A Randomized Controlled Trial Comparing the Socket Seal Technique to Guided Bone Regeneration in Alveolar Ridge Preservation.

AIM: To compare alveolar ridge preservation (ARP) with the socket seal technique (SS) to the Guided Bone Regeneration technique (GBR). MATERIALS AND METHODS: Systemically healthy patients planned for ARP at two sites with an intact bone wall or a buccal dehiscence of up to 6 mm were included. Following socket grafting with collagen-enriched deproteinized bovine bone mineral, sites were randomly allocated to the SS group using a collagen matrix or GBR group using a collagen membrane. The primary outcome was soft tissue thickness at the center of the site at 4 months. Secondary outcomes include

Fibroblasts: a diverse population of cells balancing homeostasis, wound healing, regeneration, inflammation, fibrosis, and cancer across organs.

As the principal ECM-producing cell type, fibroblasts are essential regulators of tissue architecture and function in development, homeostasis, and disease. While their disease-promoting functions in fibrosis have long been the center of attention, it is increasingly recognized that fibroblasts exert critical homeostatic roles across organs, acting as sentinels that regulate the function, proliferation, and recruitment of epithelial, endothelial and immune cells in health and disease. Here, we will review the roles of fibroblasts and fibroblast-like cells in tissue maintenance, physiological w

Screening cross-injury responses in skin organoids identifies a THBS2-enriched pro-repair fibroblast state during regeneration.

INTRODUCTION: Skin injuries impose a substantial global health burden. Conventional wound healing models fail to distinguish evolutionarily conserved regenerative mechanisms from injury specific responses because they cannot capture microenvironmental complexity. OBJECTIVES: This study established a high-fidelity mouse skin organoid platform to simulate clinically relevant stressors and identify the core regulatory components of the regenerative program. METHODS: Mature skin organoids were individually exposed to hypoxia, hyperthermia, or hyperinflammation. Transcriptomic analysis identified

Multi-scale assessment of scaffold-guided regeneration in human extraction sockets using an ultra-porous TiO₂ biomaterial.

Ultra-porous TiO₂ scaffolds are architected non-degradable ceramic graft substitutes, but human evidence relating their structural integration to local tissue maturation remains limited. This exploratory study assessed scaffold-associated healing in a prospective clinical cohort registered at ClinicalTrials.gov (NCT06269497; 10 patients, 11 treated sites). Cone-beam computed tomography (CBCT)-based segmentation at baseline and follow-up was used to quantify changes in bone volume, radiographic low-density/non-mineralized volume, and radiographically detectable scaffold volume. Eleven scaffold-

Water-soluble yeast β-glucan accelerates oral ulcer healing in rats via collagen synthesis and epithelial regeneration.

BACKGROUND: Oral ulcer is a common, painful lesion that heals slowly and easily becomes secondary-infected. β-glucan, a well-known immunomodulator, has shown potential in promoting wound repair. However, the therapeutic efficacy of its water-soluble form on oral mucosal wounds remains insufficiently studied. Therefore, this study aimed to evaluate the effects of water-soluble yeast β-glucan on the healing of oral ulcers in a rat model. METHODS: Forty rats were randomly assigned to five groups (n = 8): (1) negative control (2), standard care control (0.12% chlorhexidine), and (3–5) experimental

AF6 orchestrates macrophage polarization via JAK2-STAT3 signaling and supports intestinal regeneration by stimulating stem cell proliferation.

INTRODUCTION: Macrophages play a central role in regulating intestinal inflammation, resolution, and tissue repair. However, the upstream regulators that govern their polarization during colitis remain poorly defined. METHODS: In this study, we used a dextran sulfate sodium (DSS)-induced colitis model to investigate the role of the scaffold protein AF6 in colitis pathogenesis. RESULTS: We identify AF6 as a key regulator that promotes pro-inflammatory macrophage polarization and exacerbates colitis. Mechanistically, AF6 expression promoted JAK-STAT3 complex formation. In turn, its deletion in

Alveolar ridge preservation in defect sockets with simultaneous periodontal regeneration in adjacent teeth: a retrospective cohort study.

OBJECTIVE: To evaluate the effectiveness of alveolar ridge preservation (ARP) in periodontally compromised extraction sockets with simultaneous periodontal regeneration (PR) in adjacent teeth. MATERIALS AND METHODS: A total of 69 extraction sites were retrospectively evaluated and allocated into two groups according to the presence of interdental bone defects at adjacent teeth. The test group received ARP combined with simultaneous PR of adjacent teeth, whereas the control group underwent ARP alone. All sites were managed using an open wound healing protocol. Linear dimensional changes were a

Immediate icing disrupts the very early MCP-1/CCR2 recruitment window and delays muscle regeneration in rats.

Icing is a widely used initial intervention for skeletal muscle injury in sports settings. However, accumulating evidence suggests that icing impairs muscle regeneration, potentially via delayed monocyte/macrophage accumulation. To elucidate the mechanisms underlying this icing-induced delay, we investigated whether icing modulates the monocyte chemoattractant protein 1 (MCP-1)/CC chemokine receptor 2 (CCR2) axis governing monocyte/macrophage recruitment. We comprehensively characterized inflammatory cell dynamics in a rodent crush-injury model using immunostaining and flow cytometry. During t

A Polydopamine-coated Engineered Type II Collagen Scaffold for Motor Neuron Organization and Spinal Cord Injury Regeneration.

Spinal cord injury (SCI) leads to severe motor neuron depletion and glial scar formation, limiting endogenous regeneration and motor functional recovery. While engineered collagen scaffolds offer a promising strategy for supporting neural repair, the optimal scaffold composition and functionalization remain to be explored. This study investigates the efficacy of a novel polydopamine-coated engineered type II collagen (Col 2-PDA) scaffold and evaluates its efficacy for inducing differentiation of motor neural progenitors (MNPs) into motor neurons and for promoting motor functional recovery in t

Recent advances in electroactive biomaterials and electrical stimulation for skeletal muscle regeneration: materials, strategies, and mechanistic insights.

Skeletal muscle tissue plays a crucial role in strength generation, body movement, posture maintenance, and the protection of internal organs. Volumetric muscle loss not only causes significant physical and psychological distress and disability for patients, but also poses a serious societal challenge due to the substantial financial burden it places on healthcare systems. Currently, available treatment methods for muscle tissue injury have inherent limitations. Tissue engineering has emerged as a promising therapeutic strategy to address this issue. Bioelectric activity has been widely recogn

Recombinant humanized type VII collagen for skin repair and regeneration: prospects for reconstructing the dermal-epidermal junction.

Type VII collagen (Col VII) is a key component of the anchoring fibrils at the dermal-epidermal junction (DEJ). It plays a crucial role in maintaining stable adhesion between the epidermis and dermis, promoting wound re-epithelialization, and ensuring the structural integrity of the skin following repair. With advancements in genetic engineering, synthetic biology, and recombinant protein expression technologies, recombinant humanized type VII collagen (rhCol VII) has gradually emerged as a novel functional biomaterial of interest in the fields of skin repair and regenerative medicine. Multi-biomimetic poly(amino acid)-based Janus membrane integrating barrier and osteoinductive functions for cranial bone regeneration.

Guided bone regeneration (GBR) membranes play a critical role in repairing bone defects, yet conventional collagen-based membranes suffer from immunogenicity, insufficient mechanical strength, and lack of osteoinductivity. Herein, a multi-biomimetic poly(γ-benzyl-L-glutamate) (PBLG)-based Janus membrane (PP-S/MH-P) is constructed that integrates a robust barrier function with potent osteoinductive capacity. The membrane is fabricated with a Janus architecture: PP-S side consists of a dense poly(γ-benzyl-L-glutamate) (PBLG) barrier layer, physically blended with PEGDA and surface-grafted with z

Hierarchical Silica-Cellulose Nanoarchitectures from Rice Straw: A Waste-to-Value Platform for Autonomous Osteogenic Bone Regeneration.

Current bone regeneration strategies face significant constraints, relying either on synthetic scaffolds with slow degradation that require biochemical supplements or on bioactive fillers. Recently, the focus has shifted towards functional natural biomaterials with inherent osteoinductive potential. This study presents a promising candidate based on a silica-containing plant-derived scaffold fabricated from rice straw, a sustainable resource with >20 wt.% silica. Morphological analyses revealed that the decellularised rice straw scaffold exhibits a unique surface pattern with a three-dimension

Interlayer Hydroxyl Self-Regeneration in Nickel-Aluminum Layered Double Hydroxides for Electrochemical Uranium Extraction from Nuclear Wastewater.

Electrochemical extraction of uranium from nuclear wastewater offers the dual benefit of resource recovery and environmental remediation. However, in a high-fluoride real environment, the formation of extremely stable uranyl-fluoride complexes (UO2Fx) severely impedes uranium recovery due to competitive coordination. Here, we propose an interlayer hydroxyl (-OH) enrichment strategy to confine UO2Fx and facilitate its conversion to insoluble diuranates. A nickel-aluminum layered double hydroxide grown on nickel foam (Ni-Al LDH@NF) was designed, which achieves 93.8% uranium extraction within 100

Sustained and bioactive BMP-2 delivery via nanoconfinement-affinity coupled silk fibroin microparticles for enhanced bone regeneration.

Achieving sustained and bioactive drug delivery remains a formidable challenge in regenerative medicine, primarily due to the inherent instability and rapid clearance kinetics of therapeutic agents. Here, we report a biomimetic delivery platform based on nanoporous biomineralized silk fibroin microparticles, engineered through a unified in situ biomineralization and critical-point drying process. This architecture employs a coupled nanoconfinement-affinity strategy: the interconnected nanofibrous network imposes physical confinement, while the deposited hydroxyapatite coating provides strong a

A multifunctional pearl powder-incorporated scaffold for bone regeneration: Simultaneous enhancement of osteogenesis, immune modulation, and angiogenesis.

Bone defect regeneration requires precise spatiotemporal orchestration of inflammatory response, blood vessel formation, and bone formation processes. A major unresolved issue is the fabrication of biomaterial scaffolds that can synergistically combine immunomodulation with angiogenic-osteogenic coupling to hierarchically regulate this regenerative cascade. This study developed a novel biomimetic composite scaffold that structurally and compositionally replicates natural bone by integrating methacrylic anhydride modified gelatin (GelMA) with pearl powder (PP) and dimethyloxalylglycine (DMOG) t

Two axolotl-adapted cell-ablation platforms reveal macrophage-dependent processes essential for spinal-cord and skeletal regeneration.

The axolotl (Ambystoma mexicanum) has emerged as the premier model organism for studying scarless repair and adult tissue regeneration, supported by an expanding collection of tissue-specific transgenic lines and translucent skin that enables high-quality live imaging and cell tracking. However, functional characterization of specific cell types during regeneration has been limited by the absence of validated cell-specific ablation systems. Here, we developed and rigorously compared two independent inducible genetic cell-ablation platforms - bacterial nitroreductase (NTR 2.0) and mammalian ind

CO2-assisted selective lithium extraction and closed-loop regeneration of spent NCM cathodes toward cleaner battery recycling.

The growing volume of spent lithium-ion batteries (LIBs) has increased the need to recover valuable materials while limiting environmental impacts. Conventional hydrometallurgical recycling of spent NCM cathodes generally dissolves Li, Ni, Co, and Mn together, resulting in complex downstream separation. Selective Li recovery and direct cathode regeneration are usually developed separately. Integrating them requires efficient Li removal while retaining a transition-metal matrix suitable for subsequent reconstruction. In this study, lignite carbothermal reduction was coupled with CO2-assisted aq

A biosilica nanocage-based injectable hydrogel to trap endogenous TGF-β1 for in-situ hyaline cartilage regeneration.

Regeneration of hyaline cartilage remains a major clinical challenge. Current first-line treatments for focal articular cartilage defects, typically induce fibrocartilage instead of hyaline cartilage, resulting in inferior mechanical and biological outcomes. Here we report a biomimetic strategy inspired by the biosilicification process of diatoms, for the de novo synthesis of biosilica nanocages exhibiting effective binding to the hyaline chondrogenic cytokine transforming growth factor β1 (TGF-β1). To enhance clinical applicability, an injectable biosilica nanocage-based hydrogel (Si-aGel hyd

A 3D-printed spider silk-based functional mesh for ventral hernia repair via endogenous regeneration-promoting strategies.

Ventral hernia, a prevalent condition especially in the elderly, is primarily treated with mesh implantation. However, high postoperative recurrence persists due to mismatched degradation rates of biological meshes and tissue remodeling. We engineered an electrospun composite mesh (CPCM) integrating chitosan, polylactic acid, and chimeric mini-spidroin recombinant collagen to address this challenge.
Methods: The CPCM was fabricated by combining 3D printing with bilateral electrospinning. Fourier-transform infrared spectroscopy, scanning electron microscopy, water contact-angle measurements
