Immunoglobulin A (IgA) is a crucial component of the human immune system, and its interaction with receptors is essential for immune regulation. Fc-receptor-like 4 (FcRL4) is an IgA receptor that selectively binds systemic IgA containing the joining chain (J-chain). The molecular mechanism of this interaction has remained unclear. Here, we present a cryo-EM structure of FcRL4 complexed with the dIgA core (Fcα dimer and J-chain), revealing a 1:1 binding stoichiometry. FcRL4 primarily interacts with the J-chain but can nevertheless discriminate against J-chain-containing IgM through an entropic

Amyloid oligomers of the human islet amyloid polypeptide (hIAPP) are a likely cytotoxic species driving β-cell death in type 2 diabetes, but their transient nature has precluded atomic-level structural characterization. We obtained a high-resolution structure of a physiologically relevant hIAPP oligomer. Using 2D IR spectroscopy, we identified three substitutions that slowed aggregation sufficiently for comprehensive 2D/3D NMR analysis while retaining the key wild-type structural features and cytotoxicity. The structural model reveals a dimeric assembly with N-terminal helices and a kink that

Macromolecular complexes are cells' functional units, and their correct and efficient assembly is critical to life's processes. Complex assembly was classically described as the encounter of fully synthesized, mature protein subunits, yet an amalgam of current studies shows that cotranslational assembly is prevalent, in which nascent proteins vectorially form interfaces with their partners during translation. In this review, we examine the advances in this emerging field. We discuss the thermodynamic and kinetic principles underlying different modes of assembly and highlight how the specific s

The advent of machine learning structural prediction tools has largely accelerated bottom-up structural biology and provides static Anfinsenian models with near-experimental accuracy, complementing X-ray crystallography and cryogenic electron microscopy. However, these techniques often overlook two fundamental aspects of biomolecular structure-function relationships: (i) conformational equilibria and (ii) transient noncovalent interactions. Indeed, these considerations are particularly important for integral membrane proteins (MPs), frequently serving mechanistic roles, such as conformational

The giant E2-E3 chimera BIRC6 specifically engages ubiquitin-activating E1 enzyme UBA6, but not UBA1, enabling UBA6 to transfer activated ubiquitin to BIRC6, which then ubiquitinates downstream substrates such as caspases to regulate apoptosis. However, the molecular mechanism that underlies UBA6-mediated specific engagement of BIRC6 remains elusive. Here we use a UbDha probe to trap the transient E1-E2 transthioesterification intermediate and determine cryo-EM structures of singly and doubly loaded UBA6-BIRC6UBC complexes at 3.3 and 3.4 Å resolution. Structural analysis reveals that an insert

Glycosaminoglycan (GAG)-binding lectins represent a rare class of carbohydrate-binding proteins with the ability to recognize and organize linear polysaccharide chains. Here, we describe XN-IL, a novel calcium-dependent lectin from the Gram-negative bacterium Xenorhabdus nematophila, which exhibits an unusual specificity for glycosaminoglycans. X. nematophila is an entomopathogenic bacterium and a symbiont of insect-parasitic Steinernema nematodes. Glycan array screening, analytical ultracentrifugation, and differential scanning fluorimetry revealed that XN-IL selectively binds hyaluronan and

Protein-ligand binding is largely determined by multiple noncovalent interactions (NCIs), which may exhibit synergistic or antagonistic effects. Here, we applied an approach combining PDB statistics with QM/MM calculations to quantify relationships among five major NCIs: halogen bonds (XB), hydrogen bonds (HB), cation-π interactions (CP), π-π interactions (PP), and salt bridges (SB). Statistical analysis showed that 69.28% of the ligands with SB acceptors interact with protein residues, followed by HB acceptors (65.21%) and XB donors (62.76%). Energy analysis revealed the strongest antagonism

Corrinoid iron‑sulfur protein (CFeSP) is the methyltransferase in the Wood-Ljungdahl pathway, yet the geometric and electronic structure that enables its higher catalytic efficiency relative to small-molecule analogs remains a subject of debate. While axial ligation is known to tune the reactivity of B12-dependent enzymes, weak interactions in "base-off" species like methylated CFeSP (Me-CFeSP) have been difficult to define using standard structural probes, despite that solution-phase spectroscopy has suggested their presence. Here we utilize Co K-edge X-ray absorption spectroscopy (XAS) and t

Cisplatin remains a widely used anticancer agent; however, its effectiveness can be influenced by systemic toxicity, resistance mechanisms, and interactions with exogenous compounds. Nicotinamide (vitamin B3), an NAD+ precursor and a commonly used dietary supplement, is involved in cellular metabolism, redox homeostasis, and DNA repair pathways, which may potentially modulate the cellular responses to Platinum-based agents. Here, we combine chemical synthesis, computational studies, spectroscopic analysis, and biological assays to investigate the molecular and biological aspects of Cisplatin-N

Exploring the natural diversity of phylogenetically distant nitrogenases is crucial for gaining new insights into the mechanism of atmospheric N2 fixation and unlocking biotechnological developments in sustainable ammonia production. Here, we investigated the N2-fixing system of Methanocaldococcus infernus, a deep-sea hyperthermophilic archaeon growing diazotrophically at 92 °C. This natively isolated nitrogenase has a melting temperature close to the water boiling point, with an extrapolated specific activity superior to mesophilic homologues. The crystal structures obtained at near-atomic re

Metal nanoclusters supported on metal oxides are known to exhibit high catalytic performance in a range of reactions, with their shape and oxidation state, key factors influencing catalytic behavior, often governed by the support material. Herein, density functional theory (DFT) is employed to investigate the adsorption and activation of carbon dioxide (CO2) on an icosahedral Cu13 cluster supported on the (110) surface of γ-alumina (γ-Al2O3), with a focus on the effects of nickel and platinum (NiCu12 and PtCu12) doping. CO2 adsorption was first examined on isolated clusters. Enhanced binding o

Artificial enzymes constructed by introducing abiological catalytic cofactors into protein scaffolds are emerging as promising biocatalysts for reactions that have no natural equivalents. However, the design of artificial enzymes for in cellulo stereoselective biosynthesis remains largely unexplored because of the numerous challenges posed by the complex cellular environment. Here, we present an efficient strategy for the intracellular assembly of artificial enzymes via covalent anchoring by incubating cells that are expressing target protein with a synthetic catalytic cofactor, in the manner

Histone deacetylase inhibitors (HDACi) are an emerging class of epigenetic anticancer drugs that exert their activity through coordination to the catalytic Zn2+ ion within the active site of histone deacetylases (HDACs). Due to the limited isoform-selectivity of hydroxamic acid-based inhibitors, benzamide-based HDACi (BBHDACi) have been developed as subtype-selective alternatives. Clinically relevant representatives include Chidamide, Entinostat, Mocetinostat, Zabadinostat, and Tacedinaline. Although these compounds share a conserved o-aminoanilide zinc-binding group (ZBG), they differ in link

Adenylyl cyclases (ACs) convert ATP into the second messenger cAMP, thus directly influencing cellular signaling in response to a wide variety of stimuli. Despite their physiological importance, structural studies of isoform-specific AC regulation are compounded by difficulties in AC expression and purification. Here, we designed a chimeric construct AC95, combining human AC9 as a molecular scaffold and incorporating the catalytic-allosteric core of human AC5. Cryo-EM analysis of AC95 at 3.5 Å resolution revealed a state of AC95 bound to both ATPαS and forskolin, demonstrating that the chimera

The recent discovery of the isonitrile biosynthetic enzyme ScoE expanded the catalytic repertoire of the Fe(II)/αKG-dependent dioxygenase enzyme family. ScoE synthesizes an isonitrile functional group from a glycyl-fatty acid adduct, with both the isonitrile nitrogen and carbon atoms coming from the glycyl moiety. This challenging chemistry cannot be performed in a single step. Instead, the mechanism appears to require two half reactions, each involving αKG cleavage to generate a highly reactive iron-oxygen species. Here, we report sixteen crystal structures that provide snapshots along the re

In mycobacteria, the F1Fo-ATP synthase enzyme comprises five polypeptide chains (Chain α, β, γ, ε, δ) and three subunits (Subunit a, b, and c), respectively. ATP is fundamental for the survival and growth of mycobacterial strains. In tuberculosis, this enzyme survives in harsh conditions like hypoxia, fluctuations in pH, and low nutrition in the host, further coercing it to enter the dormant state. Depletion of ATP stopgap the mycobacterial strains strenuous to survive. The discovery of Bedaquiline in 2012 has validated mycobacterial ATP synthase as a substantial target to combat resistance de

Legionella pneumophila is an opportunistic human pathogen that causes atypical pneumonia called Legionnaires' Disease. To replicate within host cells, L. pneumophila injects up to 330 effector proteins into the host cytosol via a Dot/Icm type IV secretion system. Several effectors, called metaeffectors, regulate the activity of other effectors within infected host cells through direct protein-protein interactions. LegA11 (AnkJ/Lpg0436) has been identified as a metaeffector of SidL (Ceg14/Lpg0437), one of eight L. pneumophila effectors that inhibit host mRNA translation. LegA11 binds and suppre

Mycobacteria have long posed a threat to human health, being responsible for a range of hard-to-treat infections such as tuberculosis, leprosy, and other chronic diseases caused by nontuberculous mycobacteria. Their intrinsic resistance to antibiotics and resilience when exposed to environmental stress complicate treatment and underscore the need for discovering novel drug targets and developing new therapeutic strategies. Mycobacterium hassiacum, a thermophilic mycobacterium, is a promising source of stable proteins suitable for detailed structural analysis. In this study, we investigated its

Common bean (Phaseolus vulgaris) encodes three class 2 L-asparaginase enzymes: two potassium-dependent enzymes [PvAIII(K)-1 and PvAIII(K)-2] and a potassium-independent enzyme (PvAIII). Here, we present the crystal structure of PvAIII, which displays a rare P2 space-group symmetry and a unique pseudosymmetric 41-like double-helical packing. The asymmetric unit contains 32 protein chains (16 αβ units labeled A-P) organized into two right-handed coiled arrangements, each consisting of four PvAIII (αβ)2 dimers. Detailed analysis of the crystal structure revealed that this unusual packing originat

Human inflammation-related CC chemokine ligand 5 (hCCL5) has significant self-assembly property under physiological conditions. The mechanism and function of hCCL5 oligomerization remain unclear. Different intermolecular interactions, such as E66-K25 or E66-R44/K45, have been reported to mediate hCCL5 oligomerization. This complexity makes structural determination difficult. Based on a K25S mutation to eliminate the E66-K25 interaction, we observed hCCL5 forming a helical-sharped filament in transmission electron microscopy (TEM). The filamentous polymerization is a dominant process when the c
