Molecular mechanisms of alcohol-induced liver injury. An Update.

Alcoholic liver disease has a known etiology but a complex pathogenesis. The understanding of how alcohol damages the liver has expanded substantially over the last decade. In particular the genesis of fatty liver, the effect of the metabolites of ethanol oxidation, the interaction between endotoxin and kupffer cells, and the genetic predisposition to develop severe liver disease have been the focus of a great deal of research. Recent studies have demonstrated that in addition to the altered redox state ethanol induced lipid accumulation by altering the expression and transcriptional regulatio

Association between Alcohol Consumption and Metabolic Dysfunction-Associated Steatotic Liver Disease Based on Alcohol Flushing Response in Men: The Korea National Health and Nutrition Examination Survey 2019-2021.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is distinguished by the buildup of excessive liver fat unrelated to alcohol consumption. However, the role of alcohol consumption on disease progression is debatable. Recently, alcohol flushing syndrome in Asian populations has gained interest, and its role in the risk of developing MASLD is unknown. Therefore, in this cross-sectional study, we investigated the association between alcohol consumption and MASLD in Korean men, considering their alcohol flushing response and utilizing the lipid accumulation product (LAP) score. Data

Purification and characterization of alcohol dehydrogenase reducing N-benzyl-3-pyrrolidinone from Geotrichum capitatum.

(S)-N-Benzyl-3-pyrrolidinol is widely used in the synthesis of pharmaceuticals as a chiral building block. We produced 30 mM (S)-N-benzyl-3-pyrrolidinol (enantiometric excess > 99.9%) from the corresponding ketone N-benzyl-3-pyrrolidinone with more than 99.9% yield in 28 h of the resting-cell reaction of Geotrichum capitatum JCM 3908. NAD(+)-dependent alcohol dehydrogenase reducing N-benzyl-3-pyrrolidinone from G. capitatum JCM 3908 was purified to homogeneity by ammonium sulfate fractionation and a series of DEAE-Toyopearl, Butyl-Toyopearl, Superdex 200, and Hydroxyapatite column chromatograp

Morphometry of liver parenchyma in needle biopsy specimens from patients with alcoholic liver disease: preliminary variables for the diagnosis and prognosis of cirrhosis.

The diagnosis of liver cirrhosis depends on assessing fibrosis and architectural alterations of the liver. In a needle biopsy specimen the connective tissue is often inadequately sampled, which leads to an uncertain diagnosis. Parenchymal features alone are currently insufficient. We, therefore, carried out a comprehensive morphometric study to detect parenchymal structures that may be useful in the diagnosis and prognosis of cirrhosis. Five human liver biopsy specimens were selected from each of four alcoholic disease groups: fatty liver, alcoholic hepatitis, cirrhosis with greater-than-5-yr

Use of the triolein breath test in alcoholics with liver damage.

The triolein breath test (TBT) is a simple, noninvasive technique for the evaluation of steatorrhea. However, because it depends on intermediary hepatic processes, and because both liver damage and pancreatic dysfunction often co-exist in the alcoholic, the overall usefulness of the test in patients with liver injury was reassessed. We found that even in the absence of steatorrhea, a majority of patients with advanced liver injury (alcoholic hepatitis, cirrhosis, or both) had abnormal TBT results that failed to correct with pancreatic extract. In contrast, patients with less severe lesions (st

[The effect of caerulein on intraluminal pressures of the common bile duct and duodenum in man (author's transl)].

Caerulein is a dekapeptide related to gastrin and cholecystokinin with profound effects on gastrointestinal motility. In the present study we examined the effect of intravenously administered caerulein (0.1 microgram/kg body weight) on the intraluminal pressure of common bile duct and duodenum in 12 patients. The pressure recordings were obtained by endoscopic perfusion manometry. The tip of the perfused catheter was positioned under radiological control and the measurements were carried out under standardized conditions. Following the injection of caerulein there was a rise in pressure in the

Aspartate treatment ameliorates metabolic dysfunction-associated fatty liver disease through upregulation of CBS and suppression of ferroptosis.

Dysregulation of non-essential amino acids (NEAAs) contributes to metabolic dysfunction-associated fatty liver disease (MAFLD) progression. Aspartate (ASP), an NEAA serving as a critical precursor for purine, asparagine, and arginine biosynthesis, exhibits unexplored pathophysiological significance in MAFLD. Mass spectrometry profiling identified hepatic ASP as the sole NEAA consistently depleted in both human patients with MAFLD and animal models. Dietary or orally administered ASP supplementation in mice significantly improved insulin sensitivity, suppressed de novo lipogenesis, and enhanced

METTL16 promotes MASLD progression through the regulation of lipid synthesis and immune response.

BACKGROUND: Methyltransferase-like 16 (METTL16) is a recently identified m6A RNA methyltransferase. Although its role in hepatocellular carcinoma has been explored, its function in metabolic dysfunction-associated steatotic liver disease (MASLD) remains elusive. METHODS: Expression profiles of methylation-related genes were analyzed in a cohort comprising 206 MASLD patients and 10 healthy controls. Functional assays were conducted using MASLD cell models and diet-induced mouse models. Bioinformatic analyses were performed to evaluate immune cell infiltration and signaling pathway activation. P

Cytokeratin-18 increase as a signal for early liver cell changes in high-fat diet-induced obesity before overt steatohepatitis.

OBJECTIVE: The major cause of non-alcoholic fatty liver disease (NAFLD) is obesity; in fact, this condition has been defined as metabolic dysfunction-associated steatotic liver disease (MASLD). Non-invasive diagnosis and grading of NAFLD are the subjects of the studies. Cytokeratin-18 (CK-18) is particularly highlighted as an indicator of liver cell death and steatohepatitis, but its relationship with obesity remains unclear. This study aimed to evaluate the metabolic parameters, CK-18 levels, and liver histopathology in high-fat diet-induced obesity in rats. METHODS: Rats were fed either a n

MASLD as a systemic metabolic disease: expanding the scope of cardiovascular-kidney-metabolic (CKM) syndrome.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a highly prevalent and progressive liver condition that is increasingly recognized for its systemic cardiometabolic impacts. MASLD increases the risk of cardiovascular and renal complications mainly through shared mechanisms, such as insulin resistance, low-grade inflammation, oxidative stress, atherogenic dyslipidemia, and a procoagulant state. Although these interrelated processes drive multisystem damage, MASLD remains often underdiagnosed in cardiology and nephrology settings and is excluded from the recently proposed fram

Decreased degree of adipocyte differentiation in visceral adipose tissue contributes to metabolic dysfunction-associated steatotic liver disease.

The mechanisms connecting the human fat depots, subcutaneous adipose tissue (SAT) and visceral adipose tissue (VAT), to metabolic dysfunction-associated steatotic liver disease (MASLD) remain elusive. We hypothesize that in individuals with obesity, a decreased degree of adipocyte differentiation may contribute to ectopic fat accumulation in the liver, seen in MASLD. Here we show, using single nucleus RNA-sequencing from adipose tissue biopsies, that the predicted degree of VAT adipocyte differentiation is decreased in individuals with MASLD, with an attenuated impact observed in SAT adipocyte

Thyroid-Liver Axis: The Role of Thyroid Dysfunction in the Pathogenesis and Progression of MASLD: A Comprehensive Review.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is one of the leading causes of chronic liver disease worldwide with an estimated prevalence ranging between 30% and 38% of the global population. Increasing evidence highlights the complex role of thyroid dysfunction in MASLD pathogenesis. Although many aspects remain unclear, thyroid dysfunction is emerging as a potential risk factor. Thyroid hormones (THs) regulate crucial metabolic processes, including lipid metabolism and mitochondrial function, both central to the development and progression of MASLD. Dysregulated thyroid s

A precision-constrained framework for evaluating noninvasive biomarkers in MASLD and beyond.

In patients with Metabolic Dysfunction-associated Steatotic Liver Disease (MASLD), accurate and low-cost non-invasive risk stratification remains a major unmet need. We developed a clinical records-based neural network integrating patient history, routine laboratory tests, and ultrasound imaging features. Here we show that in an internal test set (n = 209), the model achieved receiver operating characteristic area under the curve (ROC-AUC) values of 0.85 vs 0.82 (F ≥2), 0.90 vs 0.86 (F ≥3), 0.96 vs 0.89 (F  = 4) compared with Fibrosis-4 (FIB-4). To address limitations of ROC-AUC, we applied RP
