Enhanced susceptibility of B lymphoma cells to measles virus by Epstein-Barr virus type III latency that upregulates CD150/signaling lymphocytic activation molecule.

Measles virus (MV) is one of the candidates for the application of oncolytic virotherapy (OVT). Although an advanced clinical study has been reported on a T-cell lymphoma, the potential of MV OVT against B-cell lymphomas remains to be clarified. We found that an EBV-transformed B lymphoblastoid cell line, a model for diffuse large B-cell lymphoma, and EBV-positive Burkitt's lymphoma cells bearing type III latency were highly susceptible to the cytolysis induced by an MV vaccine strain CAM-70. As analyzed by EBV-positive and -negative counterparts of the same cytogenetic background, type III EB

CXCR6+ natural killer cell immunotherapy preserves CD4+ T helper cells in humanized mice.

Human immunodeficiency virus (HIV) persists despite antiretroviral therapy because long-lived viral reservoirs are not eliminated, and ongoing or rebound infection contributes to progressive loss of CD4+ T helper cells. Natural killer (NK) cells can acquire adaptive, antigen-experienced functions, including recall responses to HIV envelope protein, suggesting that defined NK-cell subsets may be therapeutically useful against HIV. Because HIV-responsive adaptive NK-cell activity is enriched among CXC chemokine receptor 6-positive (CXCR6+) NK cells, we tested whether CXCR6+ NK cells provide enha

A comprehensive immune repertoire study for patients with pulmonary tuberculosis.

BACKGROUND: Tuberculosis (TB) is a major global health problem and has replaced HIV as the leading cause of death from a single infectious agent. METHODS: Here, we applied high throughput sequencing to study the immune repertoire of nine pulmonary tuberculosis patients and nine healthy control samples. RESULTS: Tuberculosis patients and healthy controls displayed significantly different high express clones and distinguishable sharing of CDR3 sequences. The TRBV and TRBJ gene usage showed higher expression clones in patients than in controls and we also found specific high express TRBV and TR

Tuberculosis Therapy Modifies the Cytokine Profile, Maturation State, and Expression of Inhibitory Molecules on Mycobacterium tuberculosis-Specific CD4+ T-Cells.

BACKGROUND: Little is known about the expression of inhibitory molecules cytotoxic T-lymphocyte antigen-4 (CTLA-4) and programmed-death-1 (PD-1) on Mycobacterium tuberculosis (Mtb)-specific CD4 T-cells and how their expression is impacted by TB treatment. METHODS: Cryopreserved PBMCs from HIV-TB co-infected and TB mono-infected patients with untreated and treated tuberculosis (TB) disease were stimulated for six hours with PPD and stained. Using polychromatic flow cytometry, we characterized the differentiation state, cytokine profile, and inhibitory molecule expression on PPD-specific CD4 T-

Early depletion of Mycobacterium tuberculosis-specific T helper 1 cell responses after HIV-1 infection.

BACKGROUND: The acid-fast bacillus Mycobacterium tuberculosis is often the first manifestation of acquired immunodeficiency syndrome in patients infected with human immunodeficiency virus (HIV). This study was conducted to better understand the mechanism underlying M. tuberculosis-specific pathogenicity early after onset of HIV infection. METHODS: M. tuberculosis-specific T helper 1 (Th1) cells were studied in HIV negative (n=114) and chronically HIV infected (n=68) Tanzanian subjects by using early secreted antigenic target 6 (ESAT6) protein or tuberculin (purified protein derivative) with i

Endothelial Protein C Receptor and Its Impact on Rheumatic Disease.

Endothelial Protein C Receptor (EPCR) is a key regulator of the activated protein C anti-coagulation pathway due to its role in the binding and activation of this protein. EPCR also binds to other ligands such as Factor VII and X, γδ T-cells, plasmodium falciparum erythrocyte membrane protein 1, and Secretory group V Phospholipases A2, facilitating ligand-specific functions. The functions of EPCR can also be regulated by soluble (s)EPCR that competes for the binding sites of membrane-bound (m)EPCR. sEPCR is created when mEPCR is shed from the cell surface. The propensity of shedding alters dep

Current treatment approach and future perspectives in B cell lymphoma.

Diffuse large B cell lymphoma (DLBCL) and follicular lymphoma (FL) represent the two major subtypes of mature B cell lymphoma. A deeper understanding of tumor biology, as well as molecular classification characterized by targetable gene alterations, and the introduction of novel treatment options, including targeted drugs (e.g., antibody-drug conjugates and small molecules [e.g., Bruton tyrosine kinase inhibitor]) and immune therapies (e.g., chimeric antigen receptor [CAR] T cell therapy and bispecific antibody [BsAb]), has changed the treatment paradigms for DLBCL and FL. In clinical practice

Dynamics and function of Langerhans cells in vivo: dermal dendritic cells colonize lymph node areas distinct from slower migrating Langerhans cells.

Langerhans cells (LCs) are prominent dendritic cells (DCs) in epithelia, but their role in immunity is poorly defined. To track and discriminate LCs from dermal DCs in vivo, we developed knockin mice expressing enhanced green fluorescent protein (EGFP) under the control of the langerin (CD207) gene. By using vital imaging, we showed that most EGFP(+) LCs were sessile under steady-state conditions, whereas skin inflammation induced LC motility and emigration to lymph nodes (LNs). After skin immunization, dermal DCs arrived in LNs first and colonized areas distinct from slower migrating LCs. LCs

Spatial Transcriptomic and Proteomic Analysis Identifies Novel Immune-Associated Biomarkers of Anti-PD1 Response in Metastatic Melanoma.

Resistance to anti-PD1 therapy remains a major challenge in metastatic melanoma, despite long-term benefit in some patients. Since a functional dendritic cell (DC)-CD8+ T cell axis is essential for anti-PD1 response, biomarkers associated with this axis may help characterize underexplored immune states linked to resistance. Here, we further characterized poor survival predictor (PSP) biomarkers previously negatively correlated with DC-CD8+ T cell axis in melanoma, evaluating their associations with anti-PD1 response, immune infiltration, and transcriptional and proteomic signatures. Pre-treatm

Explainable graph learning for multimodal single-cell data integration.

BACKGROUND: Understanding cellular heterogeneity and identifying functionally distinct subpopulations are central goals in single-cell analysis. Integrating paired multi-omic data, such as transcriptomic and proteomic profiles from the same cells, offers a more comprehensive view of cell states. However, current integration methods often struggle to balance expressiveness with interpretability, largely due to the complex and non-linear relationships across modalities. RESULTS: We present Single-Cell PROteomics Vertical Integration (SCPRO-VI), a new algorithm designed to integrate paired singl

Ceramides induce a form of apoptosis in human acute lymphoblastic leukemia cells that is inhibited by Bcl-2, but not by CrmA.

The generation of ceramides by the action of acidic and/or neutral sphingomyelinases has been implicated in many forms of apoptosis. We investigated whether exposure to ceramides is sufficient to induce apoptosis in human leukemia cells and, if so, what the characteristics of this form of apoptosis might be. Treatment of the acute lymphoblastic T-cell line CEM-C7H2 with short- and medium-chain ceramide analogs (C2-, C6-, and C8-ceramide) resulted in apoptosis, whereas the inactive C2-dihydroceramide had no effect on cell survival. Induction of apoptosis was relatively slow (approximately 40% a

Intercellular Transfer of CD155 Mediated by Melanoma Extracellular Vesicles Downregulates DNAM-1 Expression on NK Cells.

Melanoma is a highly immunogenic cancer, and immunotherapy is the first-line treatment for patients with advanced disease. However, CD155 overexpression in melanoma has been associated with resistance to immunotherapy. Whether CD155 can be transferred from tumour cells via extracellular vesicles (EVs), and how this transfer affects immune responses in melanoma, remains unclear. To investigate melanoma cell transfer of CD155 in vivo, B16F10-WT and B16F10-CD155KO melanoma cells were subcutaneously injected into CD155-/- mice. We show that CD155 is transferred from melanoma cells to several immun

Follow-up of the T-cell clonality in Sezary patients treated by extracorporeal photopheresis using a new assay: the immunoscope technique.

Sezary syndrome is a leukemic form of epidermotropic cutaneous T-cell lymphoma related to the malignant proliferation of clonal CD4+ T-cells. Extracorporeal photochemotherapy (ECP) may induce a transient improvement of the clinical signs but it's efficiency is discussed. In order to investigate the T-cell clonality in the peripheral blood of patients with Sezary syndrome and to monitor its evolution in 8 patients treated by ECP, we used the Immunoscope technique. In one patient, we observed a decrease of the T-cell clonality from 15.6% to 0%, paralleling a complete remission of the clinical di

Lack of muco-cutaneous signs of toxic shock syndrome when T cells are absent: S. aureus shock in immunodeficient adults with multiple myeloma.

Staphylococcal toxic shock syndrome (TSS) is an acute life threatening disease. The diagnosis can be made clinically based on diagnostic criteria. The clinical manifestations are caused in large part by there lease of high levels of T-cell-derived cytokines as a result of potent toxins, also called superantigens (SAg), produced by Staphylococcus aureus, but it is not clear which clinical symptoms/signs are strictly T-cell dependent. Here, we report on three adults with multiple myeloma (MM) presenting with S.aureus sepsis/shock, and two patients with typical TSS. The MM patients had compromise

Serglycin Drives LAG3+ Treg Differentiation and Immunosuppression in Gastric Cancer.

UNLABELLED: The enrichment of CD4+ regulatory T cells (Treg) drives gastric cancer immunosuppression. Revealing the underlying mechanisms driving Treg differentiation could help develop strategies to stimulate antitumor immunity. Serglycin (SRGN), derived from tumor cells, facilitates tumor progression in gastric cancer primarily via its receptor CD44, which is highly expressed in T cells, especially Tregs. This study investigated the immunomodulatory functions of tumor cell-derived SRGN in gastric cancer. Analysis of an integrated single-cell RNA sequencing dataset of 23 paired gastric cancer

Research progress on the molecular mechanisms of PD-1 and LAG-3 synergy in regulating T cell exhaustion and immunotherapy.

BACKGROUND: PD-1 and LAG-3 are immune checkpoint molecules frequently co-expressed in the tumor microenvironment, where they synergistically drive T-cell exhaustion and immune escape. Dual blockade of these pathways represents a promising strategy to overcome immunotherapy resistance. METHODS: This review systematically synthesizes mechanistic studies on PD-1/LAG-3 synergy and summarizes preclinical and clinical advances in dual blockade therapies, with emphasis on bispecific antibodies and combination trial data. RESULTS: PD-1 and LAG-3 cooperatively suppress T-cell function via complementa

Single-Cell Immune Profiling Suggests Non-Classical Monocyte Is Associated Immunodepression in Tuberculosis Treatment Non-Responders.

Sputum culture conversion (SCC) at 2 months is an early indicator of tuberculosis (TB) treatment response, yet the associated immune alterations remain incompletely defined. We performed single-cell RNA sequencing on peripheral blood mononuclear cells from eight TB patients, stratified by 2-month SCC status. Non-responders showed a relative enrichment of non-classical monocytes and higher TB progression risk scores. CellChat analysis inferred altered IL16 and TRAIL communication involving these cells. CD4+ Tregs in non-responders showed higher LGALS9 expression and inferred GALECTIN signalling

Buffering Mechanism in Aortic Arch Artery Formation and Congenital Heart Disease.

BACKGROUND: The resiliency of embryonic development to genetic and environmental perturbations has been long appreciated; however, little is known about the mechanisms underlying the robustness of developmental processes. Aberrations resulting in neonatal lethality are exemplified by congenital heart disease arising from defective morphogenesis of pharyngeal arch arteries (PAAs) and their derivatives. METHODS: Mouse genetics, lineage tracing, confocal microscopy, and quantitative image analyses were used to investigate mechanisms of PAA formation and repair. RESULTS: The second heart field (

Cationic antimicrobial peptides in psoriatic skin cooperate to break innate tolerance to self-DNA.

Psoriasis is a T-cell-mediated skin autoimmune disease characterized by the aberrant activation of dermal dendritic cells (DCs) and the sustained epidermal expression of antimicrobial peptides. We have previously identified a link between these two events by showing that the cathelicidin antimicrobial peptide LL37 has the ability to trigger self-nucleic acid mediated activation of plasmacytoid DCs (pDCs) in psoriatic skin. Whether other cationic antimicrobial peptides exert similar activities is unknown. By analyzing heparin-binding HPLC fractions of psoriatic scales, we found that human beta-
