Comparative transcriptome analysis reveals significant differences in the regulation of gene expression between hydrogen cyanide- and ethylene-treated Arabidopsis thaliana.

BACKGROUND: Hydrogen cyanide (HCN) is a small gaseous molecule that is predominantly produced as an equimolar co-product of ethylene (ET) biosynthesis in plants. The function of ET is of great concern and is well studied; however, the function of HCN is largely unknown. Similar to ET, HCN is a simple and diffusible molecule that has been shown to play a regulatory role in the control of some metabolic processes in plants. Nevertheless, it is still controversial whether HCN should be regarded as a signalling molecule, and the cross-talk between HCN and ET in gene expression regulation remains u

Adsorption isotherms, degradation kinetics, and leaching behaviors of cyanogen and hydrogen cyanide in eight texturally different agricultural soils from China.

Cyanogen (C2N2) is a new and effective alternative soil fumigant to methyl bromide. The effects of soil properties on the fate of C2N2 and its degradation products, including hydrogen cyanide (HCN), are not fully understood. The objectives of this study were to determine the adsorption kinetics, adsorption isotherms, and degradation kinetics of C2N2 and HCN in texturally different soils and evaluate their leaching potentials using soil columns. Eight agricultural soils were collected throughout China: Luvisols (Hebei Province), Phaeozems (Heilongjiang Province), Gleysols (Sichuan Province), An

Protective effect of hydrogen-rich saline on pressure overload-induced cardiac hypertrophyin rats: possible role of JAK-STAT signaling.

BACKGROUND: Molecular hydrogen has been shown to have antioxidant effect and have been used to prevent oxidative stress-related diseases. The goal of this study was to explore if hydrogen-rich saline (HRS) plays a cardioprotective effect on abdominal aortic constriction (AAC) induced cardiac hypertrophy in rats. 60adult Sprague-Dawley rats received surgically the AAC for 6-week. After the surgery, the rats were randomly divided into 4 groups (15 for each):1: sham-operated (sham); 2: AAC-model; 3: AAC + Low HRS (LHRS); and 4: AAC + High HRS (HHRS). The rats in sham and AAC-model groups were tre

Berberine Protects Human Retinal Pigment Epithelial Cells from Hydrogen Peroxide-Induced Oxidative Damage through Activation of AMPK.

Age-related macular degeneration (AMD) is the leading cause of central vision loss in the elderly with less effective treatment, especially for dry AMD (90% of AMD). Although the etiology of this disease is not well elucidated, increasing evidences indicate that excessive reactive oxygen species (ROS) impairing the physiological functions of retinal pigment epithelium (RPE) cells may be one of the main causes. Therefore, it could be a great strategy to find some drugs that can effectively protect RPE cells from oxidative damage which is desired to treat and slow the process of AMD. In the pres

Duox1 Regulates Primary B Cell Function under the Influence of IL-4 through BCR-Mediated Generation of Hydrogen Peroxide.

Engagement of the BCR with Ags triggers signaling pathways for commitment of B lymphocyte responses that can be regulated, in part, by reactive oxygen species. To investigate the functional relevance of reactive oxygen species produced in primary B cells, we focused on the role of the hydrogen peroxide generator Duox1 in stimulated splenic B cells under the influence of the TH2 cytokine IL-4. We found that H2O2 production in wild type (WT) and Nox2-deficient CD19+ B cells was boosted concomitantly with enhanced expression of Duox1 following costimulation with BCR agonists together with IL-4, w

Hydrogen sulfide formation control and microbial competition in batch anaerobic digestion of slaughterhouse wastewater sludge: Effect of initial sludge pH.

High sulfur content in excess sludge impacts the production of biomethane during anaerobic digestion, meanwhile leads to hydrogen sulfide (H2S) formation in biogas. Effect of initial sludge pH on H2S formation during batch mesophilic anaerobic digestion of slaughterhouse wastewater sludge was studied in this paper. The results demonstrated that when the initial sludge pH increased from 6.5 to 8.0, the biogas production increased by 10.1%, the methane production increased by 64.1%, while the H2S content in biogas decreased by 44.7%. The higher initial sludge pH inhibited the competition of sulf

Hydrogen sulfide attenuates oxidative stress-induced NLRP3 inflammasome activation via S-sulfhydrating c-Jun at Cys269 in macrophages.

Oxidative stress and inflammation are closely related to cardiovascular diseases. Although hydrogen sulfide (H2S) has been shown to have powerful anti-oxidative and anti-inflammatory properties, its role in macrophage inflammation was poorly understood. The aim of this study was to investigate the role of H2S in the regulation of macrophage NLRP3 inflammasome activation. We reported here that H2S attenuated hydrogen peroxide (H2O2)-induced NLRP3 inflammasome activation, which led to caspase-1 activation and IL-1β production in macrophages. Moreover, H2S exerted its protective effects by loweri

Exogenous hydrogen sulfide protects from endothelial cell damage, platelet activation, and neutrophils extracellular traps formation in hyperhomocysteinemia rats.

Hydrogen sulfide (H2S) prevents endothelial cells damage and P-selectin of platelets promotes neutrophils extracellular traps (NETs) formation. However, how sodium hydrosulfide (NaHS), a donor that produces H2S regulates the activation of platelets and whether H2S inhibits the formation of neutrophils extracellular traps in hyperhomocysteinemia rats have not been previously investigated. The morphological and ultrastructural alterations of endothelial cells (ECs) and platelets were tested by transmission electron microscopy. The expressions of P-selectin of platelets were determined by flow cy

Hydrogen sulfide protects against acetaminophen-induced acute liver injury by inhibiting apoptosis via the JNK/MAPK signaling pathway.

Acetaminophen (APAP) is a widely used over-the-counter analgesic and antipyretic. It can cause hepatotoxicity. Recent studies demonstrated that hydrogen sulfide (H2 S) exhibits cell protection in several cell types. This study was designed to investigate whether H 2 S ameliorated APAP-induced acute liver injury and to elucidate its mechanisms. In this study, we analyzed the detailed biological and molecular processes of APAP-induced hepatotoxicity using a bioinformatics analysis, which showed that apoptosis and the c-Jun N-terminal kinase (JNK)/mitogen-activated protein kinase pathway were con

Hydrogen Sulfide Mediates K+ and Na+ Homeostasis in the Roots of Salt-Resistant and Salt-Sensitive Poplar Species Subjected to NaCl Stress.

Non-invasive micro-test techniques (NMT) were used to analyze NaCl-altered flux profiles of K+, Na+, and H+ in roots and effects of NaHS (a H2S donor) on root ion fluxes in two contrasting poplar species, Populus euphratica (salt-resistant) and Populus popularis (salt-sensitive). Both poplar species displayed a net K+ efflux after exposure to salt shock (100 mM NaCl), as well as after short-term (24 h), and long-term (LT) (5 days) saline treatment (50 mM NaCl, referred to as salt stress). NaHS (50 μM) restricted NaCl-induced K+ efflux in roots irrespective of the duration of salt exposure, but

Lactobacillus and dietary sunflower meal supplementation in layer diets: Effects on specific serum content and hydrogen sulfide concentration in layer manure.

Studies show that in poultry houses, hydrogen sulfide (H2S) is present in a much higher concentration than its odor threshold of 10 ppb for humans and has a short-lived deleterious effect on White Leghorns at concentrations of 0.2% H2S. A possible way to reduce H2S emissions (often exhausted into the environment) is to decrease its concentration in manure by adding high fiber byproducts and probiotics in diets. Use of mature compost as filter media and the effect of packing depth on hydrogen sulfide removal from composting exhaust gases by biofiltration.

A study was conducted to investigate the utilization of mature compost as a biofilter medium for the removal of hydrogen sulfide (H2S) from the exhaust gases of the composting process. Source-selected kitchen waste from municipal solid waste was composted in a reactor, and the exhaust gas was passed through a biofilter packed with a 1:4 (wet weight) mixture of mature compost and sand. Two treatments were applied under sterilized and unsterilized conditions to quantify the contribution of microbial activity. The effect of packing depth on H2S removal efficiency was also studied. Solvomorphs of tyraminium 5,5-diethylbarbiturate: a rare example of the barbiturate R33(12) hydrogen-bond motif and a crystal structure with Z' = 4.

In the past two decades, the solvomorphism phenomenon in organic materials has attracted much attention, especially in the pharmaceutical and materials industries. Cocrystallization with solvent molecules can lead to modified physical and chemical properties of materials. We present here two new solvomorphs (pseudopolymorphs) of tyraminium 5,5-diethylbarbiturate [2-(4-hydroxyphenyl)ethanaminium 5,5-diethyl-2,4,6-trioxotetrahydro-2H-pyrimidin-1-ide, C8H12NO+·C8H11N2O3-] with unusual structural features. Pseudopolymorph (I) follows the symmetry of the P21/n space group and has four tyraminium ca

Paper-based fluorescent sensor via aggregation induced emission fluorogen for facile and sensitive visual detection of hydrogen peroxide and glucose.

Hydrogen peroxide (H2O2), an important reactive oxygen species (ROS), is related to the oxidative stress in organisms, and plays important roles in a variety of cellular activities as well. So it is of crucial importance to develop sensitive and accurate sensing strategies to detect H2O2 in biological systems. Herein, by taking advantage of the unique emission characteristics of aggregation induced emission (AIE) fluorogens, we proposed a non-enzymatic fluorescence platform for facile and sensitive detection of H2O2, both in solution state using fluorescence spectrometer and on paper-based sen

Early Aerobic Exercise Combined with Hydrogen-Rich Saline as Preconditioning Protects Myocardial Injury Induced by Acute Myocardial Infarction in Rats.

It has been reported that hydrogen-rich saline (HRS) water reduces oxidative stress, and early aerobic exercise (eAE) acts an efficient exercise preconditioning (EP) against cardiac I/R injury. However, whether early aerobic exercise combined with hydrogen-rich saline (eAE-HRS) water can more effectively protect myocardial damage induced by acute myocardial infarction (MI) is still unknown. This study was aimed to evaluate the effect of eAE-HRS in preventing MI-induced myocardial damage and explore the possible underlying mechanisms. After Sprague-Dawley (SD) rats were given a intragastric adm

Self-Catalyzing Chemiluminescence of Luminol-Diazonium Ion and Its Application for Catalyst-Free Hydrogen Peroxide Detection and Rat Arthritis Imaging.

We report the unique self-catalyzing chemiluminescence (CL) of luminol-diazonium ion (N2+-luminol) and its analytical potential. Visual CL emission was initially observed when N2+-luminol was subjected to alkaline aqueous H2O2 without the aid of any catalysts. Further experimental investigations found peroxidase-like activity of N2+-luminol on the cleavage of H2O2 into OH• radical. Together with other experimental evidence, the CL mechanism is suggested as the activation of N2+-luminol and its dediazotization product 3-hydroxyl luminol by OH• radical into corresponding intermediate radicals, a

Activation of autophagic flux and the Nrf2/ARE signaling pathway by hydrogen sulfide protects against acrylonitrile-induced neurotoxicity in primary rat astrocytes.

Hydrogen sulfide (H2S), the third gasotransmitter, has been shown to act as a neuroprotective factor in numerous pathological processes; however, its underlying mechanism(s) of action remain unclear. It is widely accepted that activation of moderate autophagy and the Nrf2/ARE signaling pathway play important roles in the biological self-defense systems. In the present study, we investigated whether exogenous H2S protects against the cytotoxicity of acrylonitrile (AN), a neurotoxin, in primary rat astrocytes. We found that pretreatment for 1 h with sodium hydrosulfide (NaHS), a donor of H2S (20

Hydrogen sulfide protects against endoplasmic reticulum stress and mitochondrial injury in nucleus pulposus cells and ameliorates intervertebral disc degeneration.

It has been suggested that excessive apoptosis in intervertebral disc cells induced by inflammatory cytokines, such as interleukin (IL)-1β, is related to the process of intervertebral disc degeneration (IVDD). Hydrogen sulfide (H2S), a gaseous signaling molecule, has drawn attention for its anti-apoptosis role in various pathophysiological processes in degenerative diseases. To date, there has been no investigation of the correlation of H2S production and IVDD or of the effects of H2S on IL-1β-induced apoptosis in nucleus pulposus (NP) cells. Here, we found that the expression levels of cystat

Hydrogen Sulfide Prevents Formation of Reactive Oxygen Species through PI3K/Akt Signaling and Limits Ventilator-Induced Lung Injury.

The development of ventilator-induced lung injury (VILI) is still a major problem in mechanically ventilated patients. Low dose inhalation of hydrogen sulfide (H2S) during mechanical ventilation has been proven to prevent lung damage by limiting inflammatory responses in rodent models. However, the capacity of H2S to affect oxidative processes in VILI and its underlying molecular signaling pathways remains elusive. In the present study we show that ventilation with moderate tidal volumes of 12 ml/kg for 6 h led to an excessive formation of reactive oxygen species (ROS) in mice lungs which was
