing the area under the concentration-time curve (PAP-AUC) as the gold standard, we screened 458 consecutive methicillin-resistant S. aureus (MRSA) bloodstream isolates to determine the most accurate and cost-effective testing strategy to detect the presence of heteroresistance. All isolates were also tested using the macromethod Etest (MET) and glycopeptide resistance detection (GRD) Etest. The MIC was determined by several met

Bacillus anthracis, the bacterium that causes anthrax, is an agent of bioterrorism. The most effective antimicrobial therapy for B. anthracis infections is unknown. An in vitro pharmacodynamic model of B. anthracis was used to compare the efficacies of simulated clinically prescribed regimens of moxifloxacin, linezolid, and meropenem with the "gold standards," doxycycline and ciprofloxacin. Treatment outcomes for isogenic spore-forming and non-spore-forming strains of B. anthracis were compared. Against spore-forming B. anthracis, ciprofloxacin, moxifloxacin, linezolid, and meropenem reduced t

BACKGROUND: There is bidirectional communication between the gut microbiota and the brain. Empirical evidence has demonstrated sex differences in both the gut microbiome and the brain. However, the effects of sex on the gut microbiota-brain associations have yet to be determined. We aim to elucidate the sex-specific effects of gut microbiota on brain and cognition.
METHODS: One hundred fifty-seven healthy young adults underwent brain structural, perfusion, functional and diffusion MRIs to measure gray matter volume (GMV), cerebral blood flow (CBF), functional connectivity strength (FCS) and w

Type 2 diabetes mellitus (T2DM) is typically accompanied by sudden weight loss, dyslipidemia-related indicators, decreased insulin sensitivity, and altered gut microbial communities. Fagopyrum tataricum possesses many biological activities, such as antioxidant, hypolipidemic, and hypotensive activities. However, only a few studies have attempted to elucidate the regulatory effects of F. tataricum ethanol extract (FTE) on intestinal microbial communities and its potential relationships with T2DM. In this study, we established a T2DM mouse model and investigated the regulatory effects of FTE on

We previously discovered new dimerization compounds of 3-amino-2-naphthoic acid (3-A-2-N) produced by the cultivation of Serinicoccus marinus KDM482 with 3-A-2-N. In the present study, we investigated metabolites with a 3-A-2-N moiety and obtained two new metabolites, designated as serinaphthoic acids A (1) and B (2). A structural analysis using one-dimensional (1D) and 2D-NMR, IR, UV, and MS data revealed that 1 and 2 were both hybrid compounds consisting of the reagent 3-A-2-N and the natural moieties 2-thioformylbenzoic acid and 1-butene. In biological assays, 1 weakly inhibited the growth

Antibiotic resistance in Gram-negative bacteria is a major global health threat, in part because of the permeability barriers of their double-membrane envelope and active efflux systems. While porins, lipopolysaccharides, and efflux pumps have been extensively studied, the role of membrane energetics, particularly the Donnan potential (DP), in antibiotic susceptibility remains poorly understood. The DP arises from negatively charged osmoregulated periplasmic glucans (OPGs), which generate an electrostatic potential in the periplasm that is capable of influencing how molecules traverse the oute

Nontuberculous mycobacteria (NTM) are important pathogens of fish, causing chronic infections that are difficult to control in aquaculture. In this study, we evaluated the in vitro antimicrobial activity of twelve plant-derived compounds against six fish-associated NTM strains using a broth microdilution assay. Minimum inhibitory concentrations (MICs) ranged from 0.25 to >32 µg/mL. Our results demonstrated that cinnamaldehyde exhibited the strongest antimycobacterial effect, followed by sesamol and farnesol, while several other phytochemicals showed moderate activity and salicylic acid was ina

Bacillus cereus is a spore-forming, toxin-producing bacterium responsible for the spoilage and contamination of a wide variety of foods, posing a risk to consumer health. Its spores are more resistant than vegetative cells and can survive in food production environments even after decontamination, representing a high food safety challenge. Thermal treatments of contaminated foods cause bacterial cell lysis and toxin release, while altering their organoleptic and nutritional properties. This is why non-thermal methods are increasingly favored to meet the demands of consumers seeking fresher and

The antimicrobial resistance crisis urgently calls for antibiotics with novel modes of action (MoAs). While growth inhibition assays can identify antibiotic molecules, they miss promising compounds below inhibitory concentrations and cannot reveal their MoA. Imaging-based profiling of drug-treated bacteria can inform on MoA, but current approaches generally require fluorescent labelling and/or inhibitory concentrations and it remains unclear whether compounds with novel MoAs can be robustly detected. Here, we demonstrate a deep learning approach to recognise antibiotic MoAs from unlabelled ima
