Fatigue Damage-Tolerant Hydrogel Engineered via Synergy of Kinetically Distinct Metal-Ligand Crosslinks.

Biological tissues resist fatigue by possessing high fatigue threshold and extremely slow crack growth above the threshold. In contrast, most synthetic hydrogels rely solely on increasing fatigue thresholds and fracture rapidly once cracks start to propagate. Here, we realize fatigue damage tolerance design (FDT) for hydrogels via the synergy of two kinetically distinct metal-ligand crosslinks. Under cyclic loads, the strong and irreversible crosslinks break to dissipate energy, shielding the weak and reversible crosslinks to re-associate upon unloading. The synergistic kinetic effects between

Shape-engineered multi-wavelength lasing in GaN-based microcavity arrays with lateral nanoporous DBRs.

Compact multi-wavelength laser sources are essential for high-capacity optical communication, optical computing, and multi-channel sensing in integrated photonic circuits. Here, we demonstrate a low-threshold GaN-based multi-wavelength microcavity laser array by engineering cavity shapes with lateral nanoporous distributed Bragg reflectors (DBRs). Circular, hexagonal, and square microcavities exhibit distinct lasing wavelengths of 459, 444, and 428 nm, covering a spectral span of 31 nm, with corresponding lasing threshold average power densities of 0.43, 0.99, and 3.75 W/cm2, respectively. The…

Precision Engineered Dissolving Microneedles Enable Green-Light Activated Chemo-Photodynamic Therapy for Psoriasis.

Psoriasis exhibits limited response to monotherapy, and customized combination strategies for synergistic treatment are constrained by skin permeability and insufficient control of drug activation. This study proposes a dissolving microneedle (MN) platform that integrates photodynamic therapy and chemotherapy for synergistic psoriasis treatment. The platform utilizes high-precision projection microstereolithography (PμSL) combined with a template replication strategy, coloading methotrexate (MTX) and a novel photosensitizer, 8-Phenyl-2,6-diiodo-1,3,5,7-tetramethyl BODIPY (I-BDP), into a poly(v

Oxygen Vacancy-Engineered CaCu3Ti4O12 Nanocatalyst for Piezoelectrically Driven Cascade Apoptosis/Cuproptosis/Ferroptosis Therapy.

Ultrasound (US) induced piezoelectric catalytic therapy is an emerging cancer treatment method. However, the development and optimization of piezoelectric catalyst remain the major challenge. Herein, we have converted the classical dielectric material CaCu3Ti4O12 (CCTO) into an efficient piezoelectric material through oxygen vacancy (VO) engineering, enabling piezoelectric catalytic-driven cascade tumor therapy. The introduction of VO results in strong polarization and a robust piezoelectric coefficient. Density functional theory (DFT) calculations reveal that VO acts as an electron trap to su…

Tissue-engineered blood vessels for clinical translation: Design logic, representative advances and persistent barriers.

Tissue-engineered blood vessels are promising alternatives for small-diameter vascular reconstruction, but clinical translation remains limited by thrombosis, compliance mismatch, delayed endothelialization and unstable host remodeling. This review synthesizes representative advances in scaffold design, seed-cell selection, bioactive regulation and biofabrication, emphasizing how these components interact under hemodynamic load. Rather than cataloguing technologies, we argue that translational success depends on matching material architecture, cellular phenotype, immune remodeling and manufact

Genetically engineered extracellular vesicles-based collagen-targeting bioinspired scaffold for tendon regenerative repair.

Tendon injuries, particularly massive tendon defects, pose significant clinical challenges due to the limited regenerative capacity of tendons and suboptimal outcomes of current therapies. This study presents a bioinspired scaffold that integrates genetically engineered extracellular vesicles (EVs) with collagen-targeting capabilities into a decellularized bovine tendon sheet (DBTS) for tendon regenerative repair. Rat tendon-derived stem cells (TDSCs) were genetically modified to overexpress biglycan (Bgn) and fibromodulin (Fmod), producing bioactive EVs that drive tenogenic differentiation vi

Targeted Lactic Acid Regulation via Engineered Exosomes for Synergistic Metabolic and Photodynamic Tumor Therapy.

Tumor cells exhibit a hyper-glycolytic phenotype, resulting in massive lactic acid (LA) production that acidifies the tumor microenvironment (TME) and fosters immunosuppression. Current lactate-targeted therapies often lack synergistic dual-directional regulation. Herein, we engineer an exosomal nanoplatform, PpIX/siRNA@EXO-LOD, to simultaneously disrupt intra- and extracellular LA homeostasis, thereby enabling synergistic metabolic and photodynamic therapy (PDT). The system integrates three key components: protoporphyrin IX (PpIX) for PDT, siRNA targeting monocarboxylate transporter 4 (MCT4)

Morphology-Engineered Ceria Nanoparticles Ameliorate Periodontitis via Enhanced Antioxidant and Anti-Inflammatory Activities.

Periodontitis is a chronic inflammatory oral disease featured with continuous alveolar bone resorption, where a ccumulated reactive oxygen species (ROS) aggravate tissue damage and local inflammation. Ceria nanoparticles (CNPs) act as promising redox nanozymes with excellent ROS-scavenging and anti-inflammatory capacities. Their catalytic performance is highly dependent on specific surface area (SSA), surface Ce3+ fraction and oxygen vacancy (Ov) concentration, all of which are closely regulated by particle morphology. Nevertheless, the correlation between CNP morphology and therapeutic effica

Hierarchically Engineered Nanocellulose-Halloysite Composite Foams for Integrated Thermal Insulation and Intrinsic Fire Resistance.

Achieving the simultaneous integration of thermal insulation, mechanical robustness, hydrophobicity, and intrinsic flame retardancy in bio-based porous materials remains a major challenge. Here, we report a hierarchically engineered nanocomposite foam composed of nanofibrillated cellulose (NFC), alkyl ketene dimer (AKD), and halloysite nanotubes (HNT), designed via a multiscale structure-property approach. Incorporation of HNT (1-10 wt%) reinforces the fibrillar network and introduces a hierarchical macro-mesoporous architecture, while AKD imparts durable hydrophobicity with water contact angl

The redox architecture of gestational diabetes mellitus: from cellular stress engine to epigenetic and mitochondrial rewiring.

Gestational diabetes mellitus (GDM) is a common pregnancy complication with a rising global prevalence, posing serious short-term and long-term health threats to both mothers and offspring. This review repositions GDM as a systemic disorder in which oxidative stress acts as a proposed mechanistic hub, linking upstream risk factors to downstream pathophysiology. We first examine how "upstream" factors-including genetic susceptibility, pre-conception status, and environmental exposures-converge to promote a state of pathological redox imbalance. We then examine key mechanistic pathways through w

Dual-Template Engineered Low-Tortuosity Hydrogels with Gradient Microchannels for On-Demand Management of Refractory Wounds.

Chronic refractory wounds present a significant clinical challenge due to the vicious cycle of excessive exudate and bacterial infections. Herein, we develop a smart hydrogel dressing via a dual-template strategy that creates a low-tortuosity structure with gradient microchannels. Compared with the random porous hydrogel, the optimized structure accelerates the absorption rate of viscous liquid by ∼17 times. Crucially, the hydrogel incorporates a thermal feedback loop: the increase in wound temperature caused by infection stimulates the hydrogel to rapidly release antibacterial agents, thereby

Engineered bacterial outer membrane vesicle vaccine prevents T cell exhaustion for antibacterial immunotherapy.

Gram-negative pathogens evade immune clearance and promote chronic infections by residing intracellularly. While outer membrane vesicles (OMVs) hold promise as antibacterial vaccine platforms, their clinical potential is limited by lysosomal entrapment of antigens and endotoxin-induced toxicity. Herein, we present an innovative nano-encapsulation strategy to engineer OMVs and overcome these two obstacles. We engineered Porphyromonas gingivalis (P. gingivalis) OMVs by incorporating metal ion adjuvants, coordinated with phenolic ligands, to form a rigid, acid-responsive nanoshell. This shell enh

Evaluation of the delivery of an anti-Listeria endolysin via CRISPR-Cas9 engineered probiotic Saccharomyces boulardii.

Listeriosis is a foodborne infection caused by Listeria monocytogenes that causes febrile gastroenteritis and central nervous system infections and that can often lead to fatality. Upon consumption of contaminated food, Listeria is able to survive a number of gastrointestinal stressors, including competition with the host microbiota. The emergence of antibiotic-resistant clones of L. monocytogenes, together with the side effects of antibiotic treatment, highlights the need for alternatives or additives for its treatment and prevention. Saccharomyces boulardii is a probiotic yeast that is often
