The rise of multidrug-resistant (MDR) pathogens in aquaculture poses a significant threat to food safety and public health by facilitating the transfer of resistance across the food chain, underscoring the need for sustainable, non-antibiotic control measures. Trans-cinnamaldehyde (TC), a phytochemical with antimicrobial activity, is a promising alternative. While previous publications have established the antibacterial efficacy of TC against Edwardsiella ictaluri, the agent of enteric septicemia of catfish, its mechanism of action and the potential for bacterial adaptation after prolonged exp

Dental caries is a classic biofilm-induced disease characterized by an imbalance between the demineralization and remineralization of the hard tissues of teeth. Effective anticaries materials and techniques should possess both remineralizing and antibacterial properties. However, a hydrogel that is simultaneously antibacterial, compositionally safe, economical, and prepared through a simple method for caries treatment is still lacking. In this study, we designed a hydrogel system (C17/PL-CA-H) composed of poly-L-lysine-based microspheres and recombinant human type XVII collagen (C17), fabricat

BACKGROUND: Biofilms formed by multidrug-resistant (MDR) Klebsiella spp. present a significant clinical challenge due to elevated antibiotic tolerance. Bacteriophages (phages) represent a promising alternative, particularly in combination with antibiotics, where phage-antibiotic synergy (PAS) can increase antibiofilm activity. Evaluating treatment efficacy in these complex structures requires real-time, noninvasive viability analysis.
METHODS: To address this, we used light-sheet fluorescence microscopy (LSFM), a high-resolution, minimally invasive approach, for dynamic tracking of PAS in int

Sustainable strategies for revalorizing food industry by-products are increasingly relevant in the development of modern experimental dermato-cosmetic formulations. In this study, two semisolid cosmetic creams (R10 and EM-R10) were designed using recycled palm oil-physically purified after intensive frying-as the lipid phase. The recycled oil was incorporated strictly within a controlled experimental framework and does not imply cosmetic-grade regulatory compliance. The formulations incorporated distinct bioactive profiles: R10 combined apricot and pineapple extracts with lime essential oil, w

Peptidomic studies in HeLa cells identified the antitumoral activity of Pep5, and our group observed that coupling Pep5 to a carrier peptide (Cpp) provides antimicrobial potential. This study evaluated the antimicrobial activity of Pep5-Cpp and its derivatives (ΔC3-Pep5-Cpp and ΔN-Pep5-Cpp) and investigated their potential mechanisms of action. For Staphylococcus aureus, MIC values were 6.25 µM for Pep5-Cpp and ΔC3-Pep5-Cpp and 12.5 µM for ΔN-Pep5-Cpp. Against Candida albicans, Pep5-Cpp showed a MIC of 3.125 µM, while both derivatives presented 6.25 µM. For Aspergillus niger, MIC values were 1

Acinetobacter baumannii is a multidrug-resistant nosocomial pathogen responsible for infections that are often difficult to treat. Here, we show that exposure of A. baumannii to the last-resort antibiotic colistin, which disrupts the outer membrane of Gram-negative bacteria, results in inner membrane permeabilization and depolarization, ultimately inhibiting ATP synthesis. Nevertheless, under these conditions, colistin-resistant mutants are rapidly and frequently selected. In addition, A. baumannii is able to tolerate colistin, most likely due membrane depolarization and ATP depletion, which a

The activity-toxicity trade-off remains a central challenge for developing membrane-active antimicrobial peptide-mimicking macromolecules. Increasing hydrophobicity enhances bactericidal potency but often leads to elevated host-cell toxicity and loss of selectivity, as hydrophobic moieties indiscriminately interact with both bacterial and mammalian lipid membranes. Here, we introduce a polarity-induced stable aggregation strategy that effectively decouples hydrophobicity from toxicity, establishing a new design principle for selective macromolecular antibacterials. We engineered maleimide-base

Biofilm-associated infections exhibit high tolerance to antibiotics and antimicrobials, posing a significant challenge to wound management. Pseudomonas aeruginosa and Staphylococcus aureus are among the most prevalent pathogens in biofilm-associated wound infections. Here, we report a main-chain cationic antimicrobial polymer, poly(butylimidazolium) (PIM1), whose imidazolium units feature an acidic C2-H carbon center. PIM1 demonstrated potent broad-spectrum activity against multidrug-resistant (MDR) bacteria with minimal cytotoxicity toward multiple mammalian cell lines in vitro. PIM1 retained

UNLABELLED: Vancomycin-resistant Enterococcus faecium (VREfm) is a major cause of invasive healthcare-associated infections. Daptomycin is an important treatment option, but its efficacy may be compromised for isolates with minimum inhibitory concentrations (MICs) of 4-8 mg/L, even at high doses. EUCAST has therefore assigned an "insufficient evidence" category for Enterococcus spp., making accurate MIC determination essential. We compared the performance of UMIC Daptomycin, several automated broth microdilution (BMD) systems, and E-test against the reference BMD method. Eighty-nine VREfm isol

AIMS: Pasteurella multocida (Pm) is one of the main pathogens causing bovine respiratory disease in China. The prevention and control measures against Pm are traditionally based on the use of broad-spectrum antibiotics. Previous studies have found that Pm is prone to developing antibiotic resistance and tolerance-related mutations when exposed to low concentrations of antibiotics, ultimately leading to challenges in the prevention and control of Pm. This study aimed to explore the role of the recO gene in Pm in mediating resistance and tolerance to fluoroquinolones.
METHODS AND RESULTS: Highl

Self-assembling phage-inspired carriers based on macrocyclic molecules have the potential to overcome antibiotic resistance and extend the antimicrobial effect of drugs. In this study, we have synthesized a new water-soluble pillar[5]arene containing thioglycoside fragments. This compound had an affinity for a model lipid membrane, underwent self-assembly, and associated with the fluoroquinolone antibiotic ciprofloxacin hydrochloride (Cipro) to form biocompatible supramolecular nanostructures. UV-vis and fluorescence spectroscopies were used to assess the ability of the pillar[5]arene/antibiot
