Navigating the Parasitic Landscape: Insights into Infection Patterns and Public Health Strategies in West Africa.

Parasitic infections remain a significant public health challenge in West Africa, contributing to high morbidity and mortality rates, economic burdens, and healthcare system strain. Malaria, soil-transmitted helminths, schistosomiasis, and other parasitic diseases persist due to environmental, socio-economic, and healthcare barriers. A systematic literature search was conducted using databases such as PubMed, Scopus, Web of Science, and Science Direct. Studies published between 2014 and 2024 were screened using predefined eligibility criteria. Cross-sectional and case-control studies reporting

Population genetic structure of Dactylogyrus vistulae, a gill parasite of cyprinoid fish in the Western Palearctic.

Monogenean parasites with a broad host range and direct life cycle provide insight into population genetic structure and dispersal processes. Dactylogyrus vistulae infects over 50 cyprinoid fish species across the western Palearctic, making it an ideal model for assessing host and geographic influences on parasite population structure. Herein, we utilized 20 microsatellite loci and mitochondrial COI sequences to analyze the genetic variability and structure of D. vistulae populations, with a deeper focus on Czech river systems. Low allelic variance was observed across most populations, except

Human keratinocytes exposed to a clinical strain of Leishmania tropica can result in parasite internalization.

Cutaneous Leishmaniasis (CL) due to Leishmania tropica is a public health burden in Morocco. The increasing clinical polymorphism challenges its proper diagnosis and treatment. Although the immunopathogenesis of CL due to L. tropica has been documented, the role of keratinocytes, a critical cell type in skin immunity, has never been investigated in this pathology. Overall, keratinocytes play a crucial role in the recognition and early immune response to Leishmania parasites upon skin inoculation, influencing the local immune response by producing early cytokines that shape anti-leishmanial imm

An ER-resident lipid scramblase is crucial for biogenesis and function of apicomplexan parasite secretory organelles.

Apicomplexan parasites, including Plasmodium falciparum and Toxoplasma gondii, contain specialized secretory organelles such as micronemes, rhoptries, and dense granules, which are essential for parasite motility, host cell invasion, development, and egress. DedA superfamily proteins are implicated in lipid mobilization, which is a key requirement for organelle biogenesis. Herein, we identify and investigate the vacuole membrane protein 1 (VMP1), a DedA superfamily member, of P. falciparum (PfVMP1) and T. gondii (TgVMP1). PfVMP1 and TgVMP1 are ER-localized lipid scramblases. TgVMP1 depletion a

GSK3β/HIF-1α signaling-dependent anti-parasite effect of Cynanchi atrati Radix.

Medicinal plants yield bioactive compounds with potential for parasite control. We examined Cynanchi atrati Radix (C. atrati) and its component 4'-hydroxyacetophenone (4'HAP) for activity against Toxoplasma gondii (T. gondii) using cultured cells and mouse infection models. C. atrati extracts limited parasite growth with minimal host-cell toxicity. Chemical screening pinpointed 4'HAP as the active constituent that suppresses T. gondii proliferation in vitro and in vivo. Mechanistically, C. atrati and 4'HAP activated GSK3β, destabilized HIF-1α, and curtailed parasite fitness; pharmacologic GSK3

Nanosuspensions of fenbendazole and ivermectin induce parasite death and low inflammatory response in experimental neurocysticercosis.

Neurocysticercosis (NCC) is the most common preventable infectious disease of the central nervous system (CNS) worldwide. Its treatment is performed with anthelmintics, albendazole, or praziquantel, associated with corticosteroids due to the intense inflammatory response after the parasite's death. Also, the current treatment does not achieve full parasitic clearance and as it is used for over 40 years, there are a few reports on parisitic resistance and drug refractoriness. Other anthelminthic drugs in their commercial or nanoformulation form may present important activity against cysticerci

Desmuramylpeptide, a NOD2 agonist, enhanced immune response and parasite clearance in Leishmania donovani infection.

INTRODUCTION: Visceral leishmaniasis is a life-threatening infectious disease caused by the intracellular protozoan Leishmania, where interactions between the parasite and the host's intracellular components play a crucial role in tracking the incursion of infection. Nucleotide-binding oligomerization domain-containing protein 2 (NOD2) plays a critical role in regulating inflammation and eliciting a defensive immune response to the pathogen. Consequently, we aimed to explore the role of a desmuramylpeptide (NOD2 agonist) in visceral leishmaniaisis caused by Leishmania donovani. METHODOLOGY: I

Host-parasite interaction in latent cerebral toxoplasmosis: The role of cytokine response in a mouse model.

BACKGROUND: Toxoplasma gondii is a widespread intracellular protozoan that can cause chronic infection in immunocompetent hosts, leading to subclinical neuroinflammation. Understanding the immunopathogenesis of chronic cerebral toxoplasmosis requires well-characterized animal models. AIM: This study aimed to evaluate infection severity, affected immune cell populations, and cytokine response in both early and late chronic phases of T. gondii infection. METHODS: A murine model was developed by infecting C57BL/6 mice with a sub-infective dose (104 tachyzoites) of the ME49 strain. Forty mice we

Paragonimiasis Diagnosed Through Detection of Parasitic Ova in Pleural Effusion: A Clinical Case Report.

Paragonimiasis, a chronic zoonotic parasitic infection caused by Paragonimus species, is frequently misdiagnosed due to its nonspecific clinical presentation and low disease awareness. We report a case of a 42-year-old male who was admitted following head and chest trauma. Initial chest CT revealed bilateral pulmonary contusions, a right rib fracture, and a significant pleural effusion, consistent with traumatic lung injury. Routine blood tests showed elevated white blood cells and neutrophils, but eosinophil counts remained within the normal range. Closed thoracic drainage was performed, and

Infection caused by a parasite most closely related to Hepatozoon luiperdjie in cats from Hungary.

Feline hepatozoonosis, due to Hepatozoon felis, has been recently reported in wild cats from Central Europe. At the same time, only two isolated cases were reported in domestic cats, one in Austria, and another in Hungary. In the southern part of Central Europe, Hungary, three cats were presented at two small animal clinics in the course of 1.5 months, their most important clinical findings included anaemia (for all three cats), malignant neoplasia (case #1), chyloabdomen, splenomegaly and hepatic nodules (case #2), and dyspnoea (case #3). Hepatozoon gamonts measuring 12.3 × 6 μm were present

Serological assessment of pediatric parasite exposure in two Senegalese districts using multiplex serology.

Although pediatric parasitic diseases cause significant morbidity and mortality in regions with high rates of co-infection, this overlap may offer opportunities for integrated control strategies. This study aimed at a serological assessment of exposure to multiple parasitic infections among children aged 1-14 years in two Senegalese districts, Saraya (Kédougou Region) and Diourbel (Diourbel Region), to inform integrated control strategies. We analysed 883 dried blood spot samples. A multiplex bead-based immunoassay quantified IgG antibody against Plasmodium falciparum, helminths (Necator ameri

Contact exposure to ivermectin induces acute mortality and inhibits parasite development in malaria vectors.

Resistance to insecticides and associated behavioural shifts are being increasingly reported in malaria vectors. To counter these adaptations, there is a pressing need to explore novel control tools and interventions. In line with this, the present study evaluates the potential of ivermectin as a contact-toxin for both malaria vectors and parasite. Laboratory reared female An. culicifacies and An. stephensi mosquitoes were exposed to different concentrations of ivermectin through topical and bottle bioassays. Mortality data was used to calculate the LD50 and LD90 values. Infection studies with

Vector microbiota and parasite infection: bacteria-mediated effects on Leishmania in Phlebotomus papatasi and culture.

AIM: Microbes are increasingly being explored as tools to control leishmaniasis and sandfly populations. This study investigated the interactions between sandflies, Leishmania, and their microbiota. METHODS: The in vitro and in vivo survival and development of Leishmania major in the presence of seven bacterial species were assessed. In vitro experiments included Leishmania co-cultures with bacteria applied at different concentrations (102-108 CFU/ml), monitoring parasite survival/development at early (30-120 min) and late (12-240 h) time points. In vivo experiments in Phlebotomus papatasi ex

Parasite responses to resource provisioning can be altered by within-host co-infection interactions.

Anthropogenic changes to the environment significantly impact wildlife infectious diseases by modifying food resources and impacting host-parasite interactions through changes in host demography, behaviour and immune defences. Supplemental resource provisioning has been found to both enhance and mitigate parasite transmission; however, the role of co-infecting parasites in mediating these effects remains understudied. We developed a mathematical model to explore these dynamics, motivated by the empirical system of wood mice (Apodemus sylvaticus) infected with the nematode Heligmosomoides polyg

Toxoplasma gondii chromatin remodeler SWI/SNF controls parasite division and gene expression.

Toxoplasma gondii is a zoonotic apicomplexan parasite that relies on highly orchestrated gene expression programs to coordinate its cell cycle progression. Although epigenetic mechanisms are recognized as pivotal drivers of developmental gene regulation in parasitic life cycles, the contributions of chromatin remodeling complexes to these processes remain largely unexplored. In this study, we focus on two core ATPase subunits of the SWI/SNF chromatin remodeling complex and investigate their roles in parasite biology and gene regulation. Our findings reveal that these SWI/SNF ATPases work coord

Immunoprophylactic potential of parasite-derived antigens for controlling Lernaea cyprinacea infection in Ctenopharyngodon idella.

Lernaea cyprinacea, an invasive ectoparasitic copepod, poses a significant threat to freshwater aquaculture due to its high pathogenicity and association with severe epizootics leading to mass fish mortalities. The present study evaluated the immunogenic potential of parasite-derived antigens in Ctenopharyngodon idella (grass carp) as a immunoprophylactic strategy against L. cyprinacea infection. Crude antigens isolated from L. cyprinacea were administered intraperitoneally to the experimental group, whereas the control group received phosphate-buffered saline (PBS), and a naturally infected g

Key discoveries from research in anisakiasis: From parasite biology to pathology in the human host.

Anisakidosis is a fish-borne zoonosis characterised by mild to severe gastrointestinal (GI) symptoms and potentially linked to the onset of allergic reactions and cancer. The disease follows the ingestion of marine products infected with third-stage (L3) larvae of the genera Anisakis and Phocanema. Epidemiologically, anisakiasis is most prevalent in countries where fish consumption is high, such as Japan and the Mediterranean regions; however, the globalization of dietary habits has contributed to the expansion of its incidence worldwide. In this Chapter, we provide an overview of aspects rela

Exploring the Diagnostic Potential of Parasite-Derived miRNAs in Cystic Echinococcosis: Are We Getting Closer?

Cystic echinococcosis (CE) is the neglected zoonotic illness caused by a parasite. Non-invasive imaging methods, most notably ultrasound, are used in clinical practice to diagnose CE. Small, non-coding RNA molecules known as microRNAs (miRNAs) function as post-transcriptional regulators in various biological processes. The identification of parasite-derived miRNAs has raised significant interest, as these molecules hold promise as potential biomarkers for CE diagnosis and subsequent monitoring. This study investigates the diagnostic potential of certain parasite-derived miRNAs namely egr-let-7

Heme processing in the malaria parasite, Plasmodium falciparum: a time-dependent basal-level analysis.

Malaria is a deadly disease for which therapeutic options are threatened by the rise of antimalarial resistance. Inhibiting the formation of hemozoin (the product of heme detoxification) in the digestive vacuole (DV) is the mechanism of action of numerous antimalarial drugs, including those in development as new therapies. This drug target remains attractive as hemozoin is an abiotic and non-mutable molecule, unique to the parasite. The underlying parasite biology of the heme detoxification pathway is complex and requires a deeper understanding. This study focuses on the DV of Plasmodium falci
