Preclinical Animal Models for Q Fever Vaccine Development.

Coxiella burnetii is a zoonotic pathogen responsible for the human disease Q fever. While an inactivated whole cell vaccine exists for this disease, its widespread use is precluded by a post vaccination hypersensitivity response. Efforts for the development of an improved Q fever vaccine are intricately connected to the availability of appropriate animal models of human disease. Accordingly, small mammals and non-human primates have been utilized for vaccine-challenge and post vaccination hypersensitivity modeling. Here, we review the animal models historically utilized in Q fever vaccine deve

[Modulation of Host Immune System by Staphylococcal Superantigen-like (SSL) Proteins].

Staphylococcus aureus is a common pathogen causing a wide range of infectious diseases in humans and animals. This bacterium secretes a variety of exoproteins, including toxins known as superantigens, such as toxic shock syndrome toxin-1 (TSST-1) and enterotoxins. Staphylococcal superantigen-like (SSL) proteins are a family of exoproteins showing structural similarities with superantigens but no superantigenic activity. This family is composed of 14 members (SSL1-SSL14), and recent studies have revealed that these members exhibit various immunomodulatory activities: e.g., inhibition of antibod

Vaccination against infectious bronchitis virus: A continuous challenge.

Infectious bronchitis virus (IBV) is a significant respiratory pathogen of commercial poultry that causes millions of dollars in lost revenue worldwide each year. Even though the poultry industry extensively vaccinates against IBV, emergence of new serotypes and variants continually occur, making control of the disease difficult. Current mass application strategies for IBV vaccines are inefficient and frequently result in vaccination failures. Novel vaccine technology development has been slow, and is hindered by the constraints of large-scale poultry production. Further complicating the situa

Bacterial gene regulation by alpha-hydroxyketone signaling.

Bacteria produce diffusible, small signaling molecules termed autoinducers to promote cell-cell communication. Recently, a novel class of signaling molecules, the alpha-hydroxyketones (AHKs), was discovered in the facultative human pathogens Legionella pneumophila and Vibrio cholerae. In this review, we summarize and compare findings on AHK signaling in these bacteria. The L. pneumophila lqs (Legionella quorum sensing) and V. cholerae cqs (cholera quorum sensing) gene clusters synthesize and detect Legionella autoinducer 1 (3-hydroxypentadecan-4-one) or cholera autoinducer-1 (3-hydroxytridecan

Helicobacter pylori-induced epithelial cell signalling in gastric carcinogenesis.

Helicobacter pylori represents a highly successful human microbial pathogen that infects the stomach of more than half of the world's population. H. pylori induces gastric inflammation, and the diseases that can follow such infection include chronic gastritis, peptic ulcers and, more rarely, gastric cancer. The reasons why a minority of patients with H. pylori develops gastric cancer could be related to differences in host susceptibility, environmental factors and the genetic diversity of the organism. This review examines the features of H. pylori-induced epithelial cell signalling in gastric

Regulatory T cells restrain IL-15-mediated cytotoxic and bystander T cell activity in mucosal tissue without compromising antigen-driven memory.

Many pathogenic human infections enter the host via a mucosal surface. These nonlymphoid tissues are abundantly populated by polyclonal memory CD8 T cells that persist following infections to protect the host in the event of repeat exposure. Memory T cells can be triggered via T cell receptor (TCR) interaction with their cognate antigen upon re-infection to exert effector functions, including cytotoxicity and cytokine production, and assist in pathogen elimination. Alternatively, some T cells are 'bystander activated' by cytokines without antigenic signal. This layered approach boosts efficien

Wbm0152, an outer membrane lipoprotein of the Wolbachia endosymbiont of Brugia malayi, inhibits yeast ESCRT complex activity.

Human pathogenic filarial nematodes of the family Onchocercidae, including Brugia malayi and Onchocerca volvulus, cause debilitating filarial diseases such as lymphatic filariasis and river blindness. These mosquito-borne pathogens are obligately colonized by the gram-negative intracellular alphaproteobacterium, Wolbachia pipientis, which is essential for nematode sexual reproduction, long-term survival, and pathogenicity in the mammalian host. Like many intracellular bacteria, Wolbachia likely uses numerous surface-exposed and secreted effector proteins to regulate its ability to persist and

Lipid droplet accumulation occurs early following Salmonella infection and contributes to intracellular bacterial survival and replication.

Salmonellosis is a public health problem caused by Salmonella sp., a highly adapted facultative intracellular pathogen. After internalization, Salmonella sp. Manipulates several host processes, mainly through the activation of the type III secretion system (T3SS), including modification of host lipid metabolism and lipid droplet (LD) accumulation. LDs are dynamic and complex lipid-rich organelles involved in several cellular processes. The present study investigated the mechanism involved in LD biogenesis in Salmonella-infected macrophages and its role in bacterial pathogenicity. Here, we repo

Cryptic Interactions in a Marine Microcosm: Sponges Shape the Microbiome of a Resident Goby.

Microbial symbionts influence ecological and evolutionary processes in their multicellular hosts; yet they remain poorly characterised in marine systems. Sponges host exceptionally diverse microbiomes, potentially providing rich microbial reservoirs for their residents. The tusked goby (Risor ruber), an obligate sponge-dweller, and its sponge hosts provide an ideal model to investigate the causes and consequences of shared microbial taxa in host-resident systems. Using 16S rRNA sequencing, we characterised the microbiomes of R. ruber skin and gut, their sponge hosts, a non-obligate, sponge-ass

MicroRNA expression profiling in fowl adenovirus serotype 4-infected Leghorn male hepatoma cells using small RNA deep sequencing.

Fowl adenovirus serotype 4 (FAdV-4) is the causative agent of hydropericardium-hepatitis syndrome (HHS), a disease that causes severe economic losses to the global poultry industry. MicroRNAs (miRNAs) are key regulators of host-pathogen interactions; yet, their temporal expression dynamics and functional roles during FAdV-4 replication in chicken-derived cells remain poorly defined. This study characterized the temporal expression profile of host miRNAs during FAdV-4 infection in Leghorn male hepatoma (LMH) cells to elucidate their regulatory roles in viral replication through small RNA deep s…

The Role of Wildlife and Climate Change in the Emergence of One Health-Related Viral Diseases in Australia.

Wildlife-origin viral diseases are an increasing One Health challenge in Australia, with climate change emerging as a key driver of disease emergence, transmission, and spillover. Australian wildlife, particularly marsupials, birds, and bats, serve as important reservoirs of zoonotic and vector-borne viruses. However, the combined influence of wildlife reservoirs and climate change on viral disease ecology in Australia has not been comprehensively synthesised. This review examines the epidemiology, significance, and climate sensitivity of wildlife-associated viral diseases and evaluates how cl

Nuclear receptor 4A1 is critical for neutrophil-dependent pulmonary immunity to Klebsiella pneumoniae infection.

INTRODUCTION: Bacterial pneumonia is a burdensome, costly disease and increasingly challenging to treat due to antibiotic resistance. Complex host-pathogen interactions regulate protective immunity. Neutrophils play a central role in pulmonary bacterial immunity, and mechanistic understanding of neutrophil functions in bacterial pneumonia has potential clinical and fundamental application. Nuclear receptor 4a1 (Nr4a1), a member of the nuclear orphan receptor family, has been described to regulate inflammation and immune development in a cell type-specific manner, but its role in pulmonary host…

MFSD6 is an entry receptor for enterovirus D68.

With the near eradication of poliovirus due to global vaccination campaigns, attention has shifted to other enteroviruses that can cause polio-like paralysis syndrome (now termed acute flaccid myelitis)1-3. In particular, enterovirus D68 (EV-D68) is believed to be the main driver of epidemic outbreaks of acute flaccid myelitis in recent years4, yet not much is known about EV-D68 host interactions. EV-D68 is a respiratory virus5 but, in rare cases, can spread to the central nervous system to cause severe neuropathogenesis. Here we use genome-scale CRISPR screens to identify the poorly character

Changes in the Transcriptome and Long Non-Coding RNAs but Not the Methylome Occur in Human Cells Exposed to Borrelia burgdorferi.

Lyme disease, caused by infection with members of the Lyme borreliosis group of Borrelia spirochete bacteria, is increasing in frequency and distribution worldwide. Epigenetic interactions between the mammalian host, tick, and bacterial pathogen are poorly understood.
