A Solar-Powered Hydrogel Platform for Precise Thermal Control and On-Demand Antibiotic Release in Infected Wound Management.

Bacterial-infected wounds pose a critical clinical challenge, exacerbated by antibiotic resistance (AMR) and the limitations of photothermal therapy. Herein, we report a pioneering solar-powered hydrogel with phase-change material (PCM)-engineered 3D ordered macroporous carbon (OMC), enabling laser-independent PTT, precise thermal control, and on-demand antibiotic release. The OMC exhibits 96.5% solar absorptivity (250-800 nm) and 74.6% solar-to-thermal conversion efficiency, serving as a renewable photothermal agent and stimuli-responsive reservoir. Infiltrated PCM endows autonomous thermal r

Repairing the Infarcted Heart: Preclinical Outcomes of Cardiac Tissue Engineering.

Myocardial infarction (MI) remains a leading cause of cardiovascular mortality, driven by irreversible cardiomyocyte loss and the limited regenerative capacity of the adult heart. Conventional pharmacological and interventional therapies reduce acute injury yet rarely restore functional myocardium, motivating bioengineering strategies to enhance repair. This review synthesizes recent preclinical advances in biomaterial-based cardiac tissue engineering, with an emphasis on engineered cardiac patches and injectable hydrogels evaluated in vivo, alongside early human translation where available. Living Probiotic Microneedles Coordinate Antiandrogen and Nitric Oxide Therapy for Androgenetic Alopecia.

Androgenetic alopecia (AGA) is characterized by follicular miniaturization mediated by androgens along with chronic inflammatory and vascular disturbances. These alterations suggest broader dysregulation involving endocrine pathways, immune activity, and vascular function. Current therapies such as finasteride and minoxidil provide limited and often transient improvement because they target only single components of this multifactorial process. In this study, we developed a living probiotic-based microneedle system that integrates nitric oxide-generating Weissella cibaria (WC) with clascoteron

FOXP3 Stability-Adaptation Paradox: Reshaping Treg-Based Therapies for Intestinal Diseases.

Regulatory T cells (Tregs) maintain intestinal immune homeostasis, but their therapeutic potential is constrained by a fundamental paradox: the same plasticity that enables tissue repair renders FOXP3 vulnerable to degradation in chronic inflammation. Mechanistically, microbial metabolites (short-chain fatty acids, bile acids) and retinoic acid stabilize FOXP3 and induce RORγt⁺/GATA3⁺ Treg specialization. In contrast, inflammatory cytokines and succinate accumulation drive ER stress and post-translational FOXP3 degradation, leading to lineage instability in inflammatory bowel disease, colorect

Inulin-Butyrate Nanogel for Modulation of Gut Microbiome, Intestinal Barrier, and Regulatory T-Cells in Colitis.

Inflammatory bowel diseases (IBD) arise from a vicious cycle of intestinal barrier dysfunction, gut microbiome dysbiosis, and dysregulated immune responses. Current therapies predominantly suppress immunity but fail to address root causes or break this cycle. While inulin, a prebiotic, restores microbial diversity and enables colon-targeted drug delivery, they lack specificity for inflamed tissue. On the other hand, even though butyrate, a microbial metabolite, is a potent enhancer of intestinal barrier integrity and anti-inflammatory Treg cell differentiation, their clinical applications are

Therapeutic challenges in central nervous system viral infections: advancing mesenchymal stem cell-based strategies for treating neuroinflammation and promoting tissue repair.

Although significant progress has been made in medicine and antimicrobial research, viral infections continue to pose a critical global health challenge, particularly when they involve the central nervous system (CNS). Despite advances in vaccines, antiviral agents, and small molecule therapeutics, current strategies remain insufficient to address the complex consequences of many CNS infections fully. Notably, many viruses are neurotropic and can invade the CNS, triggering infectious neuroinflammation that often lead to chronic neurological disorders and lasting morbidity. Current therapeutic

Development and preclinical evaluation of next-generation ΔsigH-based live candidate vaccines.

To radically diminish tuberculosis (TB) incidence and mortality by 2035, as set out by the WHO End TB Strategy, there is a desperate need for improved TB therapies and a more effective vaccine against the deadly pathogen Mycobacterium tuberculosis. Aerosol vaccination with the MtbΔsigH mutant protects 2 species of nonhuman primates against lethal TB challenge by invoking vastly superior T and B cell responses in the lungs through superior antigen presentation and interferon conditioning. Since the Geneva Consensus on essential steps toward the development of live mycobacterial vaccines recomme

Dynamic immunoediting by macrophages in homologous recombination deficiency-stratified pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease, notably resistant to existing therapies. Current research indicates that PDAC patients deficient in homologous recombination (HR) benefit from platinum-based treatments and poly-ADP-ribose polymerase inhibitors (PARPi). However, the effectiveness of PARPi in HR-deficient (HRD) PDAC is suboptimal, and significant challenges remain in fully understanding the distinct characteristics and implications of HRD-associated PDAC. We analyzed 16 PDAC patient-derived tissues, categorized by their homologous recombination deficiency (HRD) scores

Photothermal therapeutic effect by gold nanostars/extracellular vesicles nanocomplex on melanoma cells.

Photothermal therapy (PTT) is a method for treating cancer using the heat generated by light irradiation, often in combination with light-absorbing materials. Efficient PTT requires a drug delivery system to deliver light-absorbing materials to cancerous tissues. Gold nanostars (GNSs) enable efficient PTT through absorbing long-wavelength light. In this study, we established a platform for preparing GNSs-loaded extracellular vesicles (EVs), that were expected to provide the combined biological functionality of EVs and photothermal conversion properties. By electrostatically binding cationic po

Revisiting the role of hyperbaric oxygen therapy in knee injuries: Potential benefits and mechanisms.

Knee injury negatively impacts routine activities and quality of life of millions of people every year. Disruption of tendons, ligaments, and articular cartilage are major causes of knee lesions, leading to social and economic losses. Besides the attempts for an optimal recovery of knee function after surgery, the joint healing process is not always adequate given the nature of intra-articular environment. Based on that, different therapeutic methods attempt to improve healing capacity. Hyperbaric oxygen therapy (HBOT) is an innovative biophysical approach that can be used as an adjuvant treat

Inflammation and thrombosis in patients with COVID-19: A prothrombotic and inflammatory disease caused by SARS coronavirus-2.

Coronavirus disease 2019 (COVID-19) caused by 'Severe Acute Respiratory Syndrome Coronavirus-2' (SARS-CoV-2) infection emerged in Wuhan, a city of China, and spread to the entire planet in early 2020. The virus enters the respiratory tract cells and other tissues via ACE2 receptors. Approximately 20% of infected subjects develop severe or critical disease. A cytokine storm leads to over inflammation and thrombotic events. The most common clinical presentation in COVID-19 is pneumonia, typically characterized by bilateral, peripheral, and patchy infiltrations in the lungs. However multi-systemi

Photothermal Nano-antibiotic for Effective Treatment of Multidrug-Resistant Bacterial Infection.

Bacteria's antibiotic resistance is one of the major challenges in the treatment of infectious diseases. With increasing difficulty in discovering antibiotics, there is an urgent need to develop antibiotic-free therapeutic strategies to address this grand challenge. In this report, we developed a polypyrrole (PPy)-based photothermal nano-antibiotic (PTNA) for effective treatment of multidrug-resistant (MDR) bacterial infection. PTNA was fabricated by polymerizing pyrrole onto an anionic vesicle to integrate the cationic and photothermal properties of PPy for combinatory killing against bacteri

Breast Cancer Metabolomics Using NMR.

Continued progress is being made in understanding the breast cancer metabolism using analytical magnetic resonance (MR)-based methods like nuclear magnetic resonance (NMR) and in-vivo MR spectroscopy (MRS). Analyses using these methods have enhanced the knowledge of altered biochemical pathways associated with breast cancer progression, regression, and pathogenesis. Comprehensive metabolic profiling of biological samples like tissues, cell lines, fine needle aspirate, and biofluids such as sera and urine enables identification of new biomarkers and abnormalities in biochemical pathways. Clinical significance of receptor shedding-platelet GPVI as an emerging diagnostic and therapeutic tool.

Platelet membrane bedecked with a wide array of receptors offers a platform to regulate platelet responsiveness, thrombotic propensity, inflammatory disposition, and immune reactivity under diverse pathophysiological conditions. Ectopic proteolytic cleavage of such receptors irreversibly inactivates receptor-mediated intracellular signaling governing cellular functions, further releases soluble fragments into circulation which might modulate functions of target cells. Glycoprotein VI-(GPVI) is a membrane glycoprotein expressed in platelets and megakaryocytes. Platelet GPVI surface expression i

Antioxidant properties of probiotics and their protective effects in the pathogenesis of radiation-induced enteritis and colitis.

Radiation therapy has become one of the most important treatment modalities for human malignancy, but certain immediate and delayed side-effects on the normal surrounding tissues limit the amount of effective radiation that can be administered. After exposure of the abdominal region to ionizing radiation, nearly all patients experience transient symptoms of irradiation of the bowel. Acute-phase symptoms may persist for a short time, yet long-term complications can represent significant clinical conditions with high morbidity. Data from both experimental studies and clinical trials suggest the

Efficacy of pyrvinium pamoate against Cryptosporidium parvum infection in vitro and in a neonatal mouse model.

No effective approved drug therapy exists for Cryptosporidium infection of immunocompromised patients. Here we investigated the nonabsorbed anthelmintic drug pyrvinium pamoate for inhibition of the growth of the intestinal protozoan parasite Cryptosporidium parvum. The concentration of pyrvinium that effected 50% growth inhibition in human enterocytic HCT-8 cells by a quantitative alkaline phosphatase immunoassay was 354 nM. For comparison, in the same assay, 50% growth inhibition was obtained with 711 microM paromomycin or 27 microM chloroquine. We used a neonatal mouse model to measure the a

Suppression of transforming growth factor beta/smad signaling in keloid-derived fibroblasts by quercetin: implications for the treatment of excessive scars.

BACKGROUND: Keloids are characterized by abnormal proliferation and overproduction of extracellular matrix. Quercetin, a dietary compound, has strong antioxidant and anticancer properties. Previous studies by the authors have shown that quercetin inhibits fibroblast proliferation, collagen production, and contraction of keloid and hypertrophic scar-derived fibroblasts. Quercetin also blocks the signal transduction of insulin-like growth factor-1 in keloid fibroblasts. This study assessed the effects of quercetin on the transforming growth factor (TGF)-beta/Smad-signaling pathway in keloid-deri
