A Single-Cell Atlas of Transcriptional and Immunoglobulin Repertoire Evolution in Early B Cell Development.

The development of human B cells in the bone marrow can be separated into functionally and transcriptionally distinct subsets, which while canonically described have yet to be profiled deeply and individually and with NGS and bioinformatic techniques. In this study, single-cell RNA sequencing was performed on 65,110 B cells from six healthy donors to observe the foundation of the diversity observed in mature immune repertoires. Following the established narrative of development, committed B cells undergo heavy and light chain recombination, which together form the functional BCR. However, a gr

Morphological and Spectroscopic Characterization of Multifunctional Self-Healing Systems.

Multifunctional self-healing supramolecular structural toughened resins, formulated to counteract the insulating properties of epoxy polymers and integrating auto-repair mechanisms, are morphologically and spectroscopically characterized using Tunneling Atomic Force Microscopy (TUNA) and Fourier transform infrared spectroscopy (FT-IR), respectively. Specifically, the multifunctional resin comprises self-healing molecular fillers and electrically conductive carbon nanotubes (CNTs) embedded in the matrix. The selected self-healing molecules can form non-covalent bonds with the hydroxyl (OH) and

Immunogenic Switch of RPE Cells.

The barrier function of the retinal pigment epithelium (RPE) secures a highly selective exchange of molecules between the blood stream of the choroid and retina but also maintenance of the immune privilege of the retina. The latter function includes a mechanical barrier through the tight junctions and immune barrier of either membrane bound or secreted immune-suppressive factors in response to increasing inflammatory activities in the outer retina. However, in disease, both physical and immune barriers are compromised to allow accumulation of immune cells in the subretinal space or even to pas

Furfural Valorization to γ‑Valerolactone over Zr/Sn Zeolite-Supported Catalysts in a Liquid-Phase Continuous Flow Reactor.

This work presents for the first time the combined effect of Sn and Zr in the conversion of furfural (FU) to γ-valerolactone (GVL) by using a single liquid-phase continuous flow reactor. To address the high costs and environmental impact of this cascade reaction, catalytic transfer hydrogenation is a promising approach, utilizing alcohols as hydrogen donors in place of molecular H2. This process, which requires both Lewis and Bro̷nsted acidity, when coupled with heterogeneous catalysts, offers a potentially more cost-effective and environmentally friendly alternative. The production of GVL in

Narrative review on ethical and psychological issues raised by genetic and genomic testing in pediatric oncology care.

In pediatric oncology, genetic and genomic tests are proposed throughout the care pathway for many reasons (e.g., cancer characterization, identification of the most appropriate treatment, patient selection for clinical trials, identification of tissue/organ donors, or risk of relapse prediction). Despite the many different approaches (somatic or germline testing, targeted gene or genome sequencing), the implicated individuals are confronted with situations that may intersect and that are interesting to compare. No study has identified and analyzed the available works on these new practices in

Characterisation of the HLA-DQ alpha eplet 52SK targeting the DQA1*01 alleles family.

Eplet 52SK is unique in the HLA eplet registry as targeting the whole family of DQA1*01 alleles. It is proposed as an antibody-verified eplet but has not been validated enough to deserve this label. Especially, confusion can occur with reactivity targeting the 52PQ eplet which is present on the DQB1*05 and DQB1*06 alleles families, as DQ molecule stability imposes DQA1*01 to selectively associate with these DQ-β families only. Using two Luminex single antigen (LSA) assays from two vendors, beads bearing DR-α/DQ6 heterodimers, a special build LSA panel of additional DQ beads, and an adsorption/

Fabrication of Self-Expanding Metal-Organic Cages Using a Ring-Openable Ligand.

Metal-organic cages (MOCs), which are formed via coordination-driven assembly, are being extensively developed for various applications owing to the utility of their accessible molecular-sized cavity. While MOC structures are uniquely and precisely predetermined by the metal coordination number and ligand configuration, tailoring MOCs to further modulate the size, shape, and chemical environment of the cavities has become intensively studied for a more efficient and adaptive molecular binding. Herein, we report self-expanding MOCs that exhibit remarkable structural variations in cage size and

Small-for-size syndrome in a 9.7 kg pediatric recipient with a whole liver graft.

BACKGROUND: Small-for-size syndrome (SFSS) in pediatric liver transplant recipients, particularly those weighing less than 10 kg, is rare. This report describes a case of a 15-month-old whole liver transplant recipient who suffered SFSS, and systematic literature review was performed to identify outcomes of such cases and potential risk factors for SFSS. CASE PRESENTATION: A 15-month-old toddler with a history of biliary atresia underwent a deceased donor whole liver transplant. The graft weighed 160 g, resulting in a graft-to-recipient weight ratio (GRWR) of 1.6%. The post-operative course w

Probing the Origin of Affinity in the GM1-Cholera Toxin Complex through Site-Selective Editing with Fluorine.

Carbohydrates regulate an inimitable spectrum of biological functions, yet successfully leveraging this therapeutic avenue continues to be frustrated by low affinities with glycan-specific proteins. A conspicuous exception is the interaction of monosialotetrahexosylganglioside (GM1) with the carbohydrate-recognition domain of cholera toxin from Vibrio cholerae: this is one of the strongest protein-carbohydrate interactions known. To establish the importance of a long-discussed key hydrogen bond between C2 of the terminal galactose of GM1 and the B subunit pentamer of cholera toxin (CTB5), the

Expression of Androgen and Estrogen Receptors in the Human Lacrimal Gland.

Lacrimal gland dysfunction causes dry eye disease (DED) due to decreased tear production. Aqueous-deficient DED is more prevalent in women, suggesting that sexual dimorphism of the human lacrimal gland could be a potential cause. Sex steroid hormones are a key factor in the development of sexual dimorphism. This study aimed to quantify estrogen receptor (ER) and androgen receptor (AR) expression in the human lacrimal gland and compare it between sexes. RNA was isolated from 35 human lacrimal gland tissue samples collected from 19 cornea donors. AR, ERα, and ERβ mRNA was identified in all sampl

Developing our knowledge of the quinolone scaffold and its value to anticancer drug design.

INTRODUCTION: The quinolone scaffold is a bicyclic benzene-pyridinic ring scaffold with nitrogen at the first position and a carbonyl group at the second or fourth position. It is endowed with a diverse spectrum of pharmacological activities, including antitumor activity, and has progressed into various development phases of clinical trials for their target-specific anticancer activity. AREAS COVERED: The present review covers both classes of quinolones, i.e. quinolin-2(H)-one and quinolin-4(H)-one as anticancer agents, along with their possible mode of binding. Furthermore, their structure-a

Endothelium-Mimicking Bilayer Vascular Grafts with Dual-Releasing of NO/H2S for Anti-Inflammation and Anticalcification.

Vascular complications caused by diabetes impair the activities of endothelial nitric oxide synthase (eNOS) and cystathionine γ-lyase (CSE), resulting in decreased physiological levels of nitric oxide (NO) and hydrogen sulfide (H2S). The low bioavailability of NO and H2S hinders the endothelialization of vascular grafts. In this study, endothelium-mimicking bilayer vascular grafts were designed with spatiotemporally controlled dual releases of NO and H2S for in situ endothelialization and angiogenesis. Keratin-based H2S donor was synthesized and electrospun with poly(l-lactide-co-ε-caprolacton

Inverting the Electronic Structure of Diylidylgermylenes by Backbone Modification.

Owing to the strong electron-donating ability of ylide substituents, diylidyltetrylenes are usually highly nucleophilic species with strong donor capacities. Here, we demonstrate that their electronic properties are in fact highly flexible and can be effectively tuned through variation of the substituent in the ylide backbone. Initial density functional theory studies showed that cyano groups are particularly capable in lowering the LUMO energy of diylidyl germylenes thus turning these usually highly nucleophilic species into electrophilic compounds. This was confirmed by experimental studies.

Biomass-Derived Core-Shell Carbon Dots with Embedded Tripodal Receptors for the Selective Recognition of Mefenamic Acid in Pharmaceutical Formulations and Urine.

A tripodal amine (TPA) with -OH, N, and S donors is synthesized to functionalize a core-shell carbon dot composite (FCDs@SiO2-TPA) for sensing application. The TPA is characterized by spectroscopic and spectrometric techniques, and the composite is characterized by Fourier transform infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), Brunauer-Emmett-Teller (BET), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and energy-dispersive X-ray spectra (EDS) techniques. The composite has the ability to recognize mefenamic acid (MFA) selectively even in the pre

Inhibition of spleen tyrosine kinase decreases donor specific antibody levels in a rat model of sensitization.

Antibody mediated rejection is a major cause of renal allograft loss. Circulating preformed donor specific antibodies (DSA) can result as a consequence of blood transfusion, pregnancy or prior transplantation. Current treatment strategies are limited due to partial or transient efficacy, adverse side-effects or patient unsuitability. Previous in vivo studies exploring autoimmune diseases have shown that spleen tyrosine kinase (SYK) signalling is involved in the development of pathogenic autoantibody. The role of SYK in allogenic antibody production is unknown, and we investigated this in a rod

Meteorites as Food Source on Early Earth: Growth, Selection, and Inhibition of a Microbial Community on a Carbonaceous Chondrite.

Meteoritic material accumulated on the surface of the anoxic early Earth during the Late Heavy Bombardment around 4.0 Gya and may have provided Earth's surface with extraterrestrial nutrients and energy sources. This research investigates the growth of an anaerobic microbial community from pond sediment on native and pyrolyzed (heat-treated) carbonaceous chondrite Cold Bokkeveld. The community was grown anaerobically in liquid media. Native Cold Bokkeveld supported the growth of a phylogenetically clustered subset of the original pond community by habitat filtering. The anaerobic community on

Nitrogen-doped magnetic porous carbon material from low-cost anion-exchange resin as an efficient adsorbent for tetracyclines in water.

In this work, nitrogen-doped magnetic porous carbon material (N-MPC) was prepared through the high-temperature calcination of low-cost [Fe(CN)6]3--loaded anion-exchange resin, which was experimentally demonstrated to have significant adsorption performance for tetracycline (TC) in water. The N-MPC adsorbent with a large specific surface area (781.1 m2 g-1) was able to maintain excellent performance in a wide pH range from 4 to 10 or in high ionic strength solution. The adsorption of TC on N-MPC was found to be more consistent with the pseudo-second-order model and Langmuir adsorption model, an

A novel phenanthridine and terpyridine based D-π-A fluorescent probe for the ratiometric detection of Cd2+ in environmental water samples and living cells.

A "turn-on" Donor-π-Acceptor (D-π-A) containing phenanthridine-functionalized extended π-conjugate terpyridine, 5-(4'-([2,2':6',2''-terpyridin]-4'-yl)-[1,1'-biphenyl]4-yl)7,8,13,14-tetrahydrodibenzo [a, i] phenanthridine (TBTP) was synthesised. It shows strong selectivity for the detection of toxic Cd2+ without interference from other metal ions. In the presence of Cd2+, the absorption of the TBTP changes dramatically along with the fluorescent emission with the large Stokes shift of 6300 cm-1. When the compound TBTP is exposed to UV light, its colour changes from blue to orange over the addit
