Autologous hematopoietic stem cell product contaminated with Salmonella due to occult salmonellosis in an asymptomatic donor.

Recent advancements in infectious disease testing methods and pathogen reduction technologies have greatly reduced the incidence of microbial contamination of allogeneic blood products. Despite this significant reduction, contamination of autologous cellular therapy products remains a challenging issue, as many of these mitigation strategies are not feasible for such products. Most microorganisms isolated from cellular therapy products are Gram-positive normal skin flora, and studies have demonstrated that adverse effects are infrequent when these contaminated products are infused. However, no

Design, synthesis and antibacterial activity evaluation of moxifloxacin-amide-1,2,3-triazole-isatin hybrids.

In this work, a series of novel moxifloxacin-amide-1,2,3-triazole-isatin hybrids 7a-l were designed and synthesized. The in vitro antibacterial activity against a panel of clinically important Gram-positive and Gram-negative bacteria including drug-resistant pathogens was also evaluated. All hybrids showed considerable activity against the tested pathogens with MIC values of ≤0.03 to 128 μg/mL, and some of them such as hybrids 7e, 7g and 7j were comparable to or better than the parent moxifloxacin (MIC: ≤0.03-8 μg/mL). Moreover, hybrids 7e, 7g and 7j also demonstrated low cytotoxicity towards …

Long-Chain Alkyl Cyanides: Unprecedented Volatile Compounds Released by Pseudomonas and Micromonospora Bacteria.

The analysis of volatiles from bacterial cultures revealed long-chain aliphatic nitriles, a new class of natural products. Such nitriles are produced by both Gram-positive Micromonospora echinospora and Gram-negative Pseudomonas veronii bacteria, although the structures differ. A variable sequence of chain elongation and dehydration in the fatty acid biosynthesis leads to either unbranched saturated or unsaturated nitriles with an ω-7 double bond, such as (Z)-11-octadecenenitrile, or methyl-branched unsaturated nitriles with the double bond located at C-3, such as (Z)-13-methyltetradec-3-eneni

Snake venom-like waprin from the frog of Ceratophrys calcarata contains antimicrobial function.

A 255-bp cDNA encoding an 84-amino acid residue (aa) precursor protein containing 8 half-cysteines was cloned from the skin of the frog, Ceratophrys calcarata. By sequence comparison and signal peptide prediction, the precursor was predicted to release a 63-aa mature peptide with amino acid sequence, NVTPATKPTPSKPGYCRVMDELILCPDPPLSKDLCKNDSDCPGAQKCCYRTCIMQCLPPIFRE. The mature was named ceratoxin. Ceratoxin shares significant sequence similarity with the toxin family of waprins containing the whey acidic protein-type (WAP) four-disulfide core domain found in snake venoms. Antimicrobial and tryps

Culture of body fluids using the bact/alert system.

The BacT/Alert system was evaluated for its utility to recover clinically significant bacteria from several sterile body fluids, other than blood. Of the1500 specimens processed, 530 (35.33%) cultures were recorded positive by the BacT/Alert bottles while only 379 (25%) cultures were recovered on the plate cultures. The mean time to detection was < 12 hours for Gram-negative organisms and 12 - 16 hours for the Gram-positive organisms. The rapid detection of important pathogens like Streptococcus pneumoniae, Streptococcus pyogenes and Salmonella typhi, within 12 hours of inoculation, is of part

Haemophilus parainfluenzae biliary tract infection: rationale for an ascending route of infection from the gastrointestinal tract.

Haemophilus parainfluenzae was isolated from a bile specimen of a 56-year-old patient with elevated liver enzymes and chronic cholecystitis. Several factors contributing to nonbacteremic H. parainfluenzae infection of the biliary tract include (i) the not infrequent occurrence of H. parainfluenzae in stool specimens, (ii) the presence of an outer membrane protein similar to those found in enteric bacteria which facilitates adherence to the colonic mucosa, and (iii) an adequate supply of V factor (nicotinamide adenine dinucleotide) supplied from bile and the local flora, which is necessary for

Genomic analysis of prophages from Burkholderia gladioli exposes extensive genetic diversity.

The Burkholderia genus of Gram-negative bacteria are known for their extensive metabolic capabilities, their relatively plastic genomes, their high intrinsic resistance to antibiotics, and their ability to cause various diseases in humans, animals, and plants. Burkholderia gladioli is one such species, that is an opportunistic pathogen in people, causing sometimes fatal pneumonia, or in plants, causing onion bulb rot, mushroom soft rot, or rice panicle blight. Therapies that utilize bacterial viruses (bacteriophages, or phages) to counter or control pathogenic bacteria are a promising alternat…

Synthesis and Characterization of DOTAM-Based Sideromycins for Bacterial Imaging and Antimicrobial Therapy.

The rise of antimicrobial resistance, especially in Gram-negative bacteria, calls for novel diagnostics and antibiotics. To efficiently penetrate their double-layered cell membrane, we conjugated the potent antibiotics daptomycin, vancomycin, and sorangicin A to catechol siderophores, which are actively internalized by the bacterial iron uptake machinery. LC-MS/MS uptake measurements of sorangicin derivatives verified that the conjugation led to a 100- to 525-fold enhanced uptake into bacteria compared to the free drug. However, the transfer to the cytosol was insufficient, which explains thei…

Review of phenotypic assays for detection of extended-spectrum β-lactamases and carbapenemases: a microbiology laboratory bench guide.

BACKGROUND: Infections caused by gram-negative antibiotic-resistant bacteria continue to increase. Despite recommendations by the Clinical Laboratory Standards Institute (CLSI) and the European Committee on Antimicrobial Susceptibility Testing (EUCAST) with regards to detection of antibiotic degrading enzymes secreted by these bacteria, the true prevalence of extended-spectrum β-lactamase (ESBL) and carbapenemase producers remains a difficult task to resolve. Describing of previously designed phenotypic detection assays for ESBLs and carbapenemases in a single document avails a summary that al

Effect of High Intensity Blue Light on Fusobacterium nucleatum Membrane Integrity.

Oral malodour is considered to be caused mainly by the production of volatile sulfide compounds (VSC) by anaerobic gram-negative oral bacteria. Previous studies showed that these bacteria were susceptible to blue light phototoxicity mediated by the production of reactive oxygen species (ROS). In the present study, we tested the effect of blue light on the integrity Fusobacterium nucleatum's membrane, cellular proteins and DNA. Bacterial samples were exposed to high intensity blue light for 0, 70, 140 and 280 s (i.e. fluences of 0, 96, 192 and 384 J cm-2 , respectively). Following light exposur

Neisseria gonorrhoeae evades autophagic killing by downregulating CD46-cyt1 and remodeling lysosomes.

The Gram-negative human pathogen N. gonorrhoeae (Ngo) quickly attaches to epithelial cells, and large numbers of the bacteria remain on the cell surface for prolonged periods. Ngo invades cells but few viable intracellular bacteria are recovered until later stages of infection, leading to the assumption that Ngo is a weak invader. On the cell surface, Ngo quickly recruits CD46-cyt1 to the epithelial cell cortex directly beneath the bacteria and causes its cleavage by metalloproteinases and Presenilin/γSecretease; how these interactions affect the Ngo lifecycle is unknown. Here, we show Ngo ind

Whole-genome sequencing of a large collection of Myroides odoratimimus and Myroides odoratus isolates and antimicrobial susceptibility studies.

The genus Myroides comprises several species of Gram-negative, non-motile, and non-fermenting bacteria, which have been regarded as non-pathogenic for decades. Multiple recent reports, however, underscore the pathogenic potential that Myroides sp. possesses for humans. These bacteria seem to be resistant to a wide range of antibiotics (including ß-lactams and aminoglycosides). Therefore, treatment options are limited. Knowledge of antimicrobial resistance, however, is based on only one meaningful comprehensive study and on data published from case reports. This lack of data motivated us to tes

Commensal pathogen competition impacts host viability.

While the structure and regulatory networks that govern type-six secretion system (T6SS) activity of Vibrio cholerae are becoming increasingly clear, we know less about the role of T6SS in disease. Under laboratory conditions, V. cholerae uses T6SS to outcompete many Gram-negative species, including other V. cholerae strains and human commensal bacteria. However, the role of these interactions has not been resolved in an in vivo setting. We used the Drosophila melanogaster model of cholera to define the contribution of T6SS to V. cholerae pathogenesis. Here, we demonstrate that interactions be

A single point mutation at a splice site generates a silent RH50 gene in a composite heterozygous RHnull blood donor.

Vibrio cholerae is a Gram-negative bacterium that occurs naturally in aquatic environment. Only V. cholerae O1 and V. cholerae O139 produce cholera toxin and cause cholera, other serogroups can cause gastroenteritis, open wounds infection, and septicaemia. V. cholerae O1 and V. cholerae O139 grow and survive inside Acanthamoeba castellanii. The aim of this study is to investigate the interactions of the Swedish clinical isolates V. cholerae O3, V. cholerae O4, V. cholerae O5, V. cholerae O11, and V. cholerae O160 with A. castellanii. The interaction between A. castellanii and V. cholerae strai

A 16-year survey of changes in bacterial isolates associated with chronic suppurative otitis media.

Bacteria were isolated and cultured from the ears of outpatients with chronic suppurative otitis media (OMC) who attended the otologic clinics of Kyushu University Hospital from 1976 to 1991. Bacterial isolates were analyzed retrospectively to determine their incidences and antibiotic sensitivities. Staphylococcus aureus and Pseudomonas aeruginosa were the most commonly isolated organisms. The incidence of S. aureus infections gradually rose over 16 years, while the incidence of Proteus infections gradually declined. The frequency of isolation of glucose non-fermenting gram-negative rod (NF-GN

Quantitative definition of bacteriuria.

Quantitative criteria distinguish bacterial infection (or colonization) of the urine from contamination. These criteria depend on the fact that the density of bacteria in infected urine is usually several orders of magnitude higher than the density of bacteria in contaminated urine. Most research on quantitative definitions of infection has concerned Gram-negative rod infections in women. For asymptomatic bacteriuria, the most prevalent urinary tract infection, and for pyelonephritis, a criterion of 1 X 10(5) cfu/ml provides optimal separation of infection from contamination of voided urine.
