Role of dopaminergic DAD1 and DAD2 receptors in the sensitization of amphetamine-suppressed schedule-induced polydipsia in rats.

Effects of dopaminergic D1 (DAD1) and D2 (DAD2) receptors were examined in the sensitization of amphetamine (AMPH)-suppressed schedule-induced polydipsia (SIP). After training under a fixed-interval 60 sec schedule of food presentation in the presence of a water tube, rats received injections of different doses of AMPH 10 min prior to the test. It was found that AMPH at 2.0 mg/kg significantly to reduced licks and water intake during the SIP. The AMPH-suppressed SIP manifested again following 5-days of pretreatment with a sub-threshold dosage of AMPH (1.0 mg/kg) and a period of withdrawal. The…

Chronic psychostimulant exposure and vulnerability to Parkinson's disease: is there a role for brain iron?

Alterations in brain iron homeostasis have been reported in Parkinson's disease (PD) and Attention Deficit/Hyperactivity Disorder (ADHD). Re … t to contribute to PD pathophysiology. In this perspective paper, we describe the interplay between dopamine autoxidation, inhibition of mitochondrial complex I, and increased cellular iron accumulation. We then discuss how alterations in membrane transporters following chronic MPH exposure may disrupt dopamine compartmentalization and promote dopamine aut …

Mu opioid receptors expressed in striatal D2 medium spiny neurons have divergent contributions to cocaine and morphine reward.

While our understanding of the neurobiological mechanisms underlying cocaine and opiate reward has historically been dopamine-focused, evidence from genetic and pharmacological approaches indicates that µ-opioid receptors (MORs) in the striatum are important contributors. Within the striatum, MORs are expressed in both dopamine D1-receptor and D2-receptor expressing GABAergic medium spiny neurons (MSNs), as well as in interneurons and various afferents. Thus, it remains unclear how these distinct MOR populations regulate drug reward. To address this, we generated mice with a targeted deletion …

Machine Learning Identifies a Rat Model of Parkinson's Disease via Sleep-Wake Electroencephalogram.

Alpha-synuclein induced degeneration of the midbrain substantia nigra pars compact (SNc) dopaminergic neurons causes Parkinson's disease (PD). Rodent studies demonstrate that nigrostriatal dopamine stimulates pallidal neurons which, via the topographical pallidocortical pathway, regulate cortical activity and functions. We hypothesize that nigrostriatal dopamine acting at the basal ganglia regulates cortical activity in sleep and wake state, and its depletion systemically alters electroencephalogram (EEG) across frequencies during sleep-wake state. Compared to control rats, 6-hydroxydopamine i…

Reduced D2 /D3 Receptor Binding and Glucose Metabolism in a Macaque Model of Huntington's Disease.

BACKGROUND: Dopamine system dysfunction and altered glucose metabolism are implicated in Huntington's disease (HD), a neurological disease caused by mutant huntingtin (mHTT) expression. OBJECTIVE: The aim was to characterize alterations in cerebral dopamine D2 /D3 receptor density and glucose utilization in a newly developed AAV-mediated NHP model of HD that expresses mHTT throughout numerous brain regions. METHODS: Positron emission tomography (PET) imaging was performed using [18 F]fallypride to quantify D2 /D3 receptor density and 2-[18 F]fluoro-2-deoxy-d-glucose ([18 F]FDG) to measure ce…

New and emerging treatments for schizophrenia: a narrative review of their pharmacology, efficacy and side effect profile relative to established antipsychotics.

Schizophrenia is associated with substantial unmet needs, highlighting the necessity for new treatments. This narrative review compares the pharmacology, clinical trial data and tolerability of novel medications to representative antipsychotics. Cariprazine, brexpiprazole and brilaroxazine are partial dopamine agonists effective in acute relapse. Lumateperone (serotonin and dopamine receptor antagonist) additionally benefits asocial and depressive symptoms. F17464 (D3 antagonist and 5-HT1A partial agonist) has one positive phase II study. Lu AF35700 (dopamine and serotonin receptor antagonist)…

The neuroscience of positive memory deficits in depression.

Adults with unipolar depression typically show poor episodic memory for positive material, but the neuroscientific mechanisms responsible for this deficit have not been characterized. I suggest a simple hypothesis: weak memory for positive material in depression reflects disrupted communication between the mesolimbic dopamine pathway and medial temporal lobe (MTL) memory systems during encoding. This proposal draws on basic research showing that dopamine release in the hippocampus is critical for the transition from early- to late-phase long-term potentiation (LTP) that marks the conversion of…

Cardiac failure following inadvertent administration of high-dose epinephrine subcutaneously.

Our aim is to report the consequences of epinephrine toxicity leading to cardiac failure in a child and the successful management with dopamine and milrinone. A previously healthy 13-year-old girl undergoing a left tympanomastoidectomy was inadvertently administered 10 mL of 1:1000 epinephrine subcutaneously (0.175 mg/kg) on the left post auricular region in lieu of lidocaine. She developed sudden supraventricular tachycardia, hypertension and flash pulmonary edema. She was initially treated with propofol, nitrogycerin and increased peak end-expiratory pressure. Within 4 h, she remained tachyc…

Enhanced striatal cholinergic neuronal activity mediates L-DOPA-induced dyskinesia in parkinsonian mice.

Treatment of Parkinson disease (PD) with L-3,4-dihydroxyphenylalanine (L-DOPA) dramatically relieves associated motor deficits, but L-DOPA-induced dyskinesias (LID) limit the therapeutic benefit over time. Previous investigations have noted changes in striatal medium spiny neurons, including abnormal activation of extracellular signal-regulated kinase1/2 (ERK). Using two PD models, the traditional 6-hydroxydopamine toxic lesion and a genetic model with nigrostriatal dopaminergic deficits, we found that acute dopamine challenge induces ERK activation in medium spiny neurons in denervated striat…

Predictors of punding in Parkinson's disease: results from a questionnaire survey.

Dopamine replacement therapy (DRT) for Parkinson's disease (PD) has recently been linked to the development of a number of nonmotor behavioral control problems. Punding, one of these nonmotor problems, is a term used to describe complex, purposeless stereotyped behaviors such as the repetitive handling or sorting of objects. A self-report questionnaire was adapted to assess punding in the context of dysfunctional hobby-related activities. We report the results of a survey of PD outpatients from a PD research clinic (n = 141) and non-PD controls (n = 103); conducted to identify clinical and psy…

Excessive daytime sleepiness in parkinsonism.

Excessive daytime sleepiness (EDS) can affect 20-50% of patients with Parkinson's disease (PD), whereas sleep attacks (SA), which are sleep episodes without prodroma, seem infrequent. EDS is associated with more advanced disease, higher doses of levodopa-equivalent, and sometimes the use of dopamine agonists. Patients at risk for SA have higher Epworth sleepiness scores (ESS) (although an important subset of patients under-score on this scale) and a more frequent use of ergot or non-ergot dopamine agonists. Polysomnography is a valuable tool in patients with PD, because sleep apnea may occur i…

Potential of gene therapy for Parkinson's disease: neurobiologic issues and new developments in gene transfer methodologies.

Gene transfer techniques have been explored as therapeutic modalities and neurobiologic tools to understand the role of various genes in animal models of Parkinson's disease. The gene for tyrosine hydroxylase, the rate-limiting step of dopamine synthesis, has been transferred into animal models by viral vectors or by implantable cells that have been modified by retrovirus vectors. The role of additional genes such as GTP cyclohydrolase 1 and aromatic L-amino acid decarboxylase in optimal delivery of dopamine in animal models is reviewed. Gene therapy also allows goals beyond replacement of dop…

A study of effects of putative neurotransmitters injected into the lateral cerebral ventricle of man.

1. The study was carried out in adult patients having normal cardiovascular reflexes and no brain stem lesions. They were exposed to ambient temperature of 72-74 degrees F. Injections of agonists and antagonists of receptors were made into the lateral cerebral ventricles of these patients through diagnostic burr hole in the skull. 2. Noradrenaline, adrenaline and dopamine evoked hypotension and bradycardia. While the core temperature was reduced by nor-adrenaline and adrenaline, dopamine evoked hyperthermia. Isoprenaline elicited hypertension, tachycardia and hyperthermia. Opposite cardiovascu…

Refractory bradycardia and hypotension in patients with autonomic dysfunction treated with pseudoephedrine.

We describe a man in his 40s with a history of chronic intranasal cocaine use and C5-C7 incomplete quadriplegia complicated by neurogenic orthostatic hypotension, admitted to the intensive care unit for worsening bradycardia and hypotension requiring initiation of dopamine and an increase of his home midodrine dose. The patient experienced refractory bradycardia and hypotension with weaning of dopamine, and therefore a recommendation was made to add pseudoephedrine to his current regimen. This case describes the addition of pseudoephedrine to facilitate weaning off intravenous vasopressors wit…

Treatment of Tardive Dyskinesia.

Tardive dyskinesia (TD) is an iatrogenic condition that encompasses a wide phenomenological spectrum of movement disorders caused by exposure to dopamine receptor blocking agents (DRBAs). TD may cause troublesome or disabling symptoms that impair quality of life. Due to frequent, often inappropriate, use of DRBAs, TD prevalence rates among patients exposed to DRBAs continue to be high. The judicious use of DRBAs is key to the prevention of TD, reduction of disease burden, and achieving lasting remission. Dopamine-depleting vesicular monoamine transporter type 2 inhibitors are considered the tr…

The hidden side of Parkinson's disease: Studying pain, anxiety and depression in animal models.

Parkinson's disease is a neurodegenerative disease leading to the loss of midbrain dopamine neurons. It is well known and characterized by motor symptoms that are secondary to the loss of dopamine innervation, but it is also accompanied by a range of various non-motor symptoms, including pain and psychiatric disorders such as anxiety and depression. These non-motor symptoms usually appear at early stages of the disease, sometimes even before the first motor symptoms, and have a dramatic impact on the quality of life of the patients. We review here the present state-of-the-art concerning pain, …

Oculogyric crises: A review of phenomenology, etiology, pathogenesis, and treatment.

Oculogyric crises are a rare movement disorder characterized by paroxysmal, conjugate, tonic, usually upwards, deviation of the eyes. Causes for oculogyric crises are limited and include complications of dopamine-receptor blocking medications and neurometabolic disorders affecting dopamine metabolism, suggesting that an underlying hypodopaminergic state is important to the pathogenesis. Mimickers of oculogyric crises exist, and we propose diagnostic criteria to distinguish true oculogyric crises. Recognition of oculogyric crises is important for the diagnosis and appropriate treatment of rare …

Latent inhibition in an insect: the role of aminergic signaling.

Latent inhibition (LI) is a decrement in learning performance that results from the nonreinforced pre-exposure of the to-be-conditioned stimulus, in both vertebrates and invertebrates. In vertebrates, LI development involves dopamine and serotonin; in invertebrates there is yet no information. We studied differential olfactory conditioning of the proboscis extension response in the honeybee Apis mellifera, and we compared LI in individuals treated with antagonists of biogenic amines (dopamine, octopamine, and serotonin). An antagonist of octopamine receptors and two antagonists of serotonin re…

Does dysregulation of catechol-O-methyltransferase predispose to opioid-induced hallucinations? A report of a patient with microdeletion of chromosome 22 and opioid-associated hallucinations.

Velocardiofacial syndrome is a genetic disorder associated with a microdeletion on the long arm of chromosome 22, and this segment is responsible for coding catechol-O-methyltransferase, an enzyme involved in dopamine degradation. We submit a case of velocardiofacial syndrome and Madelung deformity of the wrists presenting with hallucinatory phenomena associated with opioid exposure. Overactivity of the dopaminergic system has been postulated to cause schizophrenia in this population, and here we speculate that dysregulation of dopamine metabolism may have predisposed our patient to an increas…

[Correlation between the levels of calcium in the blood and catecholamines in the brain during memory formation and fixation in hypoparathyreosis].

Relationship between the blood level of calcium and the level of catecholamines in the brain limbic structures was studied in passive avoidance conditioning and extinction in rats with hypoparathyreosis. After parathyroidectomy, conditioning processes were shown to be impaired as a result of a disorder of calcium supply. In hypoparathyreosis, not only the basic dopamine and noradrenalin levels change, but catecholamine dynamics in learning and forced extinction of a passive avoidance reaction shifts. The results point to the deranged functioning of dopamine and noradrenaline brain systems as a…
