Interleukin-8 expressed in the granulocytes of the eye in a patient with Behçet's disease complicated by lens-induced endophthalmitis.

BACKGROUND: Interleukin-8 (IL-8) is believed to be involved in the progression of intraocular inflammation. We sought the source of IL-8 in the enucleated eye of the present patient. CASE: A 40-year-old Japanese man was diagnosed as having Behçet's disease. His vision deteriorated due to persistent uveitis and secondary glaucoma. His left eye had lens-induced endophthalmitis. OBSERVATIONS: The left eye had to be enucleated, and it was investigated by an immunohistochemical analysis using antibodies for CD 1a (dendritic cells), CD 3 (T cells), CD 68 (monocytes/macrophages), interferon-gamma,

Effects of long-term administration of roxithromycin on neutrophil count and interleukin-8 level in endometrial cavity subjected to pyometra.

We investigated the effect of long-term administration of roxithromycin (RXM) on clinical responses and immune responses to chronic intrauterine infection, pyometra. One hundred and fifty milligrams of RXM were orally administered once a day for 3 months to patients with pyometra. Neutrophils and interleukin-8 (IL-8), which is a neutrophil chemotactic factor, in the lavage fluid of the uterine endometrial cavity obtained from 5 normal women and 18 patients with pyometra were investigated. Sixteen (88.9%) of the 18 patients showed a clinical improvement at the end of treatment. The immune respo

[Interferon and diseases].

Interferon (IFN) is part of the body's natural defense system. IFN production is a cellular response to the foreign constituents of microbes, tumors and antigens. The three types (alpha, beta and gamma) of IFN proteins differ both structurally and antigenically and have molecular weights ranging from 16,000-25,000 daltons. The IFN proteins induce antiviral, antimicrobial, antitumor, and immunomodulatory actions. We reviewed the recent studies on the relation between IFN and diseases concerning; 1) the IFN-alpha and -gamma producing capacity in patients with cancers, liver cirrhosis, diabetes m

Expression induced by interleukin-6 of tissue-type transglutaminase in human hepatoblastoma HepG2 cells.

We examined the effect of interleukin-6 (IL-6) on the expression of transglutaminase in human hepatoblastoma HepG2 cells. Treatment of cells with IL-6 increased their transglutaminase activity in a time- and dose-dependent way. Dexamethasone strengthened the stimulation by IL-6. Half-maximum stimulation of transglutaminase activity in the cells occurred at a dose of 40 pM IL-6 regardless of the presence of dexamethasone. Based on its immunoreactivity, the transglutaminase induced was identified as tissue-type transglutaminase. Immunoblot analysis showed that the increase in transglutaminase ac

In vitro interleukin-1 and tumor necrosis factor-alpha production by macrophages from chickens infected with either Eimeria maxima or Eimeria tenella.

We measured the in vitro production of interleukin-1 (IL-1) and tumor necrosis factor-alpha (TNF- ) by macrophages from chickens during and immediately following an infection with one of two different species of Eimeria, E. maxima and E. tenella. Quantitatively, the amounts of IL-1 produced during each infection were nearly identical regardless of the oocyst dose of each parasite. TNF production followed a biphasic pattern of increased production with the first peak associated with the pathogenesis of disease and the second peak associated with the development of protective immunity. These exp

Mechanisms of pathogenesis in scleroderma. I. Overproduction of interleukin 6 by fibroblasts cultured from affected skin sites of patients with scleroderma.

Fibroblasts were cultured from affected skin sites of patients with progressive systemic sclerosis (PSS), from unaffected skin sites of the same patients, and from a healthy donor. The concentration of interleukin 6 (IL-6) in culture medium conditioned by the growth of early passage cells was determined by radioimmunoassay and by quantitative bioassay. Results demonstrated that fibroblasts from affected PSS skin produce from 6 to 30-fold higher levels of biologically active IL-6 compared to unaffected and control cells. In contrast, serum IL-6 concentrations in 6 of 8 patients examined were no

B cell function in acquired "common-variable" hypogammaglobulinemia: proliferative responses to lymphokines.

We have compared the proliferative responses of an enriched population of B lymphocytes from patients with acquired (common variable) hypogammaglobulinemia (CVH) with the responses of cells from normal individuals. The uptake of [3H]thymidine into DNA was measured on stimulation with a range of interleukins (IL-2, IL-4, and IL-6) and solid-phase anti-IgM. Flow cytometry using CD19 and surface IgM showed that the "non-T" preparations from the peripheral blood of CVH patients either contained B cells within the normal range (30-40% of the cells) or in a minority group no B cells (less than 3% of

Endotoxin enhancement of toxic shock syndrome toxin 1-induced secretion of interleukin 1 by murine macrophages.

The purpose of this study was to determine whether endotoxin could augment toxic shock syndrome toxin 1 (TSST-1)-induced production of interleukin 1 (IL-1) by murine macrophages. Macrophages from C3H/HeJ or C57Bl/6 mice were stimulated with purified TSST-1 alone or in combination with lipopolysaccharide (LPS). A dramatic synergistic thymocyte-proliferative response was induced by supernatants from C57Bl/6 macrophages stimulated with both TSST-1 and LPS. No enhanced response was induced by supernatants from C3H/HeJ macrophages. A portion of the enhanced response induced by C57Bl/6 macrophage su

Gender, Inflammation, and Cognition Affect the Sleep Trajectories of Subarachnoid Hemorrhage Survivors.

Longitudinal trajectories of sleep in subarachnoid hemorrhage (SAH) survivors are not well known. We identified subgroups of SAH survivors based on distinct trajectories of sleep during the first 6 months post-SAH and compared sociodemographic characteristics, clinical characteristics, inflammatory biomarkers (Toll-like receptor 4, tumor necrosis factor [TNF]-α, interleukin [IL]-1β, and IL-6), and symptom-related characteristics (depression/anxiety, fatigue, sleep quality, and cognition) among the subgroups. We conducted a 6-month longitudinal study of 40 SAH survivors (mean age = 56.0). Multi

Sigma-1 receptor regulates HIV-1 and methamphetamine-induced endothelial/pericyte barrier impairment via strain-specific inflammatory responses and mitochondrial dysregulation.

BACKGROUND: Methamphetamine (METH) use disorder (MUD) is a frequent comorbidity among people with HIV-1 (PWH). While both METH and HIV-1 independently disrupt the brain microvascular system and promote neuroinflammation, the molecular mechanisms underlying their synergistic neurotoxicity remain unclear. The sigma-1 receptor (Sigma-1R) signaling pathway has been suggested as a potential target in METH misuse; however, its involvement in the impact of METH and/or HIV-1 infection on brain pericytes remains unknown. METHODS: Transwell co-cultures of human brain microvascular endothelial cells (HB

Investigations of remibrutinib in models pertinent to multiple sclerosis.

Inhibitors of Bruton tyrosine kinase (BTK) are actively being pursued as potential disease-modifying therapies for multiple sclerosis (MS), as attested by several completed or ongoing Phase 3 clinical trials. Yet, key aspects of BTK inhibitors biology remain unclear. Here, we assessed the effects of remibrutinib, a BTK inhibitor in Phase 3 MS trials, in preclinical models of MS. Remibrutinib was evaluated in the myelin oligodendrocyte glycoprotein (MOG)35-55 model of experimental autoimmune encephalomyelitis (EAE) and in the lysolecithin model of demyelination. In culture, we assessed its abil

Protective effect of obeticholic acid on cyclophosphamide-induced thyroid toxicity in rats by inhibiting TXNIP/NLRP3/ASC/caspase-1-dependent pyroptosis and p53/BAX/caspase-3-dependent apoptosis.

Cyclophosphamide (CYC) is an alkylating agent that is widely used in cancer chemotherapy and immunosuppressive therapy; however, its clinical application is limited because it causes multiple organ toxicities, including thyroid dysfunction. Obeticholic acid (OCA), a farnesoid X receptor (FXR) agonist, possesses antioxidant, anti-inflammatory and anti-apoptotic properties, suggesting potential protective effects. This study aimed to evaluate the efficacy of OCA in reducing CYC-induced thyroid toxicity and to investigate its underlying molecular mechanisms in rats. Rats were randomly assigned to

Stereotypic immune response in Mycobacterium avium ssp. paratuberculosis infection among different Swiss caprine genotypes.

Paratuberculosis is an infection with Mycobacterium avium ssp. paratuberculosis (MAP) causing chronic enteritis in domestic and wild ruminants worldwide. In goats, the infection is caused by C (cattle)-type and S (sheep)-type strains. In this study, the correlation between different MAP strains and histomorphological lesions in the small and large intestines, as well as the mesenteric lymph nodes, in Swiss goats (Caprae aegagrus hircus) was investigated. Ten Swiss caprine MAP isolates were characterized using polymerase chain reaction (PCR) and enzymatic restriction-based single nucleotide pol

Proliferating macrophages in human tumours show characteristics of monocytes responding to myelopoietic growth factors.

Macrophages play essential roles in maintaining tissue homeostasis and immune defence. However, their extensive infiltration into tumours has been linked to adverse outcomes in multiple human cancers. Within the tumour microenvironment (TME), tumour-associated macrophages (TAMs) promote tumour growth and metastasis, making them prime targets for cancer immunotherapy. Recent single-cell analysis suggest that proliferating TAMs accumulate in human cancers, yet their origins and differentiation pathways remain uncertain. Here, we show that a subpopulation of CD163+ TAMs proliferates in situ withi
