Primary Sjogren's Disease (SjD) is characterized by features of dryness arising from inflammation in the secretary glands, particularly the salivary and lachrymal glands. Generalized symptoms of fatigue and limb pain are very common but, in addition, about 40% of patients have one or more features of organ-specific systemic disease. This review goes through the background of SjD including diagnosis and classification, epidemiology, impact and investigations such as ultrasound and lip biopsy. It then focuses in detail on each of the systemic organ-specific features, principally using the Europe

Dosage-sensitive transcription factors (TFs) underlie altered gene regulation in human developmental disorders, and cell type-specific gene regulation is linked to the reorganization of three-dimensional (3D) chromatin during cellular differentiation. In this work, we show dose-dependent regulation of chromatin organization by the congenital heart disease (CHD)-linked, lineage-restricted TF TBX5 in human cardiomyocyte differentiation. Genome organization, including compartments, topologically associated domains, and chromatin loops, was sensitive to reduced TBX5 dosage in a human model of CHD,

Macrophages play a major role in immune responses and in the maintenance of tissue homeostasis. Most of our knowledge of macrophage biology comes from mouse studies, which have revealed the existence of distinct subpopulations differing in origin, life cycle and tissue-specific features, as well as the influence of tissue perturbations on macrophage heterogeneity. Here, we review the evidence supporting the conservation of these principles in human macrophages, along with the methodological approaches employed, including omics analyses of macrophages from clinical samples, human transplantatio

Understanding how a gene variant may influence antifungal resistance, or other phenotypic characteristics, is an important step in determining or dissecting resistance mechanisms. The influence of specific genes or gene alleles on a phenotype can initially be assessed within the model organism, Saccharomyces cerevisiae. S. cerevisiae exhibits efficient rates of homologous recombination making it amendable for heterologous expression and represents a susceptible organism that can be used to determine changes in antifungal susceptibilities. Many groups have developed different methodologies for

BACKGROUND: Legionella bacteria is a common cause of pneumonia, but the infection may affect several organs in the most serious cases. A systemic involvement ab initio could be non-specific, leading to a diagnostic misinterpretation.
CASE PRESENTATION: A 33-year-old woman had been complaining of mental confusion, restlessness, aggressiveness, and, subsequently, hirsutism. After 3 weeks, the patient developed pneumonia and died during the hospitalization. The autopsy examination revealed a multi-organ necrotizing exudative disease involving the lung, the heart and the brain. The microbiologica

Activation of self-reactive T cells is a major driver to autoimmunity and is suppressed by mechanisms of regulation. In a humanized model of autoimmune thyroiditis, we investigated the mechanism underlying break of tolerance. Here, we found that a human TCR specific for the self-antigen thyroid peroxidase (TPO) is positively selected in the thymus of RAG KO mice on both T effector (Teff) and T regulatory (Treg) CD4+Foxp3+ cells. In vivo Teff are present in all immune organs, whereas the TPO-specific Treg are present in all lymphoid organs with the exception of the thyroid-draining lymph nodes.

We tested the hypothesis that brain-derived chemical stimuli contribute to direct endocrine regulation of peripheral organs during ontogeny before blood-brain barrier closure. Dopamine and gonadotropin-releasing hormone present in high concentration in peripheral blood only before blood-brain barrier closure were chosen as the chemical stimuli. It was shown than dopamine in concentrations equal to its level in the peripheral blood inhibits prolactin secretion in organotypic culture of the pituitary gland from newborn rats via specific receptors. Experiments on organotypic culture of neonatal r

Living donor kidney transplantation is regulated in Spain by the transplantation law 30/1979 and developed by Royal Decree 2070/1999. These policies permit the living donation of an organ while its function is compensated by the rest of the body. It further specifies that the destination of the organ must be the transplantation in a specific person (this specification allows for the donation, not only among emotionally related, but between people that do not know each other, provided it is done altruistically, without any purpose of profit and without coercion). The donor must be competent, he

Hashimoto's thyroiditis, the most common form of autoimmune thyroid disease, is characterised by lymphocytic infiltration of the thyroid gland, gradual destruction of the organ and production of thyroid specific auto antibodies (antithyroid peroxidase and antithyroglobulin antibodies). There are evidences that cast doubt on the pathogenetic role of these antibodies in thyroid autoimmunity. It is very likely that cellular destruction is mediated by other cellular mechanisms, such as auto reactive T-lymphocytes, natural killer and cytokines. However, other studies performed in animal models have

Routine examinations of conventional outbred Wistar rats in our laboratory showed increased serum levels of alkaline phosphatase, aspartate aminotransferase, alanine aminotransferase, and urea. Electron microscopy and specific reactions showed C. pneumoniae and M. pulmonis in lung, liver, spleen, heart, and kidney sections. We could not exclude the fact that other infectious microorganisms detected through routine health surveillance affected the Wistar rat colony; however, we have not identified any of those microorganisms by electron microscopy of the organs listed. Natural coinfection of C.

A case study of an acute tuberculous bronchiolitis (a rarely encountered form of generalized miliary tuberculosis), whose aggravated progressing resulted in fatal outcome 2 months after the onset of the initial disease signs, is described. The prevalence of non-specific infectious-and-allergic manifestations is a clinical-and-morphological peculiarity within the case study, which confirms a diversity of morphological changes occurring in the organism in tuberculosis infection. The etiological- and pathogenetic specific features of acute tuberculous bronchiolitis in children and in adults are u

A group of 49 patients with non-small cell lung carcinoma was investigated at the Clinic of Pulmonary Diseases of the Military Medical Academy in the period 01.01.1995.-01.03.1999. In all patients were performed bronchoscopy, ultrasound of the abdomen and computed tomography (CT) of the thorax. The patients suspected to have locoregionally disseminated disease were selected. The aim of the investigation was to estimate the reliability of CT of the thorax in the evaluation of the possibility for lung carcinoma resection. The findings were confirmed either by invasive diagnostic procedures and/o

Previous data have indicated that organ-specific and non-organ-specific autoimmune diseases may occur in the same patient. We report here our study on the type and prevalence of endocrine autoimmune diseases in undifferentiated connective tissue disease (UCTD). A retrospective analysis revealed five out of 75 UCTD cases (6.6%) with cytology-verified autoimmune thyroiditis (associated with insulin-dependent diabetes mellitus in one case). Other UCTD patients had Graves' disease (one case), non-toxic multinodular goitre (two cases) and central hypothyroidism (one case). In a prospective study, t

The use of animals as a source of organs and tissues for humans has been an enduring goal of transplantation. Xenotransplantation, as such, would overcome a shortage of human donors and allow for biochemical or genetic approaches to modification of transplants. The use of animal organs and tissue, however, is hindered by an intense immune response of the recipient against the graft. The molecular basis for this immune response has recently been elucidated, at least in part, and specific approaches to therapy, including the genetic engineering of source animals, have been developed. Other hurdl

Tissue kallikrein in the rat pineal gland was immunocytochemically investigated with the aid of specific antiserum against rat urinary kallikrein. We also compared the tissue kallikrein immunoreactivity of the pineal gland with that of the submandibular gland and kidney, which have been well established as tissue kallikrein-synthesizing organs. The cytoplasm of pinealocytes from both the superficial and the deep portion of the gland exhibited specific immunolabeling for tissue kallikrein, but the immunoreaction was weaker than that observed in exocrine organs. Two types of tissue kallikrein-im

Jejunal segments from fetal rats of 18 days gestation were maintained in an organ culture system for up to 72 h. During this period, villi developed within the intestinal lumen and the epithelium changed from stratified to simple columnar. Peristaltic activity was observed during in-vitro culture. Alkaline phosphatase specific activity of the bowel segments fell after 48 hours culture, compared with pre-culture values (P less than 0.05), but that of alpha-glucosidase increased. The addition of Frazer's gluten fraction III to the culture medium caused slowing in the rate of morphological matura

Analysis of 7,966 hospital admissions and other available data suggest that disease associated with autoimmunity are rare in Ethiopia. Sera from 107 patients with dyspepsia and 80 healthy Ethiopians were studied for autoantibodies. Using the immunofluorescence technique and anti-Fab-FITC conjugate, a search for organ-specific autoantibodies against thyroid (microsomal and thyroglobulin) cells as well as for non-organ-specific autoantibodies against nuclear material, smooth muscle, mitochondria, cardiolipin, glomerular basement membrane and connective tissue (reticulin antibodies) revealed that

Hypertension-mediated organ damage (HMOD) represents a critical and dynamic stage on the cardiovascular continuum, serving as the primary mechanistic link between sustained arterial pressure elevation and major adverse cardiovascular and renal events. HMOD is often subclinical at first occurrence and is a systemic, cumulative process, representing structural and functional injury of the heart, brain, kidneys, and vasculature. This narrative review synthesises current evidence on the pathogenesis and clinical sequelae of HMOD, specifically focusing on the interplay between chronic haemodynamic

Sepsis is a systemic infection characterized by the dissemination of microbial pathogens through the bloodstream, and subsequently triggers inflammation, organ failure and concurrent immune suppression. The dysregulation of the innate and adaptive immune systems results in life-threatening organ failure or recurrent and often fatal infections. There are no effective and specific therapeutics due to the poorly understood underlying mechanisms. G-protein coupled receptors (GPCRs) are membrane-bound receptors that have emerged as important upstream regulators of immune system homeostasis and exce
